台盼蓝 (Trypan blue) 又称锥蓝或锥虫蓝等，分子式为 C₃₄H₂₄N₆Na₄O₁₄S₄，分子量为 960.81，CAS Number 72-57-1，是细胞活性染料，常用于检测细胞膜的完整性与细胞的存活率，是组织和细胞培养中最常用的死细胞鉴定染色方法之一。正常的活细胞细胞膜结构完整，能够排斥台盼蓝，细胞不会被染成蓝色；而丧失活性或细胞膜不完整的细胞可被台盼蓝染成蓝色。通常认为细胞膜完整性丧失，即可认为细胞已经死亡。

台盼蓝可被巨噬细胞吞噬，故可用于巨噬细胞的活体染色剂。凋亡小体也有台盼蓝拒染现象。台盼蓝染色后，通过显微镜下直接计数或显微镜下拍照后计数，就可以对细胞存活率进行比较精确的定量。染色时间只需3-5分钟，操作简单。

使用说明 (参考)：

a. 收集细胞：

对于贴壁细胞先用胰酶消化处理。对于悬浮细胞，则可以直接收集细胞。把收集的细胞在1000-2000rpm离心3分钟，弃上清，制备单细胞悬液，并做适当稀释。

b. 台盼蓝染色：

用0.2%的台盼蓝悬浮细胞沉淀，染色3分钟 (染色3分钟时间已经足够，染色时间可以更长一些，但不宜超过10分钟)。

c. 细胞计数：

吸取少量经过染色的细胞，用血细胞计数板计数。死亡细胞着蓝色并膨大，无光泽；活细胞不着色并保持正常形态，有光泽。通常要比较精确地进行定量，每个细胞样品至少500个细胞。

细胞存活率＝(细胞总数－蓝色细胞数)/细胞总数×100%

注意事项：

1. 本产品长时间存放可能会出现少量颗粒状沉淀，可在37℃水浴约10分钟以充分溶解沉淀。沉淀完全溶解后即可正常使用。

2. 台盼蓝对人体有毒，操作时请特别小心，并注意有效防护以避免直接接触人体或吸入体内。

Trypan blue, also known as Trypan Blue, has a molecular formula of C₃₄H₂₄N₆Na₄O₁₄S₄, a molecular weight of 960.81 and CAS Number 72‑57‑1. It is a cell‑viability dye widely used to assess cell‑membrane integrity and cell survival rate, and represents one of the most common staining methods for dead‑cell identification in tissue and cell culture. The intact cell‑membrane of healthy living cells can exclude trypan blue, so these cells will not turn blue. In contrast, cells that have lost viability or possess damaged cell‑membranes are stained blue by trypan blue. Loss of membrane integrity is generally regarded as an indicator of cell death.

Since trypan blue can be phagocytosed by macrophages, it can serve as a vital stain for macrophages. Apoptotic bodies also exhibit trypan‑blue exclusion. After trypan‑blue staining, cell‑viability can be quantified relatively accurately either by direct microscopic counting or by counting cells from captured microscopic images. The staining procedure is simple and only requires 3‑5 minutes.

Instructions for Use (For Reference):

a. Cell collection

Adherent cells should first be digested with trypsin. Suspension cells can be collected directly. Harvested cells are centrifuged at 1000‑2000 rpm for 3 min, the supernatant is discarded, and a single‑cell suspension is prepared with appropriate dilution.

b. Trypan‑blue staining

Resuspend the cell pellet in 0.2 % trypan‑blue solution and incubate for 3 min. Three minutes is sufficient for staining; longer incubation is permissible but should not exceed 10 minutes.

c. Cell counting

Aspirate a small volume of stained‑cell suspension and count the cells using a hemocytometer. Dead cells appear blue, swollen and dull; live cells remain unstained, retain their normal morphology and are bright. For reliable quantitative results, count at least 500 cells per sample.

Cell viability = (Total cell count − Blue‑stained cell count) / Total cell count × 100 %

Precautions:

1. A small amount of granular precipitate may form during long‑term storage. Place the product in a 37 °C water bath for approximately 10 minutes to fully dissolve the precipitate. The reagent may be used normally once all precipitate has dissolved.

2. Trypan blue is toxic to humans. Exercise extreme caution during handling and take adequate protective measures to avoid skin contact or inhalation.

3. For scientific‑research‑use‑only by qualified personnel. Not for clinical diagnosis or therapy, and not for food‑ or pharmaceutical‑related applications.

4. Wear a laboratory coat and disposable gloves throughout the procedure for your safety and health.
