酸性磷酸酶（ACP）广泛分布于各种组织细胞中，主要定位于溶酶体内，是溶酶体的标志酶，少量存在于胞质和内质网中。不同物种来源的ACP在生化特性上有所差异，其最适pH范围一般为4.5～5.5。抗酒石酸酸性磷酸酶（TRAP）是ACP的一种同工酶，主要存在于白血病人脾脏、正常人肺泡巨噬细胞及破骨细胞中，血液中的TRAP主要来源于破骨细胞，因此可通过检测血清或血浆中的TRAP活性反映破骨细胞的功能状态。 本试剂盒基于TRAP的酶学特性，以萘酚AS-BI为底物，在酸性条件下被ACP水解释放出萘酚与磷酸，萘酚与重氮盐偶联生成不溶性有色沉淀，定位于酶活性位点。若细胞内ACP具有抗酒石酸的活性，则呈阳性反应，形成酒红色沉淀。该染色液适用于细胞涂片、新鲜血涂片、石蜡切片及冰冻切片等多种样本类型。
Acid phosphatase (ACP) is widely distributed in various tissues and cells, primarily localized in lysosomes, where it serves as a lysosomal marker enzyme, with minor amounts present in the cytoplasm and endoplasmic reticulum. ACP exhibits species-specific biochemical variations, with an optimal pH range generally between 4.5 and 5.5. Tartrate-resistant acid phosphatase (TRAP) is an isoenzyme of ACP found mainly in the spleen of leukemia patients, alveolar macrophages of normal individuals, and osteoclasts. Circulating TRAP in the blood originates predominantly from osteoclasts; therefore, TRAP activity in serum or plasma can be measured to assess osteoclast functional status. This kit is based on the enzymatic properties of TRAP, using naphthol AS-BI as the substrate. Under acidic conditions, ACP hydrolyzes the substrate to release naphthol and phosphate. The released naphthol couples with a diazonium salt to form an insoluble colored precipitate localized at the enzyme active site. Cells exhibiting ACP activity resistant to tartrate inhibition show a positive reaction, appearing as a wine-red precipitate. This staining solution is suitable for various sample types, including cell smears, fresh blood smears, paraffin sections, and frozen sections.
