Teratogenicity‑Test Fixative (Visceral Examination) is mainly composed of PA solution, formaldehyde and glacial acetic acid. It is a yellow‑clear liquid with a slight pungent odour.Principle of Action: It can rapidly fix the visceral soft tissues of fetuses, completely preserve the original morphology and subtle lesions of organs including the brain, heart, lungs, liver, kidney and genital organs, prevent tissue autolysis and atrophy, and facilitate subsequent free‑hand sectioning for visceral malformation screening.Scope of Application: It is applied to the toxicological safety evaluation of food, cosmetics, health‑care products and other products, and specially used for the fixation of fetal visceral tissue specimens in teratogenicity tests.
Operating Procedure (For Reference Only):
1. Sampling and Grouping (Applicable to Rats and Mice)
1) Divide the live fetuses of each litter equally into two parts. One‑half of the fetuses are used for skeletal staining, and the remaining half are fixed with this product for visceral examination.
2) No skinning or bone removal is required. Place the intact fetus directly into the fixative solution.
Note: Half‑sampling is not allowed for rabbit teratogenicity tests. All fetuses shall be fixed synchronously.
2. Fixation of Visceral Specimens:
1) Fully submerge all intact fetuses to be tested in this fixative. Seal the container, protect it from light, and incubate at room temperature for no less than 14 days.
2) Keep the container sealed during fixation. Inspect specimens periodically to ensure complete submersion and avoid autolysis of exposed tissues.
3. Washing Pretreatment after Fixation:
1) After 14‑day fixation, take out the fetuses and rinse thoroughly under running water to remove yellow pigments on the specimen surface.
2) After rinsing, perform free‑hand sections of the head, thorax and abdomen in accordance with national standards for visceral malformation observation.
4. Statistical Requirements for Visceral Microscopic Examination:
1) Observe the olfactory bulb, cerebral ventricle, eyeball, heart, lung, liver, kidney, reproductive system and other organs sequentially along 5 standard cutting planes.
2) Record all types of visceral malformations, such as anencephaly, microphthalmia, ventricular septal defect, diaphragmatic hernia, hydronephrosis, organ absence / ectopia, etc.
3) All visceral malformation indicators shall be counted on a litter‑basis to calculate the visceral malformation rate.
Precautions:
1. This reagent is intended for research‑use only. It is not suitable for clinical diagnosis or other purposes.
2. The fixation period shall not be shorter than 14 days. Insufficient fixation will soften visceral tissues and impair the identification of malformed lesions.
3. For fetal visceral tissue fixation only. Not for pre‑fixation of skeletal specimens.
4. Recommended test scheme: Divide fetuses from each litter into two subgroups. One subgroup is fixed with Teratogenicity‑Test Fixative (Skeletal Examination) for skeletal staining; the other subgroup is fixed with this product for visceral sectioning, so as to complete dual evaluation of skeletal and visceral teratogenicity simultaneously.
5. For your safety and health, please wear a lab coat and disposable gloves during operation.
