The Gram staining method, developed by the Danish physician Christian Gram in 1884, is a widely used differential staining technique in bacteriology. Through this staining method, bacteria can be classified into two major groups: Gram-positive bacteria (G⁺) and Gram-negative bacteria (G⁻). The differential color reaction of bacteria is primarily attributed to the differences in cell wall permeability to ethanol and resistance to decolorization, which are mainly determined by the thickness and structure of the peptidoglycan layer. Under the same staining conditions, utilizing the different isoelectric points of bacteria (Gram-positive bacteria have an isoelectric point of pH 2.0–3.0, while Gram-negative bacteria have an isoelectric point of pH 4.0–5.0), Gram-positive bacteria carry more negative charges than Gram-negative bacteria and thus bind more firmly to positively charged basic dyes such as crystal violet. Upon addition of a mordant (iodine), which enters the bacterial cell, it combines with the dye to form a water-insoluble crystal violet-iodine-protein complex. This complex further binds with magnesium ribonuclease within Gram-positive bacteria, making the stained bacteria resistant to decolorization. The differentiating agents (aniline, acetone, etc.) cannot easily penetrate the cell wall of Gram-positive bacteria, thus Gram-positive bacteria are not readily decolorized; however, these differentiating agents readily enter Gram-negative bacteria, dissolving the dye-iodine complex and causing decolorization of Gram-negative bacteria.
The Aladdin Gram-Weigert Gram Staining Solution (Aniline Crystal Violet Method) is an improvement upon the classic Gram staining formulation, incorporating hematoxylin and Weigert's iodine solution to enhance the staining. This method has evolved from the ammonium oxalate crystal violet method. When applied to direct smears of clinical specimens, it produces a clean background with clearly visible intracellular phagocytic bodies, and the bacteria exhibit typical staining characteristics, allowing clear differentiation between Gram-positive and Gram-negative bacteria. It is particularly suitable for distinguishing bacteria from non-bacterial blue granular materials (such as calcium salts).
This reagent is intended for research use only and is not suitable for clinical diagnosis or other applications.

Materials Required (User-Supplied):
1.10% Formalin fixative solution
2.Incubator or water bath
3.Microscope
Operating Procedure (For Reference Only):
1.Fix tissues in 10% formalin fixative solution, then perform routine dehydration and embedding.
2.Cut sections at 4–5 μm thickness. Deparaffinize through routine xylene or dewaxing clearing solution and rehydrate to water.
3.Counterstain the nuclei with Hematoxylin Staining Solution for 3–5 min. Discard the staining solution and rinse under running tap water for 10 min.
4.Immerse in Eosin Staining Solution, cover, and stain at 56°C for 5–10 min. Discard the staining solution and rinse briefly with water.
5.Apply Aniline-Crystal Violet Staining Solution dropwise onto the sections and stain for 5 min. Discard the staining solution and use filter paper to gently absorb the excess liquid around the sections.
6.Apply Weigert's Iodine Solution dropwise onto the sections and stain for 2–3 min. Discard the iodine solution and repeatedly absorb the moisture from the sections with filter paper.
7.Differentiate with aniline-xylene solution (1:2) while gently shaking occasionally, until no purple color elutes from the sections. Immediately wash with xylene or dewaxing clearing solution and examine under a microscope.
8.If differentiation is insufficient, apply additional aniline differentiation solution dropwise until the Gram-positive bacteria on the sections are clearly visible.
9.Wash repeatedly with fresh xylene or dewaxing clearing solution several times to thoroughly remove the aniline. Mount with neutral mounting medium.

Precautions:
1.After treatment with Weigert's Iodine Solution and water washing, the moisture on the sections must be repeatedly absorbed with filter paper before applying the aniline-xylene solution (1:2). Failure to do so may result in uneven differentiation.
2.After washing with xylene, examine under a microscope. If differentiation is insufficient, add additional aniline-xylene solution (1:2) dropwise and continue differentiation until the positive bacteria are clearly visible. However, take care to avoid over-differentiation.
3.Finally, wash the sections repeatedly with xylene or dewaxing clearing solution to completely remove the aniline. Residual aniline on the sections may cause subsequent fading.
4.For your safety and health, please wear a lab coat and disposable gloves during operation.
