本产品主要由Tricine、Tris、SDS调配而成，是Tricine- SDS- PAGE专用分离胶缓冲体系。常规Tris- 甘氨酸电泳体系对10kDa以下小分子蛋白分离效果差，Tricine- SDS- PAGE电泳系统能够较好地分离分子量在10kDa以下的蛋白或多肽，是电泳法分离小分子多肽的主要方法。

Tricine凝胶缓冲液 (3mol/L, pH8.45)是一种以Tris- HCl为基础、含SDS的凝胶缓冲液，pH值为8.45。电泳分离后可直接进行考马斯亮蓝染色、银染等。

该试剂仅用于科研领域，不适用于临床诊断或其他用途。

操作步骤（仅供参考）：

1、配制Tricine分离胶参考配比（以9 ml胶液为例）：取3mol/L Tricine凝胶缓冲液3.0 ml，搭配Acr- Bis、APS、促凝剂、纯水混合配胶。

2、充分混匀胶液后快速灌胶，插上梳齿室温静置聚合。

3、胶板聚合完成后拔出梳子，加入Tricine电泳缓冲液，上样蛋白样品进行电泳。

4、电泳至溴酚蓝指示剂接近胶底即可停止，转膜或染色。

注意事项：

1、本品为澄清浓缩缓冲液，常温放置即可，低温出现微量结晶时室温摇晃溶解后使用，不影响性能。

2、仅用于Tricine- SDS- PAGE分离胶配制，不可替代Tris- 甘氨酸缓冲液、浓缩胶缓冲液、电泳缓冲液使用。

3、本产品仅限于科学实验研究使用，不得用于临床诊断、治疗等领域。

This product is mainly prepared with Tricine, Tris and SDS, serving as a dedicated resolving gel buffer system for Tricine‑SDS‑PAGE. Conventional Tris‑glycine electrophoresis systems exhibit poor separation performance for small proteins below 10 kDa. The Tricine‑SDS‑PAGE system enables favorable separation of proteins or peptides with molecular weights under 10 kDa, and it is a major electrophoretic method for isolating small‑molecular‑weight peptides.

Tricine Gel Buffer (3 mol/L, pH 8.45) is a Tris‑HCl‑based gel buffer containing SDS, with a pH of 8.45. After electrophoretic separation, gels can be directly stained with Coomassie Brilliant Blue, silver stain, etc.

This product is for research use only. It is not intended for clinical diagnosis or other applications.

Operating Procedures (For reference only):

1. Reference formula for Tricine resolving gel (for a total gel volume of 9 mL): Take 3.0 mL of 3 mol/L Tricine Gel Buffer, then mix with Acr‑Bis, APS, accelerator and purified water to prepare the gel solution.

2. Mix thoroughly, cast the gel rapidly, insert the comb and allow polymerization at room temperature.

3. After polymerization, remove the comb, add Tricine electrophoresis buffer, load protein samples and start electrophoresis.

4. Stop electrophoresis when the bromophenol blue tracking dye migrates near the bottom of the gel, then proceed to blotting or staining.

Precautions:

1. This product is a clear concentrated buffer and can be stored at ambient temperature. Minor crystals formed at low temperature can be dissolved by shaking at room temperature before use, with no impairment to performance.

2. It is only used for preparing resolving gels for Tricine‑SDS‑PAGE. Do not substitute it for Tris‑glycine buffer, stacking gel buffer or running electrophoresis buffer.

3. This product is limited to scientific research and shall not be used for clinical diagnosis, treatment or related purposes.

4. For your safety and health, wear a lab coat and disposable gloves during operation.
