800rpm 离心5min(除去冻存液中DMSO)。
Vero serum-free medium is a universal adherent medium for mammalian cell lines, widely used in adherent vaccine production.
Medium Adaptation
Direct adaptation: In the initial culture phase, cells are passaged at a split ratio of 1:3, and the medium is directly replaced with Aplo VVP1 for culture. After 2–3 passages, when cell growth is stable, subsequent experiments can be carried out. For stable adherent cell growth, cells are passaged at 1:3; after 72 hours, microscopic observation should show good cell growth, an adherent monolayer, and a confluence of approximately 80%.
Indirect adaptation: Aplo VVP1 and the original cell medium are mixed at a 1:1 ratio for culture. Cells are passaged continuously for 2–3 generations. After stable growth is achieved, the medium is replaced with Aplo VVP1, and cells are passaged for another 2–3 generations. Once growth is stable, subsequent experiments can be performed. For stable adherent cell growth, cells are passaged at 1:3; after 72 hours, microscopic observation should show good cell growth, an adherent monolayer, and a confluence of approximately 80%.
Precautions
1.Choose direct or indirect adaptation based on the specific condition of the cells.
2.During the early stage of cell acclimatization, if unstable conditions such as low confluence or floating cells occur, select the appropriate subculture method (medium‑change subculture or centrifugation subculture) based on the day’s cell growth status.
3.Medium‑change subculture: If confluence is ≥30%, discard the old cell suspension and add fresh corresponding medium.
4.Centrifugation subculture: If confluence is <30%, collect the entire cell suspension, centrifuge at 800 rpm to remove the supernatant, resuspend the cells in fresh medium, and replenish with fresh corresponding medium.
5.Normal cell subculture after recovery: Adherent cells are passaged at a 1:3 ratio; after 72 h, microscopic observation should show good cell growth, an adherent monolayer, and a confluence of approximately 80%.
Cryopreservation of Adherent Cells
1.After cells return to normal status, expand the culture volume for cryopreservation and banking. Prepare as many cell banks as possible to ensure sufficient backups.
2.Select cells in logarithmic growth phase with viability >90% for cryopreservation. It is recommended to cryopreserve 3–4 vials per T75 flask.
3.Prepare the freezing container: Add an appropriate amount of isopropanol to the controlled‑rate freezing container and pre‑cool at 4°C.
4.Collect the cell suspension, centrifuge at 800 rpm for 5 min, discard the supernatant, and resuspend the cells in an appropriate volume of cryopreservation medium to adjust to the target density.
5.Quickly aliquot the cell suspension into cryovials.
6.Place the cryovials into the controlled‑rate freezing container, store at –80°C for 24 h, then transfer to liquid nitrogen. After a period of time, thaw one vial for testing.
Thawing of Adherent Cells
1.Pre‑warm medium: Place 15 mL of the corresponding medium in a square flask and pre‑warm in a 37°C CO₂ incubator for at least 30 min (to equilibrate the pH) to ensure thorough pre‑warming.
2.Remove the cryovial from liquid nitrogen or –80°C storage and quickly transfer to a 37°C water bath with rapid shaking until completely thawed.
3.Transfer 5 mL of pre‑warmed medium into a sterile centrifuge tube, then transfer the thawed cell suspension into this tube. Centrifuge at 800 rpm for 5 min (to remove DMSO).
