Engineered 3D-Printed Polyvinyl Alcohol Scaffolds Incorporating β-Tricalcium Phosphate and Icariin Induce Bone Regeneration in Rat Skull Defect Model

Zhimin Xu,Yidan Sun,Huanyan Dai,Yujie Ma,Han Bing

The skull defects are challenging to self-heal, and autologous bone graft repair has numerous drawbacks. The scaffolds for the rapid and effective repair of skull defects have become an important research topic. In this study, polyvinyl alcohol (PVA)/β-tricalcium phosphate(β-TCP) composite scaffolds containing icariin (ICA) were prepared through direct-ink three-dimensional (3D) printing technology. β-TCP in the composite scaffold had osteoconductive capability, and the ICA molecule had osteoinductive capacity. The β-TCP and ICA components in the composite scaffold can enhance the capability to repair skull defects. We show that ICA exhibited a slow-release behaviour within 80 days. This behaviour helped the scaffold to continuously stimulate the formation of new bone. The results of in vitro cell compatibility experiments showed that the addition of ICA molecules contributed to the adhesion and proliferation of MC-3T3-E1 cells. The level of alkaline phosphatase secretion demonstrated that the slow release of ICA can promote the osteogenic differentiation of MC-3T3-E1 cells. The introduction of ICA molecules accelerated the in situ bone regeneration in in vivo. It is concluded that the 3D-printed PVA scaffold with β-TCP and ICA has a wide range of potential applications in the field of skull defect treatment.Keywords:three-dimensional printing;polyvinyl alcohol;icariin;β-tricalcium phosphate;skull base defects

A chitosan based scaffold with enhanced mechanical and biocompatible performance for biomedical applications

Xianzhen Dong,Qiao Cheng,Yanpiao Long,Chao Xu,Honglin Fang,Yuzhe Chen,Honglian Dai

Chitosan exhibiting excellent functional properties was found to be potential in a wide variety of industrial and biomedical applications. The lack of biological activities and the poor mechanical properties somewhat limit its applications. To address these limitations, chemical modification has been proved to be an efficient method. Here, we immobilized arginine-glycine-aspartic acid (RGD) on chitosan molecules and incorporated the chitosan with Poly(L-lactide-co-glycolide)-Poly(ethylene glycol) (PLGA-PEG) and β-tricalcium phosphate (β-TCP) nanoparticles to obtain a chitosan-based scaffold with enhanced mechanical and biocompatible performance. XPS and FT-IR confirmed the successful grafting of RGD peptides onto the chitosan chain. The mechanical properties, hydrophilicity and degradation behaviors of the membrane were characterized by stress-strain, water contact angle, and weight loss, respectively. Cytotoxicity, cellular adhesion, and cellular viability of the scaffolds were studied by MTT assay, cell adhesion, and cell survival experiments, respectively. The advantageous mechanical properties and the hydrophilic surface of the chitosan-based scaffold were achieved by blending with PLGA-PEG. The preliminary biocompatibility studies revealed that all the scaffolds were cell compatible, and the hydrophilic surface was more suitable for cell adhesion. The scaffolds grafted with RGD exhibited high cell adhesion rate than the pure chitosan. The results suggested that the surface hydrophilicity and biocompatibility of the chitosan-based scaffolds play an important role in enhancing cell adhesion and growth.

Nanosized-Ag-doped porous β-tricalcium phosphate for biological applications

Junjie Yuan,Baoxin Wang,Chen Han,Xiaoyan Huang,Haijun Xiao,Xiao Lu,Jianxi Lu,Dong Zhang,Feng Xue,Youzhuan Xie

The treatment of infectious or potentially infective bone defects remains a major problem in clinical practice. Silver has the ability to potentiate antibiotics against resistant bacterial strains. In order to reduce the risk of long-term infections, it is necessary for the biomaterial scaffold to release Ag + in a controlled manner during the entire healing process. In this study, given the antimicrobial characteristics of nanosized Ag (NSAg), we synthesized β-tricalcium phosphate (β-TCP) doped with 5 and 10 wt% NSAg (5 wt% NSAgTCP and 10 wt% NSAgTCP, respectively). The NSAgTCP composites exhibited similar macroporous structures to pure β-TCP. The NSAgTCP samples were examined by scanning electron microscopy at 10,000-times magnification, which revealed that silver was still present at the nanometer scale. X-ray diffraction revealed that silver does not change the crystalline properties of β-TCP. In addition, we observed that the mechanical strength of NSAgTCP increased with increasing amounts of added Ag. The antibacterial, physical, and chemical properties of NSAgTCP were investigated in vitro. We found that NSAgTCP is effective at inhibiting the growth of Staphylococcus aureus and Escherichia coli and is not cytotoxic to human bone marrow mesenchymal stem cells. Moreover, it does not hinder liver or kidney function when tested in vivo. As the bioceramic degrades, Ag ions are slowly released and new bone is formed. No significant cytotoxic effects were observed even when 10 wt% NSAgTCP was used. NSAgTCP has the ability to simultaneously repair bone defects and act as an anti-infective agent; hence, we expect that this material, with its good bone-repairing and anti-infective properties, will find wide spread use as a novel bone substitute.

Fabrication and characterization of honeycomb β-tricalcium phosphate scaffolds through an extrusion technique

Shenglei Feng,Fupo He,Jiandong Ye

In this study, for the first time honeycomb β-tricalcium phosphate (β-TCP) scaffolds were fabricated through an extrusion technique. The physicochemical properties and cell behaviors of the honeycomb β-TCP scaffolds were investigated. The results showed that scaffolds were characterized by ordered channel-like macropores and unidirectional interconnection. The pore structure and mechanical strength could be tailored by changing the parameters of extrusion molds. The pore size of scaffolds was in the range of 400–800   µm approximately, while their compressive strength parallel to the pore direction and porosity ranged from 14 to 20   MPa and 60–70%, respectively. The in vitro cell behavior demonstrated that cells could well attach on the surfaces and grow into the inner channel-like pores of the scaffolds; the scaffolds with higher porosity showed better cell proliferation but poorer cell differentiation. The honeycomb scaffolds fabricated by extrusion technique are potential candidate for bone tissue engineering.

DLP 3D printing porous β-tricalcium phosphate scaffold by the use of acrylate/ceramic composite slurry

Sa Liu,Lina Mo,Gangyuan Bi,Shenggui Chen,Diwei Yan,Junzhong Yang,Yong-Guang Jia,Li Ren

Digital light processing (DLP) 3D printing has been utilized to fabricate controlled porous β-tricalcium phosphate (β-TCP) scaffolds, which promote cell adhesion and angiogenesis during bone regeneration. However, the current limitation of DLP 3D printing for the fabrication of β-TCP scaffold is how to prepare a low viscosity ceramic slurry and remove the toxicity of residual non-polymerized slurry. The present study has developed a low viscosity ceramic slurry system by mixing β-TCP with photosensitive acrylate resin, and the viscosity of slurry is about 3 Pa s and the solid content of β-TCP can be as high as 60 wt%. After optimizing the ratio of slurry, printing, degreasing and sintering processes, the maximum compressive strength of the DLP printed scaffolds reaches up to 9.89 MPa, while the porosity keeps ca. 40%. According to the proliferation of cells, it confirms the preserved biocompatibility of DLP-fabricated β-TCP scaffolds. These porous scaffolds made by DLP 3D printing technology is of great significance for bone regeneration, and will also help to expand the application of DLP technology in biomedical field.

A 3D-printed Sn-doped calcium phosphate scaffold for bone tissue engineering

Liang Hong,Fu Gaosheng,Liu Jinrui,Tang Yueting,Wang Yujue,Chen Shan,Zhang Yanjie,Zhang Chen

Recent developments in 3D printing technology have been applied in the field of tissue engineering to fabricate customized bone repair scaffolds. β-tricalcium phosphate (β-TCP) is a bioceramic material with excellent potential as a scaffold foundation. Doping metallic ions with β-TCP will significantly enhance the mechanical property and bone regeneration performance compared with pure β-TCP specimens. In this study, we proposed a protocol for the fabrication of a Sn-doped β-TCP (Sn@TCP) scaffold using 3D printing technology, and the effect of Sn-doping on the physicochemical properties of the material and its in vitro bioactivity were investigated. Polyethylene glycol and polyvinyl alcohol were used as binder to construct Sn@TCP scaffolds which have good biocompability and can break down into H2O and CO2 after scaffolds sintering. The appearance of the scaffold constructed by 3D printing technology closely matched the computer design. The incorporation of Sn into β-TCP improved the compressive strength of the scaffold. Moreover, the Sn@TCP scaffold retained the inherently good biocompatibility of β-TCP and exhibited better osteoinduction capability than pure β-TCP scaffolds. Notably, the osteoinduction ability of Sn@TCP scaffolds were dependent on the Sn content. In conclusion, the 3D printing of Sn@TCP scaffolds with enhanced mechanical properties and osteoblast-inducing activity show great promise as scaffold materials in bone tissue engineering applications.

This work investigated a novel Poly(vinyl alcohol)-polyetheretherketone/Poly(vinyl alcohol)-β-tricalcium phosphate (PVA-PEEK/PVA-β-TCP) bilayered hydrogels by freezing-thawing with biomimetic properties for articular cartilage and subchondral bone is developed. The bilayered hydrogels microarchitecture consists of a highly porous and dense structure, and the internal structure were analyzed by micro-CT. The morphology of the resulting hydrogels was analyzed by scanning electron microscopy (SEM), the enhancement of the mechanical properties of the PVA-PEEK/PVA-β-TCP bilayered hydrogels were demonstrated by mechanical testing. The bilayered structure indicate that a good bonding exist between the two layers, which is known to be a requisite necessary to assure a good integrity and functionality of the osteochondral construct. In addition, in vitro cell culture studies revealed that the hydrogels has no negative effect on the cell viability and proliferation and possess good biocompatibility. Then, the bilayered hydrogels were implanted into the knee joint defect of rabbits and hematoxylin and eosin and immunohistochemical staining. The PVA-PEEK/PVA-β-TCP bilayered hydrogels show good potential for use in the field of articular cartilage repair.

Three-dimensional printing of $$\varvec{\upbeta }$$ -tricalcium phosphate/calcium silicate composite scaffolds for bone tissue engineering

Dong Yifan,Duan Haibo,Zhao Naru,Liu Xiao,Ma Yijuan,Shi Xuetao

Bioactive scaffolds with interconnected porous structures are essential for guiding cell growth and new bone formation. In this work, we successfully fabricated three-dimensional (3D) porous \(\upbeta \) -tricalcium phosphate ( \(\upbeta \) -TCP)/calcium silicate (CS) composite scaffolds with different ratios by 3D printing technique and further investigated the physiochemical properties, in vitro apatite mineralization properties and degradability of porous \(\upbeta \) -TCP/CS scaffolds. Moreover, a series of in vitro cell experiments including the attachment, proliferation and osteogenic differentiation of mouse bone marrow stromal cells were conducted to testify their biological performances. The results showed that 3D printed \(\upbeta \) -TCP/CS scaffolds possessed of controllable internal porous structures and external shape. Furthermore, the introduction of CS decreased the shrinkage of scaffolds and improved the in vitro apatite formation activity and degradation rate. Meanwhile, compared with pure \(\upbeta \) -TCP scaffold, the \(\upbeta \) -TCP/CS composite scaffolds were more conducive to promote cell adhesion, spread and osteogenesis differentiation. However, when the content of CS was increased to 45%, the ions dissolution rate of the composite scaffolds was so high that leaded to the increase in pH value, which inhibited the proliferation of cells. Our results suggested that the introduction of appropriate CS into \(\upbeta \) -TCP bioceramic is an effective strategy to prepare bioactive 3D printed bioceramic scaffolds for hard tissue regeneration.

Kang Wang,Zexin Liu,Lin Liu,Tiantian Wang,Kun Chen,Runhua Yao,Ruiqiang Hang,Xiaohong Yao

Zinc-based materials have been extensively researched as biodegradable bone repair materials in recent years, but their low degradation rate and limited bioactivity need to be further improved. In this study, a new-type dual-sized macro-micro porous Zinc-Magnesium-β-tricalcium phosphate (DMMP Zn-Mg-β-TCP) scaffolds were designed and fabricated by vacuum heating-press sintering. The mechanical properties, degradation behaviors, in vitro biocompatibility, in vivo osteogenesis, and biological safety of the scaffolds were systematically evaluated. The results show that DMMP Zn-Mg-β-TCP has suitable mechanical properties, degradation properties, and excellent osteogenic properties. The compressive yield strength range of the scaffolds is 12.90–25.46 MPa, and the elastic modulus range is 0.31–0.44 GPa, meeting the mechanical properties required for implant materials in cancellous bone. In vitro degradation tests showed that the corrosion rate of the scaffolds ranges from 2.25 to 2.61 mm/y. Furthermore, the scaffolds exhibited excellent in vitro cell compatibility. After a 24-week animal experiment, the scaffold demonstrated excellent osteogenic capacity during bone healing, while maintaining biomechanical stability throughout its degradation process. In summary, the DMMP Zn-Mg-β-TCP has the potential to become a novel biodegradable bone implant material.

Study on the high-temperature tribological performance of biodegradable ultrafine β-tricalcium phosphate reinforced barium complex grease

Shuai Li,Haopeng Cai,Haichao Liu,Zhuang Xu,Wenjing Lou

Lubricating additives are essential for enhancing the tribological properties of grease. The development of low-cost lubricant additives with excellent biodegradability is expected to be the prevailing trend for future advancements. In this study, biodegradable ultrafine β-tricalcium phosphate (β-TCP) was utilized to enhance the friction reduction, anti-wear and extreme pressure properties of barium complex greases at high temperatures. At 150 °C and a load of 150 N, compared to the base grease, the average friction coefficient (AFC) of barium complex grease with 4.5% β-TCP was reduced by 21.82%. Meanwhile, the wear volume was reduced by 88.43%. Moreover, the extreme pressure properties of the barium complex grease were increased by 3.75 times. The lubrication mechanism of β-TCP is the formation of a protective film of P and Ca compounds on the surface of the friction partner.

Competitive adsorption of bovine serum albumin and lysozyme on a beta-tricalcium phosphate nanocoating

Lanxiang Ma,Dongwen Pang,Chunlin Deng

The competitive and cooperative adsorptions of proteins on the surface of implanted materials can affect the biocompatibility and osteogenesis of the material. The adsorption processes are related to the properties of the materials, as well as to the nature of the proteins. Competitive adsorption of bovine serum albumin (BSA) and lysozyme (LSZ), both sequentially, and from a mixture, was investigated on a beta-tricalcium phosphate (β-TCP) nanocoating using quartz crystal microbalance with dissipation technique. In experiments with individual proteins, the BSA adlayer showed higher adsorption quality and viscoelasticity than the LSZ on the β-TCP surface. In the sequential adsorption of LSZ + BSA, protein exchange was observed but not in the BSA + LSZ sequence, because of the larger molecular and higher surface affinity of BSA relative to LSZ. The substitution rate increased as the increasing concentration of the BSA solution. With a BSA&LSZ mixture, the total amount of protein adsorbed was greater than that for individual or sequential adsorption, owing to the formation of BSA and LSZ agglomerates. The electrostatic force and affinity were dominant factors governing the adsorption behavior between BSZ and LSZ on the β-TCP surface. This work will be beneficial to understand the protein adsorption mechanism on β-TCP surface and provide information to design better osteogenic materials.

Preparation and properties of chitosan quaternary ammonium salt/α-calcium sulfate hemihydrate/β-tricalcium phosphate composite bone cement

Shaobing Zhang,Zhi Lian,Xingtao Chen,Ke Duan,Cheng Liang,Xin Duan,Jiyuan Yan,Jianhua Ge

Objective The effective treatment of bone tissue defects is a significant challenge in clinical orthopaedics. This study investigated the preparation and properties of composite bone cements comprising 2-hydroxypropyltrimethyl ammonium chloride chitosan (HACC)-modified α-calcium sulfate hemihydrate (α-CSH)/β-tricalcium phosphate (β-TCP) for potential applications in bone regeneration. Methods α-CSH was synthesised via a hydrothermal method, while β-TCP was prepared through a chemical reaction involving calcium salts and phosphates. HACC was dissolved in deionised water to formulate curing solutions with concentrations ranging from 0 to 4 wt% HACC. The α-CSH and β-TCP powders (7:3 mass ratio) were homogeneously blended with the HACC solution (1:0.3 w/v) to produce HACC/α-CSH/β-TCP composite bone cements. To assess the biocompatibility, osteogenic potential, and antimicrobial activity, extracts of the composite bone cements were used to culture rat bone marrow mesenchymal stem cells. A critical bone defect model in the femoral condyle of male Sprague–Dawley rats was employed to evaluate the in vivo osteogenic repair efficacy. Results The self-curing time met clinical requirements, and the compressive strength approached that of normal human cancellous bone. The in vitro degradation rate was consistent with the rate of new bone formation. Increasing the HACC concentration enhanced the injectability and washout resistance of the composites. The materials exhibited good in vitro biocompatibility; composites containing HACC upregulated osteogenesis-related gene expression and significantly inhibited the growth of Staphylococcus aureus and Escherichia coli . In vivo, the 3 wt% HACC/α-CSH/β-TCP composite showed the best overall performance. Conclusions HACC/α-CSH/β-TCP composite bone cements demonstrate promising clinical potential owing to their improved setting time, mechanical properties, injectability, and cytocompatibility. This material is a strong candidate as a substitute for artificial bone to treat infectious bone defects.
