Preparation and characterization of temperature/pH/CO2-triple-responsive homopolymers and their substituent determined response

Ke Wang,Zilong Wang,Meiyu Si,Xiaofang Liu,Guiyan Liu,Yongfei Zeng

Three novel temperature/pH/CO2-triple-responsive homopolymers namely P(Et-N-EO2MA), P(THP-N-EO2MA) and P(Ph-N-EO2MA) were prepared by reversible addition–fragmentation chain transfer (RAFT) polymerization. They have the same backbone structure but different types of end substituents i.e., ethyl, tetrahydropyran, and the benzene ring, respectively. Subsequently, their responsive properties were studied. P(Et-N-EO2MA) has a lower critical solution temperature (LCST)-type cloud point (CP) in aqueous solution and is affected by many factors such as the molecular structure and weight, concentration, additives, pH and CO2. P(THP-N-EO2MA) is soluble in water, but in different buffer solutions, it shows a soluble–LCST–insoluble transition, and its LCST-type CP can also be adjusted by molecular weight and concentration. P(Ph-N-EO2MA) is insoluble in water, but reversible phase transformation can be achieved by adjusting the pH of the solution or injecting CO2, and it also shows different properties in buffer solutions and an upper critical solution temperature (UCST)-type CP in n-butanol. In addition, since P(Ph-N-EO2MA) is amphiphilic, it can be self-assembled in solution, and the release of Nile Red (NR) can be achieved under the stimulation of acid. The different properties of these homopolymers show the effect of substituents on the responsiveness of multistimuli-responsive polymers, which provides ideas for the design of stimuli-responsive polymers, and have good application prospects in the fields of self-assembly and controlled release.

A novel triple responsive smart fluid for tight oil fracturing-oil expulsion integration

Ming-Wei Gao,Ming-Shan Zhang,Heng-Yi Du,Ming-Wei Zhao,Cai-Li Dai,Qing You,Shun Liu,Zhe-Hui Jin

The traditional multi-process to enhance tight oil recovery based on fracturing and huff-n-puff has obvious deficiencies, such as low recovery efficiency, rapid production decline, high cost, and complexity, etc. Therefore, a new technology, the so-called fracturing-oil expulsion integration, which does not need flowback after fracturing while making full use of the fracturing energy and gel breaking fluids, are needed to enable efficient exploitation of tight oil. A novel triple-responsive smart fluid based on “pseudo-Gemini” zwitterionic viscoelastic surfactant (VES) consisting of N -erucylamidopropyl- N , N -dimethyl-3-ammonio-2-hydroxy-1-propane-sulfonate (EHSB), N , N , N′ , N′ -tetramethyl-1,3-propanediamine (TMEDA) and sodium p -toluenesulfonate (NaPts), is developed. Then, the rheology of smart fluid is systematically studied at varying conditions (CO 2 , temperature and pressure). Moreover, the mechanism of triple-response is discussed in detail. Finally, a series of fracturing and spontaneous imbibition performances are systematically investigated. The smart fluid shows excellent CO 2 -, thermal-, and pressure-triple responsive behavior. It can meet the technical requirement of tight oil fracturing construction at 140 °C in the presence of 3.5 MPa CO 2 . The gel breaking fluid shows excellent spontaneous imbibition oil expulsion (∼40%), salt resistance (1.2 × 10 4  mg/L Na + ), temperature resistance (140 °C) and aging stability (30 days).

Self-Adhesive, Self-Healable, and Triple-Responsive Hydrogel Doped with Polydopamine as an Adsorbent toward Methylene Blue

Sisi Li,Jun Xu,Guohong Yao,Hui Liu

As a kind of attractive biomaterial, stimuli-responsive hydrogel had been widely used in wound dressings and wearable devices. However, it remained challenging to prepare a hydrogel with both excellent self-adhesion and self-healing performances. Inspired by the adhesion mechanism of mussel, we prepared a self-adhesive, self-healing, and triple-responsive hydrogel by a hybrid system based on tunable composition of polydopamine (PDA) and polymer network composed of poly(N,N-diethylacrylamide-co-acrylic acid). It was found that the resulting hydrogel could be directly adhered to a variety of substrates without the need of external adhesives on account of the existence of enough free catechol groups in the hydrogel. Due to the synergistic effect of PDA nanoparticles and polymer network, hydrogel also showed excellent self-healing property. Moreover, we demonstrated that the triple-responsive hydrogel as an adsorbent could efficiently remove methylene blue, and the maximum adsorption capacity was 305.4 mg/g when the optimum mass fraction of PDA was 0.4 wt % in hydrogel. The adsorption of the hydrogel toward methylene blue could be well simulated by pseudo-second-order model and Langmuir isothermal adsorption model. From the viewpoint of mechanism, methylene blue molecules could be directly adsorbed to these carboxyl, amino and catechol functional groups in the molecular structure of polymeric hydrogels by electrostatic force, π–π interaction, Van Der Waals force, hydrogen bonding, etc. Ultimately, we hoped to extend this hybrid strategy to develop multifunctional hydrogels and consequently broaden the application prospects of hydrogels in biomedical treatment and environmental protection.

Magnetic Nanocarriers for pH/GSH/NIR Triple-Responsive Drug Release and Synergistic Therapy in Tumor Cells

Di Zhang,Wanyu Wei,Tianxiang Xie,Xue Zhou,Xu He,Jie Qiao,Rui Guo,Gang Jin,Ningbo Li

In this study, the mesoporous Fe3O4 nanodrug carriers containing disulfide bonds (CHO-SMNPs) were successfully synthesized and characterized. Doxorubicin (DOX) was loaded onto the CHO-SMNPs as a model drug and gatekeeper through the formation of imine bonds with the aldehyde groups on the surface of the mesoporous materials. This drug carrier demonstrates effective drug release triggered by pH, glutathione (GSH), and near-infrared (NIR) light, along with satisfactory photothermal conversion efficiency under NIR irradiation at 808 nm. Furthermore, CHO-SMNPs exhibit excellent blood compatibility and biodegradability. They also show good biocompatibility and efficient cellular uptake in HeLa and MCF-7 cancer cells. Most importantly, the CHO-SMNPs/DOX has shown significant effectiveness in killing both HeLa and MCF-7 cancer cells. Consequently, CHO-SMNPs/DOX presents substantial potential as a magnetic-targeted, pH/GSH/NIR triple-triggered drug delivery system for synergistic chemo-photothermal therapy in tumor treatment.

Rational design and facile fabrication of biocompatible triple responsive dendrimeric nanocages for targeted drug delivery

Dan Zhong,Huayu Wu,Yahui Wu,Yunkun Li,Xianghui Xu,Jun Yang,Zhongwei Gu

Multi-responsive polymeric nanoparticles have shown great promise in the sufficient site-specific delivery of drugs in heterogeneous and complicated biological microenvironments, but without great success due to many problems such as sophisticated manufacture process, high cost and cytotoxicity. In this work, a novel triple responsive dendrimeric nanocage (TDN) is fabricated through co-assembling and cross-linking of lipoic acid modified low generation dendrimers with lipoic acid modified polyethylene glycols (PEGs). This nanocage exhibits improved drug loading capacity (about 2 times higher) at a lower temperature and stimuli-responsive drug release profile upon the stimulation of temperature, acid pH and reducing agent. More importantly, the nanocage promotes drug internalization, conduces endosomal escape, and realizes intracellular controlled drug release. Furthermore, the nanocage significantly improves the pharmacokinetics and biodistribution of antitumor drugs, confirming the potent in vivo therapeutic effect with reduced side effects. The rational design and facile fabrication of multi-responsive dendrimeric nanocages provide a “proof-of-concept” for precise targeted drug delivery, and may have great potential for clinical use in the future.

Triply Responsive Films in Bioelectrocatalysis with a Binary Architecture: Combined Layer-by-Layer Assembly and Hydrogel Polymerization

Huiqin Yao,Naifei Hu

In this work, triply responsive films with a specific binary architecture combining layer-by-layer assembly (LbL) and hydrogel polymerization were successfully prepared. First, concanavalin A (Con A) and dextran (Dex) were assembled into {Con A/Dex}5 LbL layers on electrode surface by the lectin-sugar biospecific interaction between them. The poly(N,N-diethylacrylamide) (PDEA) hydrogels with entrapped horseradish peroxidase (HRP) were then synthesized by polymerization on the surface of LbL inner layers, forming {Con A/Dex}5–(PDEA-HRP) films. The films demonstrated reversible pH-, thermo-, and salt-responsive on–off behavior toward electroactive probe Fe(CN)63– in its cyclic voltammetric responses. This multiple stimuli-responsive films could be further used to realize triply switchable electrochemical reduction of H2O2 catalyzed by HRP immobilized in the films and mediated by Fe(CN)63– in solution. The responsive mechanism of the films was explored and discussed. The pH-sensitive property of the system was attributed to the electrostatic interaction between the {Con A/Dex}5 inner layers and the probe at different pH, and the thermo- and salt-responsive behaviors should be ascribed to the structure change of PDEA hydrogels for the PDEA-HRP outermost layers under different conditions. The concept of binary architecture was also used to fabricate {Con A/Dex}5–(PDEA-GOD) films on electrodes, where GOD = glucose oxidase, which was applied to realize the triply switchable bioelectrocatalysis of glucose by GOD in the films with ferrocenedicarboxylic acid as the mediator in solution. This film system with the unique binary architecture may establish a foundation for fabricating a novel type of multicontrollable biosensors based on bioelectrocatalysis with immobilized enzymes.

Gold Nanorods@Mesoporous SiO2@Hyaluronic Acid Core–Shell Nanoparticles for Controlled Drug Delivery

Zhengyin Liang,Cheng Cao,Jun Gao,Wenrong Cai,Junyao Li,Datong Wu,Yong Kong

To improve the therapeutic efficacy of breast cancer (BC), an intelligent tri-responsive vehicle based on core–shell structured gold nanorods (GNR)@mesoporous SiO2 (MS)@hyaluronic acid (HA) is proposed for controlled delivery of methotrexate (MTX). The characterization of the resultant GNR@MS@HA is systematically investigated. GNR is used for photothermal conversion under near-infrared light (NIR) irradiation, and the produced hyperthermia can lead to the ablation of cancer cells. MTX can be encapsulated in the mesopores of MS with the aid of HA, which is grafted to the surface of aminated MS (MS-NH2) through amidation reaction. The proposed vehicle can be used for MTX release in the tri-response of pH, NIR, and hyaluronidase (HAase) and integrates photothermal therapy and chemotherapy so as to improve the inhibition rate of cancer cells. The results of cell viability experiments demonstrate that the developed tri-responsive vehicle displays excellent biocompatibility to L929 mouse fibroblasts while exhibiting high chemo-photothermal synergistic therapy efficiency against murine BC cells (4T1).

Dual-Functional Emulsifying/Antifogging Coumarin-Containing Polymeric Micelle

Feng Wang,Hui Liu

Dual-functional emulsifying/antifogging polymeric micelles have been prepared by the self-assembly of poly(2-(dimethylamino)ethyl methacrylate-co-methyl methacrylate-co-7-acryloxy-4-methylcoumarin) P(DMAEMA-co-MMA-co-AOM). On one hand, the polymeric micelles have spherical morphology and exhibit triple responsive behaviors. The sample with the DMAEMA/MMA/AOM molar ratio of 5/3/3 (T2) is chosen to investigate the influence of pH on the emulsifying performance, and the configuration of polymeric micelles on the oil/water interface is investigated. The amount of micelles arranged on the surface of solidified polymerized beads gradually decreases as the pH changes from 12 to 5, and then total phase separation and demulsification occur at pH 3. On the other hand, we prepare an effective antifogging coating by forming an interchain network (ICN) resulting from self-assembled micelles and photo-cross-linked coumarin units. The ICN-T2 coating can not only absorb water vapor rapidly but also avoid excessive swelling of the amphiphilic terpolymer, thus leading to excellent antifogging performance. To the best of our knowledge, the photo-cross-linkable coumarin derivative is first introduced into an antifogging polymer coating at the molecular level, successfully avoiding the use of chemical cross-linking agents. The coumarin-containing polymeric micelle may be applied in many industrial processes and will provide new insight into the interface or surface property of macromolecular materials.

Triple-Responsive Cross-Linked Micelles for Synergistic Chemo-Photothermal Cancer Therapy

Yaning Ou,Yudong Wang,Kanglin Chen,Wenxin Liao,Zhaomin Tang

We have developed a novel triple-responsive (pH/reduction/near-infrared (NIR) light) cross-linked polymer micelle PEG-P(LL/LL-LA)-PCL-IR820 for enhanced synergistic cancer therapy. This micelle was synthesized using a triblock amphiphilic polymer based on polylysine, functionalized with maleic anhydride (LA) as a cross-linking site and polycaprolactone (PCL) for encapsulating the anticancer drug doxorubicin (DOX). The photosensitizer IR820 was grafted onto PCL to enable photothermal and photodynamic effects under NIR irradiation. The DOX-loaded cross-linked micelles (DCMs) demonstrated exceptional stability under physiological conditions, effectively preventing premature drug release. Rapid DOX release was triggered by the intracellular high glutathione (GSH) level and acidic pH in tumor cells. Under NIR irradiation, DCM micelles exhibited significant photothermal and photodynamic effects, which markedly enhanced the cytotoxicity against B16 tumor cells. In B16 tumor-bearing mice, the DCM-treated group achieved superior tumor growth inhibition and prolonged survival under NIR irradiation compared to un-cross-linked micelles (DUCM) or free DOX, with minimal systemic toxicity. The tumor volume in the DCM + NIR group was significantly inhibited compared with other groups, reaching only about 300 mm3 within 15 days, while for the PBS-treated group, it reached approximately 1400 mm3. The survival rate of the DCM + NIR group was also significantly higher, with an over 70% survival rate within a 40-day observation period. This work presents a robust nanoplatform that combines stability, microenvironment responsiveness, and multimodal therapy, offering a promising strategy for targeted cancer treatment.

Construction of triple-responsive nanogel embedded with upconversion nanoparticles via microemulsion polymerization for near-infrared-controlled drug delivery

Yuanpeng Cai,Danyang Li,Yufei Ling,Huihui Jiang,Hui Liu

It was generally accepted that ultra-violet (UV) light was prone to suffer from shallow tissue penetration and potential cell phototoxicity, limiting its further application in biomedical field. To overcome this shortcoming, a novel triple-responsive nanogel embedded with upconversion nanoparticles (UCNPs), β-NaYF 4 :Yb,Tm, was constructed by emulsion polymerization of photo-responsive SPMA, temperature-sensitive N-isopropylacrylamide (NIPAM), and pH-responsive methacrylic acid (MAA). A series of characterizations were used to prove successful synthesis of the composite nanogel. The system exhibited reversible volume change in response to pH, temperature, and near-infrared (NIR) light, enabled by UCNPs-mediated photoactivation. With doxorubicin (DOX) as a model molecule, drug loading efficiency of composite nanogel could attain relatively high value of 53.06 ± 1.58 %. High release efficiency of 90.53 % could be achieved under UV light, 73.64 % under NIR light, and 82.17 % at pH = 3 & 30 °C, with minimal release efficiency (<6%) under normal physiological condition (pH = 7, 36 °C), confirming controllable releasing behaviors of the nanogel. Also, the drug releasing process could be well fitted with Weibull model. Cytocompatibility assays confirmed the nanogels’ safety (87.6 % viability at 500   μ g/mL), while DOX-loaded nanogels showed significant cytotoxicity toward A549 cells (≤10 % viability, p ≤ 0.001). This study highlighted the potential of UCNPs-doped nanogels for smart, on-demand, and minimally invasive cancer therapy.

One-pot synthesis of triple responsive composite hydrogel based on hyaluronic acid and its targeted controlled drug release

Danyang Li,Yuanpeng Cai,Hui Liu

Owing to the complexity of tumor microenvironment, drug carriers will be required to achieve controlled targeted drug release according to different patient sites. With hyaluronic acid (HA), acrylic acid (AA), and N-isopropylacrylamide (NIPAM) as the main raw materials, a triple-responsive HA-AA-NIPAM composite hydrogel with active targeting was synthesized by one-pot method. The resulting hydrogel was characterized using Fourier infrared spectroscopy, scanning electron microscopy, and X-ray photoelectron spectroscopy. Using doxorubicin (DOX) as a model drug, drug loading content and drug loading efficiency of the hydrogel could attain about 20.22% and 72.39%, respectively. The results demonstrated that sustained release of loaded DOX from HA-AA-NIPAM hydrogel reached approximately 84% after 6 h at pH = 2, t  = 40 °C, and in the presence of HAase environment. Also, six kinetic models were used to fit the releasing data of optimal drug release environment, and the releasing behaviors of the drug-loaded hydrogel could be well described by Ritger-Peppas model with R 2 of >0.99. Besides, after co-cultivation of MDA-MB-231 cell with HA-AA-NIPAM hydrogel, cell viability could maintain >90% and excellent cell adhesion was observed. Laser confocal results revealed that more pronounced green (DOX) fluorescence was displayed in HA-AA-NIPAM hydrogel compared to binary AA-NIPAM hydrogel. Therefore, HA-AA-NIPAM hydrogel was promising to be potentially used in biomedical fields to achieve targeted controlled release of chemotherapeutic drugs.

Micro-environment Triple-Responsive Hyaluronic Acid Hydrogel Dressings to Promote Antibacterial, Collagen Deposition, and Angiogenesis for Diabetic Wound Healing

Wenquan Wang,Jingxia Zheng,Xiaojing Hong,Jiaying Zhou,Yuwen Xiong,Hailong Yang,Shengnan Li,Guoqi Chen,Qiao Su,Wenwen Li,Bin Cheng,Jun Fu,Tong Wu

The clinical treatment of chronic diabetic wounds is a long-standing thorny issue. Strategies targeting the diabetic micro-environment have been developed to promote wound healing. However, it remains challenging to reverse the adverse conditions and re-activate tissue regeneration and angiogenesis. In this work, we develop injectable hydrogels that are responsive to the acid, reactive oxygen species (ROS), and high glucose levels in diabetic wound micro-environment to sustainably deliver tannic acid (TA) to augment antibacterial, anti-inflammation, and anti-oxidation potency. The triple-responsive mechanisms are designed by introducing dynamic acylhydrazone and phenylboronic ester bonds to crosslink modified hyaluronic acid (HA) chains. At the diabetic wound, the acylhydrazone bonds may be hydrolyzed at low pH. Meanwhile, glucose may compete with TA and ROS may oxidize the C-B bond to release TA. Thus, sustained release of TA is triggered by the diabetic micro-environment. The released TA effectively scavenges ROS and kills bacteria. In vivo experiments on diabetic mice demonstrate that the hydrogel dressing highly promotes angiogenesis and extracellular matrix (ECM) deposition, leading to eventual full healing of diabetic skin wound healing. This micro-environment-triggered triple-responsive drug release provides a promising method for diabetic chronic wound healing.
