Ginsenoside Rh1 (Rh1) has anti-inflammatory effects in asthma mice, but the underlying mechanism remains unclear. BALB/c mice were sensitized and challenged with ovalbumin (OVA) to construct asthma model. Mice received Rh1 or tiotropium bromide 0.5 h before OVA challenge. Airway morphology and airway remodeling were assessed by HE staining and Masson's trichrome staining, respectively. Th1/Th2 cytokines in serum or broncho alveolar lavage fluid (BALF) were measured by ELISA kits. Rh1 significantly alleviated the lung resistance and airway resistance, and reduced the number of total inflammation cells, eosinophils, neutrophils, and lymphocytes in BALF of the asthmatic mice. The morphological changes and collagen deposition of airway were also reduced by Rh1 in asthmatic mice. The increase of Eotaxin, IL-4, IL-5, IL-13, and IL-33 and the decrease of IL-12 and IFN-γ in both BALF and serum of OVA exposed mice were reversed by Rh1. Rh1 attenuates OVA-induced asthma in the mice model by regulating Th1/Th2 cytokines balance.

Deep learning-based drug-target interaction (DTI) prediction methods have demonstrated strong performance; however, real-world applicability remains constrained by limited data diversity and modeling complexity. To address these challenges, we propose SCOPE-DTI, a unified framework combining a large-scale, balanced semi-inductive human DTI dataset with advanced deep learning modeling. SCOPE-DTI is constructed from 13 public repositories and expands data volume by up to 100-fold compared to common benchmarks such as the Human dataset. The SCOPE model integrates three-dimensional protein and compound representations, graph neural networks, and bilinear attention mechanisms to effectively capture cross domain interaction patterns and outperform state-of-the-art methods across various DTI prediction tasks. Additionally, SCOPE-DTI provides a user-friendly interface and database. We further demonstrate its effectiveness by experimentally identifying anticancer targets of two bioactive natural compounds. By offering comprehensive data, advanced modeling, and accessible tools, SCOPE-DTI accelerates drug discovery research.

Graphene oxide enhanced hydrogel enables tunable mechanics. • Sustained Rh1 release promotes angiogenesis via VEGF/CD31 upregulation. • Hydrogel boosts anti-inflammatory cytokines (IL-4, IL-10) for faster healing. • The water gel dressing has an antibacterial rate of up to 98.69% against S. aureus . During the healing process of skin wounds, there are challenges such as slow tissue regeneration and bacterial infections. This paper has developed a drug-loaded composite hydrogel dressing that can promote angiogenesis, inhibit bacteria, and reduce inflammation. The hydrogel is composed of gelatin (Gel) and acrylamide (AM), and is formed into a biomimetic cell membrane structure through cross-linking. By adding different concentrations of graphene oxide (GO), the tensile strength of the hydrogel dressing can be significantly increased (from 57.54 KPa to 95 KPa), and the adhesion strength (shear strength) can be effectively enhanced (16.44 KPa). Additionally, Panax ginsenoside Rh1 has been successfully encapsulated in the hydrogel, achieving local controlled release. In vivo evaluation shows that the enhanced matrix by graphene oxide and the sustained release of Panax ginsenoside Rh1 have an antibacterial rate of up to 98.69% against S. aureus . It also stimulates the proliferation, migration, and tube formation of endothelial cells, thereby achieving strong vascularization at the wound site. Moreover, it can enhance the secretion capacity of anti-inflammatory factors (IL-4 from 19.54 to 105.61 pg/mL, IL-10 from 151.94 to 339.98 pg/mL), thereby accelerating the wound healing process. This hydrogel dressing provides a promising strategy for achieving high-quality wound repair. Download: Download high-res image (223KB) Download: Download full-size image

Serving as a world-renowned tonic, ginseng contains various types of bioactive metabolites. The comprehensive profiling of these metabolites may help explore the nutritional value of ginseng. Due to high variety in chemical structures, simultaneous monitoring of these metabolites remains a challenge. Herein, a high-throughput and high-selectivity online derivatization mass spectrometry imaging strategy targeting C C was developed. As a widely existed chemical group, C C acts like a bridge connecting different kinds of metabolites. [ d 0 ]/[ d 10 ]-Bis(pyridine) iodine tetrafluoroboride reagent was chosen for the derivatization of C C, the detection sensitivity of which increased about 3 magnitudes after derivatization. Assisted by laser ablation carbon fiber ionization mass spectrometry, the spatial distribution of bioactive metabolites in mountain-cultivated and garden-cultivated ginseng were visualized. The correlation heatmap results revealed that metabolites in mountain-cultivated ginseng hold higher correlation than those in garden-cultivated ginseng. The proposed method showed potential in providing comprehensive information on the nutrient content of foods.
