Establishment, application and comparison of three immunoaffinity pretreatment techniques for mycotoxins systematically

Tian Ye,Liu Zhaoying,Sun Mingna,Duan Jinsheng,Verheecke-Vaessen Carol,Du Daolin,Li Ming

To further promote the pretreatment technique, the immunoaffinity column (IAC), immunoaffinity magnetic (IAM) and multi-component immunoaffinity magnetic (multi-IAM) for mycotoxins were developed and evaluated systematically. Anti-mycotoxin-McAbs had immobilized on the CNBr-Sepharose 4B, carboxyl magnetic bead and protein G magnetic bead to prepare the IAC/IAM/multi-IAM, respectively. The capture efficiencies of mycotoxins (100 ng) reached 92.8% for IAC and exceeded 94.0% for IAM/multi-IAM. The multi-IAM showed more beneficial features in multiple targets, pretreatment time and recycling time than IAC/IAM, which could simultaneously pretreat AFB 1 /DON/ZEN in 15 min with three recycling times. The recoveries of IAC/IAM/multi-IAM coupled with ELISA ranged from 85.2 to 105.1%, with RSD between 4.0 and 13.2%. Moreover, the mycotoxin-positive authentic samples were detected from 1.1 to 749.6 ng/g by ELISA, and 1.2 to 762.7 ng/g by LC-MS/MS, with correlated R 2 exceeding 0.9955. The proposed IAC/IAM/multi-IAM owned the desired performance, such as specificity, accuracy, simpleness, efficiency, cost-effectiveness, simultaneity and environmentally friendly. This study can provide the theoretical basis for three immunoaffinity pretreatment techniques systematically, which may guide their application under different requirements and scenarios. Self-driven nanoprodrug platform with enhanced ferroptosis for synergistic photothermal-IDO immunotherapy

Ping Huang,Yao Yang,Wenyan Wang,Zimu Li,Nansha Gao,Hongzhong Chen,Xiaowei Zeng

Insufficient immune stimulation and stubborn immune resistance are the critical factors limiting tumor immunotherapy. Here, we report a multifunctional nanoprodrug platform with self-driven indoximod (IND) release and oxidative stress amplification. The aim is to awaken immune responses and block the indoleamine 2,3-dioxygenase (IDO) pathway through a combination of ferroptosis , photothermal therapy, and immunotherapy. This nanosystem improved the delivery efficiency of IND due to click chemistry linked ROS responsive prodrug and self-driven drug release. Meanwhile, the tactic of simultaneously increasing ROS and eliminating GSH amplified oxidative stress and strengthened ferroptosis, which further enhanced immunogenicity along with polydopamine-based photothermal therapy. IDO immunization combined with ferroptosis as well as photothermal therapy not only stimulated immune response, but also reversed immune suppression with enhanced immune memory . Therefore, primary tumor, distant tumor, and cancer metastasis were inhibited. This study provides a perspective on immunotherapeutics for cancer treatment.

The Importance of Particle Shape: Effect of Nonspherical Particles and Their Stabilized Pickering Emulsions on Immunization Efficacy

Yanan Li,Jie Teng,Qiuting Chen,Qingze Fan,Hiroyuki Oku,Guanghui Ma,Jie Wu

Adjuvants based on spherical particles and stabilized Pickering emulsions represent a significant area of research in the adjuvant field. However, the immune effects of adjuvants containing nonspherical particles and their stabilized Pickering emulsions remain largely unexplored. Particle shape plays a critical role in influencing particle–cell interactions and antigen storage efficiency. In this study, it is aimed to synthesize nonspherical particles with diverse morphologies and sizes and successfully utilize one type to stabilize Pickering emulsions. In this research, their immunological effects are further evaluated by examining the activation of antigen-presenting cells (APCs) and their impact on both cellular and humoral immunity. In these findings, it is demonstrated that nonspherical particles extend the in vivo residence time of vaccines and enhance APCs activation, thereby improving cellular immunity. Additionally, Pickering emulsions stabilized by nonspherical particles exhibit superior flexibility, higher antigen uptake by APCs, and more robust APCs activation compared to those stabilized by spherical particles. These advantages ultimately result in enhanced humoral and cellular immune responses.

Anti-SARS-CoV-2 IgG and IgM detection with a GMR based LFIA system

Qiaoge Bayin,Lei Huang,Chunhui Ren,Yusheng Fu,Xing Ma,Jinhong Guo

Since December 2019, Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) has caused millions of deaths and seriously threatened the safety of human life; indeed, this situation is worsening and many people are infected with the new coronavirus every day. Therefore, it is very important to understand patients’ degree of infection and infection history through antibody testing. Such information is useful also for the government and hospitals to formulate reasonable prevention policies and treatment plans. In this paper, we develop a lateral flow immunoassay (LFIA) method based on superparamagnetic nanoparticles (SMNPs) and a giant magnetoresistance (GMR) sensing system for the simultaneously quantitative detection of anti -SARS-CoV-2 immunoglobulin M (IgM) and G (IgG). A simple and time-effective co-precipitation method was utilized to prepare the SMNPs, which have good dispersibility and magnetic property, with an average diameter of 68 nm. The Internet of Medical Things-supported GMR could transmit medical data to a smartphone through the Bluetooth protocol, making patient information available for medical staff. The proposed GMR system, based on SMNP-supported LFIA, has an outstanding advantage in cost-effectiveness and time-efficiency, and is easy to operate. We believe that the suggested GMR based LFIA system will be very useful for medical staff to analyze and to preserve as a record of infection in COVID-19 patients.

Incorporation of a Toll-like receptor 2/6 agonist potentiates mRNA vaccines against cancer and infectious diseases

Gu Yangzhuo,Yang Jingyun,He Cai,Zhao Tingmei,Lu Ran,Liu Jian,Mo Xianming,Wen Fuqiang,Shi Huashan

mRNA vaccines have emerged rapidly in recent years as a prophylactic and therapeutic agent against various diseases including cancer and infectious diseases. Improvements of mRNA vaccines have been underway, among which boosting of efficacy is of great importance. Pam2Cys, a simple synthetic metabolizable lipoamino acid that signals through Toll-like receptor (TLR) 2/6 pathway, eliciting both humoral and cellular adaptive immune responses, is an interesting candidate adjuvant. To investigate the enhancement of the efficacies of mRNA vaccines by Pam2Cys, the adjuvant was incorporated into mRNA-lipid nanoparticles (LNPs) to achieve co-delivery with mRNA. Immunization with the resulting mRNA-LNPs (Pam2Cys) shaped up the immune milieu in the draining lymph nodes (dLNs) through the induction of IL-12 and IL-17, among other cytokines. Antigen presentation was carried out mainly by migratory and dLN-resident conventional type 2 DCs (cDC2s) and significantly more potent antitumor responses were triggered in both prophylactic and therapeutic tumor models in a CD4 + and CD8 + T cell-dependent fashion. Accompanying memory antitumor immunity was also established. Moreover, the vaccine also stimulated much more robust humoral and cellular immunity in a surrogate COVID-19 prophylactic model. Last but not the least, the new vaccines exhibited good preliminary safety profiles in murine models. These facts warrant future development of Pam2Cys-incorporated mRNA vaccines or relevant mRNA therapeutics for clinical application.

Non-invasive transdermal delivery of biomacromolecules with fluorocarbon-modified chitosan for melanoma immunotherapy and viral vaccines

Zhu Wenjun,Wei Ting,Xu Yuchun,Jin Qiutong,Chao Yu,Lu Jiaqi,Xu Jun,Zhu Jiafei,Yan Xiaoying,Chen Muchao,Chen Qian,Liu Zhuang

Transdermal drug delivery has been regarded as an alternative to oral delivery and subcutaneous injection. However, needleless transdermal delivery of biomacromolecules remains a challenge. Herein, a transdermal delivery platform based on biocompatible fluorocarbon modified chitosan (FCS) is developed to achieve highly efficient non-invasive delivery of biomacromolecules including antibodies and antigens. The formed nanocomplexes exhibits effective transdermal penetration ability via both intercellular and transappendageal routes. Non-invasive transdermal delivery of immune checkpoint blockade antibodies induces stronger immune responses for melanoma in female mice and reduces systemic toxicity compared to intravenous injection. Moreover, transdermal delivery of a SARS-CoV-2 vaccine in female mice results in comparable humoral immunity as well as improved cellular immunity and immune memory compared to that achieved with subcutaneous vaccine injection. Additionally, FCS-based protein delivery systems demonstrate transdermal ability for rabbit and porcine skins. Thus, FCS-based transdermal delivery systems may provide a compelling opportunity to overcome the skin barrier for efficient transdermal delivery of bio-therapeutics.

An Antigen-Free Nanovaccine-Enabled Holo-Immunotherapy for Establishing Robust Memory Immune Tolerance in Autoimmune Diseases

Shuo Wang,Yuequan Wang,Yantong Li,Xiaoxue Lai,Miao Hu,Meng Chen,Cong Li,Xinrong Liu,Yanzhi Song,Yihui Deng

Inflammation-related immune cell subsets with abnormal activation, impaired tolerance, and complex intercellular interactions drive the progression and incurability of autoimmune diseases (AIDs). Here, we present an antigen-free nanovaccine-enabled holo-immunotherapy that comprehensively modulates inflammation, innate immunity, and adaptive immunity. The nanovaccine, composed of dual NF-κB inhibitors (budesonide and bortezomib) and comodified with sialic acid (SA) and enzyme-cleavable polyethylene glycol (PEG), leverages the SA-Siglec axis for targeted delivery to inflammation-related macrophage, dendritic cell, and CD4+ T cell subsets. By bridging innate and adaptive immunity, this multitargeted strategy synergistically reshapes the inflammatory microenvironment, establishes robust memory immune tolerance, and promotes immune homeostasis restoration, leading to sustained efficacy and delayed relapse in the collagen-induced arthritis (CIA) mouse model. Unlike traditional vaccines, the antigen-free design eliminates reliance on exogenous antigens, minimizes off-target immune risks, and holds promise for broader clinical applicability. The nanovaccine outperforms anti-TNF-α antibodies in treating models of CIA, adjuvant-induced arthritis, and chronic ulcerative colitis, highlighting its potential as a universal vaccine platform for AID therapy. Overall, this work provides a strategy to address AID challenges through precise immune regulation and durable therapeutic benefits.

PD-L1-Targeting Autophagy Modulator to Upregulate MHC-I and Activate Photo-Immunotherapy for Metastatic Tumor Eradication

Zhen-Ming Lu,Zi-Wen Qiu,Yan-Mei Li,Ke-Yan Zhang,Ye-Yang Wu,Ni Yan,Hong Cheng

Breast cancer cells are characterized by heightened autophagy, which impairs tumor-associated antigen presentation and represents a significant barrier to the antitumor immunity. In this study, a PD-L1-targeting autophagy modulator (PFC@CQ) is fabricated to activate the photoimmunotherapy against breast cancer. Specifically, the hydrophobic photosensitizer protoporphyrin IX (PpIX) is covalently linked to the hydrophobic peptide FFVLK and a PD-L1-targeting peptide sequence CLQKTPKQC, resulting in the formation of an amphiphilic photosensitizer-peptide conjugate (PpIX-FFVLK-CLQKTPKQC, called PFC), which is capable of encapsulating the autophagy inhibitor chloroquine (CQ). PFC@CQ can not only facilitate the targeted drug codelivery to PD-L1-overexpressing breast cancer cells, but also effectively disrupt their immune evasion by blocking PD-1/PD-L1 pathway. Upon light irradiation, the photodynamic therapy (PDT) of PFC@CQ induces tumor cell destruction and immunogenic cell death (ICD), causing the release of damage-associated molecular patterns (DAMPs). Simultaneously, PFC@CQ can inhibit autophagy pathway to mediate the upregulation of MHC-I, thereby enhancing antigen presentation. This cascade immunomodulation promotes the dendritic cell maturation and CD8+ T cell activation, leading to a synergistic suppression of both primary and metastatic tumors. This work introduces an innovative autophagy modulation strategy with potent immunomodulatory capability, demonstrating a potential to trigger systemic antitumor immune responses through local treatment.

3D Printing Scaffold Vaccine for Antitumor Immunity

Yue Zhang,Jialu Xu,Ziying Fei,Huaxing Dai,Qin Fan,Qianyu Yang,Yitong Chen,Beilei Wang,Chao Wang

Cancer vaccine platform has attracted great interest in the field of cancer immunotherapy. Here, 3D printed scaffolds loaded with immunoregulators are developed for enhanced cancer immunotherapy. The rapid manufacturing and precise molding based on 3D printing can realize the mass manufacturing of cancer vaccines and personalized design. Meanwhile, compared to the traditional hydrogel, the 3D-scaffold with porous structure endows its similar functions compared with real lymphoid organs by recruitment of a great number of immune cells, leading to the formation of “artificial tertiary lymphoid structures,” where there is a promising site to enhance both humoral and cellular immune responses. Efficient anticancer immunity is induced when combined with immune checkpoint blockade to inhibit the tumor growth. Personalized antitumor scaffold vaccines are further demonstrated for filling of tumor site after surgery to prevent cancer metastasis. Taken together, these results promise the 3D printing scaffold vaccine as the potential strategy for cancer vaccine therapy in the future.

Tumor-Adhesive Chitosan-Derived Multi-Immune Agonist Unleashes Strong and Durable Anti-Cancer Immunity

Huilan He,Liang Liu,Yun Zheng,Jinlong Ji,Li Cao,Chunlian Ye,Yu Sun,Ying Zhang,Zhiyuan Zhong

The immunomodulation of the tumor microenvironment is critical for effective cancer immunotherapy, particularly for tumors that exhibit limited responses to conventional treatments. However, current immune agonists developed for tumor immunomodulation face several challenges, such as poor intratumoral retention, inadequate biocompatibility, and restricted cellular targets, which ultimately hamper their therapeutic efficacy and clinical application. In this study, a tumor-adhesive chitosan-tethered immune agonist construct (TACTIC) is introduced, which demonstrates good biocompatibility and robust immunostimulatory effects, enhancing the immunogenicity of tumor cells while simultaneously stimulating pro-inflammatory responses in various immune cell populations. Mechanistic investigations reveal that TACTIC targets multiple signaling pathways, conferring it to effectively remodel the irradiated tumor microenvironment, improve tumor control on murine cancer models post-radiotherapy, and elicit systemic immune responses with memory effects. The findings highlight the potential of TACTIC as a powerful macromolecular immune adjuvant, paving the way for its broader application in innovative cancer immunotherapies.

A combination of PD-L1-targeted IL-15 mRNA nanotherapy and ultrasound-targeted microbubble destruction for tumor immunotherapy

Xiaoxuan Wang,Fangxuan Li,Jialu Zhang,Lu Guo,Mengmeng Shang,Xiao Sun,Shan Xiao,Dandan Shi,Dong Meng,Yading Zhao,Chao Jiang,Jie Li

PD-1/PD-L1-based immune checkpoint blockade therapy has shown limited benefits in tumor patients, partially attributed to the inadequate infiltration of immune effector cells within tumors. Here, we established a nanoplatform named DPPA/IL-15 NPs to target PD-L1 for the tumor delivery of IL-15 messenger RNA (mRNA). DPPA/IL-15 NPs were endowed with ultrasound responsiveness and contrast-enhanced ultrasound (CEUS) imaging performance. They effectively protected IL-15 mRNA from degradation and specifically transfected it into tumor cells through the utilization of ultrasound-targeted microbubble destruction (UTMD). This resulted in the activation of IL-15-related immune effector cells while blocking the PD-1/PD-L1 pathway. In addition, UTMD could generate reactive oxygen species (ROS) that induce endoplasmic reticulum (ER) stress-driven immunogenic cell death (ICD), initiating anti-tumor immunity. In vitro and in vivo studies revealed that this combination therapy could induce a robust systemic immune response and enhance anti-tumor efficacy. Thus, this combination therapy has the potential for clinical translation through enhanced immunotherapy and provides real-time ultrasound imaging guidance.

Chiral polypeptide hydrogels regulating local immune microenvironment and anti-tumor immune response

Ding Junfeng,Wang Tianran,Lin Zhiqiang,Li Zhenyu,Yang Jiaxuan,Li Fujiang,Rong Yan,Chen Xuesi,He Chaoliang

The impact of chirality on immune response has attracted great interest in cancer vaccine research recently. However, the study of chiral synthetic polypeptide hydrogels as cancer vaccines as well as of the impact of biomaterials themselves for antitumor immunotherapy has rarely been reported. Here, we show the key role of residue chirality of polypeptide hydrogels in antitumor immunity and local immune microenvironment regulation. Compared to poly(γ-ethyl-L-glutamate)-based hydrogels (L-Gel), poly(γ-ethyl-D-glutamate)-based hydrogels (D-Gel) induces enhanced level of immune cell infiltration. However, D-Gel causes higher levels of suppressive markers on antigen-presenting cells and even induces stronger T cell exhaustion than L-Gel. Finally, D-Gel establishes a local chronic inflammatory and immunosuppressive microenvironment and shows insufficient anti-tumor effects. Conversely, the milder host immune responses induced by L-Gel leads to more effective tumor inhibition. This study provides insights on the role of residue chirality in the regulation of local immune microenvironment and affecting antitumor immune response.
