In Vitro Assessment of Biodegradable and Biocompatible Brain-Derived Neurotrophic Factor-Mesenchymal Stem Cells Incorporated Chitosan/Gelatin Scaffold

Dai Liming,Zhu Hongtao,Zhang Chaoguang,Liu Tao,Wang Yi

In this paper, the biocompatible chitosan/gelatin scaffolds at different chitosan/gelatin ratios were fabricated by a facile method. The important physical parameters of the scaffolds, including density, porosity and tensile strength were measured. MSCs were transfected to introduce brain-derived neurotrophic factor (BDNF) gene into the cells. To evaluate the bioactivity of the scaffolds, the gene-modified BDNF-MSCs were cultivated on the scaffolds, and the cell proliferation and BDNF expression level were determined.

Sheddable, degradable, cationic micelles enabling drug and gene delivery

Yongyong Li,Xue Lei,Haiqing Dong,Tianbin Ren

In this work, sheddable, degradable, cationic micelles were designed and developed based on intermediate disulfide-linked poly(ε-caprolactone)-b-poly(N,N-dimethylamino-2-ethylmethacrylate) (PCL-SS-PDMA) diblock copolymers, for glutathione (GSH)-mediated intracellular delivery of anticancer drug doxorubicin (DOX) and gene. The PCL-SS-PDMA diblock polymers with different PDMA block lengths were prepared by a combination of ring opening polymerization (ROP) and atom transfer radical polymerization (ATRP). Driven by hydrophobic interaction, these polymers self-assembled into nanoscaled micelles with size ranging from 70 to 200 nm, and positive surface charges from +24 to +37 mV. The PDMA length was found to determine the surface charge and in turn affect the physiochemical properties and cumulative drug release. More importantly, glutathione mediated intracellular drug release was observed by intracellular fluorescence arising from DOX for the GSH treated cells. The accelerated drug release was induced by the structural disassembly after disulfide cleavage. The PCL-SS-PDMA block polymers exhibited high DNA binding affinity and cellular uptake efficiency. The gene transfection efficiency of PCL-SS-PDMA/DNA complex showed relatively low transfection efficiency in 293 T and HeLa cells, compared to PEI 25K. However, the transfection efficacy dramatically outperformed PEI 25K in human oral carcinoma cell lines (KB and CAL-27 cells). A ten-fold population of the transfected cells was found for the PS-SS-PDMA polymer, compared with PEI. The experimental results show the great potential of the new micelles in gene and drug delivery for cancer therapy.

A biodegradable and flexible neural interface for transdermal optoelectronic modulation and regeneration of peripheral nerves

Sun Pengcheng,Li Chaochao,Yang Can,Sun Mengchun,Hou Hanqing,Guan Yanjun,Chen Jinger,Liu Shangbin,Chen Kuntao,Ma Yuan,Huang Yunxiang,Li Xiangling,Wang Huachun,Wang Liu,Chen Shengfeng,Cheng Haofeng,Xiong Wei,Sheng Xing,Zhang Milin,Peng Jiang,Wang Shirong,Wang Yu,Yin Lan

Optoelectronic neural interfaces can leverage the photovoltaic effect to convert light into electrical current, inducing charge redistribution and enabling nerve stimulation. This method offers a non-genetic and remote approach for neuromodulation. Developing biodegradable and efficient optoelectronic neural interfaces is important for achieving transdermal stimulation while minimizing infection risks associated with device retrieval, thereby maximizing therapeutic outcomes. We propose a biodegradable, flexible, and miniaturized silicon-based neural interface capable of transdermal optoelectronic stimulation for neural modulation and nerve regeneration. Enhancing the device interface with thin-film molybdenum significantly improves the efficacy of neural stimulation. Our study demonstrates successful activation of the sciatic nerve in rodents and the facial nerve in rabbits. Moreover, transdermal optoelectronic stimulation accelerates the functional recovery of injured facial nerves.

Healable, Recyclable, and Multifunctional Soft Electronics Based on Biopolymer Hydrogel and Patterned Liquid Metal

Xing Peng Hao,Chuan Wei Zhang,Xin Ning Zhang,Li Xin Hou,Jian Hu,Michael D. Dickey,Qiang Zheng,Zi Liang Wu

Recent years have witnessed the rapid development of sustainable materials. Along this line, developing biodegradable or recyclable soft electronics is challenging yet important due to their versatile applications in biomedical devices, soft robots, and wearables. Although some degradable bulk hydrogels are directly used as the soft electronics, the sensing performances are usually limited due to the absence of distributed conducting circuits. Here, sustainable hydrogel-based soft electronics (HSE) are reported that integrate sensing elements and patterned liquid metal (LM) in the gelatin–alginate hybrid hydrogel. The biopolymer hydrogel is transparent, robust, resilient, and recyclable. The HSE is multifunctional; it can sense strain, temperature, heart rate (electrocardiogram), and pH. The strain sensing is sufficiently sensitive to detect a human pulse. In addition, the device serves as a model system for iontophoretic drug delivery by using patterned LM as the soft conductor and electrode. Noncontact detection of nearby objects is also achieved based on electrostatic-field-induced voltage. The LM and biopolymer hydrogel are healable, recyclable, and degradable, favoring sustainable applications and reconstruction of the device with new functions. Such HSE with multiple functions and favorable attributes should open opportunities in next-generation electronic skins and hydrogel machines.

Biodegradable oxygen-evolving metalloantibiotics for spatiotemporal sono-metalloimmunotherapy against orthopaedic biofilm infections

Su Zheng,Xu Dongdong,Hu Xianli,Zhu Wanbo,Kong Lingtong,Qian Zhengzheng,Mei Jiawei,Ma Ruixiang,Shang Xifu,Fan Wenpei,Zhu Chen

Pathogen-host competition for manganese and intricate immunostimulatory pathways severely attenuates the efficacy of antibacterial immunotherapy against biofilm infections associated with orthopaedic implants. Herein, we introduce a spatiotemporal sono-metalloimmunotherapy (SMIT) strategy aimed at efficient biofilm ablation by custom design of ingenious biomimetic metal-organic framework (PCN-224)-coated MnO 2 -hydrangea nanoparticles (MnPM) as a metalloantibiotic. Upon reaching the acidic H 2 O 2 -enriched biofilm microenvironment, MnPM can convert abundant H 2 O 2 into oxygen, which is conducive to significantly enhancing the efficacy of ultrasound (US)-triggered sonodynamic therapy (SDT), thereby exposing bacteria-associated antigens (BAAs). Moreover, MnPM disrupts bacterial homeostasis, further killing more bacteria. Then, the Mn ions released from the degraded MnO 2 can recharge immune cells to enhance the cGAS-STING signaling pathway sensing of BAAs, further boosting the immune response and suppressing biofilm growth via biofilm-specific T cell responses. Following US withdrawal, the sustained oxygenation promotes the survival and migration of fibroblasts, stimulates the expression of angiogenic growth factors and angiogenesis, and neutralizes excessive inflammation. Our findings highlight that MnPM may act as an immune costimulatory metalloantibiotic to regulate the cGAS-STING signaling pathway, presenting a promising alternative to antibiotics for orthopaedic biofilm infection treatment and pro-tissue repair.
