Responses of Scirpus triqueter, soil enzymes and microbial community during phytoremediation of pyrene contaminated soil in simulated wetland

Xinying Zhang,Xiaoyan Liu,Shanshan Liu,Fahui Liu,Lisha Chen,Gang Xu,Chenglin Zhong,Pengcheng Su,Zhengnan Cao

The aim of this study was to determine the enhancement of Scirpus triqueter in the dissipation of pyrene and the interaction of pyrene with plant, soil enzymes and microbial community. The results indicated that the dissipation ratios of pyrene in the rhizospheric and non-rhizospheric soil were 64.65   ±   3.86% and 54.49   ±   2.74%, respectively, and were higher than that in the unplanted soil (42.60   ±   0.71%) at 80d after planting S. triqueter . The pyrene was toxic to S. triqueter , as evidenced by growth inhibition in height, diameter, shoot number and biomass during the planting period. The activities of dehydrogenase decreased significantly at the presence of pyrene in soils, and increased remarkably with the introduction of S. triqueter . It was found that the pyrene addition increased the ratios of fungal/total fatty acids and gram-positive/gram-negative, but the presence of S. triqueter decreased the ratios of gram-positive/gram-negative. A larger stress level was found in the pyrene treated soils without S. triqueter . The ratio of aerobic/anaerobic bacteria decreased with increasing pyrene concentration, but increased when S. triqueter was planted. The principal analysis of phospholipid fatty acid signatures revealed that microbial community structures in the rhizospheric and non-rhizospheric soil were similar, but different from those in the unplanted and control soil.

Effects of interaction between enrofloxacin and copper on soil enzyme activity and evaluation of comprehensive toxicity

Rui Yang,Jinhua Wang,Lusheng Zhu,Jun Wang,Lili Yang,Shushuai Mao,Jeremy L. Conkle,Yangyang Chen,Young Mo Kim

Antibiotics are detected in association with heavy metals in the soil. However, interactions between antibiotics and heavy metals on soil enzyme activity have yet to been studied thoroughly. In this study, soil enzyme activity (urease, sucrase, phosphatase , and Rubisco) were measured after exposure to soils dosed with copper (Cu) and/or enrofloxacin (ENR) over 28 days. Enzyme responses to ENR only treatments varied, but Cu exhibited a strong negative response from all soil enzymes except Rubisco. An interaction between the effects of the two pollutants on soil enzymes was observed in the combined contamination treatments. Greater comprehensive toxicity to soil enzyme activity was observed in combined treatment groups compared to other groups. We anticipate our studies can provide a scientific theoretical basis for the combined pollution of antibiotics and heavy metals in soil.

Comparable effects of manure and its biochar on reducing soil Cr bioavailability and narrowing the rhizosphere extent of enzyme activities

Shibin Liu,Shengyan Pu,Daili Deng,Hongyan Huang,Chun Yan,Hui Ma,Bahar S. Razavi

Chromium (Cr) contamination is especially hazardous to soil biota. Application of manure and biochar has been frequently proposed to remediate Cr-contaminated soil. However, the understanding of mechanisms behind manure and biochar impacts on soil enzyme activities requires advanced visualization technologies. For the first time, we compared manure and its biochar influence on the spatial distribution of β-glucosidase, N-acetyl-glucosaminidase and phosphomonoesterase activities in Cr-contaminated soil using direct zymography. Maize was planted for 45 days in (a) soil mixed with manure, (b) soil mixed with manure-derived biochar and (c) soil without any addition. Soil pH decreased over 45 days, inducing an increase in acid soluble Cr. The concomitant decrease in β-glucosidase and N-acetyl-glucosaminidase activities explained the narrowing rhizosphere extent of enzyme activities by 13–44%, indicating that increased Cr bioavailability decreases microbial activities. A larger maize performance index and the greatest plant shoot/root ratio after biochar application suggested enhanced maize growth ( p  < 0.05). In contrast, manure induced the narrowest extent of β-glucosidase and phosphomonoesterase activities due to the addition of labile organic compounds and nutrients following its application. Our study emphasizes the importance of pH on Cr bioavailability and enzyme activities and demonstrates that biochar application is more ideally suited for remediating Cr-contaminated soil.

Effects of Cd2+ and Pb2+ on enantioselective degradation behavior of α-cypermethrin in soils and their combined effect on activities of soil enzymes

Jiang Wenqi,Yao Guojun,Jing Xu,Liu Xueke,Liu Donghui,Zhou Zhiqiang

Heavy metals may coexist with pesticides in farmland through wastewater irrigation, application of pesticides and chemical fertilizers, or unappropriated waste disposal. Heavy metals are toxic to soil microorganism, which may influence the environmental behavior of pesticides subsequently. In this study, the influence of Cd 2+ and Pb 2+ on the degradation of α-cypermethrin and its metabolites, 3-phenoxphenoxybenzoic acid (3-PBA) and 3-(2’,2’-dichlorovinyl)-2,2-dimethylcyclopropane carboxylic acid (DCCA), were investigated through soil incubation experiment. It was found heavy metals like Cd 2+ and Pb 2+ will inhibit the degradation of α-cypermethrin, especially at high concentrations. Pb 2+ has a stronger inhibitory effect on the degradation of α-cypermethrin than Cd 2+ in the same concentration. With the presence of 10 mg/kg Pb 2+ , the half-life of α-cypermethrin increased from 41.1 to 99.9 days, even the half-life was 129.3 days with 50 mg/kg of Pb 2+ . Besides, heavy metals influenced the chiral selective degradation of α-cypermethrin. The enantiomer fraction was near 0.5 when 10 mg/kg of heavy metals existed. Furthermore, the adverse effects of heavy metals on soil urease, catalase, and sucrase activity were assayed. In tested concentrations (10 and 50 mg kg -1 ), the heavy metals result in strong inhibition of the activity of the enzymes present on soil, jeopardizing the biodegradation by the microbiome and which may inhibit the degradation of α-cypermethrin.

Catalytic efficiency of soil enzymes explains temperature sensitivity: Insights from physiological theory

Chaoyang Liu,Haixia Tian,Xiaoyue Gu,Ni Li,Xiaoning Zhao,Mei Lei,Hattan Alharbi,Mallavarapu Megharaj,Wenxiang He,Yakov Kuzyakov

Soil enzymes are crucial for carbon and nutrient cycling and are highly sensitive to warming. Biochemical reaction rates increase with temperature according to the Arrhenius law, but changes in microbial physiology may partially counteract this warming-induced acceleration that leads enzymatic rates to deviate from Arrhenius law. Here, we attempt to reconcile disparate views on the enzyme responses to warming based on the Arrhenius law and physiological theory by enzyme catalytic efficiency. In this study, we tested the kinetic parameters of five key enzymes of C, N, and P cycling to warming (from 0 to 40 °C) in cropland soils originating from 5 different temperate zones . The soils were incubated for one month at 0, 10, 20, 30, and 40 °C (±0.5 °C) with 60% water holding capacity (WHC). The kinetic parameters were calculated and measured at a range of 4-methyumbelliferone (MUB)-substrate concentrations. We found that catalytic efficiency (V max /K m ) of individual enzymes ranged from 0.05 to 27 s −1 between 0 and 40 °C. Maximum reaction rate (V max ) increased with warming, while V max /K m of most enzymes remained stable by warming at low temperatures (up to 10 °C), and it raised from 20 to 40 °C. Most enzymes had lower substrate affinities (K m ) and increased their efficiency with warming. Consistent with studies considering Arrhenius law solely, the temperature sensitivity (Q 10 ) of V max decreased with warming. However, the Q 10 of V max /K m displayed a lower value in the cold but a higher value in warmer temperature, which confirmed microbial adaptation based on physiological theory, consequently encouraging its linking with the Arrhenius law. Therefore, Arrhenius linked with physiological theory could correct explanation of enzyme activities by warming. Considering the microbial adaptation to temperature, the present predicted warming-induced acceleration of soil organic matter decomposition might be overestimated in cold and underestimated in warm environments.

Environmentally relevant concentrations of microplastic exhibits negligible impacts on thiacloprid dissipation and enzyme activity in soil

Zhenlan Xu,Xiaoting Qian,Chao Wang,Chunrong Zhang,Tao Tang,Xueping Zhao,Lingxiangyu Li

Microplastics (MPs) as a type of emerging contaminant in the environment have attracted extensive attentions in recent years, and understanding the impacts of MPs on soil biodiversity and functioning are thus increasingly urgent. Nevertheless, few studies were performed to investigate potential effects of MPs on decay of soil organic pollutants in particular pesticides and enzyme activities . Herein, three types of MPs including polystyrene fragments (PS-50) and polyvinyl chloride beads (PVC-42000 and PVC-10) were added to soil at environmentally relevant concentrations (0.2 and 1.0%) to study their impacts on dissipation of thiacloprid and activities of urease, acid phosphatase , invertase and catalase . MPs exhibited negligible impacts on thiacloprid dissipation regardless of MPs type and content, being probably attributed to the unaltered bioavailability of thiacloprid in soil even after an addition of MPs, which was documented by using the hydroxypropyl-β- cyclodextrin (HPCD) extraction method. Batch sorption experiments also exhibited the comparable adsorption capacity of thiacloprid to soil with and without MPs, along with K f valuses of 3.44–3.77. Besides, MPs exerted negligible effects on enzyme activities of soil. Taken together, this study showed negligible impacts of MPs at environmentally relevant concentrations on thiacloprid dissipation and enzyme activity, expanding our knowledge on impacts of MPs at the environmentally relevant concentrations on pesticide dissipation in soil.

Nanosilicon-based vermicompost leachate and Trichoderma harzianum promote the growth of Panax quinquefolius L. cultivated under forests by improving soil enzyme activity

Qiaoran Ma,Guobing Tian,Shengchao Yang,Junwen Chen,Wei Fan,Ping Zhao,Yan Wang,Jiamin Liu,Yonglin Liu,Shuhui Zi,Shuran He

Planting vegetation under forests in agroforestry systems fosters sustainable agricultural development. However, Limited availability of biostimulants for agroforestry and unclear mechanisms of plant growth promotion. This study synthesized and evaluated a novel biostimulant, nanosilicon-based vermicompost leachate (NSVCL), using Panax quinquefolius L. as the research plant species for forest cultivation. Trichoderma harzianum (TH) was chosen to represent a biostimulant with broad-spectrum properties, and its application effects were compared with NSVCL. The regulatory effects of both on the physiological characteristics and rhizosphere soil microenvironment of P. quinquefolius were investigated, with untreated plants serving as controls. Compared to the control, NSVCL and TH increased the dry weight of P. quinquefolius roots 129.33 % and 23.50 %, respectively. NSVCL was applied more effectively than TH. Additionally, NSVCL markedly improved leaf anatomical traits, including palisade and spongy tissue thickness, overall leaf thickness, chloroplast number, and cuticle thickness. Application of NSVCL and TH significantly elevated the net photosynthetic rate (Pn) by 86.55 % and 60.92 %, respectively, and increased total chlorophyll content (TChl) by 24.91 % and 11.76 %. Biostimulants facilitated nutrient uptake and boosted antioxidant enzyme activity in P. quinquefolius . Partial least squares path modeling (PLS-PM) further demonstrated that both NSVCL and TH promoted plant growth by enhancing soil enzyme activity in forest environments. These findings underscore NSVCL's efficacy in improving P. quinquefolius growth under forest conditions and provide a practical foundation for advancing organic forest cultivation and sustainable forest-medicine integration.

Dose and time-dependent response of single and combined artificial contamination of sulfamethazine and copper on soil enzymatic activities

Rui Yang,Xiaoming Xia,Jinhua Wang,Lusheng Zhu,Jun Wang,Zulfiqar Ahmad,Lili Yang,Shushuai Mao,Yangyang Chen

The widespread contamination of antibiotics and heavy metals results in imbalance in the ecosystem. However, the effect of the interaction between sulfamethazine (SM2) and copper (Cu) on soil enzymatic activities is unclear. Therefore, this study investigated the effect of single and combined artificial contamination of SM2 and Cu (0, 1.6 mmol kg −1 Cu and 0, 0.05, 0.2, 0.8 mmol kg −1 SM2) on soil enzymatic activities (urease, sucrose, phosphatase , and RubisCO). A single application of Cu at a concentration of 1.6 mmol kg −1 inhibited the urease, phosphatase and sucrase activities, while a stimulating effect on RubisCO activity was observed on day 7, 21, and 28 of incubation. The individual application of SM2 at higher concentration exhibited significant inhibition of sucrase, phosphatase, and urease activities while a stimulatory effect on RubisCO activity was observed on day 14 and 21 of incubation. The combined contamination of SM2 and Cu significantly inhibited the activities of urease, sucrase, and phosphatase. The effect of combined contamination of SM2 and Cu on the activity of RubisCO was different. The analysis results of interaction types show that there are synergistic or antagonistic effects between Cu and SM2, and these effects can amplify or reduce the effect of Cu or SM2 on soil enzyme activities . Integrated biological responses version 2 (IBRv2) analysis showed that the combined contamination of Cu and SM2 had a greater inhibitory or stimulatory effect on soil enzyme activities than the single contamination of Cu and SM2, depending upon dose and time.

Influence of root components of celery on pyrene bioaccessibility, soil enzymes and microbial communities in pyrene and pyrene-diesel spiked soils

Jing Wei,Xinying Zhang,Xiaoyan Liu,Xia Liang,Xueping Chen

Though phytoremediation is deemed as a promising approach to restore polycyclic aromatic hydrocarbon (PAHs) contaminated sites, studies about how the biodegradation of PAHs is enhanced still remains incomprehensive. Effects of root components on pyrene bioaccessibility , soil enzymes and microbial communities were explored in the paper, and their interactions in simulated pyrene and pyrene-diesel spiked microcosms were tried to give a reasonable explanation. Results indicated that root components enhanced the pyrene removal of bioaccessible and adsorbed fractions by 16.10 and 1.80   mg   kg −   1 , respectively, in pyrene-spiked soils at the end of the experiment. By contrast, root components increased the degradation of bioaccessible fraction by only 3.3   mg   kg −   1 in pyrene-diesel spiked soils. Although the bound fractions of pyrene increased over time in treatments without root components, they remained relatively stable, ranging from 0.02 to 0.03   mg   kg −   1 , in root components amended treatments. Activities of soil enzymes (polyphenol oxidase , catalase , invertase, urease and alkaline phosphatase) varied differently in response to pollutants and root components. Analysis of phospholipid fatty acids revealed that root components increased the biomass of soil microorganisms and altered the microbial structure. Pearson correlation analysis proved positive correlations between all the microbial subgroups and pyrene removal in pyrene-spiked soils, but the degradation of bioaccessible pyrene was only positively related with microorganisms confirmed by monounsaturated fatty acids in pyrene-diesel spiked soils.

Regulatory roles of indole-3-acetic acid in the physiological and biochemical responses of Cinnamomum camphora seedlings under cadmium stress

Jing Jiang,Chenyang Zhang,Meng Na,Shangqi Xu,Xiaoping Li,Shoubiao Zhou,Jihai Zhou

Soil cadmium (Cd) pollution has become increasingly severe, negatively affecting plant growth, soil safety, the ecological balance, and public health. Phytoremediation technology is an effective method for remediating soil Cd contamination because of its low cost, environmentally friendly nature, and minimal soil disturbance. However, phytoremediation efficiency is affected by factors such as the plant species, Cd concentration, and soil environment. In this study, a 180-day indoor potted experiment was conducted to explore the influences of various indole-3-acetic acid (IAA) levels on the growth, physiological and biochemical responses, Cd uptake, and soil enzyme activity of Cinnamomum (C.) camphora seedlings using 20 mg kg −1 Cd-contaminated soil with or without IAA treatments. The results showed that Cd stress significantly suppressed C. camphora seedling growth, whereas IAA application improved the basal diameter of seedlings by 22.22–133.33 %. This effect of IAA was observed primarily at low concentrations, while higher concentrations showed diminished effects. Compared to the Cd pollution treatment, adding 20 mg kg −1 IAA significantly enhanced the activities of antioxidant enzymes (SOD, POD, CAT), reduced the malondialdehyde (MDA) content, and decreased soil basal respiration (SBR). Moreover, IAA treatment promoted Cd uptake in the roots, stems, and leaves, and reduced the soil Cd content by 22.90–41.17 %. In conclusion, exogenous IAA addition effectively alleviates Cd stress by regulating the plant’s physiological and biochemical characteristics and soil enzyme activities. These findings suggest that the combined use of IAA and C. camphora represents a promising remediation method for soil heavy metal pollution.

Single and combined effect of polyethylene microplastics (virgin and naturally aged) and cadmium on pakchoi (Brassica rapa subsp. chinensis) under different growth stages

Cong Men,Zhenwen Xie,Kaihe Li,Xin Xing,Zifu Li,Jiane Zuo

To protect agro-systems and food security, study on the effect of microplastics and heavy metals on edible plants is of great significance. Existing studies mostly used virgin microplastics to evaluate their effects on plants, effects of naturally aged microplastics and their combined effects with heavy metals are rarely explored. In this study, single and combined effect of polyethylene microplastics (PE, both virgin and naturally aged) and cadmium (Cd) on pakchoi under seedling and mature stages were analyzed from perspectives of growth inhibition, oxidative damage, nutrition content and soil enzyme activities. Results showed that inhibiting effects of naturally aged PE (PEa) on the growth of pakchoi were stronger than virgin PE (PEv), whereas co-contamination of PEa and Cd was less toxic than that of PEv and Cd. The co-contamination of PE and Cd could inhibit pakchoi dry biomass by over 85 %. Both single and combined contamination of PE and Cd promoted soil fluorescein diacetate hydrolase (FDA) activities, which were 1.11 to 2.04 times of that in control group. Soluble sugar contents under co-contamination of PEa and Cd were 14 % to 22 % higher than those in control group. PEa and PEv showed different effects on oxidative damage of pakchoi. Compared with PEv, catalase (CAT) activities were more sensitive with PEa, whereas PEa had lower effect on superoxide dismutase (SOD) activities. The response of pakchoi to PE and Cd changed with growth stage. Chlorophyll contents in pakchoi under seedling stage were generally higher than those under mature stage. For Cd contaminated soils, PE benefited pakchoi growth under seedling stage, i.e. antagonistic effect between Cd and PE but hindered their growth under mature stage, i.e. synergistic effect. The results unraveled here emphasized PE, especially PEa, could trigger negative effects on agro-systems, whereas PE could be beneficial for heavy metal contaminated agro-systems under specific situations.

The dissipation of organophosphate esters mediated by ryegrass root exudate oxalic acid in soil: Analysis of enzymes activities, microorganism

Yaxi Deng,Hongxia Zhao,Xiaonuo Zhang,Xintong Li,Goujian Chi

Complex rhizoremediation is the main mechanism of phytoremediation in organic-contaminated soil. Low molecular weight organic acids (LMWOAs) in root exudates have been shown to increase the bioavailability of contaminants and are essential for promoting the dissipation of contaminants. The effects of root exudates on the dissipation of organophosphate esters (OPEs) in soil are unclear. Consequently, we studied the combined effects of root exudates, soil enzymes and microorganisms on OPEs (tri (1-chloro-2-propyl) phosphate (TCPP) and triphenyl phosphate (TPP)) dissipation through pot experiments. Oxalic acid (OA) was confirmed to be the main component of LMWOAs in root exudates of ryegrass. The existence of OA increased the dissipation rate of OPEs by 6.04%–25.50%. Catalase and dehydrogenase activities were firstly activated and then inhibited in soil. While, urease activity was activated and alkaline phosphatase activity was inhibited during the exposure period. More bacteria enrichment (e.g., Sphingomonas , Pseudomonas , Flavisolibacter , Pontibacter , Methylophilus and Massilia ) improved the biodegradation of OPEs. In addition, the transformation paths of OPEs hydrolysis and methylation under the action of root exudates were observed. This study provided theoretical insights into reducing the pollution risk of OPEs in the soil.
