Chalcone synthases (CHSs) of the type III polyketide synthases (PKSs), catalyze the formation of a tetraketide intermediate from a CoA-tethered starter and malonyl-CoA but use different cyclization mechanisms to produce distinct chemical scaffolds. Herein, we characterized CHS and CHS-like enzymes (designated MpCHS and MpCHSL1, 2 and 3) from Marchantia paleacea and determined the crystal structure of MpCHSL1. MpCHS catalyzed a Claisen condensation to form chalcone , while MpCHSLs catalyzed the formation of lactonized α-pyrones in vitro. Based on the structural, mutational and in vitro biochemical analyses, we established that MpCHSL1 is structurally and functionally closer to prototype CHS than stilbene synthase , and characterized the structural basis for the functional diversity of the type III PKSs. A chalcone-forming mutant of MpCHSL1 was build directed by the structural information. These findings pave the way for future studies to elucidate the functional diversity of type III PKSs in liverwort .
Marchantia polymorphaL. is a representative bryophyte used as a traditional Chinese medicinal herb for scald and pneumonia. The phytochemicals inM. polymorphaL. are terpenoids and flavonoids, among which especially the flavonoids show significant human health benefits. Many researches on the gametophyte ofM. polymorphaL.have been reported. However, as the reproductive organ ofM. polymorphaL., the bioactivity and flavonoids profile of the archegoniophore have not been reported, so in this work the flavonoid profiles, antioxidant and acetylcholinesterase inhibition activities of the extracts from the archegoniophore and gametophyte ofM. polymorphaL.were compared by radical scavenging assay methods (DPPH, ABTS, O2−), reducing power assay, acetylcholinesterase inhibition assay and LC-MS analysis. The results showed that the total flavonoids content in the archegoniophore was about 10-time higher than that of the gametophyte. Differences between the archegoniophore and gametophyte ofM. polymorphaL. were observed by LC-MS analysis. The archegoniophore extracts showed stronger bio-activities than those of the gametophyte. The archegoniophore extract showed a significant acetylcholinesterase inhibition, while the gametophyte extract hardly inhibited it.Keywords:Marchantia polymorphaL.;flavonoids;antioxidant potential;archegoniophore;gametophyte;acetylcholinesterase inhibition
Liverwort secondary metabolites play an important role in the peaceful relationship between liverwort endophytic fungi and the host. This study identified potential antifungal agents based on interactions between host plants and endophytic fungi. Two endophytic fungi strains and 25 metabolites, including nine new compounds, were isolated from the Chinese liverwort Herbertus herpocladioides. Endophytic fungi were identified using internal transcribed spacer and whole-genome sequencing, and the compound structures were determined using comprehensive spectroscopic analysis coupled with electronic circular dichroism calculations. Among these compounds, compounds 10–13 exhibited potent antifungal activities. Compound 10, the most potent antifungal agent, disrupted fungal mitochondrial respiration by inhibiting the activity of mitochondrial complexes I and IV and resulted in the intracellular ATP content of endophytic fungi being significantly reduced. The in vivo results show that compound 10 protected fruits and animals from infection by phytopathogen Alternaria citriarbusti and human pathogen Candida albicans, respectively.
