Nanozymes with multienzyme activity for reactive oxygen species (ROS) generation and intracellular redox imbalance are attractive strategy for cancer therapy. However, it is severely limited by low biocompatibility and catalytic efficiency, hypoxic and high levels of GSH in the tumor microenvironment. To address these issues, a copper doping carbon nanozyme (CC) with multienzyme activity was designed and integrated with photosensitizer Ce6 and gelatin to fabricate ROS amplifier (CCC). Gelatin endowed CCC with good biocompatibility, low hemolysis, and enzyme responsive degradation. CCC with high CAT-like, POD-like, OXD-like, and GSHox-like activities can induce the intracellular ROS storm formation to eliminate the cancer cells. The OXD-like activity and PDT performance mediated 1 O 2 generation was markedly potentiated by the CAT-like activity of CCC via catalyzing high expression of H 2 O 2 to generate O 2 . At the same time, a large amount of ·OH were produced through POD-like activity of CCC and GSH was depleted by the GSHox-like activities of CCC, resulting in a destructive ROS storm formation and cellular redox homeostasis disruption. Both in vivo and in vitro experiments showed that CCC displayed satisfactory anti-tumor activity and biocompatibility. Our work provides a novel strategy for the development of nanozyne enhanced photodynamic therapy of cancer.

Rheumatoid arthritis is a chronic autoimmune disorder characterized by the pathological accumulation of reactive oxygen species (ROS), which contributes to sustained inflammation and tissue damage. Current therapeutic modalities frequently exhibit low rates of clinical remission, and effectively mitigating the oxidative stress microenvironment associated with excessive ROS presents a significant challenge. Herein, we present a novel carbon dots (CDs) with multi-enzyme activity, featuring a porphyrin iron core and surface functional groups rich in phosphorus and oxygen, providing abundant catalytic sites for enhanced scavenging of hydrogen peroxide, hydroxyl radicals, and superoxide anions. Furthermore, the phosphate groups on the surface of CDs exhibited a specific affinity for hydroxyapatite, thereby facilitating targeted delivery to the bone. The localized accumulation of CDs with multi-enzyme activity at inflammatory lesions can effectively reduce oxidative stress levels, subsequently inhibiting bone destruction in collagen-induced arthritis rat models and significantly alleviating disease symptoms. This study integrates multiple catalytic sites and targeting moieties into a single nanostructure, representing a promising strategy for the precise treatment of oxidative stress-related bone pathologies.

Nanozymes with biological enzyme activity show great promise in biochemical analysis and medicine, yet single-activity nanozymes were limited by low catalytic efficiency and strict catalytic environment requirements. Consequently, the development of nanozymes with multiple enzyme activities presents a significant challenge. In the work, a vanadium-capped polyoxometalates (POMs)-encapsulating metal-organic framework (MOF), [Cu 12 (Trz) 8 Cl][PMo 12 O 40 (VO) 2 ], was synthesized via hydrothermal synthesis, which shows multiple enzyme activities concluding oxidase (OXD), peroxidase (POD) and catalase (CAT) activities. In the catalytic procedure, a fraction of H 2 O 2 generated by the OXD is further subjected to the POD catalytic reaction and the remaining portion is transformed into O 2 through the CAT activity, in turn, supplies the driving force for the OXD-like catalytic process. This cascade reaction, in conjunction with the transformation between Cu 2+ and Cu + within the material, engenders a structure analogous to an interlocking "gear", which augments the catalytic efficacy as well as the adaptability to intricate environmental conditions of the [Cu 12 (Trz) 8 Cl][PMo 12 O 40 (VO) 2 ] enzyme. By capitalizing on this high catalytic efficiency, the rapid quantitative detection of glutathione (GSH) was established with the calculated limit of detection (LOD) 0.062 μM in the range of 5–60 μM. Surprisingly, the multi-enzymatic [Cu 12 (Trz) 8 Cl][PMo 12 O 40 (VO) 2 ] can concurrently enhance the generation of reactive oxygen species (ROS) and the depletion of GSH in the tumor microenvironment (TME), thereby inducing tumor cell apoptosis. This research holds promising application prospects in the fields of biotechnology, clinical diagnosis, and tumor therapy.

The treatment of diabetic wounds requires inhibition of bleeding, reduction of inflammation, alleviation of hypoxia, and removal of pathogenic bacteria, but the therapeutic effect is greatly limited due to the diverse and synergistic nature of drug functions and the microenvironment of diabetic wounds themselves. Therefore, in this study, copper oxide nanoparticles (CuO NPs) with multi-species enzymatic activity were modified into chrysanthemum petal biochar (BC) and loaded with taxifolin (TAX) to form the TAX@CuO-BC (TCB) system, and then TCB was doped into polyvinyl alcohol (PVA)/graphene oxide (GO) /dextran (DT) hydrogels to obtain a new nano-enzymatic hydrogel system, TAX@CuO/BC/PVA/GO/DT (TCBP), in vitro and in vivo tests showed that this hydrogel could accelerate the healing of diabetic wounds by killing wound bacteria, rapidly stopping bleeding, reducing inflammation, alleviating hypoxia, and promoting angiogenesis. Therefore, this hydrogel has great potential for the treatment of diabetic wounds.

Infected skin wounds often suffer from excessive inflammation and delayed healing due to dysregulated redox balance caused by reactive oxygen species (ROS) accumulation and bacterial colonization, which are two interconnected biological barriers in wound repair. The natural antioxidant defense system inherently regulates ROS levels via coordinated enzymatic activities such as oxidases (OXD), peroxidases (POD), and catalases (CAT) to mitigate oxidative stress while combating bacteria, maintaining a favorable microenvironment for healing. Conventional dressings generally provide limited therapeutic efficacy due to poor moisture regulation, inadequate exudate absorption, and the risk of secondary injury, which exacerbate reactive oxygen species accumulation. Inspired by the natural antioxidant defense, we developed a multifunctional sodium alginate-graphene oxide (SA-GO) hydrogel by electrochemically engineering it that mimics OXD, POD, and CAT activities to regulate redox balance. This nanozyme hydrogel also enables photothermal therapy under near-infrared (NIR) illumination. In vitro , SA-GO hydrogel achieved an antibacterial efficiency approaching 100 % against Escherichia coli and Staphylococcus aureus via synergistic ROS generation and photothermal ablation. In a mouse-infected wound model, it significantly eradicated bacteria and accelerated healing. This work highlights the potential of tri-enzyme-mimetic hydrogels as effective platforms for ROS-regulation and enhanced treatment of infected wounds.

An enzyme-like nanoplatform obtained from sodium citrate-modified iridium nanoparticles (Cit-IrNPs) was presented, which exhibits excellent peroxidase, catalase and oxidase like activities thanks to the large accessible surface area and high-index facets of the mono-dispersed nanoparticles. The morphology and structures of Cit-IrNPs were comprehensively characterized by TEM, HRTEM, XRD, IR and XPS. The surface chemistry of Cit-IrNPs reveals that the oxidase-like and peroxidase-like activities can be ascribed to the formation of O 2 ¯ from the activation of dissolved oxygen on the high-index facets of IrNPs. The oxidase-like activity of Cit-IrNPs was further manifested by the oxidation of 33.3 mM aromatic alcohols in the presence of 3 mg/mL Cit-IrNPs in ambient conditions. Over 90 % conversion rates were readily obtainable in 10 h for all the tested alcohols with an initial reaction rate of ca. 20 μmol/h. More significantly, apart from the aldehydes no other byproducts were detected. The kinetic analysis of the enzyme mimic suggests the reaction follows a classical Michaelis-Menten model. The enzyme mimic also shows a good stability, as no obvious decrease in catalytic activity was observed after recycled use for 6 times. Furthermore, DFT calculation was employed to elucidate the reaction mechanism and it was found that the alcohol is initially bond to Ir(0) and subsequently forms Ir δ + -alkoxide species and the carbonyl product. Meanwhile, the Ir δ + -hydride species reductively eliminates O 2 ¯ and returns back to Ir(0). To the best of our knowledge, this is the first report of metal nanomaterials which can effectively transform aromatic alcohols to corresponding aldehydes at ambient conditions without need of external energy input.

The composite materials of Mn oxide nanowires and ZIF-8 (Mn x O y NWs@ZIF-8-NF and ZIF-8@Mn x O y NWs-TH) with controllable sizes and morphologies (nanoflower and Tanghulu-like) were successfully synthesized using different solvents (water and dimethylformamide (DMF) under mild reaction conditions. The systematic investigation was conducted to evaluate the effects of key synthesis parameters, including precursor concentration, metal-to-ligand ratio, temperature, and time, on the microstructure and crystalline morphology of the materials. Furthermore, the synthesis mechanisms of the materials in different solvents (water and DMF) were explored, revealing the influence of solvent molecules on their crystal growth processes. The prepared materials serve as “tandem enzymes” (a single nanozyme composite can perform multiple or sequential catalytic reactions), possessing multiple enzyme-like activities (hydrolase- and oxidase (OXD)-like activities). Kinetic studies indicate that the Mn x O y NWs@ZIF-8-NF composite exhibits excellent hydrolase-like catalytic activity, characterized by the high substrate affinity and maximum reaction rate ( K m : 0.01493 mM, V max : 161.42 × 10 –8 M/s). Moreover, Mn x O y NWs@ZIF-8-NF exhibits high specificity and interference-resistance in fluorescent detection of glucose oxidase (GOX) based on its hydrolase and OXD-like activities. In short, this work investigates the role of solvents as one aspect of tailoring material properties, contributing to the development of advanced functional materials and offering a methodological reference for GOX detection. Atomic precision synthesis and modulation of nanozymes are critical for evaluating their performance and developing functional applications. Herein, we developed an in situ strategy for the controlled synthesis of gold clusterzymes (AuCEs) within the confined cavities of the protein scaffolds. Using a stable dodecameric protein (SP1) as a ligand, we fused a short peptide containing the CCY sequence (SP1-CCY) to the N-terminus of SP1, which is located within its cyclic structure. In this design, cysteine and tyrosine synergistically chelate and reduce Au ions, while the confined protein structure restricts cluster growth, leading to the formation of a 25-gold atom (Au25) nanocluster with strong fluorescence and peroxidase-like activity. Unlike chemical ligands, protein scaffolds offer atomic-level precision and allow for site-specific modulation of the local environment through rational mutations, such as L9D, L9I, and E73I/L9I. This allows precise control of both fluorescence and enzymatic activity via protein engineering. Additionally, heteroatom doping of the Au core with copper endows the AuCEs with significant superoxide dismutase-like activity. Antioxidant assays demonstrate that the optimized AuCEs exhibit potent reactive oxygen species scavenging activity, providing significant protection to Caenorhabditis elegans from oxidative stress and highlighting their potential as therapeutic candidates for aging-related diseases.

Nanozymes are nanoparticles with enzymatic activity, which are widely used in environmental and antibacterial research. Herein, we designed and synthesized novel amorphous nanozyme Cu-Im NPs with multiple enzyme-mimicking activities. Cu-Im NPs have the same active sites as natural laccase. In addition, the active center is similar to that of carbonic hydrolase, replacing the zinc ions with copper ions. Meanwhile, Cu-Im NPs also possess peroxidase-like activity. We reveal the multi-enzyme catalytic mechanisms of Cu-Im NPs. Notably, phosphate inhibits the laccase-like and peroxidase-like activities of Cu-Im NPs while activating their hydrolase activity. Based on these findings, we have developed a sensitive and selective method for detecting phosphate anions.

This work aim to study the tolerance and aroma-related enzymes activities of the non- Saccharomyces yeasts in Vidal blanc icewine from the Huanren region of China. The strains were identified by sequencing internal transcribed spacer (ITS) region and the 26S rDNA D1/D2 domain genes and all representative non- Saccharomyces yeasts were belonged to genera Metschnikowia , Hanseniaspora , Torulaspora, Candida , and Debaryomyces . A total of 28 strains were carried out for tolerance experiments and results suggested that most of them could tolerate 500 g/L glucose, 4% ethanol, 20 g/L tartaric acid, and 350 mg/L SO 2 . Finally, a total of 17 strains with better tolerance were carried out for the β-glucosidase, β-xylosidase, and pectinase activities experiments. The results showed that Candida railenensis HC08 and the strains of Hanseniaspora genus have satisfactory multi-enzyme activities, which can be used to design mixed fermentation to produce characteristic icewine. Practical applications Non- Saccharomyces yeasts produces a series of hydrolytic enzymes that are thought to have a significant contribution to the aroma complexity of wines, however, are poorly explored in icewine. In this work, most of the non- Saccharomyces yeasts screened from Chinese icewine can adapt well to the high-sugar and high-acid environment of icewine, and can secrete hydrolase. The application of these strains in mixed fermentation could provide a prospect for the production of characteristic icewine.

Nanozymes capable of inducing metabolic reprogramming and activating the immune response without external stimuli in vivo are highly pursued for malignant tumor therapy. In this paper, a PtIrFeMoZn high-entropy alloy (HEA) nanozyme is designed and synthesized via a simple one-step hydrothermal method. The HEA nanozymes not only trigger apoptosis and ferroptosis via cascade biocatalysis, thereby enhancing immunogenicity, but also enhance the immune effect by targeting the glycolytic pathway. It is worth mentioning that a simple pre-treatment of nanozymes by alternating current (AC) yielded much better therapeutic and immuno-effect. The ‘AC-treated’ nanozymes exhibit an excellent synergy of peroxidase-like (POD-like), myeloperoxidase-like (MPO-like), and glutathione peroxidase-like (GPx-like) activities, generating a sufficient reactive oxygen species (ROS) storm. Additionally, two immune pathways (ICD and the cGAS-STING) are activated simultaneously. Furthermore, the production of HClO and the depletion of NADH can regulate metabolism, further disrupting the equilibrium of the glycolysis process. This not only increases the cell death but also enhances the immune response in female tumor-bearing mice. This study proposes a multi-pronged therapeutic strategy that can significantly activate anti-tumor immunotherapeutic effects through ROS storm, GSH/NADH oxidation, and lactate/ATP depletion, triggering apoptosis/ferroptosis/immunotherapy. These findings hold significant promise for inspiring the development of HEA nanozymes for tumor immunotherapy. Nanozymes that can activate immune response without external stimuli could benefit malignant tumor therapy. Here, the authors report high-entropy alloy nanozymes that allow a multi-pronged strategy for activating anti-tumor immunotherapeutic effects, and reveal that using alternating current to pre-treat these nanozymes before in vivo administration can enhance the therapeutic and immuno-effect.

Primary liver cancer remains a significant global health challenge, with the majority of patients diagnosed at an unresectable stage, further complicated by liver dysfunction due to cirrhosis. To address these clinical hurdles, nanozyme-based catalytic therapy has emerged as a promising strategy. However, its therapeutic efficacy is often limited by self-protective mechanisms associated with the tumor microenvironment (TME), particularly the overexpression of glutathione (GSH) and hypoxia-induced metabolic adaptation. In this study, a quaternary medium-entropy layered double hydroxide (LDH) nanozyme, denoted as NiFeMnCu-LDH is developed. This nanozyme exhibits peroxidase-like, catalase (CAT)-like, and oxidase-like activities, facilitating the cascade generation of reactive oxygen species (ROS). Simultaneously, the oxygen produced by CAT mitigates hypoxia, while the glutathione peroxidase-like activity drives the depletion of tumor antioxidants, effectively reprogramming the TME. Further in vitro experiments demonstrated that efficient production of ROS and scavenging of GSH induced substantial apoptosis in liver cancer cells. Additionally, mouse liver tumor models showed robust inhibition of tumor growth without systemic toxicity. This findings provide a framework for the precise engineering of nanozymes in advanced liver cancer treatment, addressing critical barriers within the TME and offering valuable insights into a mimetic multi-enzyme synergy for translational applications.
