This study aimed to characterize the structure of polysaccharide ZOPA extracted from Zingiber officinale and its purified form (ZOPA-1), and to investigate their anti-fatigue mechanisms based on the gut-muscle axis. The study found that the backbone of ZOPA-1 is primarily composed of →3,4)-α-Glcp-1→ and →4,6)-α-Glcp-(1→ linkages, with →4)-α-Glcp(1→ serving as its side chain. In exhaustive swimming experiments with mice, both crude ZOPA and purified ZOPA-1 demonstrated significant anti-fatigue effects, including enhanced glycogen storage, improved antioxidant capacity, reduced accumulation of metabolic waste products, and regulated energy metabolism in the gastrocnemius muscles. These effects may be mediated through the activation of the Keap1-Nrf2/ARE and AMPK/PGC-1α signaling pathways. Furthermore, ZOPA and ZOPA-1 modulated the intestinal flora of mice, increasing diversity, altering abundance, and regulating short-chain fatty acid concentrations, suggesting a potential role of the gut-muscle axis in mediating the anti-fatigue effects. This study provides valuable insights into the complex interplay between polysaccharides, the gut-muscle axis, and exercise-induced fatigue.

A polysaccharide was extracted from the fresh ginger by water extraction and alcohol precipitation. A homogeneous polysaccharide was obtained by Sevage deproteinization, dialysis, DEAE-52 ion exchange chromatography and Sephadex G-100 gel column chromatography. The molecular weight of the ginger polysaccharide was determined by high performance gel permeation chromatography. The monosaccharide composition, scanning electron microscope and other physicochemical parameters of the ginger polysaccharide were further determined. The spectral properties of the ginger polysaccharide were investigated by ultraviolet and infrared spectroscopy. On this basis, methylation analysis, 1D and 2D NMR analysis were used to investigate its possible chemical structure. The results showed that the ginger polysaccharide group was divided into neutral polysaccharide components with high homogeneity, M w = 35.52 kDa, M n = 216.56 kDa and the sugar content was 93.5%. The ginger polysaccharide is mainly composed of Fuc, Ara, Rha, Gal, Glc, Xyl, Man, Gal-UA and Glc-UA, the amount of substance ratio was 1.01:1.96:9.54:14.82:64.20:3.84:2.07:1.41:1.16. To study the structure–activity relationship of ginger polysaccharide, acetylated ginger polysaccharide and phosphate ginger polysaccharide were prepared by chemical methods in this study. The results of antioxidant activity in vitro showed that the ginger polysaccharide and its derivatives had good biological activity in freeing ABTS + radical scavenging, freeing hydroxyl radical scavenging, freeing DPPH radical scavenging and reducing ability. Interestingly, the results showed that the ginger polysaccharide had a certain inhibitory effect on the growth of E. coli in vitro. In general, this study provides a certain experimental basis and reference for the further research on the activity of ginger polysaccharide and its derivatives. Graphical Abstract

Background: Ulcerative colitis (UC) is on the rise all over the world.Zingiber officinalepolysaccharide (ZOP-1) has good anti-inflammatory and antioxidant effects, but the therapeutic effect and mechanism of ZOP-1 on UC are still unclear. Methods: ZOP-1 obtained by water extraction and alcohol precipitation was analyzed by methylation and NMR. At the same time, the mechanism of ZOP-1 in the treatment of UC was clarified by hematoxylin-eosin (HE) staining, metagenomics, immunohistochemistry, and protein blot (Wb). Results: ZOP-1 was the structure of the by →4,6)-β-Glcp-1→ and →3,6)-α-Galp-(1→ constitute the main chain, there were two branched chain by →4)-β-Glcp(1→, and α-Araf(1→ as the end group. ZOP-1 significantly improved the shortening and thickening of the colon, changed the index of immune organs, inhibited the production of inflammatory factors in mice with ulcerative colitis, changed the intestinal flora of mice, increased the content of short-chain fatty acids (SCFAs) in the intestine, and controlled the TLR4/NF-κB/MAPK signaling pathway, thus preventing and treating DSS-induced ulcerative colitis in mice. Conclusions: ZOP-1 alleviated UC by controlling the expression of cytokines, thereby reducing intestinal inflammation and oxidative stress, enhancing intestinal integrity, modulating intestinal flora, and regulating the levels of SCFAs.

Green-synthesized metal nanoparticles show promise in nanomedicine and material engineering. In this study, the polysaccharide of Zingiber officinale (ZOP) was used as a raw material. Through single-factor experiments and a response surface methodology, the optimum synthesis protocol of Zingiber officinale polysaccharide silver nanoparticles (ZOP-NPs-AgNPs) was determined as follows: V(AgNO 3 ):V(ZOP) = 2.98:1, 59.79 °C, 3 h, pH 9, and 20 mL NaCl, achieving a 92.51% silver chelation rate. Structure analysis revealed that ZOP-NPs-AgNPs were spherical or quasi-spherical, with a particle size < 20 nm and a face-centered cubic crystal structure, which has good thermal stability. Subsequent studies explored the antibacterial and immunomodulatory effects of ZOP-NPs-AgNPs. The minimum inhibitory concentration (MIC) of ZOP-NPs-AgNPs against Escherichia coli and Staphylococcus aureus was determined to be 0.5000 mg/mL and 0.0310 mg/mL, respectively, while the minimum bactericidal concentration (MBC) was 0.5000 mg/mL and 0.0310 mg/mL, respectively. Additionally, ZOP-NPs-AgNPs significantly enhance RAW264.7 cell proliferation and phagocytosis and boost IL−1β, IL−6, NO, and TNF-α production. This confirms that ZOP can act as a green reductant and stabilizer, offering a new method for green nano-silver synthesis. This provides a sustainable way to produce antibacterial products and functional foods, and offers useful references for eco-friendly nano-silver applications.
