Using plant essential oil instead of chemical pesticides to control melon fly ( Bactrocera cucurbitae (Coquillett)) is still a great challenge in agricultural practices, which must effectively slow down the volatility rate of plant essential oil. In this study, citral as a natural plant essential oil was blended with environmentally friendly castor oil based waterborne polyurethane (PU). The interaction, chemical resistance, adhesion property and sustained release behavior of the composites with different citral content were clarified, and the efficacies against the melon fly under field conditions were explored. The results indicated that the composites prepared exhibited excellent adhesion behavior on bitter gourds ( Momordica charantia L.) even under rainfall washing, meanwhile the chemical resistance of composites could be tailored via controlling citral content. Low citral content composite films could be washed off by alcohol without worrying about the safety of that on crops. The composite prepared exhibited long-lasting repellent behavior against melon fly. The composite can be prepared via a simple technology and low cost, which has great value for plant essential oil applied in agriculture and can provide a new approach for melon fly control.

Odorant-binding proteins (OBPs) in insects are essential for mating and oviposition host selection. How these OBPs respond to different hosts at the mRNA level and their effects on behavior remain poorly characterized. The oriental fruit fly Bactrocera dorsalis is a highly invasive agricultural pest with an extremely broad host range and high fecundity. Based on our previously constructed B. dorsalis transcriptome, six OBP s that were differentially expressed during three different physiological adult stages were identified. A phylogenetic tree was constructed to illustrate the relationships of these six OBP s with OBP sequences from three other dipteran species ( Drosophila melanogaster , Anopheles gambiae , and Ceratitis capitata ). The spatiotemporal expression profiles of the six OBP s were analyzed using quantitative real-time PCR. Our results revealed that OBP19c, OBP44a, OBP99a , and OBP99d were abundantly expressed from the prepupa stage to the adult stage, and most of the OBP s were mainly expressed in the head, wings, and antennae. The expression levels of these OBP s were upregulated when female flies were exposed to their preferred hosts. Silencing OBP99a resulted fewer eggs being laid compared with the control group when the females were exposed to their preferred host, that is, banana, whereas more eggs were laid when a non-preferred host, that is, tomato, was used. Furthermore, silencing OBP99a led to sexually dimorphic mating behavior. dsOBP99a -injected males dramatically reduced courtship, whereas enhanced courtship was observed in the treated females. These data indicate that OBP s may participate in different biological processes of B. dorsalis . Our study will provide insight into the molecular mechanism of chemoreception and help develop ecologically friendly pest-control strategies.

The univoltine Bactrocera minax (Enderlein) is a devastating pest of citrus in Nepal, India, Bhutan, and China. To better understand the overwintering pupal diapause, we investigated the juvenile hormone III (JH III) and ecdysteroid 20-hydroxyecdysone (20E) titers across the developmental stages, and the effects of juvenile hormone analog (JHA) and 20E treatments on adult emergence patterns. The results showed that both JH III and 20E levels fluctuated from the late larval stage to the late pupal stage. JHA and 20E treatments at the late larval stage had marginal effect on the adult emergence patterns, with slightly faster adult eclosion and higher eclosion rate. Similarly, JHA treatment at the early pupal stage did not affect the adult emergence. However, 20E treatment at the early pupal stage remarkably shortened the duration of diapause, but lowered the eclosion rate, especially with relatively high dose. These findings demonstrated that 20E treatment at the early pupal stage is a suitable method to hasten the diapause termination of B. minax, which will contribute to the rearing of B. minax in the laboratory as needed for routine experiments.

Citrus fruit fly Bactrocera minax (Enderlein) (Diptera:Tephritidae) is a devastating pest of citrus plants in China. In order to characterize the behavioral mechanisms and possible chemical cues involved in host selection by female citrus fruit fly, the host plant volatiles was analyzed and Y-tube olfactometer assays were used. Host odor from citrus fruits peels elicited upwind orientation flights followed by landing and egg-laying, demonstrating the essential role of host plant volatiles in B. minax host-finding and oviposition behavior. GC–MS analysis of volatiles from the peels of three varieties citrus fruits, that were Bingtang sweet oranges ( Citrus sinensis cv. Bingtang), Satsumas mandarins ( Citrus reticulata cv. Satsuma) and Sour oranges ( Citrus aurantium ), which adult and larval of B. minax preferred to, showed that the major compounds extracted from the basal region were different from those of the distal region to some extent. Furthermore, in Y-tube olfactometer attraction assays, female flies were significantly attracted ( P < 0.05) to four out of the thirteen tested compounds from the citrus volatiles, including nonanal , citral , limonene and linalool . Conversely, furfuryl alcohol and guaiastil significantly repelled females ( P < 0.05). The percentage composition of nonanal, citral, limonene and linalool from different regions of the fruits displayed a positive correlation with the numbers of egg marks made by females, whereas furfuryl alcohol and guaiastil displayed a correspondingly negative correlation. These results suggest that the oviposition preference of B. minax females is highly related to the chemical compounds emitted from host fruit peels.

Methyl eugenol (ME) is a highly potent sexual attractant for Bactrocera dorsalis , which plays a crucial role in agricultural pest management. In this study, a composite fiber membrane was prepared by polystyrene (PS), polyethylene glycol diacrylate (PEGDA), and ME through electrostatic spinning respectively. The effective release period of ME was prolonged by triggering PEGDA with UV light. The mass ratio of PS to PEGDA affects the morphology of the fibers as well as the loading capacity (LC) and encapsulation efficiency (EE) for ME. Under optimal test conditions, the diameter of the fibers was 1.70 μm, and the ME loading capacity and encapsulation efficiency were (7.80 ± 0.07%) and (85.84 ± 7.94%). The combination of PS and PEGDA was confirmed by FTIR analysis, while its excellent thermal stability was verified through TGA testing. In the PS/PEGDA-ME-UV membrane, the cumulative release rate of ME reached 81.97% by week 8, which conformed to a first-order model. In the trapping experiments, a 53% rate of Bactrocera dorsalis capture was maintained even by week 8, demonstrating its potential for application in agricultural biocontrol.

BACKGROUND The oriental fruit fly, Bactrocera dorsalis (Hendel) is a worldwide pest damaging a wide range of hosts. Due to the long-term indiscriminate use of insecticides, B. dorsalis has developed serious resistance to several insecticides. UDP-glycosyltransferases (UGTs) are secondary metabolic enzymes involved in biotransformation and play an important role in the metabolism of plant secondary metabolites and synthetic insecticides in insects. Thus, we suspect that UGTs in B. dorsalis play an important role in insecticide tolerance. RESULTS In this study, 31 UGT genes were identified in the genome of B. dorsalis , belonging to 13 subfamilies. Real-time quantitative polymerase chain reaction (RT-qPCR) results revealed that 12 UGT genes were highly expressed in the antennae, midgut, Malpighian tubule and fat body. The mRNA expressions of 17 UGT genes were up-regulated upon exposure to λ-cyhalothrin, imidacloprid, abamectin and chlorpyrifos. Knockdown of the selected five UGT genes ( BdUGT301D2 , BdUGT35F2 , BdUGT36K2 , BdUGT49D2 , BdUGT50B5 ) by RNA interference increased the mortality of B. dorsalis from 9.29% to 27.22% upon exposure to four insecticides. CONCLUSION The abundance of UGTs in B. dorsalis is similar to other insect species, and 12 out of 31 UGTs were specifically expressed in metabolic tissues, suggesting a key role in detoxification. Down-regulation of five selected UGT genes increased the susceptibility of B. dorsalis to various insecticides, indicating that UGTs may play an important role in tolerance of B. dorsalis to multiple insecticides. © 2022 Society of Chemical Industry.

Azadirachtin exhibits excellent bioactivities against several hundred arthropods. However, current knowlege of its biochemical effect on B . dorsalis larvae is not deep enough. In this study, integrated LC-MS and GC-MS-based untargeted metabolomics were used to analyze the changes of endogenous metabolites and the biochemical effects of azadirachtin on B . dorsalis larvae. Azadirachtin has excellent bioactivities against B . dorsalis larvae in this study, leading to a longer developmental duration, lower survival rate, and low pupa weight. The effect of azadirachtin was investigated on a total of 22 and 13 differentially abundant metabolites in the LC–MS and GC–MS-based metabolomics results, are selected respectively. Pathway analysis indicated that 14 differentially enriched metabolic pathways, including seven influential pathways, are worthy of attention. Further integrated key metabolic pathway analysis showed that histidine metabolism, d -glutamine and d -glutamate metabolism, biotin metabolism, ascorbate and aldarate metabolism, pentose and glucuronate interconversions, and alanine, aspartate and glutamate metabolism in B . dorsalis larvae are significantly relevant pathways affected by azadirachtin. Although extrapolating the bioactivity results in this study to the practical project of B . dorsalis pest management in the field has limitations, it was found that azadirachtin has a significant effect on the primary metabolism of B . dorsalis larvae.

Simple SummaryThe melon fly (Zeugodacus cucurbitae) is a pest that prefers to bore into and feed on cucurbitaceous crops and that pupates in the soil. The parasitoidSpalangia endiuscan burrow into the soil to attack melon fly pupae, but it only kills about 33% of them. Since insecticides are essential for controlling melon fly populations, we assessed how five commonly used insecticides affect the toxicity, survival, reproduction, choice-making behaviour, and parasitism ability ofS. endius. Our goal was to determine how to combine insecticides withS. endiusto more effectively control melon fly pupae. Through a series of laboratory and semi-field trials, we confirmed that abamectin is suitable for combined use withS. endius. However, we must be careful about the concentration and timing of abamectin application. The best approach is to spray abamectin on the soil surface before the melon flies pupate and then release the parasitoids.Spalangia endiusWalker, a pupal parasitoid of the alien invasive pestZeugodacus cucurbitae(Coquillett), causes 33% host mortality. This study assessed whether combiningS. endiuswith insecticides (abamectin, thiamethoxam, nitenpyram, emamectin benzoate, or beta-cypermethrin)—all effective againstZ. cucurbitae—could enhance control efficacy. Among these, abamectin was the least toxic to adultS. endius. Surface contact treatments with 12 and 15 mg a.i./kg of abamectin did not significantly increaseS. endiusmortality. However, mixing 12 mg a.i./kg of abamectin into a honey solution to encourage ingestion decreased the survival, parasitism, and fecundity ofS. endius. In olfactometer assays,S. endiusadults avoided abamectin-treated host pupae, though prior exposure to abamectin mitigated this avoidance. The timing of abamectin soil application relative to host pupation andS. endiusrelease affects host mortality. The most effective timing is spraying abamectin before host pupation (to exposeZ. cucurbitaelarvae) and then releasingS. endius. Field trials confirmed that combining abamectin (12 mg a.i./kg) withS. endiusincreased host mortality more than either treatment alone. In conclusion, abamectin (12 mg a.i./kg) is a suitable insecticide for combination withS. endiusto controlZ. cucurbitae. The application sequence should be spraying abamectin before hosts pupate and, only after that, releasing the parasitoid.

Odorant-binding proteins (OBPs) are believed to play critical roles in host-seeking behavior. However, little attention was paid to its different functions in male and female. The antenna-specific OBP gene from Bactrocera dorsalis , BdorOBP13, was cloned and its expression profile was examined. The results showed that BdorOBP13 was exclusively expressed in male and female adults, which exhibited a high transcript level in antennae. After injection of BdorOBP13 dsRNA, its transcript level in males and females decreased significantly. Electrophysiological responses of RNAi-injected flies to, methyl eugenol (male attractant) and γ-octalactone (female attractant) decreased significantly. However, no significant changes in the electrophysiological response were observed in RNAi-injected flies to benzothiazole, (+),dipentene, and ethyl tiglate. The behavioral bioassay showed that males treated with RNAi significantly reduced their preference to methyl eugenol, while RNAi-injected females showed a significantly lower preference to γ-octalactone, suggesting that BdorOBP13 may have different functions between males and females: it may be involved in the detection of methyl eugenol in males but is involved in the detection of γ-octalactone in females. These findings improve our understanding of insect OBPs and their roles in insect chemosensation, which may provide us with new molecular targets in the management of B. dorsalis .

D- limonene is the main volatile gas in citrus, and its content changes significantly after the citrus becomes infected, thus making it possible to use gas-derived information to identify pest-infested citrus. In this study, a quartz crystal microbalance (QCM) sensor based on a molecularly imprinted polymers (MIPs) coating was used to identify the citrus infested with Bactrocera dorsalis . The experimental results show that within the concentration range of 300–2100 mg m −3 , the sensor’s response to the concentration follows a linear trend, and its sensitivity was 0.078 Hz (mg m −3 ) −1 ; the detection limit was 41.40 mg m −3 . The sensor’s reproducibility and repeatability were both higher than 95%, and the one-month frequency drop deviation was less than 5%. The results confirmed that the MIPs-QCM sensor can rapid detection and nondestructive of infested citrus.

BACKGROUND The melon fly, Zeugodacus cucurbitae (Coquillett), is an invasive Tephritidae pest with robust fertility. The male accessory glands (MAGs) form a vital organ that ensures insect reproductive efficiency. Most of the secreted proteins by MAGs exhibit a male bias expression. Takeout, one of these proteins, is abundantly present in the MAGs of many insects. RESULTS In this study, we identified 32 takeout genes in Z. cucurbitae . The phylogenetic analysis and multiple sequence alignment results showed that Zctakeout1 is the most related homolog to the MAGs-specific takeout in Tephritidae. The real-time quantitative PCR results showed that Zctakeout1 was exclusively expressed in the male adult stage, and its expression level gradually increased with the increase in age and then remained stable at the sexually matured stage. The distribution among tissues demonstrated the specific expression of Zctakeout1 in the MAGs, and fluorescence immunohistochemical results confirmed the presence of Zctakeout1 in close proximity to binuclear cells of the mesoderm epidermal MAGs. In continuation, CRISPR/Cas9-mediated genome editing was employed, resulting in successfully generating a homozygous strain with an +8 bp insertion. The mating experiments with the Zctakeout1 −/− males resulted in significant reductions in both the mating rate and egg production of females. CONCLUSION These findings prove that the MAGs-specific Zctakeout1 is essential in regulating fecundity in female Z. cucurbitae fruit flies. Our data suggests its utilization in future essential insect-specific gene-directed sterility insect technique (SIT) by the genetic manipulation to keep these important Tephritidae populations under control. © 2024 Society of Chemical Industry.

Azadirachtin , as the most promising and effective botanical insecticide, exhibits significant growth inhibition activity against agricultural and forestry pests. However, its biochemical effects at the metabolic level compared with those of other insect growth regulators have not been studied. Therefore, in this study, a GC–MS based untargeted metabolomics approach was applied to compare azadirachtin with pyriproxyfen (a juvenile hormone analog) and tebufenozide (a molting hormone analog) in terms of their metabolic effects on Bactrocera dorsalis larvae. The bioactivity of azadirachtin against B. dorsalis larvae was significantly different than those of pyriproxyfen and tebufenozide. A total of 693 mass features were recognized, and 112 metabolites were identified in this study. The results showed that a total of 16, 13 and 10 differentially regulated metabolites corresponding to 12, 5 and 8 pathways occur in Aza versus CK, Pyr versus CK and Teb versus CK group, respectively. Further analysis showed that 6 differentially regulated metabolites corresponding to 5 key pathways could be the primary differential metabolic response of B. dorsalis larvae to the three insect growth regulators. The pathways were myo-inositol corresponding to ascorbate and aldarate metabolism as the specific response of B. dorsalis larvae to azadirachtin; xylitol, xylulose and 3-aminopropionitrile corresponding to pentose and glucuronate interconversions, and cyanoamino acid metabolism as the common responses to azadirachtin and pyriproxyfen; and 3-hydroxypropionic acid and beta-alanine corresponding to propanoate metabolism and beta-alanine metabolism as the specific responses to tebufenozide. The results showed that the metabolic response of B. dorsalis larvae to azadirachitin is closer to that of pyriproxyfen than tebufenozide. The differentially regulated metabolites and pathways responsible for this difference are discussed.
