Kaempferol Extends Male Lifespan Under Blue Light Irradiation in Drosophila

Qimeng He,Hui Jin,Jie Shen

Short-wavelength blue light is ubiquitous in daily life and has a lasting destructive influence. Its potential harm to biological health is significant. This study used Drosophila as a model organism to investigate the protective effects of kaempferol, a flavonoid, against the toxicity of blue light. It also examined its physiological effects on Drosophila under blue light irradiation. In this experiment, fruit flies were fed with three different concentrations of kaempferol solutions (0.1, 0.01, and 0.001 mol/L) dissolved in food. The survival rate and physiological indexes of Drosophila were investigated under blue light irradiation of 2500 lux. The results showed that 0.1 mol/L kaempferol increased the activity of male flies during the day and significantly extended the male survival time under blue light irradiation. However, the study found that kaempferol did not significantly prolong the survival time of Drosophila in the oxidative stress experiment, and no significant difference was observed in the feeding experiment. In summary, our research found that kaempferol, at the concentration of 0.1 mol/L, has a protective effect on Drosophila under blue light irradiation, potentially achieved through alterations in circadian rhythm. Summary Kaempferol protects Drosophila from blue light damage. The effect of kaempferol on Drosophila exhibits sex-specific responses. Kaempferol enhances daytime activity and extends the lifespan of male Drosophila .

An ultra-sensitive kaempferol electrochemical sensor based on flower-like ZIF-8 pyrolysis-derived ZnWO4/porous nanocarbon composites

Yilin Wang,Chenxi Wang,Li Zhang,Jiaodi Li,Rongjie Li,Qian Fu,Chunyan Li,Pengcheng Zhao,Yixi Xie,Junjie Fei

Kaempferol (KA), an important polyphenolic compound, is able to combat a variety of inflammatory diseases. Here, a ZnWO 4 / porous nanocarbon based electrochemical sensor has been constructed to detect KA for the first time. Innovatively, ZnWO 4 has been synthesized by pyrolysis using ZIF-8 as a zinc source, while porous nanocarbon was introduced. The nanocomposites have been characterized by X-ray diffraction, X-ray photoelectron spectroscopy and scanning electron microscopy and demonstrated that Acidified ZnWO 4 /porous nanocarbon has been successfully prepared. Notably, the excellent catalytic properties of ZnWO 4 and the great enrichment ability and conductivity of porous nanocarbon greatly enhanced the detection performance of the sensor for KA with a linear range of 0.009–0.7 μM and a LOD of 2.03 nM. In addition, the sensor has obtained satisfactory results for the detection of KA in actual natural samples (broccoli and kaempferia powder). The reaction sites and redox mechanism of KA were shown by density functional theory. Finally, the sensor is highly sensitive and stable, making it a novel detection method with extremely high potential applications.

Transcriptomics-based Exploration of the Mechanism of Kaempferol in the Treatment of Osteoporosis in Ovariectomized Rats

Zhang-lin Pu,Chi Zhang,Qian Yan,Zhou Liang,Zhi-Wei Xu,Ji-hu Wei,Hua Liu,Feng Chen

Objective: We explore the pharmacodynamic targets of kaempferol in treating osteoporosis using transcriptomics and animal experiment. Methods: Firstly, we constructed an ovariectomized rat model (OVX). The anti-osteoporosis effect of kaempferol was evaluated by bone mineral density (BMD) and hematoxylin-eosin staining comprehensively. Moreover, differential genes between groups were screened by RNA sequencing technology (RNA-seq) for transcriptomics and Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathways enrichment analysis were performed. Finally, partial results of transcriptomics were verified to detect the expression of the relevant gene expression by immunohistochemistry. Results: The pharmacodynamic findings indicated that the administration of kaempferol resulted in an elevation in BMD (P < .01) and a notable enhancement in tibia microstructural indices in the experimental rats (P < .01). The transcriptomic analysis revealed that NTN1, LTBP4, GSN, and EBF1 were identified as the principal targets for therapeutic intervention in osteoporosis. The results of animal experiments showed that kaempferol promoted osteogenesis and inhibited bone resorption by downregulating the protein expression of NTN1, LTBP4, GSN, and EBF1 (P < .01). Conclusion: Kaempferol enhances BMD levels, retards tibial bone loss with structural deterioration in OVX model rats, and promotes bone formation while curbing bone resorption through NTN1, LTBP4, GSN, and EBF1 protein down-regulation.

Voltammetric studies of kaempferol on polyvinyl pyrrolidone cladding quantum dots CdS doped carbon paste electrode and analytical application

Kai Zhang,Ge Song,Yinfeng Li,Xianli Wu,Kunjing Li,Baoxian Ye

A facile and highly sensitive electrochemical sensor for kaempferol was fabricated by doping polyvinyl pyrrolidone cladding quantum dots to the carbon paste electrode. The quantum dots were characterized with transmission electron microscopy , ultraviolet spectrum and infrared spectrum. The electrochemical behaviors and kinetic parameters of kaempferol were investigated on the sensor by cyclic voltammetry and chronocoulometry in NaAc–HAc (pH 4.4). The proposed sensor accelerated the redox processes of kaempferol obviously, and the oxidation currents were linearly relationship with kaempferol in two concentration intervals: one was 6.0   ×   10 −8 –2   ×   10 −6   mol/L and the other was 5.0   ×   10 −6 –2.5   ×   10 −5   mol/L. The quantification limit obtained by square wave voltammetry was 6.0   ×   10 −8   mol/L. Merits such as extreme simplicity, low cost, good sensitivity, high sensitivity, good stability and reproducibility made it an excellent electrochemical sensor for kaempferol. A new electroanalytical method for kaempferol was proposed and used for determination of kaempferol in tablets with satisfactory results.

Simple quantitative analytical methods for the determination of alkaloids from medicinal and edible plant foods using a homemade chromatographic monolithic column

Mingxue Li,Ligai Bai,Shan Peng,Fanrong Sun,Laisen Wang,Haiyan Liu,Hongyuan Yan

A polymer-based chromatographic monolithic column was prepared via in-situ radical polymerization using tetrahydrofurfuryl methacrylate as the monomer . The homemade column was used for the separation and quantitative analysis of alkaloids, including piperine from Piper longum (fruit of Piper longum Linn .) and pepper (fruit of Piper nigrum L. ), hydroxy-α-sanshool, and hydroxy-γ-sanshool from zanthoxylum (fruit of Zanthoxylum bungeanum Maxim ), as well as caffeine from Wuyi rock tea. The chromatographic fractions were identified by mass spectrometry. Single factor test and orthogonal test were both carried out to optimize the extraction conditions. The method validation indicated that the accuracy represented by spiked recovery ranged in 98.89%–102.06%, the correlation coefficients in 0.99986–0.99999. These results show that the prepared monolithic column can be successfully used to quantitatively analyse alkaloids from the real medicinal and edible plant foods with reversed-phase mechanism, which can avoid the long analytical time using traditional packed C18 column. The present method is a simple, and inexpensive method for quantitatively analysing alkaloids from medicinal and edible plant foods, exhibiting good specificity and durability.

Kaempferol as an AIE-active natural product probe for selective Al3+ detection in Arabidopsis thaliana

Lu Sun,Xiaoqing Wang,Jiuzhou Shi,Shilong Yang,Li Xu

In this work, a natural product probe, kaempferol, which exhibited aggregation-induced emission (AIE) characteristic in water/tetrahydrofuran (THF) binary solvent was explored. The probe showed high resistance to photobleaching capacity and excellent selectivity towards Al 3+ in the aggregation state. Upon the addition of Al 3+ , the probe displayed more than 12-fold (I 486 /I 421 ) fluorescence intensity enhancement, accompanied by a color change, suggesting that the aggregated kaempferol can be used as a ratiometric probe for Al 3+ detecting. Notably, promising selectivity to Al 3+ within the pH range of 6–8 made the probe suitable for physiological conditions. Further Arabidopsis thaliana root imaging experiment demonstrated that the probe could image Al 3+ in the plant.

Fabrication of Kaempferol Loaded Zein Nanoparticles: Investigation of in Vitro Cytotoxicity and Apoptosis Induction in Oral Cancer Cells

Roopashree R.,Gupta Anchal,Rathore Mahendra Singh,Sharma Kamini,Chinnathambi Arunachalam,Alharbi Sulaiman Ali,Ramar Mohankumar,Kalaiarasi Giriraj,Thangavelu Indumathi,Suriyaprakash Jagadeesh

Oral cancer remains a significant health challenge, necessitating innovative therapeutic strategies to enhance treatment efficacy and minimize side effects. This study investigates the potential of kaempferol-loaded zein nanoparticles (KZNPs) for this purpose. Kaempferol, a flavonoid with anticancer properties, has poor water solubility, limiting its effectiveness. Zein nanoparticles (ZNPs) offer a promising delivery system for such bioactive compounds. UV-Vis spectroscopy identified Kaempferol’s absorption peaks at 347 and 253 nm, which shifted to 338 nm when encapsulated in ZNPs, indicating a change in π–π* conjugation. Dynamic light scattering (DLS) and scanning electron microscopy (SEM) confirmed that sodium caseinate (SC) stabilizes ZNPs, resulting in spherical particles with optimal size and stability. Fourier transform infrared (FTIR) spectroscopy suggested enhanced hydrogen bonding between Kaempferol and zein. Differential scanning calorimetry (DSC) revealed the absence of Kaempferol’s crystalline peaks in KZNPs. The encapsulation efficiency (EE) was 98.39%, and drug release studies showed a controlled release of 79% kaempferol over 8 h. In vitro assays demonstrated that KZNPs significantly increased Kaempferol’s cytotoxicity against PCI-13 oral cancer cells without affecting normal NIH3T3 cancer cells. Overall, these results demonstrate that our KZNPs enhanced biocompatibility and anticancer properties for oral cancer cells.

Kaempferol Molecularly Imprinted Monolithic Columns Prepared by a Surface Imprinting Method and Their Applications to Direct Separation of Flavonoids From Ginkgo Leaves by Liquid Chromatography

Wenlin Zhong,Fangping Liang,Lingling Zhong,Jiming Ou,Jianfeng He,Quanzhou Wu

In this study, kaempferol molecularly imprinted monolithic columns (MIMCs) featuring a homogeneous pore structure and high separation ability were successfully prepared by surface imprinting on silica monoliths. These columns were then used to separate flavonoids from the hydrolysate of Ginkgo leaves. The preparation process involved three simple steps: preparation of silica monoliths, functionalization of the silica surface, and polymerization of the imprinting system onto the silica surface. The resulting MIMCs exhibited a homogeneous pore structure, high surface area (>100 m 2 /g), high porosity (ca. 74%), and good permeability (3.0 − 3.9 × 10 −15 m 2 ). The chromatographic separation performance of the MIMCs prepared on amino-functionalized silica monoliths was significantly superior to those prepared on thiol and vinyl-functionalized silica monoliths. The MIMCs prepared on amino-functionalized silica monoliths (I.D. 4.6 × 20 mm) could nearly achieve baseline separation of four structurally similar flavonoids: genistein, kaempferol, isorhamnetin, and quercetin. In addition, these MIMCs exhibited excellent selectivity in the chromatographic separation of flavonoids from the hydrolysate of Ginkgo leaves. However, when used as SPE adsorbents, the MIMCs prepared on thiol and vinyl-functionalized silica monoliths were superior to those on amino-functionalized silica monoliths in terms of purification of flavonoids from the Ginkgo hydrolysate. This study may be of instructive significance to the facile preparation of MIMCs for the high-selectivity separation and analysis of target components in complex natural systems.

Quantitative Determination of Kaempferol by using a novel B-Z Chemical Oscillating system Catalyzed by a Cu (II)-tetraazamacrocyclic Complex

Saif Ullah,Rooh Ullah,Rasha M.K. Mohamed,Gang Hu,Ateeq ur rehman,Muhammad Yasir Nawabi,Asif Hayat,Ata Ur Rehman,Mehboob Hassan,Ikram Uddin,Syed Attaullah Shah,Muhammad Zahir Shah,Jimei Song

An appropriate analytical technique for determination of kaempferol (KMF) by its perturbation effect on Belousov-Zhabotinskii (B-Z) oscillating system was reported. The macrocyclic copper (II) complex [CuL] (ClO 4 ) 2 ] was used as a catalyst while malic acid as substrate in B-Z system. The ligand L in the macrocyclic-complex is 5, 7, 7, 12, 14, 14-hexamethyl-1, 4, 8, 11-tetraazacyclotetradeca-4, 11-diene. Experimental outcomes shown the perturbation effect of KMF could cause the change in the amplitude of oscillation (ΔA) which was directly related to its concentration (2.5 × 10 -6 to 3.0 × 10 -4 mol/L) with correlation coefficient of 0.99315. The calculated relative standard deviation (RSD) is 3.1 % by eight samples (1.8 × 10 -5 mol/L) and the observed lower limit of detection is 1.25 × 10 -6 mol/L. The cyclic voltammetry (CV) experiments were used to confirm the redox reaction between kaempferol and sodium bromate. Furthermore, the reaction perturbation mechanism was derived from the well-known FKN (Field-Koros-Noyes) oscillation mechanism.

Mechanism of Toona sinensis seed polyphenols inhibiting oxidation and modifying physicochemical and gel properties of pork myofibrillar protein under oxidation system

Jingyu Wang,Lei Zhou,Lujuan Xing,Guanghong Zhou,Wangang Zhang

This research aimed to elucidate how Toona sinensis seed polyphenols (TSSs) inhibited pork myofibrillar protein (MP) oxidation and modified physicochemical and gel properties in the Fenton oxidation system. Our result displayed that TSSs had a lower carbonyl (from 1.38 to 0.59 nmol/mg) and dityrosine contents (from 47.22 to 25.07), and higher free amino content (from 386.99 to 485.00 nmol/mg), followed by ellagic acid (EA) and quercetin-3-O-rhamnoside (QC). Meanwhile, the incorporation of phenolic compounds changed secondary and tertiary structure of proteins and then increased solubility and absolute value of zeta potential, as well as decreased turbidity and average particle size. Molecular docking indicated that MP interacted with EA primarily via hydrogen bonds and hydrophobic interaction, and with QC mostly through hydrogen bonds and electrostatic interaction. Otherwise, the incorporation of EA promoted MP gel to form a honeycomb-like microstructure after oxidation. Therefore, TSSs, EA, and QC could significantly inhibit MP oxidation as well as modify its physicochemical properties, but only EA enhanced its gel properties after oxidation. The results could be useful to improve the comprehensive utilization of plant by-products and provide theoretical references for the role of TSSs in the improvement of meat products.

Studies on the inhibition mechanism of α-glucosidase by kaempferide: Enzyme kinetic, multi-spectroscopy and molecular docking techniques

Li Zhao,Shuang Luo,Zhiyun Peng,Guangcheng Wang

α-Glucosidase (α-Glu) is an enzyme that lowers postprandial blood glucose after breaking down complex carbohydrates. Kaempferide is the principal flavonoid active ingredient in plants and is widely found in fruits, vegetables, and beverages. This study found that kaempferide has the potential to inhibit α-Glu activity to treat type 2 diabetes. The results showed that kaempferide (IC 50  = 55.35 ± 0.27 μM), serving as a mixed-type inhibitor for α-Glu, exhibited sensibly superior inhibition of α-Glu than acarbose (IC 50  = 414.08 ± 10.73 μM). In addition, the outcomes from fluorescence quenching, 3D fluorescence, synchronous fluorescence, CD spectroscopy, and molecular docking analysis showed that kaempferide can not only chelate with α-Glu by hydrogen bonding and Van der Waals forces, but also affect the secondary structure and activity of the enzyme. After oral administration of sucrose in mice, kaempferide effectively reduces postprandial blood glucose (PBG) and without any other adverse symptoms. In summary, this study has the potential to contribute to the development of functional foods for the prevention and management of type 2 diabetes (T2DM).

Multi-responsive fibroin-based nanoparticles enhance anti-inflammatory activity of kaempferol

Wenjing Yang,Dengchao Xie,Yuqi Liang,Nanxi Chen,Bo Xiao,Lian Duan,Min Wang

Kaempferol is well known for the desirable properties in radical scavenging, oxidation resistance and inflammation resistance. However, the clinical translation of kaempferol for inflammatory disease treatment is still restricted by the poor solubility, undesirable stability and dissatisfied bioavailability. In this manuscript, multi-responsive kaempferol loaded fibroin nanoparticles (KF-NPs) were fabricated to overcome these shortages in biomedical application. The experimental results suggested that the negative charged KF-NPs (−25.2 ± 0.3 mV) had favorable average particle size (151 ± 1.6 nm) with uniform distribution. Noticeably, the accumulative drug release of fibroin nanoparticles dramatically increased in environment with acidity, glutathione (GSH) or reactive oxygen species (ROS), indicating the on-demand intracellular drug release properties. In vitro experiments revealed that the KF-NPs were biocompatible and can be internalized by cells. Moreover, KF-NPs had synergistic effect in downregulating the expression of TNF-α and eliminating the intracellular reactive oxygen. These results suggested that KF-NPs were promising therapeutic platform for anti-inflammatory treatments.
