A Biotic Strategy for Enhanced Hexavalent Chromium Removal by Zero-Valent Iron under the Interference of Humic Acid

Mingxi Li,Yuhang Yang,Weiquan Li,Zhiyi Deng,Jinhua Wu

Zero-valent iron (Fe0) has been extensively used in hexavalent chromium (Cr(VI)) removal from groundwater, but its treatment suffers from interference of humic acid (HA) and ferrochrome precipitate. In this study, a biotic Fe0system was established to address these problems in Cr(VI) removal from HA-rich groundwater by introducing a combination of heterotrophic and hydrogen-autotrophic microorganisms. Due to the formation of HA-Fe complexes and ferrochrome precipitates on the Fe0surface, the HA-abiotic Fe0system obtained a slight Cr(VI) removal of 10.5%. While in the HA-biotic Fe0system, heterotrophic microbes could effectively eliminate HA through biodegradation and decrease HA-Fe complex generation; autotrophic microbes used H2from iron corrosion as electron donors for their metabolism and promoted iron corrosion and active secondary mineral generation (e.g., magnetite and green rust) for Cr(VI) adsorption and reduction. Therefore, a much higher Cr(VI) removal of 84.9% was achieved. Additionally, increasing HA content and extra electron acceptors (e.g., sulfate and nitrate) both boosted Cr(VI) removal, further proving the role of heterotrophic microbes in biodegrading HA for enhanced Cr(VI) elimination. This work presented a feasible strategy to achieve efficient Cr(VI) removal with Fe0by diminishing HA interference and ferrochrome precipitate passivation through the synergistic effect of heterotrophic and hydrogen-autotrophic microorganisms.

Enhanced Cr(VI) removal and stabilization from bioleached wastewater by zero-valent iron coupled with hetero and autotrophic bacteria

Yuhang Yang,Bing Li,Mingxi Li,Zhiyi Deng,Zuoyi Chen,Jinhua Wu

Zero-valent iron (Fe 0 ) usually suffers from organic acid complexation and ferrochrome layer passivation in Cr(VI) removal from bioleached wastewater of Cr slag. In this work, a synergetic system combined Fe 0 and mixed hetero/autotrophic bacteria was established to reduce and stabilize Cr(VI) from bioleached wastewater. Due to bacterial consumption of organic acid and hydrogen, severe iron corrosion and structured-Fe(II) mineral generation (e.g., magnetite and green rust) occurred on biotic Fe 0 surface in terms of solid-phase characterization, which was crucial for Cr(VI) adsorption and reduction. Therefore, compared with the abiotic Fe 0 system, this integrated system exhibited a 6.1-fold increase in Cr(VI) removal, with heterotrophic reduction contributing 3.4-fold and abiotic part promoted by hydrogen-autotrophic bacteria enhancing 2.7-fold. After reaction, the Cr valence distribution and X-ray photoelectron spectroscopy indicated that most Cr(VI) was converted into immobilized products such as Fe x Cr 1-x (OH) 3 , Cr 2 O 3 , and FeCr 2 O 4 by biotic Fe 0 . Reoxidation experiment revealed that these products exhibited superior stability to the immobilized products generated by Fe 0 or bacteria. Additionally, organic acid concentration and Fe 0 dosage showed significantly positive correlation with Cr(VI) removal within the range of biological adaptation, which emphasized that heterotrophic and autotrophic bacteria acted essential roles in Cr(VI) removal. This work highlighted the enhanced effect of heterotrophic and autotrophic activities on Cr(VI) reduction and stabilization by Fe 0 and offered a promising approach for bioleached wastewater treatment .

Isolation and identification of green alga Chlorella vulgaris HNUFU001 for environmental selenium enrichment in heterotrophic growth regime

Zhenyu Zhang,Li Wang,Yinqiang Wu,Chenshuo Li,Pengcheng Fu,Jing Liu

Microalgae , particularly Chlorella , have promising selenium (Se) enrichment capacity and great potential in wastewater treatment and the development of nutraceuticals. To acquire Chlorella strains with strong heterotrophic propensity and robust Se tolerance, this research utilized a heterotrophic isolation strategy along with a high Se concentration medium as selection factors. The combined sequence of the ribosomal small subunit (SSU) and internal transcribed spacer (ITS) was utilized for phylogenetic analysis , and comparative analysis of ITS secondary structures was also integrated to identify the desired strain. A novel subtropical microalgal strain, Chlorella vulgaris HNUFU001 with high Se tolerance, was isolated from water samples in a lake at Hainan University, China. Gaussian process regression was utilized to assess the strain's growth under varying Se concentrations. The intracellular accumulation and extracellular removal of Se were measured using Inductively Coupled Plasma-Mass Spectrometry (ICP-MS) and Inductively Coupled Plasma Optical Emission Spectrometer (ICP-OES), respectively. The maximum intracellular organic Se (92 % of total Se) was observed as 108.835 μg/g dry weight (DW) for 1 mg/L Se treatment. The highest Se removal rate was 40 %. The high Se tolerance and enrichment traits enable Chlorella vulgaris HNUFU001 to be a promising candidate for the heterotrophic production of organic Se-related nutraceuticals.

Man Quanli,Zhang Peilian,Huang Weiqi,Zhu Qing,He Xiaoling,Wei Dongsheng

A novel salt-tolerant heterotrophic nitrification and aerobic denitrification (HN-AD) bacterium was isolated and identified as Halomonas venusta TJPU05 ( H. venusta TJPU05). The nitrogen removal performance of H. venusta TJPU05 in simulated water (SW) with sole or mixed nitrogen sources and in actual wastewater (AW) with high concentration of salt and nitrogen was investigated. The results showed that 86.12% of NH 4 + -N, 95.68% of NO 3 − -N, 100% of NO 2 − -N and 84.57% of total nitrogen (TN) could be removed from SW with sole nitrogen sources within 24 h at the utmost. H. venusta TJPU05 could maximally remove 84.06% of NH 4 + -N, 92.33% of NO 3 − -N, 92.9% of NO 2 − -N and 77.73% of TN from SW with mixed nitrogen source when the salinity was above 8%. The application of H. venusta TJPU05 in treating AW with high salt and high ammonia nitrogen led to removal efficiencies of 50.96%, 47.28% and 43.19% for NH 4 + -N, NO 3 − -N and TN respectively without any optimization. Furthermore, the activities of nitrogen removal-related enzymes of the strain were also investigated. The successful detection of high level activities of ammonia oxygenase (AMO), hydroxylamine oxidase (HAO), nitrate reductase (NAR) and nitrite reductase (NIR) enzymes under high salinity condition further proved the HN-AD and salt-tolerance capacity of H. venusta TJPU05. These results demonstrated that the H. venusta TJPU05 has great potential in treating high-salinity nitrogenous wastewater.

Effective Two-Stage Heterotrophic Cultivation of the Unicellular Green Microalga Chromochloris zofingiensis Enabled Ultrahigh Biomass and Astaxanthin Production

Chen Qiaohong,Chen Yi,Xu Quan,Jin Hu,Hu Qiang,Han Danxiang

Chromochloris zofingiensis has obtained particular interest as a promising candidate for natural astaxanthin production. In this study, we established a two-stage heterotrophic cultivation process, by using which both the growth of C. zofingiensis and astaxanthin accumulation are substantially enhanced. Specifically, the ultrahigh biomass concentration of 221.3 g L−1 was achieved under the optimum culture conditions in 7.5 L fermenter during 12 days. When scaled-up in the 500 L fermentor, the biomass yield reached 182.3 g L−1 in 9 days, while the astaxanthin content was 0.068% of DW. To further promote astaxanthin accumulation, gibberellic Acid-3 (GA3) was screened from a variety of phytohormones and was combined with increased C/N ratio and NaCl concentration for induction. When C. zofingiensis was grown with the two-stage cultivation strategy, the astaxanthin yield reached 0.318 g L−1, of which the biomass yield was 235.4 g L−1 and astaxanthin content was 0.144% of DW. The content of the total fatty acids increased from 23 to 42% of DW simultaneously. Such an astaxanthin yield was 5.4-fold higher than the reported highest record and surpassed the level of Haematococcus pluvialis. This study demonstrated that heterotrophic cultivation of C. zofingiensis is competitive for industrial astaxanthin production.

Heterotrophic Nitrification–Aerobic Denitrification by Bacillus sp. L2: Mechanism of Denitrification and Strain Immobilization

Qiang Li,Yuehui He,Boyan Wang,Nanhai Weng,Lei Zhang,Kaichun Wang,Fengrong Tian,Mingsheng Lyu,Shujun Wang

The biological denitrification of low-C/N wastewater is a great challenge in treatment plants due to the lack of microorganisms with heterotrophic nitrification–aerobic denitrification (HN-AD) abilities. In this study,Bacillussp. L2 was isolated from aeration tank water samples using a nitrification medium and screened for its ability to perform HN-AD in low-C/N wastewater. The strain showed a maximum NH4+-N removal rate of 98.37% under low-C/N conditions. In the presence of a mixed N source, strain L2 was capable of completely removing NH4+-N within 24 h. Furthermore, optimal nitrogen removal conditions for strain L2 were found to be C/N = 9, pH = 9, and sodium acetate as the C source. Under optimal conditions, the strain was able to maintain a high NH4+-N removal rate under 0–3% salinity and an NH4+-N concentration of 200 mg/L or less. The denitrification pathways of strain L2 were NH4+→NH2OH→NO2−(↔NO3−)→NO→N2O→N2and NH4+→NH2OH→NO→N2O→N2. Furthermore, semi-continuous wastewater treatment was conducted using immobilized technology, which resulted in more than 82% NH4+-N removal after three cycles of reuse. This study demonstrates the great potential ofBacillussp. L2 in wastewater treatment applications.

Study on the Synchronous Removal of Nitrogen and Phosphorus by Autotrophic/Heterotrophic Denitrification in the Presence of Pyrite

Minyi Zhu,Minhui Ma,Shuo Chen,Rongfang Yuan,Shaona Wang

Pollution caused by N and P is a significant contributor to water eutrophication. While traditional biological treatment processes can remove some N and P elements from water, the effluent quality often fails to meet the stringent requirements of sensitive areas. The autotrophic denitrification’s simultaneous nitrogen and phosphorus removal pro-cess, known for its low operating cost and minimal sludge production, has garnered considerable attention from researchers. In this study, natural pyrite was used for the removal of nitrogen and phosphorus in a denitrification system, and the underlying mechanisms were elucidated. The results indicate that the N and P removal efficiency was influenced by empty bed contact time (EBCT) and the pH value. The highest NO 3 − -N removal rate of 90.24% was achieved at an EBCT of 8 h, while the PO 4 3− -P removal rate reached 81.58% at an EBCT of 12 h. The addition of a carbon source enhanced the synergistic autotrophic/heterotrophic denitrification, significantly improving phosphorus removal with an increasing C/N ratio. Microbial characteristics analysis revealed that, at the phylum level, Chlorobiota , Bacteroidota , and Chloroflexota played a crucial role in heterotrophic autotrophic denitrification. At the genus level, Thauera , Aridibacter , and Gemmatimonas were key players in heterotrophic denitrification, while Thiobacillus , Rhodoplanes , and Geobacter were associated with autotrophic denitrification.

Evaluation of wood vinegar as a green carbon source for heterotrophic nitrification and aerobic denitrification process from performance, mechanism and potential

Weidong Xiao,Yaoguo Wu,Sihai Hu,Zherui Zhang,Lang Ran,Hongjian Luo,Ran Sun

Carbon sources as energy sources and electron donors are essential for the nitrogen removal efficiencies of heterotrophic nitrification and aerobic denitrification (HNAD) process. However, conventional carbon sources from the chemical industry face issues of low stability of biological treatment systems and increased indirect carbon emissions in practical applications. Encouragingly, wood vinegar, a by-product of biomass pyrolysis process, promises to address these concerns. Herein, we investigated the potential of wood vinegar for green composite carbon source in HNAD process from multiple perspectives by establishing different microcosms. Regular monitoring results showed that wood vinegar-fed microcosms had the optimal cost-effectiveness with a C/N ratio of 7, and total nitrogen and COD removal efficiencies of up to 85.0 % and 93.4 % respectively. Furthermore, wood vinegar significantly improved the species diversity and optimized the microbial community structure compared to traditional carbon sources, strengthening the stability and resistance of the microcosm. Meanwhile, the enrichment analysis of key genes and enzymes indicated that both carbon and nitrogen metabolism were enhanced obviously, and the wood vinegar as a composite carbon source was expected to overcome the defects of traditional carbon sources, better balance the nitrification and denitrification capabilities. Importantly, the introduction of wood vinegar does not present a risk of secondary contamination and contributes to carbon reduction in the water treatment field. All indications are that wood vinegar as a green composite carbon sources can enhance wastewater treatment system stability and efficiency, and provide a viable strategy for promoting more sustainable wastewater management.

Removal of nitrogen by heterotrophic nitrification-aerobic denitrification of a novel metal resistant bacterium Cupriavidus sp. S1

Zhiyi Sun,Yongkang Lv,Yuxiang Liu,Ruipeng Ren

A novel heterotrophic nitrifying and metal resistant bacterium was isolated and identified as Cupriavidus sp. S1. The utilization of ammonium, nitrate and nitrite as well as the production of N 2 proved the heterotrophic nitrification and aerobic denitrification ability of S1. The ammonium, nitrate and nitrite removal efficiencies were 99.68%, 98.03% and 99.81%, with removal rates of 10.43, 8.64 and 8.36   mg/L/h, respectively. A multiple regression equation well described the relationship between carbon source utilization, cell growth and nitrification. Keeping the shaking speed at 120   rpm was beneficial for denitrification. Moreover, different forms of nitrogen source could be utilize in simultaneous nitrification and denitrification. Additionally, the efficient removal of ammonium occurred at 20.0   mg/L   Zn 2+ , or 10.0   mg/L   Ni 2+ or 8.0   mg/L   Cu 2+ or 5.0   mg/L   Cr 6+ , 33.35   mmol/L sodium pyruvate, C/N 12–28. These findings demonstrate that S1 was effective for nitrogen removal in industrial wastewater containing heavy metal.

Polystyrene nanoparticles intensify the algae-mediated negative priming effect on leaf litter decomposition

Jingjing Du,Tianying Tao,Mengxi Gao,Xueting Zhang,Junjie Zhang,Maosen Wang,Xun Zhou,Yangyi Qin,Peizhi Ji,Xiaole Hu,Tianyao Cheng

The potential threat of nanoplastics on the heterotrophic decomposition of leaf litter lies in their impact on microbial activity and community structure in streams. Therefore, it is crucial to understand the interactions between benthic algae and microbial decomposers when assessing the functioning of stream ecosystems. However, the potential influence of benthic algae on the response of heterotrophic decomposers to nanoplastics remains unknown. This study investigated the interactions between benthic algae and heterotrophic microbial decomposers in the presence of polystyrene nanoparticles (PS-NPs), focusing on their role in leaf litter decomposition and nutrient cycling. The microcosm experiment demonstrated a negative priming effect of benthic algae on leaf decomposition in the absence of PS-NPs. In contrast, the algae-mediated negative priming effect was significantly intensified under PS-NP exposures, decreasing the decomposition rate by 21.3 %. During the various ecological processes, PS-NP exposures significantly reduced the algal biomass and dissolved organic carbon, which in turn disrupted the transfer of labile carbon from benthic algae to heterotrophic microbial decomposers and consequently impeding leaf decomposition process. Additionally, the synergistic effect of benthic algae and PS-NPs decreased the fungal diversity and undermined the dominance of functional genera (i.e., Setophaeosphaeria and Tetracladium ) within the microbial decomposer community. In summary, this study offers innovative insights into the interactions among microbial communities colonizing streambed substrates under plastic pollution, highlighting the ecological implications of nanoplastic detrimental influences on aquatic ecosystems.

Enhanced denitrification of sewage via bio-microcapsules embedding heterotrophic nitrification-aerobic denitrification bacteria Acinetobacter pittii SY9 and corn cob

Yunqi Shi,Yongyou Hu,Donghui Liang,Guobin Wang,Jieyun Xie,Xiaoqiang Zhu

In this work, bio-microcapsules were prepared by embedding heterotrophic nitrification and aerobic denitrification (HN-AD) bacteria ( Acinetobacter Pittii SY9) and corn cob. Bio-microcapsules (20 g/L of corn cob and 30% v/v suspension of strain SY9) were porous (pore size 2579.74–3725.44 nm; porosity 53.6%–79.9%). Under the appropriate conditions (C/N > 2, temperature of 20–35 ℃, rotation speed of 100–120 rpm, pH of 7–9), TN removal efficiency of bio-microcapsules reached 94.4%, and 74.0% of nitrogen was converted into N 2 . The results of kinetics fitting indicated that aerobic denitrification was the limiting step during HN-AD process. Bio-microcapsules could slow the carbon release of corn cob for 120 days, which ensuring high HN-AD performance even at low C/N of 2.8. Bio-microcapsule SBR could stably run for 88 days with TN removal efficiency > 90% for synthetic sewage. Bio-microcapsules embedding strain SY9 and corn cob have prospective applications for enhancing denitrification of sewage.

Non-Equilibrium Hydrogel Demonstrates Cytocompatible and Seamless Heterotrophic Self-Growth

Xinyu Chen,Ruilin Zhang,Hanlei Zhang,Xiaofan Hu,Ruinan Wang,Qiangjun Ling,Peng Shi,Kunyu Zhang,Liming Bian

The growth of living organisms is achieved through the autonomous integration of external nutrients as building blocks under mild physiological conditions. Inspired by this biological property, researchers have developed self-growing materials based on the chemical polymerization of reactive monomers. However, the limited cytocompatibility of covalent bonding-driven self-growing materials hinders their biomedical applications. Herein, a non-equilibrium self-growing hydrogel driven is reported by cytocompatible ligand–ion coordination between calcium ion (Ca 2+ ) and bisphosphonate-modified pectin (PT-BP). Multiform self-growth of the seed PT-BP-Ca 2+ hydrogel occurs autonomously in a heterotrophic manner by seamlessly integrating supplied soluble PT-BP polymer as integral structures via the crosslinking mediated by Ca 2+ diffusion. This self-growth enables diverse cytocompatible post-gelation modifications of hydrogels, including complex surface patterning and segmental growth-induced cold welding. Unlike the gap-interfered interfaces of self-healed hydrogels, the monolithic and continuous structures of self-growing PT-BP-Ca 2+ hydrogels better support the migration and interactions of cells encapsulated in the seed hydrogel and newly grown regions. In vivo self-growth of cancer cell-seeded hydrogel in an animal model enhances macrophage infiltration, a key pathogenic feature during cancer progression. This work demonstrates a simple and cytocompatible strategy to fabricate bio-inspired heterotrophic self-growing materials for diverse biomedical applications.
