Rationale : Ischemia-reperfusion-induced acute kidney injury (IR-AKI), characterized by the abrupt decline in renal function, is distinguished by the intricate interplay between oxidative stress and inflammation. In this study, a reactive oxygen species (ROS) scavenger-CF@PDA was developed to effectively target antioxidant and anti-inflammatory pathways to disrupt the oxidative stress-inflammation cycle in IR-AKI. Methods : UV-vis absorption spectra, FTIR spectra, and TEM were employed to determine the successful construction of CF@P. ABTS, TMB, and NBT analyses were performed to detect the antioxidant ability and enzyme-mimicking ability of CF@P. In vitro and in vitro , the antioxidant/anti-inflammatory effect of CF@P was detected by MTT, qPCR, fluorescence, and flow cytometry. Multi-omics revealed the mechanism of CF@P in IR-AKI therapy, and molecular docking was further used to determine the mechanism. MRI and photoacoustic imaging were employed to explore the dual-mode imaging capacity of CF@P in IR-AKI management. Results : CF@P could disrupt the oxidative stress-inflammatory cascade by scavenging ROS, reducing pro-inflammatory cytokines, and modulation of macrophage polarization. Subsequent multi-omics indicated that the renal protective effects may be attributed to the inhibition of pyruvate dehydrogenase kinase 4 (PDK4). Metabolomics demonstrated that CF@P could improve the production of antioxidant compounds and reduce nephrotoxicity. Additionally, CF@P exhibited promising capabilities in T1-MRI and photoacoustic imaging for AKI management. Conclusions : Collectively, CF@P, possessing antioxidant/anti-inflammatory properties by inhibiting PDK4, as well as imaging capabilities and superior biocompatibility, holds promise as a therapeutic strategy for IR-AKI.

Objective To develop and validate updated classification criteria for giant cell arteritis (GCA). Methods Patients with vasculitis or comparator diseases were recruited into an international cohort. The study proceeded in six phases: (1) identification of candidate items, (2) prospective collection of candidate items present at the time of diagnosis, (3) expert panel review of cases, (4) data‐driven reduction of candidate items, (5) derivation of a points‐based risk classification score in a development data set and (6) validation in an independent data set. Results The development data set consisted of 518 cases of GCA and 536 comparators. The validation data set consisted of 238 cases of GCA and 213 comparators. Age ≥50 years at diagnosis was an absolute requirement for classification. The final criteria items and weights were as follows: positive temporal artery biopsy or temporal artery halo sign on ultrasound (+5); erythrocyte sedimentation rate ≥50 mm/hour or C reactive protein ≥10 mg/L (+3); sudden visual loss (+3); morning stiffness in shoulders or neck, jaw or tongue claudication, new temporal headache, scalp tenderness, temporal artery abnormality on vascular examination, bilateral axillary involvement on imaging and fluorodeoxyglucose–positron emission tomography activity throughout the aorta (+2 each). A patient could be classified as having GCA with a cumulative score of ≥6 points. When these criteria were tested in the validation data set, the model area under the curve was 0.91 (95% CI 0.88 to 0.94) with a sensitivity of 87.0% (95% CI 82.0% to 91.0%) and specificity of 94.8% (95% CI 91.0% to 97.4%). Conclusion The 2022 American College of Rheumatology/EULAR GCA classification criteria are now validated for use in clinical research.

Background Cerebral venous sinus thrombosis (CVST) is a rare neurovascular disorder with highly variable manifestations and clinical courses. Animal models properly matched to the clinical form of CVST are necessary for elucidating the pathophysiology of the disease. In this study, we aimed to establish a rat model that accurately recapitulates the clinical features of CVST in human patients. Methods This study consisted of a clinical analysis and animal experiments. Clinical data for two centres obtained between January 2016 and May 2021 were collected and analysed retrospectively. In addition, a Sprague–Dawley rat model of CVST was established by inserting a water-swellable rubber device into the superior sagittal sinus, following which imaging, histological, haematological, and behavioural tests were used to investigate pathophysiological changes. Principal component analysis and hierarchical clustering heatmaps were used to evaluate the similarity between the animal models and human patients. Results The imaging results revealed the possibility of vasogenic oedema in animal models. Haematological analysis indicated an inflammatory and hypercoagulable state. These findings were mostly matched with the retrospective clinical data. Pathological and serological tests further revealed brain parenchymal damage related to CVST in animal models. Conclusions We successfully established a stable and reproducible rat model of CVST. The high similarity between clinical patients and animal models was verified via cluster analysis. This model may be useful for the study of CVST pathophysiology and potential therapies.

Background Inflammatory bowel disease (IBD) is a chronic inflammatory bowel disease with no clinical cure. Excessive production of reactive oxygen species (ROS) at the inflammatory sites leads to the onset and progression of IBD. And the current non-invasive imaging methods are not ideal for the diagnosis and monitoring of IBD.Methods Herein, we developed inulin (IN)-coated cerium oxide nanoparticles (CeO2@IN NPs) for treatment and monitoring of IBD guided by computed tomography (CT). The physicochemical properties, ROS scavenging ability and CT imaging capabilities of CeO2@IN were investigated in vitro. Moreover, the therapeutic and targeted inflammation imaging effects of CeO2@IN were validated in dextran sulfate sodium (DSS)-induced colitis model. Results CeO2@IN with catalase (CAT) and superoxide dismutase (SOD) capabilities effectively scavenged ROS, thus protecting the cells against oxidative stress. In colitis model mice, orally administered CeO2@IN successfully traversed the gastrointestinal tract to reach the colon under the protection of IN, and effectively reduced intestinal inflammation, thereby maintaining the intestinal epithelial integrity. Notably, CeO2@IN performed better than conventional CT contrast agents for gastrointestinal tract imaging, particularly in detecting the inflamed areas in the colon. In addition, CeO2@IN exhibited excellent biocompatibility in vitro and in vivo. Conclusion The study provided a novel integrated diagnostic and therapeutic tool for the treatment and monitoring of IBD, presenting great potential as a clinical application for IBD.

Objectives This study aims to investigate the use of sodium iodide (NaI), dimethyl sulfoxide (DMSO), ethyl alcohol, and ethyl acetate as cone-beam CT (CBCT) contrast agents for diagnosing cracked teeth. The optimal delay time for detecting the number of crack lines beyond the dentino-enamel junction (Nd), the number of cracks extending from the occlusal surface to the pulp cavity (Np), and the depth of the crack lines was explored. Methods 14 human extracted cracked teeth were collected, 12 were used for enhanced scanning, and 2 were used for exploring the characteristic of crack lines. The teeth were scanned in 3 CBCT enhanced scanning (ES) modes: ES1 using meglumine diatrizoate (MD); ES2 using NaI and DMSO, ES3 using NaI, DMSO, ethyl alcohol and ethyl acetate. Three delay times (15mins, 30mins, and 60mins) were set for scanning. Nd, Np, and depth of crack lines were evaluated. Results There were totally 24 crack lines on 12 cracked teeth. Nd was 10 in ES1 at 60mins, 24 in ES2 at 60mins and 24 in ES3 at 15mins. Np was 1 in ES1 at 60mins, 10 in ES2 at 60mins and 21 in ES3 at 60mins, and there were significantly different among them ( p < 0.01). The average depth presented on ES3 was significantly deeper than ES1 and ES2 ( p < 0.01). Conclusion NaI, DMSO, ethyl alcohol and ethyl acetate show potential as contrast agents for enhanced CBCT scanning in diagnosis of cracked teeth and their depth in vivo . A delay time of 15 min is necessary to confirm the existence of crack lines, while a longer delay time is required to ascertain if these crack lines extend to the pulp cavity.

PEG and PEG/PEI modified superparamagnetic iron oxide nanoparticles (SPIONs) were synthesized by the thermal decomposition of iron (III) acetylacetonate (Fe(acac) 3 ) in poly (ethylene glycol) (PEG) containing poly (ethylene imine) (PEI) (0 or 0.3 g). PEG/PEI-SPIONs were coated with Tween 80 (PEG/PEI/Tween 80-SPIONs). Fourier transform infrared spectroscopy (FTIR) analyses indicated that PEG , PEG/PEI and PEG/PEI/Tween 80 were attached to the surfaces of the SPIONs. The PEG-SPIONs, PEG/PEI-SPIONs and PEG/PEI/Tween 80-SPIONs performed excellent colloidal stability in the phosphate buffered saline (PBS), and in deionized water with the mean hydrodynamic sizes of 19.5, 21.0, 24.0 nm and the zeta potentials of -5.0, 35.0, 19.0 mV, respectively. All the SPIONs showed low cytotoxicity assessed by the MTT assay. In vivo magnetic resonance imaging (MRI) of the Kunming (KM) mouse brains were performed, the PEG-SPIONs, PEG/PEI-SPIONs and PEG/PEI/Tween 80-SPIONs exhibited vascular imaging effects in bulbus olfactorius, frontal cortex, temporal, thalamus and brain stem of the mouse brains after 24 h intravenous injection of the nanoparticles . The SPIONs have potentials as MRI contrast agents in the mouse brains.

Inorganic nanoparticles have been introduced into biological systems as useful probes for in vitro diagnosis and in vivo imaging, due to their relatively small size and exceptional physical and chemical properties. A new kind of colortunable Gd-Zn-Cu-In-S/ZnS (GZCIS/ZnS) quantum dots (QDs) with stable crystal structure has been successfully synthesized and utilized for magnetic resonance (MR) and fluorescence dual modality imaging. This strategy allows successful fabrication of GZCIS/ZnS QDs by incorporating Gd into ZCIS/ZnS QDs to achieve great MR enhancement without compromising the fluorescence properties of the initial ZCIS/ZnS QDs. The as-prepared GZCIS/ZnS QDs show high T
1 MR contrast as well as “color-tunable” photoluminescence (PL) in the range of 550–725 nm by adjusting the Zn/Cu feeding ratio with high PL quantum yield (QY). The GZCIS/ZnS QDs were transferred into water via a bovine serum albumin (BSA) coating strategy. The resulting Cd-free GZCIS/ZnS QDs reveal negligible cytotoxicity on both HeLa and A549 cells. Both fluorescence and MR imaging studies were successfully performed in vitro and in vivo . The results demonstrated that GZCIS/ZnS QDs could be a dual-modal contrast agent to simultaneously produce strong MR contrast enhancement as well as fluorescence emission for in vivo imaging.

Gd3+-based/mesoporous silica-coated multifunctional core–shell (Y,Gd)2O3:Eu3+@nSiO2@mSiO2 nanoparticles (YGO-Bmnc NPs) with red luminescence, paramagnetic and mesoporous properties are developed as a novel nanomedical platform for combined multimodal diagnosis and therapy. 40 nm sized (Y,Gd)2O3:Eu3+ (YGO-Ac) nanocore is synthesized by a labor-saving solvothermal method, 20 nm sized perpendicularly aligned mesoporous silica shell is then successfully coated onto the nanocore through two-step sol–gel process and a highly efficient surfactant removal method (NH4NO3–EtOH). The surface area of nanocore can be increased from 24 m2 g−1 to 562 m2 g−1 after coating mesoporous silica shell without any core-free silica NPs. Light-emitting Eu3+ ion is chosen as a model to study the solvothermal mechanism and downconversion/upconversion luminescence properties of nanocore; Y3+ is doped into matrix to enhance energy transfer from Gd3+ to the doped Eu3+. Further investigation of luminescence property of YGO-Bmnc NPs shows that they are good red phosphors with millisecond-lever fluorescence lifetime (τ = 2.54 ms, 612 nm, 5D0–7F2). MTT assay reveals low cytotoxicity of the system with IC50 ([Gd]) > 1000 µg mL−1 (6.36 mM). The possibility of using YGO-Bmnc NPs for optical imaging in vitro has been demonstrated. Magnetic resonance imaging (MRI) in vitro exhibits a high r1 relaxivity of 5.05 mM−1 s−1 and low r2/r1 ratio of 1.216 (3.0 T) which is much lower than other existing Gd3+-based nanoscale contrast agents, manifesting that YGO-Bmnc NPs can be efficient T1 contrast agents. In light of their good performance in optical-MR imaging and highly ordered mesoporous structure, it is anticipated that they are suitable for combined multimodal diagnosis and therapy, they can load other imaging agents or drugs to provide complementary information from each imaging modality and guide individual treatment. What's more, the mesoporous silica shell can provide a large venue for the effective modification of various functional groups and biotargets with selectivity and specificity.

This study demonstrates the synergistic integration of X-ray-induced radioluminescence (RL), persistent luminescence (PersL) and radio-photoluminescence (RPL) in a single material system, which provides innovative multimodal applications for radiometric information sensing, storage and reading, anti-counterfeiting and imaging. A novel radiation-sensing material, BaMgP2O7:Sm, was prepared using a high temperature solid state method. Under X-ray irradiation, the pale pink phosphor shows distinct luminescence from defects. Under 365 nm ultraviolet (UV) light (bright-field), it emits bright red light, confirming trivalent samarium (Sm3+) reduction to divalent samarium (Sm2+). In dark-field, orange fluorescence appears, indicating defect-mediated charge storage and slow release. Notably, the material enables single-irradiation multiple imaging, effectively overcoming the limitation of conventional materials that only allow single imaging per irradiation event, thereby greatly enhancing information utilization and imaging flexibility. Moreover, the flexible film based on polydimethylsiloxane (BaMgP2O7:Sm@PDMS) demonstrates remarkable tri-mode X-ray detection and imaging capabilities, it has a high spatial resolution of 14.4 lp mm−1, integrating real-time readout, delayed readout, and long-term information storage. BaMgP2O7:Sm enables dynamic anti-counterfeiting via time-dependent luminescence and precise radiation dose monitoring through X-ray responsive emission, these features make it a promising multifunctional material.

Presented in this article is the synthesis of a new class of block copolymer, poly(ethylene oxide)-block-poly(tert-butyl acrylate-stat-acrylic acid) [PEO-b-P(AA-stat-tBA)], which can self-assemble into polymer vesicles with tuneable sizes at various conditions. The biocompatible and hydrophilic PEO chains form the vesicle coronas, while the PAA-stat-PtBA chains form the membrane. Superparamagnetic iron oxide nanoparticles (SPIONs) were generated in situ within the membrane of the polymer vesicles by nanoprecipitation. 1H NMR, GPC, DLS, TGA, VSM and TEM were employed to characterize the structure and properties of the block copolymer, polymer vesicles and Fe3O4-decorated magnetic polymer vesicles. The water-dispersible, biocompatible, drug deliverable and superparamagnetic polymer vesicles exhibited excellent colloidal stability at a range of pH conditions and very high T2 relaxivity, demonstrating ultra-sensitivity for magnetic resonance imaging and promising potential applications in nanomedicine.

Objectives:To evaluate the diagnostic accuracy using sodium iodide (NaI) and dimethyl sulfoxide (DMSO) as contrast agent in cone beam computed tomography (CBCT) scanning, and compare this with micro-CT.Methods:18 teeth were cracked artificially by soaking them cyclically in liquid nitrogen and hot water. After pre-treatment with artificial saliva, the teeth were scanned in four modes: CBCT routine scanning without contrast agent (RS); CBCT with meglumine diatrizoate (MD) as contrast agent (ES1); CBCT with NaI + DMSO as contrast agent (ES2); and micro-CT (mCT). The number of crack lines was evaluated in all four modes. Depth of crack lines and number of cracks presented from the occlusal surface to the pulp cavity (Np) in ES2 and micro-CT images were evaluated. Results:There were 63 crack lines in all 18 teeth. 45 crack lines were visible on ES2 images as against four on the RS and ES1 images (p<0.05) and 37 on micro-CT images (p>0.05). Further, 34 crack lines could be observed on both ES2 and micro-CT images, and the average depth presented on ES2 images was 4.56 ± 0.88 mm and 3.89 ± 1.08 mm on micro-CT images (p<0.05). More crack lines could be detected from the occlusal surface to the pulp cavity on ES2 images than on micro-CT images (22 vs 11).Conclusion:CBCT with NaI +DMSO as the contrast agent was equivalent to micro-CT for number of crack lines and better for depth of crack lines. NaI + DMSO could be a potential CBCT contrast agent to improve diagnostic accuracy for cracked tooth.
