In-situ synthesis of ZnO onto nanoporous gold microelectrode for the electrochemical sensing of arsenic(III) in near-neutral conditions

Zhongxin Zhuang,Yan Chen,Kankan Chen,Zhonggang Liu,Zheng Guo,Xingjiu Huang

The exploration of advanced electrochemical sensors towards highly toxic arsenic(III) has attracted growing interest in recent times. Herein, a ZnO decorated nanoporous Au (NPG) microelectrode was fabricated via a facile electrochemical alloying/dealloying method followed by thermal treatment, and developed for the electrochemical sensing of toxic As(III). Inspired by the electrochemical deposition of Zn and the formation of Au-Zn nanostructure on the surface of the Au microelectrode during the alloying/dealloying process, the in-situ evolution of the formed Au-Zn nanostructure to ZnO nanoparticles wrapped on the nanoporous Au surface was achieved. The ZnO/NPG microelectrode exhibited excellent electrochemical performance towards As(III) detection in near-neutral conditions (0.1 M phosphate buffer solution (PBS), pH: 5.0). A high sensitivity of 1.366 μA·ppb −1 ·cm −2 with a low limit of detection of 0.30 ppb (S/N = 3) was achieved at concentrations ranging from 1.0 ppb to 260 ppb. Moreover, the prepared ZnO/NPG microelectrode was successfully applied in the analysis of As(III) in tap and lake water samples with satisfactory recovery rates. The ZnO/NPG microelectrode presented exciting prospects for the electrochemical analysis of As(III) in near-neutral conditions.

Electrochemical Measurement of Cholesterol Flip-Flop in Plasma Membrane at Single Cells

Kang Liu,Shuohan Huang,Danjun Fang

Here, a microelectrode approach is established to measure the flip-flop rate of cholesterol in plasma membranes at single living cells. The initial validation is performed in a modeled phospholipid bilayer positioned at an interconnecting hole between two compartments, in which cholesterol in one compartment diffuses into the other one through a flip-flop movement in the bilayer and is then detected by a cholesterol oxidase-modified microelectrode. As compared with the time (140 ± 28 s) for free cholesterol transport in absence of the bilayer, a prolonged time (702 ± 42 s) is needed to observe the current increase in the presence of the bilayer. The difference in the time (562 s) gives the estimated flip-flop time of cholesterol in the bilayer. The position of the microelectrode in contact with a living cell and the injection of cholesterol inside the cell are further applied to measure the cholesterol flip-flop in the plasma membrane. The average time (1183 ± 146 s) is obtained to observe an additional current increase at the microelectrode, which reflects the cholesterol flip-flop rate in plasma membranes in single living cells. All these results support the establishment of this microelectrode approach for the study of the cholesterol flip-flop process in lipid membranes.

Ion Selective Electrode Mediated by Transfer Layer of Graphene Oxide for Detection of Ca2+ with Highly Sensitivity and Selectivity

Yuandong Liu,Limin Zhang,Jingwen Mo,Yi Zhou,Yang Tian

A Ca2+ selective microelectrode was proposed through electrodepositing graphene oxide (GO) on carbon fiber as an efficient ion-electron transfer layer. The gold leaves were further synthesized at GO surface to increase the specific surface area, and provide a substrate for assembly of the specific ligands for Ca2+. GO with negatively charges greatly facilitated the electron transfer process from the recognition center to electrode surface, resulting in the apparently enhanced sensitivity towards Ca2+. Moreover, the developed ion-selective microelectrode exhibited good selectivity to Ca2+, and a good linear range from 10 μM to 10 mM with a detection limit of 5.91 ± 0.46 μM.

MWCNTs/PEDOT:PSS nanocomposites-modified microelectrode array for spatial dynamics recording of epileptic discharges in multi-subregion of hippocampal slice

Enhui He,Shengwei Xu,Guihua Xiao,Yuchuan Dai,Xinrong Li,Yilin Song,Fei Gao,Yu Zhang,Shihong Xu,Xinxia Cai

The acute hippocampal slice is useful for understanding how epilepsy spatially initiates, propagates, and terminates in the hippocampus, which is vital to study the epileptic mechanism in it. However, it is challenging to detect the electrical signals of the main cell layer (the granular cell layer in the dentate gyrus (DG) and the pyramidal cell layer in CA1 and CA3a–c), which play an important role in epilepsy, because it is difficult to be accurately located by electrodes and typically show low throughput. In this study, a special in vitro microelectrode array (MEA) for rat hippocampal slices was fabricated. The recording sites of the MEA were designed along the main cell layer to record accurately the electrical signals of this layer in each hippocampal sub-region. A special “less-hydrophilic-MWCNTs/PEDOT:PSS modification method” for in vitro MEA was applied to deposit electrochemically the MWCNTs/PEDOT:PSS nanocomposites onto the surface of the microelectrodes, which significantly improved their performance with low impedance (36.41 ± 3.07 kΩ), small phase delay (−41.2 ± 4.90°), high signal-to-noise ratio (S/N = 5.4) and good biocompatibility. The application of high-K + -ACSF and D-AP5 (NMDA receptor antagonist) showed a neural signal projection pathway of the epileptic discharges in the hippocampal slice. The epileptic discharges originated in CA3b, where the neurons acted as a pacemaker, and spread to CA1 and DG bi-directionally along the main cell layer. Moreover, the epileptic discharges spread backward (CA3b to DG) faster than forward (CA3b to CA1). The MEA provides a reliable and sensitive dedicated biosensor for hippocampal slices.

PtNPs/Short MWCNT-PEDOT: PSS-Modified Microelectrode Array to Detect Neuronal Firing Patterns in the Dorsal Raphe Nucleus and Hippocampus of Insomnia Rats

Yun Wang,Mixia Wang,Yuchuan Dai,Yilin Song,Yiding Wang,Botao Lu,Yinghui Li,Xinxia Cai

Research on the intracerebral mechanism of insomnia induced by serotonin (5-HT) deficiency is indispensable. In order to explore the effect of 5-HT deficiency-induced insomnia on brain regions related to memory in rats, we designed and fabricated a microelectrode array that simultaneously detects the electrical activity of the dorsal raphe nucleus (DRN) and hippocampus in normal, insomnia and recovery rats in vivo. In the DRN and hippocampus of insomnia rats, our results showed that the spike amplitudes decreased by 40.16 and 57.92%, the spike repolarization slope decreased by 44.64 and 48.59%, and the spiking rate increased by 66.81 and 63.40%. On a mesoscopic scale, the increased firing rates of individual neurons led to an increased δ wave power. In the DRN and hippocampus of insomnia rats, the δ wave power increased by 57.57 and 67.75%. Furthermore, two segments’ δ wave slopes were also increased in two brain regions of the insomnia rats. Our findings suggest that 5-HT deficiency causes the hyperactivity of neurons in the hippocampus and DRN; the DRN’s firing rate and the hippocampal neuronal amplitude reflect insomnia in rats more effectively. Further studies on alleviating neurons affected by 5-HT deficiency and on achieving a highly effective treatment for insomnia by the microelectrode array are needed. Keywords:microelectrode array;insomnia;electrophysiology;dorsal raphe nucleus;serotonin

Uric acid sensor based on surface molecularly imprinted nanocavity coordination carbon nanotube functionalized acupuncture needle microelectrode

Quan Li,Jinhai Li,Maofeng Wang,Hongying Liu

Uric acid (UA) levels within the human body are intricately associated with a variety of diseases, underscoring the critical need for a reliable and sensitive method for UA detection to facilitate disease prevention and early diagnosis. This paper presents a novel microsensor designed to address this method. The sensor employs an alumina-based inorganic molecular-imprinted polymer (MIP) that specifically encapsulates uric acid molecules. This MIP is electropolymerized onto multi-walled carbon nanotubes (MWCNTs) anchored on a microelectrode (ANE), forming a highly sensitive platform for UA detection. The sensor’s performance was meticulously characterized using electrochemical analysis, high-resolution scanning electron microscopy (SEM), and elemental mapping techniques. Our findings reveal that the imprinted samples demonstrate significantly enhanced adsorption capacity and selectivity compared to their non-imprinted counterparts. This superior performance is attributed to the synergistic effects of chemical interactions between the target UA molecules and the precisely tailored imprinted cavities, as well as the precise size matching between the cavities and the target molecules. Additionally, the inorganic molecular-imprinted polymer membrane also addresses the instability issues associated with traditional organic molecular-imprinted layers. The sensor’s specificity is further validated by the preparation of distinct recognition sites within the electropolymerized molecular imprinting layer. Under the optimized conditions, the sensor exhibits a linear increase in peak current with UA concentration ranging from 0.5 to 500 μM, with an impressive detection limit of 0.11 μM. Moreover, the developed sensor shows promising potential for the analysis of UA levels in human serum and urine samples, thereby paving the way for broader applications in clinical diagnostics and personalized medicine.

Natural Convection in Molten Salt Electrochemistry

Jianbang Ge,Biwu Cai,Fei Zhu,Yang Gao,Xinrui Wang,Qianjin Chen,Mingyong Wang,Shuqiang Jiao

Molten salt electrochemistry has been widely used in many fields, especially for metal extraction/refinement. The understanding of mass transfer in molten salts under harsh operation conditions is of great importance to reveal reaction mechanisms and advance fine technologies. It has been generally assumed that natural convection is negligible in stagnant molten salt electrochemistry. Herein, we report an abnormal natural convection in molten LiCl–KCl, with the arising time from 2.37 s at 873 K to 10.13 s at 673 K. Using the concentration correction factor, the derived thickness of the natural convection–diffusion layer (δconv.) was found to be ranging from 128 to 163 μm, much thinner than those in aqueous solutions (∼200 μm). The simulations showed that the notable natural convection resulted from convection–diffusion layer (CDL) convection dominated over the density-driven convection even at high redox concentrations, implying the severe vibration of molten salt systems. To suppress the intense natural convection, we predicted that the use of microelectrodes (with radii less than 23.2 μm for δconv. = 150 μm) would be a promising tool, regardless of their inferior stabilities in high-temperature molten salts at this stage. These innovative findings offer insights into the impact of natural convection on mass transfer in molten salts that have not been previously revealed.

Effect of support salts on apparent performance of Li metal anode in Ethylene carbonate/Ethyl methyl carbonate solvent and dynamic mechanism analysis

Xiaodong Liu,Guanyu Li,Feng Wu,Cunzhong Zhang

Lithium metal batteries (LMB) have received widespread attention due to their high energy density and potential commercial value. The relationship between the rate performance, cycling performance, microstructure of the deposition layer, and SEI of lithium anodes has not yet been elucidated. Here, we investigated the cycling performance and rate characteristics of metal lithium anodes in classic carbonate electrolytes containing different supporting salts. And the microstructure and chemical information of the lithium anode surface were analyzed. In addition, microelectrode technology was used to study the dynamic behavior and reaction of electroplating/stripping lithium process under different polarization conditions (current density), revealing the apparent performance, microscopic visual information, and dynamic causes and mechanisms of SEI. The results indicate that in carbonate electrolytes containing LiBOB and LiPF 6 , the cycling performance of lithium anodes, the microstructure of the deposition layer, and SEI can be significantly improved. Microelectrode research has revealed a close causal relationship between the stability and coordination of each basic step, including lithium ion mass transfer, interface charge transfer, and surface crystallization kinetics parameters , and the apparent performance of lithium anodes. This correlation can serve as a dynamic evaluation basis for future selection and optimization of operating and manufacturing conditions.

Electrochemical Determination of Iron(III) in River Water by Differential Pulse Voltammetry (DPV) Using a Poly(3,4-Ethylenedioxythiophene)–Polystyrene Sulfonate co-Polymer/Gold Nanoparticle Modified Gold Microelectrode

Wenyan Tao,Mingguo Ye,Ying Liu,Zhenjia Lin,Chenchen Wang,Yujun Huang,Dawei Pan

An electrochemical sensor based upon a poly(3,4-ethylenedioxythiophene):polystyrenesulfonate (PEDOT-PSS) co-polymer film with gold nanoparticles (AuNPs) modified microelectrode is reported for the determination of Fe(III). Scanning electron microscopy images showed that the AuNPs were immobilized on gold (Au) microelectrode by formation of a nanofilm with PEDOT-PSS. The individual particles and their cluster-like aggregates provide a larger active electrochemical area and higher conductivity, which facilitates the determination of Fe(III). Electrochemical impedance spectroscopy (EIS) shows the AuNPs/PEDOT-PSS/Au microelectrode possesses high conductivity. A low detection limit of 0.01 µM Fe(III) was obtained using the modified microelectrode by differential pulse voltammetry (DPV) due to its large specific surface area, high conductivity, and the presence of more active sites. The optimized pH of the supporting acetate buffer was 2.0. The AuNPs/PEDOT-PSS/Au provided a linear range from 0.0119 μM to 7.14 μM. The modified electrode was successfully employed for the the determination of total dissolved iron in river water from Three Gorges with satisfactory results.

Understanding natural convection in low-concentration electrochemical system: Validation of the convection modes

Biwu Cai,Jianbang Ge,Fei Zhu,Yang Gao,Shuqiang Jiao

Natural convection could arise under density gradients of solutions caused by the electrochemical reaction occurring on the electrode surface. This density-driven convection decays rapidly with the decreasing redox concentration, and becomes negligible in dilute solutions (1–2 mM). However, steady-state voltammetry is still observed in these dilute solutions due to the presence of a convection–diffusion layer. Here we studied natural convection effects in low-concentration (5–30 mM) redox solutions and clarified the convection modes in these transient-concentration solutions using cyclic voltammetry and chronoamperometry. The combined theoretical–experimental work demonstrated the coexistence of convection–diffusion layer and density-driven convection. With the use of a 150-μm-radius Pt microelectrode, the convection–diffusion layer convection dominated natural convection in dilute solution (1–5 mM), reinforced mass transport with density-driven convection in low redox concentrations (5–11 mM), and suppressed the density-driven convection at high redox concentrations (>15 mM). The zone diagrams delineating the transition from diffusion to convection were then established to reveal the effects of electrode radius, thickness of the convection–diffusion layer, time scale, and redox concentration on natural convection. Consequently, the use of microelectrodes (∼25 μm radius) could greatly inhibit natural convection effects in 30 mM redox solution.

Detection of Dopamine Based on Aptamer-Modified Graphene Microelectrode

Cuicui Zhang,Tianyou Chen,Yiran Ying,Jing Wu

In this paper, a novel aptamer-modified nitrogen-doped graphene microelectrode (Apt-Au-N-RGOF) was fabricated and used to specifically identify and detect dopamine (DA). During the synthetic process, gold nanoparticles were loaded onto the active sites of nitrogen-doped graphene fibers. Then, aptamers were modified on the microelectrode depending on Au-S bonds to prepare Apt-Au-N-RGOF. The prepared microelectrode can specifically identify DA, avoiding interference with other molecules and improving its selectivity. Compared with the N-RGOF microelectrode, the Apt-Au-N-RGOF microelectrode exhibited higher sensitivity, a lower detection limit (0.5 μM), and a wider linear range (1~100 μM) and could be applied in electrochemical analysis fields.

Chemical polymerization of conducting polymer poly(3,4-ethylenedioxythiophene) onto neural microelectrodes

Xinze Yang,Weihua Pei,Chunrong Wei,Xiaowei Yang,Huiling Zhang,Yang Wang,Miao Yuan,Qiang Gui,Yuanyuan Liu,Yijun Wang,Hongda Chen

Poly(3,4-ethylenedioxythiophene) (PEDOT) is an organic material with good electrochemical activity and electrical conductivity , and it is usually electrochemically deposited on the neural probes to improve their interface properties . However, the workload for electrode modification using an electropolymerization method becomes greater with the increase in the number of electrode channels. Moreover, the integration of electrodes and amplification circuits makes electropolymerization difficult to implement. In this study, a chemical polymerization method was proposed to produce conducting polymer PEDOT on the electrode sites. Compared with electropolymerization, it does not require each electrode site to be connected to a power source. It is competitive in batch preparation and is particularly suitable for neural microelectrodes prepared by a micro-electromechanical systems (MEMS) process. The charge storage capacity, impedance, mechanical stability , and electrochemical stability of conducting polymer PEDOT prepared by the proposed method are evaluated. The results indicate that the method is feasible for batch modification of the microelectrodes, and it is also suitable in cases where energization is inconvenient.
