Rapid screening and analysis of alcohol dehydrogenase binders from Glycyrrhiza uralensis root extract using functionalized magnetic nanoparticles coupled with HPLC−MS/MS

ChenMiao,LiuLiangliang,ChenXiaoqing

Alcohol dehydrogenase (ADH) is a key enzyme that converts ethanol into acetaldehyde. Its binders could be used to efficiently treat human toxic alcohol poisoning and suppress the acetaldehyde accumulation in alcohol hypersensitive alcoholics. In the present study, a new assay based on ADH-functionalized magnetic nanoparticles coupled with high-performance liquid chromatography − tandem mass spectrometry (HPLC−MS/MS) was developed for the rapid screening and identification of ADH binders. ADH was immobilized on silica-coated Fe3O4 magnetic nanoparticles via covalent bonds and the synthesized nanoparticles were characterized by TEM, XRD, FT-IR, and VSM. The amount of bound ADH onto magnetic nanoparticles was 80.6 μg/mg. The optimum temperature and pH for the enzymatic activity of immobilized ADH were 25 °C and 7.0, respectively. The relative activity of immobilized ADH remained 68.14% after 10 times of recycle, which exhibited good durability. Nine compounds with ADH-binding activity were screened and identified in Glycyrrhiza uralensis root extracts, among which seven compounds were first screened and identified. Experimental results proved that the proposed method could rapidly screen ADH binders from complex mixtures.

Rapid screening of α-amylase inhibitors from Aloe vera based on polydopamine/L-cysteine bifunctionalized magnetic mesoporous silica immobilized α-amylase

Wang Lei,Li Yue,Wang Sikai,Lv Lin,Liu Hongmei,Zhang Guoqi,Zhao Yan

Diabetes mellitus is a metabolic disorder that impacts millions of individuals globally. In the treatment of this condition, it is imperative to explore natural resources for therapeutic agents that exhibit fewer adverse effects and enhanced efficacy. Currently, the methods employed for isolating anti-diabetic lead compounds from natural sources are often intricate and time-consuming. Therefore, there is an urgent need to develop efficient and rapid screening techniques. In this study, α-amylase was immobilized using a novel polydopamine/L-cysteine bifunctionalized magnetic mesoporous silica composite material (Fe 3 O 4 @nSiO 2 @mSiO 2 @PDA@L-Cys) for the first time. A ligand fishing approach utilizing the immobilized α-amylase was developed to rapidly screen for α-amylase inhibitors from Aloe vera . Characterization and property analysis of the immobilized enzyme showed that the immobilized α-amylase exhibited exceptional stability and reusability. Two ligands were successfully screened from Aloe vera and then characterized as aloin B and aloin A using ultra-high performance liquid chromatography tandem mass spectrometry. Their respective IC 50 values were 0.99 ± 0.09 mM and 1.14 ± 0.05 mM. Molecular docking studies confirmed the interaction of both ligands with specific amino acid residues within the active site of α-amylase. The study presents a fast and efficient approach for screening α-amylase inhibitors from intricate natural sources, thereby offering significant potential for the development of anti-diabetic agents. Rapid screening and characterization of natural antioxidants in Polygonum viviparum by an on-line system integrating the pressurised liquid micro-extraction, HPLC-DAD-QTOF-MS/MS analysis and antioxidant assay

Zheng-ming Qian,Ling Chen,Meng-qi Wu,De-qiang Li

Rapid discovery of active ingredients from complex matrices is one of great challenges for modern drug development. Traditional methods often require many sample treatment steps, including an extraction step with exclusively dedicated solvents followed by repeated separation and activities assessment. This present work described an integrated analytical setup for natural antioxidants discovery in which the online extraction (OLE) of a solid sample is directly coupled to its analysis by high performance liquid chromatography-diode array detector-quadrupole time-of-flight tandem mass spectrometry and 2,2′-Azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS) antioxidant assay (OLE-HPLC-DAD-QTOF-MS/MS-ABTS). This developed approach makes sample extraction, chromatographic separation and chemical detection, and antioxidant assay integrated into a single HPLC injection and was successfully applied for the rapid discovery of natural antioxidant bioactives from Polygonum viviparum . A total of 21 secondary metabolites were characterized according to their retention times, ultraviolet (UV) spectra, exact mass and fragmentation ions in MS/MS spectra, and 18 of them displayed antioxidant activity (response as negative peaks in antioxidant assay). This work describes a simple, green and efficient approach to minimize the sample consumption (only 0.4 mg was required) and eliminate complex sample treatment procedures. The developed OLE-HPLC-DAD-QTOF-MS/MS-ABTS system offers new perspectives for rapid chemical profiling of natural products and their antioxidants discovery.

Insights into a rapid screening method for anti-cucumber mosaic virus compounds

Yunlong Yan,Maoxi Huang,Li Wang,Wei Xue,Xin Xie,Xiangyang Li

Cucumber mosaic virus (CMV) is a detrimental plant virus in agricultural production. Traditionally, the half-leaf method using Nicotiana glutinosa has been used for screening agrochemicals to control CMV. However, this forms a time-consuming experimental bottleneck. In this study, we constructed a rapid screening model for anti-CMV compounds using CMV. In short, purified CMV particles were labeled through amine reactions and then subjected to binding studies with commercial compounds. The relative gene expression levels were then confirmed. Additionally, the rapid screening model results were verified using synthesized compounds. The commercial compounds ningnanmycin, ribavirin, and moroxydine hydrochloride bound to CMV with dissociation constants of 0.012, 2.870, and 0.069 μM, respectively, and they significantly inhibited expression of the gene for the CMV coat protein in CMV-infected tobacco leaves. This rapid screening model was assessed using our synthetic compounds N12 , N16 , and N18 through binding, which were shown to have dissociation constants 0.008, 0.025, and 70.800 μM, respectively, as well as via gene expression studies. Thus, a rapid method for screening anti-CMV commercial compounds and our synthetic compounds was constructed and confirmed.

Rapid Screening of Lipase Inhibitors From Chrysanthemum Based on Ionic Liquid/Chitosan Bifunctionalized Magnetic Multi-Walled Carbon Nanotubes Immobilized Lipase

Sikai Wang,Yue Li,Lei Wang,Lin Lv,Hongmei Liu,Guoqi Zhang,Yan Zhao

In this study, a novel ionic liquid/chitosan bifunctionalized magnetic multi-walled carbon nanotubes composite material (m-MWCNTs@CS@IL) was utilized for the first time to immobilize lipase. The immobilized lipase exhibited exceptional stability and reusability, as evidenced by its characteristic properties. A ligand fishing approach utilizing the immobilized lipase was developed to enable rapid screening of lipase inhibitors from Chrysanthemum . Three ligands were successfully screened from Chrysanthemum and then identified as cynaroside, quercitrin, and linarin by ultra-high performance liquid chromatography-tandem mass spectrometry. The IC 50 values of these three ligands were 76.77 ± 0.36, 83.01 ± 0.46, and 43.61 ± 0.77 µM, respectively. Furthermore, molecular docking analysis further confirmed the binding of three ligands to specific amino acid residues within the active site of lipase. This work presents a fast and efficient approach for screening lipase inhibitors from intricate natural sources, demonstrating promising prospects in discovering anti-obesity compounds.
