Bacitracin immobilized on the titanium (Ti) surface significantly improves anti-bacterial activity and biocompatibility in vitro . In the current study, we investigated the biologic performance (bactericidal effect and bone-implant integration) of bacitracin-modified Ti in vivo . A rat osteomyelitis model with femoral medullary cavity placement of Ti rods was employed to analyze the prophylactic effect of bacitracin-modified Ti (Ti-BC). Thirty-six female Sprague Dawley (SD) rats were used to establish the Ti implant-associated infection. The Ti and Ti-BC rods were incubated with and without Staphylococcus aureus to mimic the contaminated Ti rod and were implanted into the medullary cavity of the left femur, and sterile Ti rods were used as the blank control. After 3 weeks, the bone pathology was evaluated using X-ray and micro-computed tomography (micro-CT) analysis. For the investigation of the Ti-BC implant osseointegration in vivo , fifteen SD rats were divided into three groups (N = 5), namely Ti, Ti-dopamine immobilized (Ti-DOPA), and Ti-BC. Ti rods were implanted into the left femoral cavity and micro-CT and histological evaluation was conducted after 12 weeks. The in vivo study indicated that Ti-immobilized bacitracin owned the prophylaxis potential for the infection associated with the Ti implants and allowed for the osseointegration. Thus, the multiple biofunctionalized Ti implants could be realized via immobilization of bacitracin, making them promising candidates for preventing the Ti implant-associated infections while retaining the osseointegration effects.

In this study, a double-network hydrogel coating with silver loading was developed for antibacterial catheters using an in-situ photoinitiation method, a stepwise immersion process, and ion exchange. The silver-loading capacity, release kinetics, and in vitro antibacterial performance of the coatings were assessed. Furthermore, a silver-loaded coating was prepared on commercially available endotracheal tubes and its anti-infection capabilities were investigated using a rabbit tracheal intubation model. The results showed that Coating@Ag + exhibited a uniform and stable dispersion of silver ions (Ag + ), controllable silver loading capacity, and effective Ag + release. The coating also exhibited excellent in vitro antibacterial performance against methicillin-resistant Staphylococcus aureus (MRSA) and Pseudomonas aeruginosa (P. aeruginosa) . In vivo studies demonstrated a significant reduction in tracheal intubation-related pulmonary infections. Silver-loaded hydrogel coatings have potential applications in the development of antibacterial catheters for preventing healthcare-associated infections.

Bacterial infection has been a critic problem for implant infections. Poly(L-lactide) (PLLA) membrane has great potential for Guided bone regeneration (GBR), however, PLLA lack antibacterial property and thus may face bacterial infections. In this work, a mussel inspired method was used to treat PLLA membrane with dopamine and formed polydopamine (PDA) coated PLLA (PLLA@PDA), and then silver Nanoparticles (AgNPs) was immobilized on the surface of PLLA via the reduction effect of PDA. The XPS results showed that the silver element contents may be tuned from 1.6% to 15.4%. The AgNPs coated PLLA (PLLA@Ag) showed good antibacterial property (98.3% bactericidal efficiency may be obtained) and good biocompatibility, implying that the PLLA@Ag membrane have potential application as antibacterial GBR membrane, which may enhance the application of PLLA.

Polyetheretherketone (PEEK) is considered to be a potential material for oral implants due to its elastic modulus being similar to that of human cortical bone. However, the poor antibacterial, anti-inflammatory, and osseointegration properties of bioinert PEEK have hindered its clinical application. Therefore, this study designed and constructed a pH-responsive PEEK implant with a bilayer core–shell zeolitic imidazolate framework-8 (ZIF-8) structure loaded on its surface, with an antimicrobial peptide (KR12) encapsulated in the outer shell and an osteogenic peptide (osteogenic growth peptide ) encapsulated in its inner core. In this study, the bilayer core–shell ZIF-8 structure was confirmed to have pH-responsive properties. In vitro studies proved that the implant could promote bone marrow mesenchymal stem cells’ proliferation and differentiation and the M1 phenotype to M2 phenotype conversion of RAW 264.7 and could inhibit bacterial adhesion and proliferation. By constructing rats’ distal femur with/without infection models, it was further demonstrated that the novel implant could effectively inhibit bacterial adhesion and growth, inhibit inflammation, and promote peri-implant osseointegration, which was more substantial when the local area was infected and the pH was lower than that of normal tissue. Collectively, the results suggest that this novel pH-responsive PEEK implant loaded with a bilayer core–shell ZIF-8 structure is a promising peptide delivery implant system, which is well suited for dental applications and offers a potential solution for the prevention of infection during the early phase after implantation.

Infection prevention and bone-implant integration remain major clinical challenges. Silver nanoparticle (AgNPs) bone-implant coatings have received extensive attention. Balancing the toxicity and antibacterial properties of AgNP coatings has become a significant problem. In this study, inspired by the structure of the ant-nest, a polyetherimide (PEI) coating with ant-nest structure was prepared, aiming to realize the structural modification of the AgNPs coating. AgNPs were loaded in the inner porous area of the PEI ant-nest coating, avoiding direct contact between AgNPs and cells. The nanopores on the surface of the coating ensured the orderly release of silver ions. SEM, FTIR, XPS, and XRD experiments confirmed that the PEI ant-nest coating was successfully prepared. Interestingly, in the PEI ant-nest coating, Ag + showed a steady increase in the release trend within 28 days, and there was no early burst release phenomenon. In -vivo experiments showed a good control effect for local infection. In order to improve the osteogenic properties of the materials, 45S5 bioactive glasses (BG) were loaded to achieve further osseointegration. In general, this natural ant-nest-inspired surface modification coating for orthopedic prostheses provides a new strategy for balancing the antibacterial and toxic effects of AgNP coatings.

Largemouth bass virus (LMBV) poses a serious threat to commercial fish production, resulting in significant economic losses to the aquaculture industry. To date, no approved treatment for LMBV infection exists. In response to this urgent need, we conducted an investigation into the anti-LMBV activity of 15 natural compounds. Among these compounds, luteolin (LUT) was the most effective, with a remarkable inhibition rate of 97.27 % on LMBV replication. In vitro antiviral activity assays further demonstrated that LUT significantly inhibited LMBV replication in epithelioma papulosum cyprini (EPC) cells, and the half maximal inhibitory concentration (IC 50 ) was 2.77 μM. Additionally, viral titers and cytopathic effect (CPE) were reduced in the presence of LUT, suggesting its potential to protect infected cells. Mechanistically, LUT could directly affect LMBV particles and weaken their infectivity. Furthermore, LUT was effective in inhibiting LMBV adsorption to host cells and had preventive effects. In vivo studies revealed that LUT improved the survival rate of largemouth bass infected with LMBV. Real-time quantitative PCR (RT-qPCR) assays demonstrated that LUT significantly decreased viral loads in the liver, spleen, and kidney of largemouth bass at 1, 3, 5, and 7 days after infection. These findings suggest that LUT has the potential as a therapeutic agent to protect economically valuable fish from LMBV infection.

Bacterial infections, aseptic loosening, and poor osseointegration remain challenges for orthopedic implants. In this study, we developed a dual-network polyelectrolyte antibacterial coating for orthopedic implants to prevent bacterial infections. The coating was prepared via UV-triggered in situ polymerization, composed of polyethyleneimine (PEI) and polyacrylic acid (PAA), with enhanced mechanical properties due to electrostatic interactions between them. The antimicrobial peptide ε-poly-L-lysine (EPL) was immobilized in the coating, providing efficient antibacterial functionality. The coating's substrate was made of hydroxyapatite (HA) and polylactic acid (PLA) composites to promote bone cell growth and tissue regeneration. In vitro experimental results demonstrated that the coating exhibited excellent antibacterial properties against both Gram-negative and Gram-positive bacteria, while enhancing the adhesion, proliferation, and differentiation of osteoblasts. In vivo experiments further confirmed the coating's capability to resist infection in an infected microenvironment. This study provides a practical strategy for developing orthopedic implant surface coatings with antibacterial functions.

Skin wound healing is an important clinical challenge, and the main treatment points are accelerating epidermal regeneration and preventing infection. Therefore, it is necessary to develop a wound dressing that can simultaneously cure bacterial infections and accelerate wound healing. Here, we report a multifunctional composite wound dressing loaded with chitosan (CS)-binding bFGF (CSBD-bFGF) and antimicrobial peptides (P5S9K). First, CS was used as the dressing matrix material, and P5S9K was encapsulated in CS. Then, CSBD-bFGF was designed by combining recombinant DNA technology and tyrosinase treatment and modified on the dressing material surface. The results show that the binding ability of CSBD-bFGF and CS was significantly improved compared with that of commercial bFGF, and CSBD-bFGF could be controllably released from the CS dressing. More importantly, the prepared dressing material showed excellent antibacterial activity in vivo and in vitro and could effectively inhibit the growth of E. coli and S. aureus. Using NIH3T3 cells as cellular models, the results showed that the CSBD-bFGF@CS/P5S9K composite dressing was a friendly material for cell growth. After cells were seeded on the composite dressing surface, collagen-1 (COL-1) and vascular endothelial growth factor (VEGF) genes expression in cells were significantly upregulated. Finally, the full-thickness wound of the rat dorsal model was applied to analyse the tissue repair ability of the composite dressing. The results showed that the composite dressing containing CSBD-bFGF and P5S9K had the strongest ability to repair skin wounds. Therefore, the CSBD-bFGF@CS/P5S9K composite dressing has good antibacterial and accelerated wound healing abilities and has good application prospects in the treatment of skin wounds.

Biofilm-associated infections (BAIs) pose a serious threat to public health; in particular, a reactive oxygen species (ROS)-rich biofilm microenvironment can weaken the immunological response of antimicrobial natural killer (NK) cells. Conventional phototherapy cannot consistently inhibit the escape of planktonic bacteria from BAIs, thereby enabling persistent and recurrent infections. In this study, a thymosin β4 bio-orthogonally labeled NK (βNK) cytomembrane is proposed to decorate PtTe 2 nanozymes (βNK@PtTe 2 ) with hyperthermia and NK cells immune re-rousing to eliminate BAIs at all stages. Te-doped PtTe 2 exhibits outstanding ROS scavenging activity through an electron-enriched catalytic center with strong p-d orbital hybridization, and it generates a thermally driven force for disorganizing dense biofilm structures under laser irradiation. After biofilm elimination and ROS scavenging, the βNK cytomembrane in βNK@PtTe 2 selectively promotes chemotaxis and activates conventional NK (cNK) cells to kill planktonic bacteria that escape from biofilm disintegration at the infection site. In vivo, this therapeutic paradigm of immune microenvironment remodeling induces cNK cells to develop into long-lived tissue-resident NK (trNK) cells with an immunological memory against reinvading pathogens, thereby effectively preventing secondary infections. Overall, the as-prepared βNK@PtTe 2 combined with NK cells provides a systematic photo-immunologic strategy to inhibit recurrent biofilms and treat infectious diseases caused by such infections.

Wounds in harsh environments can face long-term inflammation and persistent infection, which can slow healing. Wound spray is a product that can be rapidly applied to large and irregularly dynamic wounds, and can quickly form a protective film in situ to inhibit external environmental infection. In this study, a biodegradable A and B combined multi-functional spray hydrogel is developed with methacrylate-modified chitosan (CSMA 1st ) and ferulic acid (FA) as type A raw materials and oxidized Bletilla striata polysaccharide (OBSP) as type B raw materials. The precursor CSMA 1st -FA/OBSP (CSOB-FA 1st ) hydrogel is formed by the self-cross-linking of dynamic Schiff base bonds, the CSMA-FA/OBSP (CSOB-FA) hydrogel is formed quickly after UV–vis light, so that the hydrogel fits with the wound. Rapid spraying and curing provide sufficient flexibility and rapidity for wounds and the hydrogel has good injectability, adhesive, and mechanical strength. In rats and miniature pigs, the A and B combined spray hydrogel can shrink wounds and promote healing of infected wounds, and promote the enrichment of fibrocyte populations. Therefore, the multifunctional spray hydrogel combined with A and B can protect irregular dynamic wounds, prevent wound infection and secondary injury, and be used for safe and effective wound treatment, which has a good prospect for development.

Infection associated with orthopedic implants is the chief cause of implant failure. An important consideration to prevent the infection at implants is to inhibit the biofilm formation for the initial 6 h. Therefore, we fabricated hyaluronidase-sensitive multilayers of chitosan (Chi)/sodium hyaluronate-lauric acid (SL) onto the surface of bone morphogenetic protein 2 (BMP2) loaded titanium nanotube (TNT) via spin-assisted layer-by-layer technique. The results of both Fourier transform infrared spectroscopy (FTIR) and nuclear magnetic resonance ( 1 H NMR) confirmed the successful synthesis of SL. The multilayer structure on BMP2 loaded TNT was characterized by field-emission scanning electron microscopy (FE-SEM), atomic force microscopy (AFM) and water contact angle, respectively. The release profiles confirmed that hyaluronidase could trigger the release of lauric acid (LA) from the SL multilayer and accelerate the release of BMP2 in the system. The hyaluronidase-sensitive-multilayer-coated BMP2-loaded TNT (TNT/BMP2/(Chi/SL/Chi/Gel) 4 ) not only demonstrated good antibacterial capability, but also showed good biocompatibility in in vitro usage, which was supported by the efficient growth inhibition of both Staphylococcus aureus and Escherichia coli , as well as higher cell viability, alkaline phosphatase activity, mineralization capability, and higher gene expression of osteoblasts on TNT/BMP2/(Chi/SL/Chi/Gel) 4 . This study developed an alternative approach to fabricate effective antibacterial implants for orthopedic implantation.

Antibacterial hydrogels have attracted much attention in skin tissue engineering due to their superior ability to prevent infection. In this study, we developed an antibacterial hydrogel using a multi-active site N- halamine - modified dopamine-hyaluronic acid hydrogel. The in vitro antibacterial efficacy of the hydrogel exhibited 99.99 % against both Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus , while also demonstrating excellent biocompatibility. Furthermore, in Escherichia coli- infected skin wounds, the hydrogels effectively suppressed infection and significantly accelerated wound healing. These findings suggest that N -halamine-modified dopamine-hyaluronic acid antibacterial hydrogels have great potential as wound dressings for preventing infection and promoting the healing of infected wounds.
