Multi-functional bioactive hydrogel based on CCM-Co-ZIF-8 nanoparticles for chronic wound healing: Synergistic antibacterial, antioxidant, and anti-inflammatory therapy

Qi Tao,Hang Yao,Zehao Gu,Xiaofei Yang,Junli Shi,Dong-An Wang,Hui Chong

The treatment of chronic wounds are characterized by a local tissue with issues such as the accumulation of free radicals and persistent inflammation, which contribute to delayed wound healing. Thus, it has emerged as a significant global challenge, profoundly impacting patients’ quality of life and even posing threats to their lives. In this study, we fabricted a CCM (Curcumin)–Co-ZIF-8 nanoparticle (CCZ)@oxidized ammonium alginate (OSA)/carboxymethyl chitosan (CMCS)/tannic acid (TA) composite multifunctional bioactive hydrogel. This nanocomposited double-crosslinked network hydrogel dressing exhibits excellent mechanical properties and self-healing abilities. Additionally, the hydrogel shows good biocompatibility. Due to the formation of dynamic Schiff base bonds between oxidized sodium alginate and CMCS, the hydrogel-TA spray system possesses injectability upon applied to a wound model in mice, allowing better adaptation to irregular wound shapes. Furthermore, it could promote synergistic antibacterial, antioxidant, and anti-inflammatory effects, thereby accelerating wound healing. Our findings highlight the potential of this multifunctional hydrogel system as an advanced therapeutic option for chronic wound management.

Electrospun robust, biodegradable, bioactive, and nanostructured sutures to accelerate the chronic wound healing

Yiran Li,Hongxing Xu,Wenwen Zhao,Li Zhang,Shaohua Wu

The design and development of advanced surgical sutures with appropriate structure and abundant bio-functions are urgently required for the chronic wound closure and treatment. In this study, an integrated technique routine combining modified electrospinning with hot stretching process was proposed and implemented to fabricate poly(L-lactic acid) (PLLA) nanofiber sutures, and the Salvia miltiorrhiza Bunge-Radix Puerariae herbal compound (SRHC) was encapsulated into PLLA nanofibers during the electrospinning process to enrich the biofunction of as-generated sutures. All the PLLA sutures loading without or with SRHC were found to exhibit bead-free and highly-aligned nanofiber structure. The addition of SRHC was found to have no significant influences on the fiber morphology, diameter, and the crystallinity of as-prepared PLLA sutures. Importantly, all the SRHC-contained PLLA nanofiber sutures possessed excellent tensile and knot strength, which were of significant importance for the surgical suture applications. Besides, the antioxidant and anti-inflammatory properties of these sutures obviously enhanced with the increasing of SRHC concentration. Furthermore, the in vitro cell tests illustrated that the high fiber orientation of the sutures was able to efficiently induce the human dermal fibroblasts (HDFs) to migrate in a rapid manner, and the sutures loaded with high content of SRHC could significantly promote the attachment and proliferation of HDFs in comparison. The in vivo diabetic mouse model experiments revealed that all the as-developed PLLA sutures could effectively close the wound, but the PLLA sutures containing high content of SRHC could dramatically promote the wound healing with high quality by shortening the healing time, improving the collagen deposition, neovascularization, and the regeneration of hair follicles, especially compared with commercial polyester (PET) suture. This study offers a simple and easily-handling strategy to develop robust, biodegradable, bioactive, and nanostructured PLLA sutures, which shows huge potential for the treatment of hard-to-heal diabetic wounds.

Accelerating chronic wound healing by breaking the pH limitations of biomolecule-loaded Nanozymes and remodeling the microenvironment in Cascade reactions

Chuanwei Shi,Peng Liu,Yeping Liu,Zhexu Gai,Fei Yang,Yanzhao Yang

The microenvironment around the wounds of chronic disease patients often exhibits weakly alkaline conditions, primarily due to defects in the patients' blood glucose regulation, tissue hypoxia, accumulation of metabolic products, and bacterial infection and growth. To regulate the wound microenvironment across a broad pH range, a Fe-doped ZIF-8 nanozyme loaded with glucose oxidase (Fe-ZIF-8@GOx) was synthesized via a one-step in situ growth method. This nanozyme, with ZIF-8 serving both as a protective matrix and immobilization scaffold, enhances the enzyme's operational microenvironment and maintains functionality across a wide pH range. It exhibits the activities of cascade glucose oxidase (GOx), catalase-like (CAT-like), and peroxidase-like (POD-like) enzymes. When Fe-ZIF-8@GOx nanozymes encounter high glucose levels at the site of weakly alkaline chronic ulcers, the GOx particles catalyze the reduction of local pH, simultaneously triggering CAT-like enzyme activity to release O₂ and improve the wound microenvironment. The subsequent pH reduction induces the controlled release of iron ions, enabling Fe 2+ to react with endogenously produced hydrogen peroxide to generate reactive oxygen species (ROS), conferring a distinctive guest–carrier synergistic antibacterial effect. Both in vitro and in vivo studies confirm that this material effectively reduces inflammation, modulates macrophage polarization, enhances collagen deposition, and promotes angiogenesis, ultimately accelerating wound healing under chronic diabetic conditions. Thus, our work not only overcomes microenvironmental limitations in antibacterial therapy but also offers a promising strategy for treating chronic diabetic infections.

Electrospun chitosan/PVA/bioglass Nanofibrous membrane with spatially designed structure for accelerating chronic wound healing

Qingchang Chen,Jun Wu,Yuan Liu,Yanqun Li,Chuqiu Zhang,Weichen Qi,Kelvin W.K. Yeung,Tak Man Wong,Xiaoli Zhao,Haobo Pan

Cutaneous wounds, especially chronic wounds, remain clinical challenges, and this is partially due to the complex healing process composed of four overlapping but distinct stages including hemostasis, inflammation, proliferation and remodeling. Therefore, wound dressings with spatially designed structures which can temporally regulate certain bioactive components to function at specific healing stages might be able to accelerate the healing process. In this study, nanobioglass incorporated chitosan-PVA (polyvinyl alcohol) trilayer nanofibrous membrane (nBG-TFM) was fabricated via sequential electrospinning. This membrane exhibited excellent biocompatibility, antibacterial activity and regeneration promotion effect. Furthermore, spatially designed structure optimized functions of each component and provided more suitable microenvironment as compared with uniform membrane. Rat full-thickness skin defects model and mice diabetic chronic wound model showed that nBG-TFM could achieve significantly accelerated and enhanced healing, in terms of complete re-epithelialization, improved collagen alignment and formation of skin appendages. It was revealed that nBG-TFM functioned through upregulating growth factors including VEGF and TGF-β. Meanwhile inflammatory cytokines such as TNF-α and IL-1β were downregulated. The technology presented in this study shed new light on designing functional wound dressings which can promote healing of chronic wounds.

Injectable Hydrogels with Integrated Ph Probes and Ultrasound-Responsive Microcapsules as Smart Wound Dressings for Visual Monitoring and On-Demand Treatment of Chronic Wounds

Da Huang,Jiahao Du,Fang Luo,Gang He,Minglang Zou,Yongming Wang,Zhenyu Lin,Decheng Wu,Zuquan Weng

Hydrogel dressings capable of infection monitoring and precise treatment administration show promise for advanced wound care. Existing methods involve embedd ingorganic dyes or flexible electronics into preformed hydrogels, which raise safety issues and adaptability challenges. In this study, an injectable hydrogel based smart wound dressing is developed by integrating food-derived anthocyanidin as a visual pH probe for infection monitoring and poly( L -lactic acid) microcapsules as ultrasound-responsive delivery systems for antibiotics into a poly(ethylene glycol) hydrogel. This straightforwardly prepared hydrogel dressing maintains its favorable properties for wound repair, including porous morphology and excellent biocompatibility. In vitro experiments demonstrated that the hydrogel enabled visual assessment of pH within the range of 5 ∼ 9.Meanwhile, the release of antibiotics could be triggered and controlled by ultrasound. In vivo evaluations using infected wounds and diabetic wounds revealed that the wound dressing effectively detected wound infection by monitoring pH levels and achieved antibacterial effects through ultrasound-triggered drug release. This led to significantly enhanced wound healing, as validated by histological analysis and the measurement of inflammatory cytokine levels. This injectable hydrogel-based smart wound dressing holds great potential for use in clinical settings to inform timely and precise clinical intervention and in community to improve wound care management.

Autonomous, Moisture-Driven Flexible Electrogenerative Dressing for Enhanced Wound Healing

Ren Yan,Xueliang Zhang,Hai Wang,Tikang Wang,Guozhang Ren,Qizeng Sun,Fei Liang,Yangzhi Zhu,Wei Huang,Hai-Dong Yu

Electrotherapy has shown considerable potential in treating chronic wounds, but conventional approaches relying on bulky external power supplies and mechanical force are limited in their clinical utility. This study introduces an autonomous, moisture-driven flexible electrogenerative dressing (AMFED) that overcomes these limitations. The AMFED integrates a moist-electric generator (MEG), an antibacterial hydrogel dressing, and concentric molybdenum (Mo) electrodes to provide a self-sustaining electrical supply and potent antibacterial activity against Staphylococcus aureus and Escherichia coli . The MEG harnesses chemical energy from moisture to produce a stable direct current of 0.61 V without external input, delivering this therapeutic electrical stimulation to the wound site through the Mo electrodes. The AMFED facilitates macrophage polarization toward reparative M2 phenotype and regulates inflammatory cytokines. Moreover, in vivo studies suggest that the AMFED group significantly enhances chronic wound healing, with an approximate 41% acceleration compared to the control group. Using a diabetic mouse wound model, the AMFED demonstrates its effectiveness in promoting nerve regulation, epithelial migration, and vasculogenesis. These findings present a novel and efficient platform for accelerating chronic wound healing.

Thiolated chitosan hydrogel combining nitric oxide and silver nanoparticles for the effective treatment of diabetic wound healing

Zeng-Jin Huang,Meng-Nan Ye,Xin-Hui Peng,Ping Gui,Fan Cheng,Guan-Hai Wang

Nitric oxide (NO) has shown significant potential in chronic wound healing due to its ability of promoting blood circulation. However, excessive NO can trigger local inflammatory response, potentially hindering wound healing. Therefore, controlled and sustained NO release to minimize pro-inflammation effects during treatment is in great demand for diabetic wounds. Herein, an injectable thiolated chitosan hydrogel loaded with NO donors (GNO) and silver nanoparticles (AgNPs) is presented for effective diabetic wound treatment, from which NO was released stably and sustainably responsive to reactive oxygen species (ROS) at the wound site. The combination of NO and AgNPs demonstrated robust antibacterial activity and biofilm dissipation. During diabetic wound treatments, the sustained release of NO promoted blood vessel regeneration while inhibiting inflammatory factors, thereby accelerating wound healing. This combined approach achieves efficient antibacterial action, biofilm prevention, inflammation suppression, vascular repair, improved local blood circulation, ultimately facilitating the reconstruction of epithelial structures at the wound site, thereby providing a promising solution for the diabetic chronic wound healing.

A novel “hot spring”-mimetic hydrogel with excellent angiogenic properties for chronic wound healing

Lili Sheng,Zhaowenbin Zhang,Yu Zhang,Endian Wang,Bing Ma,Qing Xu,Lingling Ma,Meng Zhang,Ge Pei,Jiang Chang

The treatment of chronic wounds is a major challenge in regenerative medicine , and angiogenesis is known to be critical for chronic wound healing. Hot springs with temperature in the range of 30–45 °C can promote blood circulation, and some hot spring elements including iron and silicon are also known to be active in promoting angiogenesis. Inspired by the hot spring function, we designed a novel bioactive photothermal hydrogel with “hot spring effect” based on fayalite (FA) and N, O-carboxymethyl chitosan (NOCS), which releases bioactive ions and has heating function to create hot ion environment in wound area. The hot spring-mimetic hydrogel showed significant enhancement of angiogenesis and chronic wound healing in vivo due to the in situ heating through photothermal effect combined with the bioactive ions (Fe 2+ and SiO 4 4− ) released from the hydrogel. It is further confirmed that the synergetic effect of the mild heating and bioactive ions on angiogenesis was mainly because of the activation of different angiogenic factors and signaling pathways . Our study suggests that the hot spring-mimetic approach may be an effective strategy to design bioactive materials for promoting angiogenesis and tissue regeneration .

A Natural Polyphenol-Regulated Microporous Soybean Protein Injectable Hydrogel Loaded with Placental Stem Cell-Derived Exosomes for Promoting Chronic Wound Healing

Hui Liu,Wen Jiang,Lu Han,Xiong Lu

Diabetic chronic wounds exhibit difficulties in healing due to prolonged high glucose and inflammatory conditions. Suppression of inflammation and restoration of the epithelial layer are crucial for preventing infections, reducing moisture loss, and restoring structural and functional integrity to the wound. Currently, there are numerous studies on dressings for diabetic chronic wounds, most of which can inhibit inflammation, but the effect on epithelial regeneration is limited. In this study, we developed a novel therapeutic system utilizing a microporous hydrogel regulated by the natural tea polyphenol EGCG for the treatment of chronic wounds. This hydrogel, made from soy protein isolate and dextran, exhibits anti-inflammatory properties and excellent biocompatibility, and is designed to be both injectable and biodegradable. This hydrogel facilitated the efficient loading and release of human placental stem cell-derived exosomes. Then the exosome-loaded hydrogel was used for full-thickness diabetic skin wound healing. Histological characterization confirmed that the exosome-loaded hydrogel promoted wound healing and epidermal regeneration. Immunofluorescence staining and Western blot experiments demonstrated the anti-inflammatory effect of the hydrogel. In short, this study proposed an Exo-hydrogel therapy as a promising potential for effective treatment of chronic wounds.

Microenvironment Self-adaptive Multifunctional Hydrogels Elicit Immunomodulation Mediated Wound Healing

Jintao Fang,Yicheng Wang,Ping Ling,Dian Yu,Junwen Hu,Jinwei Wang,Qin Xia,Weihan Zhu,Wenjun Xu,Jintao Shi,Xijin Wu,YuJia Li,Xiaxin Zhu,Huiyin Fu,Qiyang Shou,Wei Zhang,Shengyu Li

Herein, we report a microenvironment self-adaptive multifunctional hydrogel (GBOD-PF) dressing with intrinsic hemostasis, antimicrobial and anti-inflammatory properties that promotes collagen deposition, activates the immune response, and angiogenesis, accelerates large-scale chronic wound healing. To address these needs, aldehyde-functionalized dextran (ODT) was first synthesized through oxidation, and an injectable hydrogel was prepared by mixing Gel and ODT solutions in the presence of borax and paeoniflorin (PF) through the formation of dynamic Schiff base bonds (GBOD-PF). This multifunctional hydrogel exhibits remodeling and self-healing properties, enhanced adhesion strength and biocompatibility. Moreover, it possesses broad-spectrum antibacterial activity and superior hemostasis, providing early-stage protection for complex wounds. Notably, GBOD-PF targets the TLR4/NF-κB signaling pathway, a core mediator of inflammation, thereby dampening the inflammatory cascade. The results demonstrate the application of the GBOD-PF hydrogel in an infection wound and diabetic model wound healing model, which enhanced immune response, induced M1-to-M2 macrophage repolarization to establish anti-inflammatory microenvironment, regulated MMP-9, promoted angiogenesis, thereby inducing a pro-regenerative response. Therefore, this work provides an effective strategy for the treatment of chronic wound repair and tissue regeneration.

An NIR-responsive hydrogel loaded with polydeoxyribonucleotide nano-vectors for enhanced chronic wound healing

Yanzhen Sun,Yao Li,Xiaokang Ding,Pan Xu,Xiaodong Jing,Hailin Cong,Hao Hu,Bing Yu,Fu-Jian Xu

Chronic diabetic wounds are difficult to treat due to imbalanced inflammatory responses, high blood glucose levels, and bacterial infections. Novel therapeutic approaches based on nucleic acid analogues have been proposed, with unique advantages in improving angiogenesis, increasing collagen synthesis, and exerting anti-inflammatory effects. However, the inherent electronegativity of nucleic acids makes them less susceptible to cellular uptake. In this paper, a kind of near infrared (NIR)-responsive nanocomposite hydrogel loaded with nucleic acid vectors was proposed for promoting wound healing. The redox system composed of molybdenum disulphide nanosheets (MoS 2 NSs) initiated the copolymerization of quaternized chitosan containing double bonds and N -isopropylacrylamide (NIPAAm) to form the matrix. In addition, MoS 2 NSs with photothermal conversion performance endow the nanocomposite hydrogel to have NIR-response property and act as physical crosslinking points in the matrix. Polydeoxyribonucleotides (PDRN), which have the effect of promoting wound healing, were made into nucleic acid vectors, and loaded into the NIR-responsive hydrogel. MoS 2 NSs can convert NIR irradiation into heat, causing phase transitions of temperature-sensitive segments that trigger volume contraction of the hydrogel to extrude the nucleic acid vector. Promoting angiogenesis, slowing inflammation, and guiding tissue regeneration were demonstrated in the diabetic wound model treated with the NIR-responsive nanocomposite hydrogel.

Multiple Stimuli-Responsive MXene-Based Hydrogel as Intelligent Drug Delivery Carriers for Deep Chronic Wound Healing

Xin Yang,Changqing Zhang,Dawei Deng,Yueqing Gu,Huan Wang,Qifeng Zhong

Chronic wound healing is an important and basic issue in medical and healthcare fields. Recently, stimuli-responsive hydrogel systems have emerged as promising drug delivery carriers for wound management. However, given to the limited therapeutic outcomes, new hydrogel systems for efficient wound treatment are urgently needed. Here, the development of a 2D MXene-based hydrogel system for highly efficient photo- and magnetic-responsive drug delivery oriented to deep chronic wounds repair is presented. The intelligent responsive MXene-based hydrogel drug delivery system is composed of MXene-wrapped magnetic colloids and poly( N -isopropyl acrylamide)-alginate dual-network hydrogels. It is demonstrated that the MXene-based hydrogel system exhibits multiple response capability and controllable drug delivery ability, which can reduce the toxic side effects of drugs and promote the wound healing process as well. Notably, the practical performance of the MXene-based hydrogel drug delivery system is demonstrated by applying it to the treatment of the full-thickness cutaneous wound and subcutaneous infected wound of the rat model, which indicates the great prospect in clinical wound healing and other related biomedical fields.
