Multi-functional bioactive hydrogel based on CCM-Co-ZIF-8 nanoparticles for chronic wound healing: Synergistic antibacterial, antioxidant, and anti-inflammatory therapy

Qi Tao,Hang Yao,Zehao Gu,Xiaofei Yang,Junli Shi,Dong-An Wang,Hui Chong

The treatment of chronic wounds are characterized by a local tissue with issues such as the accumulation of free radicals and persistent inflammation, which contribute to delayed wound healing. Thus, it has emerged as a significant global challenge, profoundly impacting patients’ quality of life and even posing threats to their lives. In this study, we fabricted a CCM (Curcumin)–Co-ZIF-8 nanoparticle (CCZ)@oxidized ammonium alginate (OSA)/carboxymethyl chitosan (CMCS)/tannic acid (TA) composite multifunctional bioactive hydrogel. This nanocomposited double-crosslinked network hydrogel dressing exhibits excellent mechanical properties and self-healing abilities. Additionally, the hydrogel shows good biocompatibility. Due to the formation of dynamic Schiff base bonds between oxidized sodium alginate and CMCS, the hydrogel-TA spray system possesses injectability upon applied to a wound model in mice, allowing better adaptation to irregular wound shapes. Furthermore, it could promote synergistic antibacterial, antioxidant, and anti-inflammatory effects, thereby accelerating wound healing. Our findings highlight the potential of this multifunctional hydrogel system as an advanced therapeutic option for chronic wound management.

Ceria Centre to Enhance Multi-Active and Photothermal Response Janus Patch for Chronic Wound Senescence Inhibition and Repair

Yilin Ding,Zhipeng Sun,Zilin Wu,Yulin Zhang,Jiuzhi Ma,Huitong Luo,Xiaodong Cao

Bacterial infections and complex microenvironments induce cellular oxidative stress and cellular senescence severely affect chronic wound healing. However, the antibacterial effect of most dressings is single, which is only against the wound-breeding bacteria but cannot resist further invasion of bacteria in the environment into the wound. Few dressings can simultaneously fulfill antibacterial, antifouling, oxidative stress regulation, and cellular protection. Therefore, a Janus patch (SBMA/OHA/GelMA/TA-CeO 2 @HA (HGT-C@HA)) is prepared, in which the top superhydrophilic SBMA zwitterionic layer resists bacterial adhesion, and CeO 2 coated with hyaluronic acid (HA) is loaded into the bottom gel as the functional center. After immersion in tannic acid (TA), Ce-TA chelate formation in situ confers photothermal antimicrobial properties to the hydrogel. It is further validated that HA-coated CeO 2 enable the dressing to obtain higher antioxidant properties by regulating the proportion of Ce 3+ on the nanoparticle surface, which protects cells from the high glucose and high H 2 O 2 environment and effectively inhibits intracellular DNA damage to mitigate cellular senescence. In vivo experiments illustrate that the material significantly accelerated wound healing and improved the quality of wound healing, demonstrating the significant potential of this dressing in diabetic wound healing.

In-situ doping of a conductive hydrogel with low protein absorption and bacterial adhesion for electrical stimulation of chronic wounds

Yuhui Lu,Yanan Wang,Jieyu Zhang,Xuefeng Hu,Zeyu Yang,Yi Guo,Yunbing Wang

Electrical stimulation (ES) via electrodes is promising for treating chronic wounds, but this electrode-based strategy is unable to stimulate the whole wound area and the therapeutic outcome may be compromised. In this study, a conductive poly(2-hydroxyethyl methacrylate) (polyHEMA)/polypyrrole (PPY) hydrogel was developed, and 3-sulfopropyl methacrylate was covalently incorporated in the hydrogel’s network to in-situ dope the PPY and maintain the hydrogel’s conductivity in the weak alkaline physiological environment. The obtained hydrogel was superior to the commercial Hydrosorb® dressing for preventing bacterial adhesion and protein absorption, and this is helpful to reduce the possibilities of infection and secondary damage during dressing replacement. The in vitro scratch assay demonstrates that ES through the hydrogel enhanced fibroblast migration, and this enhancement effect remained even after the ES was ended. The in vivo assay using diabetic rats shows that when ES was conducted with this polyHEMA/PPY hydrogel, the healing rate was faster than that achieved by the electrode-based ES strategy. Therefore, this polyHEMA/PPY hydrogel shows a great potential for developing the next generation of ES treatment for chronic wounds. Statement of Significance Electrical stimulation (ES) via separated electrodes is promising for treating chronic wounds, but this electrode-based strategy is unable to stimulate the whole wound area, compromising the therapeutic outcome. Herein, a hydrogel was developed with stable electrical conductivity in the physiological environment and strong resistance to protein absorption and bacterial adhesion. The in vitro and in vivo tests proved that ES applied through the flexible and conductive hydrogel that covered the wound was superior to ES through electrodes for promoting the healing of the chronic wound. This hydrogel-based ES strategy combines the advantages of ES and hydrogel dressing and will pave the way for the next generation of ES treatment for chronic wounds.

Electrospun robust, biodegradable, bioactive, and nanostructured sutures to accelerate the chronic wound healing

Yiran Li,Hongxing Xu,Wenwen Zhao,Li Zhang,Shaohua Wu

The design and development of advanced surgical sutures with appropriate structure and abundant bio-functions are urgently required for the chronic wound closure and treatment. In this study, an integrated technique routine combining modified electrospinning with hot stretching process was proposed and implemented to fabricate poly(L-lactic acid) (PLLA) nanofiber sutures, and the Salvia miltiorrhiza Bunge-Radix Puerariae herbal compound (SRHC) was encapsulated into PLLA nanofibers during the electrospinning process to enrich the biofunction of as-generated sutures. All the PLLA sutures loading without or with SRHC were found to exhibit bead-free and highly-aligned nanofiber structure. The addition of SRHC was found to have no significant influences on the fiber morphology, diameter, and the crystallinity of as-prepared PLLA sutures. Importantly, all the SRHC-contained PLLA nanofiber sutures possessed excellent tensile and knot strength, which were of significant importance for the surgical suture applications. Besides, the antioxidant and anti-inflammatory properties of these sutures obviously enhanced with the increasing of SRHC concentration. Furthermore, the in vitro cell tests illustrated that the high fiber orientation of the sutures was able to efficiently induce the human dermal fibroblasts (HDFs) to migrate in a rapid manner, and the sutures loaded with high content of SRHC could significantly promote the attachment and proliferation of HDFs in comparison. The in vivo diabetic mouse model experiments revealed that all the as-developed PLLA sutures could effectively close the wound, but the PLLA sutures containing high content of SRHC could dramatically promote the wound healing with high quality by shortening the healing time, improving the collagen deposition, neovascularization, and the regeneration of hair follicles, especially compared with commercial polyester (PET) suture. This study offers a simple and easily-handling strategy to develop robust, biodegradable, bioactive, and nanostructured PLLA sutures, which shows huge potential for the treatment of hard-to-heal diabetic wounds.

Accelerating chronic wound healing by breaking the pH limitations of biomolecule-loaded Nanozymes and remodeling the microenvironment in Cascade reactions

Chuanwei Shi,Peng Liu,Yeping Liu,Zhexu Gai,Fei Yang,Yanzhao Yang

The microenvironment around the wounds of chronic disease patients often exhibits weakly alkaline conditions, primarily due to defects in the patients' blood glucose regulation, tissue hypoxia, accumulation of metabolic products, and bacterial infection and growth. To regulate the wound microenvironment across a broad pH range, a Fe-doped ZIF-8 nanozyme loaded with glucose oxidase (Fe-ZIF-8@GOx) was synthesized via a one-step in situ growth method. This nanozyme, with ZIF-8 serving both as a protective matrix and immobilization scaffold, enhances the enzyme's operational microenvironment and maintains functionality across a wide pH range. It exhibits the activities of cascade glucose oxidase (GOx), catalase-like (CAT-like), and peroxidase-like (POD-like) enzymes. When Fe-ZIF-8@GOx nanozymes encounter high glucose levels at the site of weakly alkaline chronic ulcers, the GOx particles catalyze the reduction of local pH, simultaneously triggering CAT-like enzyme activity to release O₂ and improve the wound microenvironment. The subsequent pH reduction induces the controlled release of iron ions, enabling Fe 2+ to react with endogenously produced hydrogen peroxide to generate reactive oxygen species (ROS), conferring a distinctive guest–carrier synergistic antibacterial effect. Both in vitro and in vivo studies confirm that this material effectively reduces inflammation, modulates macrophage polarization, enhances collagen deposition, and promotes angiogenesis, ultimately accelerating wound healing under chronic diabetic conditions. Thus, our work not only overcomes microenvironmental limitations in antibacterial therapy but also offers a promising strategy for treating chronic diabetic infections.

An Electrostatic-Induction-Enabled Anti-Touching Hydrogel Dressing for Chronic Wound Care

Ren Yan,Zhaoqing Sun,Wanyu Du,Nian Liu,Qizeng Sun,Peng Li,Fei Liang,Yangzhi Zhu,Wei Huang,Hai-Dong Yu

Chronic wounds are inherently vulnerable to external mechanical contact owing to their impaired physiological states. Accidental touching may lead to secondary damage to delicate wound tissues, including deterioration of tissue and inflammation, thereby prolonging the healing process. In addition, patients encounter nociceptive sensations, indicative of physical discomfort and potential distress. However, the traditional dressings for chronic wounds fail to convey adequate perceptual information to prevent mechanical trauma. An electrostatic-induction-enabled anti-touching hydrogel dressing (EAHD) is proposed, which promotes wound healing and reduces touching-induced injuries. The EAHD combines a proximity sensor and a pro-healing hydrogel. The proximity sensor can detect approaching objects, enabling remote perception. More importantly, EAHD and a wireless alarm system are integrated to warn against the detected potential hazards such as unintentional touching. Meanwhile, the EAHD significantly accelerates wound healing in preclinical diabetic mice, ≈38% faster than that of the control group. This study proposes a novel strategy for multifunctional wound dressing that promotes wound healing and prevents accidental touching-induced secondary damage to the wound.

Electrospun chitosan/PVA/bioglass Nanofibrous membrane with spatially designed structure for accelerating chronic wound healing

Qingchang Chen,Jun Wu,Yuan Liu,Yanqun Li,Chuqiu Zhang,Weichen Qi,Kelvin W.K. Yeung,Tak Man Wong,Xiaoli Zhao,Haobo Pan

Cutaneous wounds, especially chronic wounds, remain clinical challenges, and this is partially due to the complex healing process composed of four overlapping but distinct stages including hemostasis, inflammation, proliferation and remodeling. Therefore, wound dressings with spatially designed structures which can temporally regulate certain bioactive components to function at specific healing stages might be able to accelerate the healing process. In this study, nanobioglass incorporated chitosan-PVA (polyvinyl alcohol) trilayer nanofibrous membrane (nBG-TFM) was fabricated via sequential electrospinning. This membrane exhibited excellent biocompatibility, antibacterial activity and regeneration promotion effect. Furthermore, spatially designed structure optimized functions of each component and provided more suitable microenvironment as compared with uniform membrane. Rat full-thickness skin defects model and mice diabetic chronic wound model showed that nBG-TFM could achieve significantly accelerated and enhanced healing, in terms of complete re-epithelialization, improved collagen alignment and formation of skin appendages. It was revealed that nBG-TFM functioned through upregulating growth factors including VEGF and TGF-β. Meanwhile inflammatory cytokines such as TNF-α and IL-1β were downregulated. The technology presented in this study shed new light on designing functional wound dressings which can promote healing of chronic wounds.

Chronic wound management: a liquid diode-based smart bandage with ultrasensitive pH sensing ability

Wang Xueqi,Cheng Jing,Wang Han

Chronic wounds, which require prolonged healing periods, pose significant impacts on individuals with diabetes, vascular diseases, and high blood pressure. Simultaneous drainage and monitoring of wound exudate are vital for advanced wound management. However, recently reported smart dressings either lack integration of wound cleaning and monitoring functions or fail to achieve dynamic in situ monitoring of wound status, which hinders their ability to meet the demands of wound care. In this study, a smart bandage is introduced, which integrates a biocompatible liquid diode membrane with an ultrasensitive 3D polyaniline mesh (M-PANI)-based pH biosensor. The smart bandage allows for unidirectional drainage of wound exudate while dynamically sensing the wound pH environment. Specifically, the proposed smart bandage effectively cleans excessive wound exudate while providing real-time information on the wound status during the drainage process. The M-PANI-based pH biosensor demonstrates a high sensitivity of 61.5 mV/pH and a wide pH detection range from 4.0 to 10.0, encompassing the pH range of normal and infected wounds. Moreover, the sensing module exhibits excellent stability after 48 hours of dynamic testing and 28 days of storage, with only a 4.8% decline in the detected signal, and high repeatability with a device-to-device relative standard deviation (RSD) of 3.1%. To evaluate the practicality of this smart bandage, simulated skin and rats have been employed, and the results indicate the immense potential of this smart bandage for clinical applications. In conclusion, the present smart bandage demonstrates considerable promise for wound exudate cleaning and monitoring in advanced wound care and offers a promising method for home-based wound management.

Bioengineered Extracellular Vesicle Hydrogel Modulating Inflammatory Microenvironment for Wound Management

Yunfei Mu,Liwen Ma,Jia Yao,Dan Luo,Xianguang Ding

Chronic wounds, frequently arising from conditions like diabetes, trauma, or chronic inflammation, represent a significant medical challenge due to persistent inflammation, heightened infection risk, and limited treatment solutions. This study presents a novel bioengineered approach to promote tissue repair and improve the healing environment. We developed a bioactive hydrogel patch, encapsulated zeolitic imidazolate framework-8 (ZIF-8) into extracellular vesicles (EVs) derived from anti-inflammatory M2 macrophages, and synthesized ZIF@EV, then embedded it in the sodium alginate matrix. This hydrogel structure enables the controlled release of therapeutic agents directly into the wound site, where it stimulates endothelial cell proliferation and promotes new blood vessel formation. These processes are key components of effective tissue regeneration. Crucially, the EV-infused patch influences the immune response by polarizing macrophages towards an M2 phenotype, shifting the wound environment from inflammation toward regenerative healing. When applied in a murine model of chronic wounds, the EV hydrogel patch demonstrated notable improvements in healing speed, quality, and tissue integration compared to traditional approaches such as growth factor therapies and foam dressings. These promising findings suggest that this bioactive hydrogel patch could serve as a versatile, practical solution for chronic wound management, providing an adaptable platform that addresses both the biological and logistical needs of wound care in clinical settings.

Injectable Hydrogels with Integrated Ph Probes and Ultrasound-Responsive Microcapsules as Smart Wound Dressings for Visual Monitoring and On-Demand Treatment of Chronic Wounds

Da Huang,Jiahao Du,Fang Luo,Gang He,Minglang Zou,Yongming Wang,Zhenyu Lin,Decheng Wu,Zuquan Weng

Hydrogel dressings capable of infection monitoring and precise treatment administration show promise for advanced wound care. Existing methods involve embedd ingorganic dyes or flexible electronics into preformed hydrogels, which raise safety issues and adaptability challenges. In this study, an injectable hydrogel based smart wound dressing is developed by integrating food-derived anthocyanidin as a visual pH probe for infection monitoring and poly( L -lactic acid) microcapsules as ultrasound-responsive delivery systems for antibiotics into a poly(ethylene glycol) hydrogel. This straightforwardly prepared hydrogel dressing maintains its favorable properties for wound repair, including porous morphology and excellent biocompatibility. In vitro experiments demonstrated that the hydrogel enabled visual assessment of pH within the range of 5 ∼ 9.Meanwhile, the release of antibiotics could be triggered and controlled by ultrasound. In vivo evaluations using infected wounds and diabetic wounds revealed that the wound dressing effectively detected wound infection by monitoring pH levels and achieved antibacterial effects through ultrasound-triggered drug release. This led to significantly enhanced wound healing, as validated by histological analysis and the measurement of inflammatory cytokine levels. This injectable hydrogel-based smart wound dressing holds great potential for use in clinical settings to inform timely and precise clinical intervention and in community to improve wound care management.

Hyaluronic acid nanofiber mats loaded with antimicrobial peptide towards wound dressing applications

Qianwen Yang,Zhigang Xie,Junli Hu,Yichun Liu

Chronic wounds are highly susceptible to bacterial infections. Previously, we loaded a natural antimicrobial peptide of low cost and high safety, ε-polylysine (EPL), into the electrospun nanofiber mat of starch. The mat showed comparable antibacterial activity but markedly better biocompatibility than the commercial silver-containing dressing. To further optimize material property, in this paper, we use hyaluronic acid (HA) to replace starch. Results show that EPL-loaded HA nanofiber mats (OHA-EPL) have suitable water vapor permeability, good biocompatibility and broad-spectrum antibacterial property similar to that of EPL-loaded starch nanofiber mat (Starch-EPL). Differently, the content of EPL in OHA-EPL nanofiber mats increases from 19.2% to 27.9%, the tensile strength rises from 0.3 MPa to 0.6 MPa, the elongation grows from 62.0% to 130.0%, and the fiber degradation and EPL release accelerates. In addition, OHA-EPL can absorb up to 26.3-times exudate, which is much higher than Starch-EPL (15.1 times). Combined with the excellent biological activity of HA, OHA-EPL may produce better therapeutic effects than Starch-EPL.

A Multifunctional Hydrogel for Infected Chronic Wounds Management: Integrated pH Sensing, Photothermal Therapy, and Ion Release

Ziqi Zhou,Liyan Zhang,Muyan Qin,Xu Cui,Deping Wang

Chronic wound healing is often compromised by bacterial infection, and wound pH changes strongly correlate with infection. Therefore, the development of wound dressings capable of real-time pH monitoring, antimicrobial activity, and tissue repair is crucial for the effective management of infected chronic wounds. This study reports a novel multifunctional composite hydrogel (GM-C-mBG@P) synthesized by incorporating microwave-synthesized carbon quantum dots (CQDs) and polydopamine-modified mesoporous bioactive glass nanoparticles (mBG@P) into modified gelatin methacryloyl (GelMA). The incorporated polydopamine (PDA) confers excellent photothermal properties to the hydrogel, enabling effective antimicrobial activity. Under 360 nm UV excitation, the fluorescence intensity of CQDs in GM-C-mBG@P exhibits a strong linear correlation with pH, enabling real-time wound pH monitoring. Furthermore, mBG@P can gradually dissolve calcium, silicon, boron, and other functional ions to enhance the bioactivity and functionality of the hydrogel and accelerate wound healing. These findings suggest that the GM-C-mBG@P hydrogel has the potential to serve as a promising wound dressing for the treatment of infected chronic wounds.
