Molecular characteristics of kappa-selenocarrageenan and application in green synthesis of silver nanoparticles

Weiping Jin,Yiran Yu,Wenfu Hou,Guozhen Wang,Zhenzhou Zhu,Jingren He,Shuiyuan Cheng,Qingrong Huang

Selenium is an essential trace element in human body, and kappa-selenocarrageenan (Se-car) is an organic source of selenium supplement. To further utilize Se-car in food packaging, biotherapy or biosensor, the molecular information of Se-car was characterized here and multi-functional Ag NPs synthesized by Se-car were fabricated. Results of GPC-MALLS, FTIR , potentiometric titration, and intrinsic viscosity showed that Se-car was polymerized by nearly 22 basic units of disaccharide . Sixty-four percentage of sulfated groups (SO 4 2− ) in carrageenan was replaced by selenium acid (SeO 3 2− ), which belonged to weak acid resulting from a gradually decrease of ζ-potential with acidity process to pH 1.0. Besides, the capacity of biosynthesis silver nanoparticles (Ag NPs) by Se-car was studied and it made a comparison with κ-carrageenan. Results exhibited that Se-car could serve as an efficient reducing and capping agent for Ag NPs fabrication (remarked as Se-car@Ag). The k app of Se-car@Ag NPs for catalyzing 4-NP degradation was 2.14 × 10 −2  s −1 . Antibacterial test revealed Se-car@Ag had an ability to inhibit the growth of Escherichia coli and Staphylococcus aureus . To combine the selenium health benefit and functional metal nanoparticles, Se-car@Ag might have potential applications in multiple areas like medicine, disease diagnostic, and drug delivery.

Modification of Silicone Hydrogel Contact Lenses for the Selective Adsorption of Ofloxacin

Wenke Xiao,Xiaojuan Zhang,Lingyun Hao,Yuan Zhao,Qing Lin,Zhao Wang,Wenli Song,Dong Liang

It is essential to design multifunctional contact lenses for selective recognition and utilization of the eye drop ofloxacin (OFL). Modified silicone hydrogel (MSL) contact lenses were firstly synthesized via ultraviolet curing by using (hydroxymethyl)methyl aminomethane, polydimethylsiloxane, methacryloxypropyl terminated polydimethylsiloxane, and N,N-dimethylacrylamide as monomers and N-vinyl-2-pyrrolidinone (NVP) as a modifier. The synthesized MSL contact lenses were characterized via Fourier transform infrared spectroscopy, scanning electron microscopy, and contact angle and mechanical property tests. The results showed that compared with silicone hydrogel (SL) contact lenses, the MSL contact lenses had a more porous structure and good swelling and mechanical properties. The molecularly imprinted polymer/ofloxacin-modified silicone hydrogel (MIP/OFL-MSL) contact lenses were firstly synthesized via free radical polymerization, using ofloxacin (OFL) as the template molecule. Moreover, compared with chlorogenic acid and myricetin molecule, the MIP/OFL-MSL contact lenses exhibited an adsorption capacity for OFL that reached 2.86 mg/g. The antibacterial test showed that the MIP/OFL-MSL contact lenses displayed an inhibition ring size of 15.2 mm and 17.8 mm for Staphylococcus aureus and Escherichia coli, respectively.

Fabrication of enzyme-responsive composite coating for the design of antibacterial surface

Liu Peng,Hao Yansha,Ding Yao,Yuan Zhang,Liu Yisi,Cai Kaiyong

In this study, a type of bacteria enzyme-triggered antibacterial surface with a controlled release of Ag ions was developed. Firstly, chitosan-silver nanocomposites (Chi@Ag NPs) were in situ synthesized via using ascorbic acid as reducing agent. Chi@Ag NPs were characterized by transmission electron microscopy, ultraviolet–visible spectroscopy, X-ray diffraction and X-ray photoelectron spectroscopy. Subsequently, Chi@Ag NPs and hyaluronic acid (HA) were used to fabricate antibacterial composite coating via Layer-by-Layer (LBL) self-assembly method. The successful construction of Chi@Ag NPs/HA composite coating was confirmed by scanning electron microscopy, energy dispersive spectroscopy and contact angle measurements, respectively. Then, the amount of released Ag ion was analyzed by inductively coupled plasma atomic emission spectrometry, which demonstrated that the release of Ag ions from the surface could be triggered by enzyme (e.g. hyaluronidase). A series of antibacterial tests in vitro, including zone of inhibition test, bacterial viability assay, antibacterial rate measurement and bacteria adhesion observation, demonstrated that the enzyme-responsive surface could inhibit the growth of bacteria. On the whole, this study provides an alternative approach for the fabrication of antibacterial surfaces on synthetic materials in various fields with the minimal side effects on surrounding environment and human body.

Facile preparation and good performance of nano-Ag/metallocene polyethylene antibacterial coatings

Li Lin,Wang Yunli,Zhu Yingchun

Metallocene polyethylene/nano-silver coatings were prepared by a facile air-spray method on polymer films. Different from the prevailing strategy to endow polyethylene with antibacterial performance, we used metallocene polyethylene sol and nano-silver as a precursor to deposit coatings on polymers at a relatively low operating temperature. Antibacterial coatings with excellent mechanical properties, water resistance, and low silver release were achieved. The composite coatings were examined in terms of surface characteristics, mechanical properties, and antibacterial activity against two representative bacterial strains including Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ). The composite coatings exhibited favorable microstructure, good mechanical properties, and suitable crystallinity. The antimicrobial tests indicated that the fabricated composite coatings showed promising antibacterial activity against E. coli and S. aureus . Furthermore, Ag ions released by the composite coating after 30 days were under 1.2 ppb. These results indicated a promising prospect of the composite coating for wide antibacterial applications.

Preparation of novel stable microbicidal hydrogel films as potential wound dressing

Sheng Huang,Hua-Jiao Chen,Yan-Ping Deng,Xiu-hua You,Quan-hui Fang,Mei Lin

This work aimed to prepare a hydrogel film with antibacterial activity and excellent stability in enzymes as potential wound dressing. Using hyaluronic acid, carboxylated chitosan and gentamicin, antibacterial film HA-CC-GS was prepared through intermolecular covalent bonding. Surface morphology of film was evaluated by scanning electron microscope images. The film exhibited good water absorption capacity and appropriate water vapor permeation rate (WVPR), outstanding mechanical properties and remarkable resistance to enzymatic hydrolysis. The biocompatibility of the film was tested by CCK-8 assay, and the results showed that the viability of NIH3T3 cell was not affected by coculture with hydrogel film. In vitro antibacterial test indicated that the synthesized hydrogel film had obvious antibacterial properties. In addition, in vivo studies showed that antibacterial hydrogel film could protect wound from infection and promote wound healing. These results indicated that the prepared antimicrobial hydrogel films by cross-linking had great potential application in skin/tissue repair, suggesting as a hopeful wound dressing.

Biomimetic SIS-based biocomposites with improved biodegradability, antibacterial activity and angiogenesis for abdominal wall repair

Guangxiu Cao,Cunyang Wang,Yubo Fan,Xiaoming Li

Small intestinal submucosa (SIS) is a widely concerned acellular material for reconstructing tissue defects, but during the restoration of abdominal wall, it has been restricted due to the fast degradation causing poor long-term mechanical properties, the infection caused by bacteria contamination, and insufficient neovascularization post-operation. In this study, we developed a biomimetic SIS-based biocomposite (CS/ES-SIS) for abdominal wall repair, in which chitosan (CS) and elastin (ES) electrospun nanofibers were used to improve the biodegradability, antibacterial activity, and angiogenesis. The CS/ES-SIS composites were examined through a series of testing experiments, especially in vitro degradation was assessed by a constant deformation loading device and the micromechanical properties during enzymatic degradation under biomechanical environment were measured by nanoindentation. In vitro antibacterial test and cytocompatibility, and in vivo biocompatibility, neovascularisation and tissue regeneration were also investigated. The main research results as follows: (1) After 7 days enzymatic degradation under biomechanical environment, the degradation rate of CS/ES-SIS composites was slower than that of SIS by about 24.5%. Moreover, the CS/ES-SIS composites could better maintain the stability of microstructure and micromechanical properties compared with SIS. (2) The antibacterial rates of CS/ES-SIS composites against E. coli and S. aureus were respectively 98.87% and 98.26% while the SIS demonstrated no obvious antibacterial capacity. (3) The CS/ES-SIS composites supported the viability and proliferation of fibroblast cell L929. In vivo studies showed that the CS/ES-SIS composites could promote tissue regeneration upon implantation without serious inflammatory reaction. Additionally, the vascular number in the CS/ES-SIS composites was as 1.69 times as that in the SIS at 4 weeks. Collectively, all the findings suggested that the newly developed CS/ES-SIS composites might be promising and attractive candidates for applications of abdominal wall repair.
