Solution-blow spinning halloysite nanotube loaded chitosan-polyethylene oxide fibrous membrane as hemostatic dressings

Jinmiao Chen,Yuxi Mo,Xiangyan Zhang,Shaohua Zhang,Aihua Cui,Xueqin Wang,Jiwei Li

In this study, chitosan/polyethylene oxide fibrous membranes (CS/PEO) with varying halloysite nanotubes (HNTs) contents were prepared by solution-blow spinning. The HNTs-CS/PEO membranes with a 12 wt% HNTs content exhibited excellent hydrophilicity, air permeability, and flexibility. The in vitro hemostasis time of the 12 % HNTs-CS/PEO was 217 s, and it was observed to form a stable thrombus. Moreover, the antibacterial efficacy of 12 % HNTs-CS/PEO against Escherichia coli and Staphylococcus aureus was 90.05 % and 69.86 %, respectively. Furthermore, the in vitro cytotoxicity study demonstrated the nontoxic nature of the 12 % HNTs-CS/PEO. The solution-blow spinning HNTs-CS/PEO fibrous membrane can potentially prevent the risk of distal thrombosis caused by separating individual HNTs and offer the advantages of a simple preparation and application process. This study presents a novel first-aid hemostatic dressing and offers a novel approach to loading inorganic nanoparticles onto fibers.

Process optimization and character evaluation of Bletilla striata polysaccharide (BSP) and chitosan (CS) composite hemostatic sponge (BSP–CS)

Zhang Yeshan,Han Xue,Zhao Jun,Gan Menglan,Chen Yaya,Zhang Jinxia,He Yu,Wu Mingkai,Liu Hai

Bletilla striata polysaccharide (BSP) and chitosan (CS) were chemically cross-linked using oxalyl chloride to prepare a composite hemostatic sponge (BSP-CS), and the process parameters were optimized using the Box–Behnken design (BBD) with response surface methodology. To optimize the performance of the hemostatic sponge, we adjusted the ratio of independent variables, the amount of oxalyl chloride added, and the freeze-dried volume. A series of evaluations were conducted on the hemostatic applicability of BSP-CS. The characterization results revealed that BSP-CS had a stable bacteriostatic effect on Escherichia coli , Staphylococcus aureus , and Pseudomonas aeruginosa within 72 h, and the bacteriostatic rate was above 30%. The CCK-8 cytotoxicity test demonstrated that BSP–CS had a certain effect on promoting cell proliferation of L929 cells. In the mouse tail-cutting experiment, the hemostasis time of BSP–CS was 463.0 ± 38.16  s, shortened by 91.3 s on average compared with 554.3 ± 34.67  s of the gauze group. The blood loss of the BSP–CS group was 28.47 ± 3.74  mg, which was 34.7% lower than that of the control gauze group ( 43.6 ± 3.83  mg). In the in vitro coagulation experiment, the in vitro coagulation index of the BSP–CS group was 97.29 % ± 1.8 % , which was reduced to 8.6% of the control group. The CT value of the BSP–CS group was 240 ± 15  s, which was 155 s lower than that of the gauze group ( 355 ± 31.22  s). All characterization results indicate that BSP–CS is an excellent hemostatic material.

Kaolin-loaded carboxymethyl chitosan/sodium alginate composite sponges for rapid hemostasis

Yannan Song,Shuo Li,Huifang Chen,Xinyi Han,Gregory J. Duns,Wubliker Dessie,Wufei Tang,Yimin Tan,Zuodong Qin,Xiaofang Luo

There are several factors that contribute to the mortality of people who suffer from unmanageable bleeding. Therefore, the development of rapid hemostatic materials is necessary. Herein, novel rapid hemostatic composite sponges were developed by incorporation of kaolin (K) into carboxymethyl chitosan (CMCS)/sodium alginate (SA) via a combination of methods that includes ionic crosslinking, polyelectrolyte action, and freeze-drying. The CMCS/SA-K composite sponges were cross-linked with calcium ions provided by a sustained-release system consisting of D-gluconolactone (GDL) and Ca-EDTA, and the hemostatic ability of the sponges was enhanced by loading the inorganic hemostatic agent-kaolin (K). It was demonstrated that the CMCS/SA-K composite sponges had a good porous structure and water absorption properties, excellent mechanical properties, outstanding biodegradability , and biocompatibility. Simultaneously, they exhibited rapid hemostatic properties, both in vitro and in vivo . Significantly, the hemostatic time of the CMCS/SA-K 60 sponge was improved by 82.76 %, 191.82 %, and 153.05 %, compared with those of commercially available gelatin sponges in the rat tail amputation, femoral vein, and liver injury hemorrhage models respectively, indicating that its hemostatic ability was superior to that of commercially available hemostatic materials. Therefore, CMCS/SA-K composite sponges show great promise for rapid hemostasis .

Injectable hemostatic hydrogel adhesive with antioxidant, antibacterial and procoagulant properties for hemorrhage wound management

Jiahao Yang,Tianjiao Wang,Liang Zhang,Peng Fan,Jiulong Zhao,Xiaoyi Zheng,Yongkang Lai,Hongrui Liu,Shige Wang

In emergencies, uncontrolled severe bleeding can result in undesired complications and even death of the injured. Designing advanced hemostatic agents is a potential solution for emergency hemostasis, yet it remains challenging to realize the persistent adhesion in a wet wound environment. In this study, based on dynamic reversible Schiff base bond and photo-initiated double-bond polymerization, a novel injectable hemostatic hydrogel ( L -COC) consisting of methacrylated carboxymethyl chitosan (CMCSMA), oxidized konjac glucomannan (OKGM) and (+)-catechin hydrate (CH) was synthesized for emergency hemostasis. To our delight, the incorporated CH imparted enhanced blood procoagulantion to the L -COC hydrogel by intensifying the hydrogel-red blood cell interactions. As a result, the hemostatic effect of the engineered L -COC hydrogel was significantly superior to that of fluid gelatin Surgiflo TM for liver bleeding wounds in rats (Blood loss: 0.62 ± 0.11 g ( L -COC), 0.90 ± 0.08 g (Surgiflo TM ); hemostasis time: 69.0 ± 2.9 s ( L -COC), 84.0 ± 2.2 s (Surgiflo TM )). With the favorable antioxidant and antibacterial activities, as well as multifunctional properties, the bio-adhesive L -COC hydrogel and the underlying design principles may facilitate further development of practical hemostatic hydrogels.
