Multidimensional exploration of essential oils generated via eight oregano cultivars: Compositions, chemodiversities, and antibacterial capacities

Yuanpeng Hao,Jiamu Kang,Rui Yang,Hui Li,Hongxia Cui,Hongtong Bai,Andrey Tsitsilin,Jingyi Li,Lei Shi

Numerous species of Origanum (Lamiaceae) have been widely used as spices to extend the shelf life of foods. Essential oils extracted from this genus have attracted much attention owing to their potential applications as bactericides. Here, we evaluated the chemical compositions of eight oregano essential oils (OEOs) using gas chromatography-mass spectrometry and assessed their antibacterial activities. The chemical compositions of OEOs were affected by the cultivar factor, and seven common compounds, including carvacrol, were identified among eight OEOs. Partial least squares discriminant analysis enabled the distinction of three groups among these OEOs, as characterized by the proportions of carvacrol, thymol , and sesquiterpenes. OEOs effectively inhibited Escherichia coli and Staphylococcus aureus with varying antibacterial activities. Spearman correlation network highlighted core antibacterial contributors in the chemical profiles of OEOs. Our results revealed that the bacteriostatic effects of OEOs could be explained by core compounds and their synergistic effects.

Cheng Zhang,Weihao Li,Ligong Chen,Zhaoliang Chen,Xuejing Wang,Qianqian Xu,Hailong Zhang,Huan Chen,Juxiang Liu

Simple SummaryDeveloping safe and effective antibiotic growth promoters (AGPs) substitutes is particularly important to improve animal health and production performance. As an essential plant oil, the oregano oil's main bioactive substance is carvacrol, which has been proven to have antioxidant, anti-inflammatory, antibacterial, and antiviral properties. The sanguinarine from macleaya cordata is the primary bioactive substance. Sanguinarine has anti-tumor, immune-enhancing, antibacterial, and anti-inflammatory effects. However, it has not been reported whether the compatibility of oregano oil and macleaya cordata extract could produce better results. This study is the first to report the effect of oregano oil combined with macleaya cordata oral solution on the growth of broilers. The oregano oil combined with macleaya cordata oral solution significantly improved the growth performance of broilers. At the same time, serum biochemical indices, serum antioxidant indices, serum immune indices, serum cytokines, and intestinal morphology were significantly improved. In summary, our results demonstrated that the mixed solution of oregano oil and macleaya cordata has substantial potential to be an alternative to AGPs for broilers to reduce costs and improve benefits. This study provides basic data and technical support for further research.AbstractThe abuse of AGPs in animal husbandry has led to severe problems such as drug resistance and ecological, and environmental destruction, which seriously threaten human health and public health security. In recent years, extracts of oregano oil and macleaya cordata have become a hot spot in the research and application of AGP substitutes for their safety and high efficiency. This study is the first to report the effect of oregano oil combined with macleaya cordata oral solution on broiler growth performance. A total of 960 one-day-old broiler chickens were randomly divided into four treatment groups (240 chickens per group). Each treatment group was divided into six replicate groups (40 birds per replicate group). There were four groups in this study: the solvent control group, the oregano essential oil combined with macleaya cordata extract oral solution group (OS group), the oregano essential oil oral solution group (OEO group), and the macleaya cordata extract oral solution group (MCE group). Two chickens from each replicate group were collected and mixed into a composite sample. Six composite samples were obtained for each treatment group. The results showed that the oregano oil combined with macleaya cordata oral solution significantly improved the growth performance of broiler chickens. At the same time, serum biochemical indices, serum antioxidant indices, serum immune indices, serum cytokines, and intestinal morphology were significantly improved by the OS group. This study shows that oregano oil combined with macleaya cordata oral solution has substantial potential to be an alternative to AGPs for broilers. Keywords:oregano oil;macleaya cordata;oral solution;broilers;growth performance

Preparation and characterization of β-cyclodextrin–oregano essential oil microcapsule and its effect on storage behavior of purple yam

Haohe Huang,Chongxing Huang,Cheng Yin,Muhammad RU Khan,Hui Zhao,Yangfan Xu,Lijie Huang,Dantong Zheng,Minghui Qi

BACKGROUND Natural plant essential oils have antimicrobial properties; however, essential oils are difficult to maintain in a system because of their volatile nature. First, we prepared microcapsules from β -cyclodextrin and oregano essential oil and characterized their properties. Second, the effect of microcapsules on the preservation of freshly cut purple yam was studied using an edible coating technique. Purple yams immersed in distilled water were used as control, and their characteristics were compared with yams coated with citric acid, citric acid + sodium alginate, and citric acid + sodium alginate + β -cyclodextrin–oregano essential oil microcapsules (CA–SA–MC) and stored at 4 °C for 5 days. RESULTS Microcapsules of oregano essential oil and β -cyclodextrin solution were successfully prepared via the inclusion method, with an optimal encapsulation efficiency of 55.14%. Scanning electron microscopy, Fourier transform infrared spectroscopy, and thermogravimetric analysis showed strong bonds between β -cyclodextrin and oregano essential oil. All edible coatings, particularly CA–SA–MC, significantly ( P ≤ 0.05) maintained firmness, total soluble solids, ascorbic acid content, and anthocyanin content compared with control treatment. This treatment also prevented browning and extended the shelf life of purple yam. CONCLUSION Oregano essential oil can be successfully encapsulated into cyclodextrin microcapsules. It has a great impact on the shelf life extension of purple yam and could be successfully applied to other fresh produce. © 2020 Society of Chemical Industry

Creation of New Oregano Genotypes with Different Terpene Chemotypes via Inter- and Intraspecific Hybridization

Meiyu Sun,Ningning Liu,Jiahui Miao,Yanan Zhang,Yuanpeng Hao,Jinzheng Zhang,Hui Li,Hongtong Bai,Lei Shi

Oregano is a medicinal and aromatic plant of value in the pharmaceutical, food, feed additive, and cosmetic industries. Oregano breeding is still in its infancy compared with traditional crops. In this study, we evaluated the phenotypes of 12 oregano genotypes and generated F1progenies by hybridization. The density of leaf glandular secretory trichomes and the essential oil yield in the 12 oregano genotypes varied from 97–1017 per cm2and 0.17–1.67%, respectively. These genotypes were divided into four terpene chemotypes: carvacrol-, thymol-, germacrene D/β-caryophyllene-, and linalool/β-ocimene-type. Based on phenotypic data and considering terpene chemotypes as the main breeding goal, six oregano hybrid combinations were performed. Simple sequence repeat (SSR) markers were developed based on unpublished whole-genome sequencing data ofOriganum vulgare, and 64 codominant SSR primers were screened on the parents of the six oregano combinations. These codominant primers were used to determine the authenticity of 40 F1lines, and 37 true hybrids were identified. These 37 F1lines were divided into six terpene chemotypes: sabinene-, β-ocimene-, γ-terpinene-, thymol-, carvacrol-, andp-cymene-type, four of which (sabinene-, β-ocimene-, γ-terpinene-, andp-cymene-type) were novel (i.e., different from the chemotypes of parents). The terpene contents of 18 of the 37 F1lines were higher than those of their parents. The above results lay a strong foundation for the creating of new germplasm resources, constructing of genetic linkage map, and mapping quantitative trait loci (QTLs) of key horticultural traits, and provide insights into the mechanism of terpenoid biosynthesis in oregano. Keywords:oregano;essential oil;terpene chemotypes;hybrid;F1lines;SSR molecular marker

Efficacy and Potential Mechanism of Essential Oils of Three Labiatae Plants against the Pathogenic Fungi of Root Rot Disease in Atractylodes chinensis

Siyuan Xie,He Si,Shenfei Zhang,Ru Zhou,Yuyan Xue,Shijie Wang,Shiqiang Wang,Yizhong Duan,Junfeng Niu,Zhezhi Wang

Atractylodes chinensishas a high medicinal value and is widely cultivated. However, root rot disease seriously affects the yield and quality ofA. chinensis. To develop green and safe pesticides, the inhibitory effect of essential oils (EOs) of three Labiatae plants on the pathogenic fungi that causes root rot disease inAtractylodes chinensiswas investigated. The results showed that theOriganum vulgareEO andThymus mongolicusEO exhibited strong inhibitory effects onFusarium oxysporum,Fusarium solani, andFusarium redolens, with 100% inhibition rate. The low MIC values of EOs and their main components against the three pathogenic fungi indicated that all of them showed strong fungicidal effects. The MIC values ofO. vulgareEO againstF. oxysporum,F. solani, andF. redolenswere 2.60 mg/mL, 3.13 mg/mL, and 1.56 mg/mL, respectively. Analyses using scanning electron microscopy (SEM) and transmission electron microscopy (TEM) showed that theO. vulgareEO severely damaged the cell wall and cell membrane of mycelial cells. TheO. vulgareEO increased cell permeability, leading to a large leakage of cell contents (DNA and proteins). In addition,O. vulgareEO inhibitedF. oxysporumby inducing ROS production and reducing the amount of intracellular GSH, leading to a large accumulation of ROS. This study showed that plant EOs have excellent fungicidal activity and can be used as novel natural and environmentally friendly pesticides for the control of root rot inA. chinensis. Keywords:Atractylodes chinensis;root rot;essential oils;Origanum vulgare;antifungal activity;mechanisms of action

Analysis of the Chemical Profiles and Anti-S. aureus Activities of Essential Oils Extracted from Different Parts of Three Oregano Cultivars

Yuanpeng Hao,Jingyi Li,Wenying Zhang,Meiyu Sun,Hui Li,Fei Xia,Hongxia Cui,Hongtong Bai,Lei Shi

The use of antibiotics in the food industry is highly regulated owing to the potential harmful effects of antibiotics on human health. Therefore, it is crucial to seek alternatives for ensuring food safety. Essential oils (EOs) extracted from plants of the genusOriganumexhibit a wide range of chemical and antibacterial activities. Species and tissue factors shape the production and accumulation processes of EOs inOriganumplants, thereby affecting their bactericidal activity. In this study, the morphologies and EO yields from the inflorescences, leaves, and stems of three oregano cultivars were evaluated. In addition, the chemical compositions and antibacterial abilities of oregano EOs (OEOs) were assessed. The results showed that OEOs from the different parts of the plant displayed only minor differences in chemical composition, whereas the yield of EOs varied considerably. Additionally, the chemical profiles of OEOs differed significantly among cultivars. The carvacrol content in the OEOs was closely related to its activity againstStaphylococcus aureus; the antibacterial properties of the OEOs were further verified using carvacrol. These findings suggested that OEOs possessing high antibacterial activity may have the potential to be developed as bactericides in the food industry. Keywords:oregano essential oil;chemical composition;carvacrol;antibacterial activity;food safety

Eco-friendly and cost-effective epoxy binder for polymer mortar utilizing oregano oil-based hardener

Mingen Fei,Xueliang Chen,Han Chen,Yongtong Fan,Yongchen Peng,Yu-Chung Chang,Wendi Liu,Renhui Qiu

Polymer mortar, consisting of a polymer binder and aggregates, offers superior properties such as enhanced strength, durability, and corrosion resistance compared to the traditional cement-based mortars. Within the polymer-based mortar, epoxy resin is one of the most commonly used resins owing to its high performance. However, its widespread adoption is hindered by costs and environmental concerns, particularly the dependence of bisphenol A-containing epoxide, a substance harmful to human health. In this work, a novel epoxy hardener derived from oregano oil (OEO) was developed to create a more eco-friendly and cost-effective binder for polymer mortar. The inclusion of OEO-based hardeners reduced the viscosity of the binder and accelerated the curing process. The Mannich base structure of the hardener induced more reactive sites in the epoxy system, resulting in a denser crosslinked network. Polymer mortars fabricated with this novel hardener exhibited improved mechanical properties, bonding strength, and corrosion resistance compared to conventional epoxy mortars. Scanning electron microscopy images also revealed a stronger interfacial bond between the epoxy resin and the aggregates. This study demonstrates the potential of OEO-based hardeners to enhance the performance of epoxy binders, providing a sustainable alternative for polymer-based mortar applications.

Fabrication and characterization of electrospun zein nanofibers incorporated with citronellol-rich Origanum vulgare essential oil

Mayire Abulitifu,Wei Qin,Wang Yingbo,Bai Xi

The citronellol-rich Origanum vulgare essential oil (OEO), which exhibits excellent antioxidant and antimicrobial activities, has limited use in food packaging owing to its high volatility and instability. This problem can be solved using electrospinning. In this study, OEO was incorporated into the zein matrix via emulsion electrospinning. OEO was obtained by hydro-distillation; gas chromatography–mass spectrometry analysis revealed that citronellol (44.34%) was a predominant compound in OEO. Scanning electron microscopy images demonstrated that the fibers had a bead-free and uniform structure with a diameter ranging from 215 ± 36 to 260 ± 72 nm. The X-ray diffractograms indicate the amorphous structure of the zein fibers. Fourier transform infrared absorption spectroscopy revealed an interaction between OEO and zein. The addition of OEO to the zein nanofibers affected its secondary structure. Thermogravimetric analysis showed that the OEO-loaded (OEO/zein) nanofibers exhibited high thermostability and that the water contact angle was enhanced. The encapsulation efficiency and loading capacity were in the range of 71.56–85.8% and 8.88–40.93%, respectively. The antioxidant activity of the OEO/zein nanofibers ranged from 45.40% to 71.50%. The OEO/zein fibers displayed antimicrobial activity against Staphylococcus aureus and Escherichia coli . In conclusion, OEO/zein nanofibers are promising alternatives for active food packaging.

Study on the stability, functional activity and preservation effect of oregano essential oil Pickering emulsion with different proportions of chicken bone gelatin/bacterial cellulose during storage

Yanlan Ma,Xinyi Yang,Zongshuai Zhu,Tianran Huang,Jichao Huang,Ming Huang

In this study, chicken bone gelatin (CBG) was extracted as a new substitute for traditional pig bone gelatin, and bacterial cellulose (BC) was used as the compound to prepare Oregano essential oil (OEO) Pickering emulsion. To explore a protein/polysaccharide emulsion system that can effectively prolong the functional activity of OEO during storage. The results indicated that the variation in CBG and BC content significantly influenced the physicochemical properties of the emulsions. The optimal formulation of OEO Pickering emulsion, prepared with a CBG-BC ratio of 6:2 ( v /v), exhibited superior characteristics including appearance, encapsulation efficiency, and stability during preservation. After 7 d of storage at 4 °C, the rheological properties remained stable, with no significant differences observed in antioxidant and antibacterial activities. It was verified in the beef fresh-keeping experiment that the shelf life of beef samples in the 6-2-2 treatment group was 6 d longer than that in the control group and 3 d longer than that in the pure OEO group. This experiment enhanced the utilization of poultry by-products and provided a valuable reference for exploring suitable protein-polysaccharide systems embedding active substances for food preservation.

Developed Chitosan/Oregano Essential Oil Biocomposite Packaging Film Enhanced by Cellulose Nanofibril

Shunli Chen,Min Wu,Caixia Wang,Shun Yan,Peng Lu,Shuangfei Wang

The use of advanced and eco-friendly materials has become a trend in the field of food packaging. Cellulose nanofibrils (CNFs) were prepared from bleached bagasse pulp board by a mechanical grinding method and were used to enhance the properties of a chitosan/oregano essential oil (OEO) biocomposite packaging film. The growth inhibition rate of the developed films with 2% (w/w) OEO againstE. coliandL. monocytogenesreached 99%. With the increased levels of added CNFs, the fibrous network structure of the films became more obvious, as was determined by SEM and the formation of strong hydrogen bonds between CNFs and chitosan was observed in FTIR spectra, while the XRD pattern suggested that the strength of diffraction peaks and crystallinity of the films slightly increased. The addition of 20% CNFs contributed to an oxygen-transmission rate reduction of 5.96 cc/m2·day and water vapor transmission rate reduction of 741.49 g/m2·day. However, the increase in CNFs contents did not significantly improve the barrier properties of the film. The addition of 60% CNFs significantly improved the barrier properties of the film to light and exhibited the lowest light transmittance (28.53%) at 600 nm. Addition of CNFs to the chitosan/OEO film significantly improved tensile strength and the addition of 60% CNFs contributed to an increase of 16.80 MPa in tensile strength. The developed chitosan/oregano essential oil/CNFs biocomposite film with favorable properties and antibacterial activity can be used as a green, functional material in the food-packaging field. It has the potential to improve food quality and extend food shelf life. Keywords:cellulose nanofibrils;chitosan;oregano essential oil;antimicrobial;oxygen barrier properties

Inhibitory effect and mechanism of oregano essential oil on Listeria monocytogenes cells, toxins and biofilms

Peng Guo,Zhenye Li,Ting Cai,Du Guo,Baowei Yang,Chunling Zhang,Zhongguo Shan,Xin Wang,Xiaoli Peng,Guorong Liu,Chao Shi,Metab Alharbi,Abdullah F Alasmari

Listeria monocytogenes ( L. monocytogenes ) is a prevalent foodborne pathogen with a remarkable capacity to form biofilms on utensil surfaces. The Listeriolysin O (LLO) exhibits hemolytic activity, which is responsible for causing human infections. In this study, we investigated the inhibitory effect and mechanism of oregano essential oil (OEO) on L. monocytogenes , evaluated the effects on its biofilm removal and hemolytic activity. The minimum inhibitory concentration (MIC) of OEO against L. monocytogenes was 0.03 % ( v / v ). L. monocytogenes was treated with OEO at 3/2 MIC for 30 min the bacteria was decreased below the detection limit (10 CFU/mL) in PBS and TSB (the initial bacterial load was about 6.5 log CFU/mL). The level of L . monocytogenes in minced pork co-cultured with OEO (15 MIC) about 2.5 log CFU/g lower than that in the untreated group. The inhibitory mechanisms of OEO against planktonic L. monocytogenes encompassed perturbation of cellular morphology, elevation in reactive oxygen species levels, augmentation of lipid oxidation extent, hyperpolarization of membrane potential, and reduction in intracellular ATP concentration. In addition, OEO reduced biofilm coverage on the surface of glass slides by 62.03 % compared with the untreated group. Meanwhile, OEO (1/8 MIC) treatment reduced the hemolytic activity of L. monocytogenes to 24.6 % compared with the positive control. Molecular docking suggested carvacrol and thymol might reduce the hemolytic activity of L. monocytogenes . The results of this study demonstrate that OEO exhibits inhibitory effects against L. monocytogenes , biofilms and LLO, which had potential as natural antimicrobial for the inhibition of L. monocytogenes .

The application of polyacrylonitrile-nano silver-oregano oil electrospun fiber membrane in controlling microbial deterioration of Grotto cultural heritage

Yulan Hu,Lichao Chen,Liwei Pan,Bingjian Zhang,Qiang Li

The study created an electrospun fiber membrane with polyacrylonitrile, nano silver, and oregano oil using the electrospinning technique. After examining the preparation parameters, an appropriate formula for the composite film spinning solution and electrospinning parameters were found: 11 wt percent polyacrylonitrile, 6 wt percent natural oregano essential oil, 15 kV of spinning voltage, 0.6 mL/h of advancing rate, and 20 cm of spinning distance. The composite nanofiber membrane demonstrated improved shape and structure, a more balanced antibacterial effect, and persistence against microbes when subjected to the prescribed formula and circumstances. In the end, the strong antimicrobial qualities of the nano-fiber material were validated by the Feilaifeng field testing. This study showed that when combined with natural plant essential oil, the composite nanofiber membrane has potential for use as a green microbial control material for Grottoes historic heritage. By preventing an intense direct contact between antimirobial agents and the cultural relics of the grottoes, the composite barrier reduced the potential harm that could be caused by the solution soaking into the rocks. Moreover, the composite nanofiber membrane provides the advantages of large-scale antibacterial activity from natural essential oil and long-lasting antimicrobial capabilities from nanosilver. It also has no effect on the cultural relics of the Grotto, is simple to remove, and leaves little trace.
