The repair of skeletal defects in maxillofacial region remains an intractable problem, the rising technology of bone tissue engineering provides a new strategy to solve it. Scaffolds, a crucial element of tissue engineering, must have favorable biocompatibility as well as osteoinductivity. In this study, we prepared berberine/polycaprolactone/collagen (BBR/PCL/COL) scaffolds with different concentrations of berberine (BBR) (25, 50, 75 and 100 μg/mL) through electrospinning. The influence of dosage on scaffold morphology, cell behavior and in vivo bone defect repair were systematically studied. The results indicated that scaffolds could release BBR stably for up to 27 days. Experiments in vitro showed that BBR/PCL/COL scaffolds had appropriate biocompatibility in the concentration of 25–75 μg/mL, and 50 and 75 μg/mL scaffolds could significantly promote osteogenic differentiation of dental pulp stem cells. Scaffold with 50 μg/mL BBR was implanted into the critical bone defect of rats to evaluate the ability of bone repair in vivo. It was found that BBR/PCL/COL scaffold performed more favorable than polycaprolactone/collagen (PCL/COL) scaffold. Overall, our study is the first to evaluate the capability of in vivo bone repair of BBR/PCL/COL electrospun scaffold. The results indicate that BBR/PCL/COL scaffold has prospective potential for tissue engineering applications in bone regeneration therapy.

Electrospun scaffolds with aligned fiber orientation are widely used in tissue engineering, such as muscle, heart, nerve, tendon, and cartilage, due to their ability to guide cell morphology and induce cellular functions. However, the dense fibrous structure of the scaffolds poses a critical obstacle to engineering highly cellular and thick 3D tissues, as it prevents cell infiltration. While many techniques have been developed to increase the pore size of electrospun scaffolds and improve cell infiltration/migration, it often leads to a decrease in direct cell-cell contact, compromising cell differentiation and tissue maturation. This study presents an alternative approach by reducing the thickness of scaffolds to the cellular scale and stacking or rolling the cell-scaffold complex into 3D constructs. We devise a series of novel tools to fabricate, characterize, and manipulate ultra-thin electrospun scaffolds, which demonstrate high reproducibility, resolution, and cellularity. Our study provides a solution to the cell infiltration issue in muscle tissue engineering and is highly versatile, and can be applied to various fields that require structures with high-resolution gradients in a layered pattern or complex spatial distribution in a rolled pattern.

Bone defects cause serious psychological and economic burden to patients. Artificially bone repairing materials bring hope to the treatment of bone defects. Electrospun technique has attracted great attention since it can fabricate fibers from nano- to micro- scale continuously. Scaffolds fabricated by electrospun can mimic the structure of extracellular matrix which is beneficial to cell adhesion and migration. Researches have showed that bioactive ions (such as silicon and calcium ions) can promote bone regeneration. In addition, physical cues can affect cellular behavior such as cell adhesion and differentiation. In this study, two kinds of calcium silicate – adopted poly (L-lactic acid) (CS-PLLA) electrospun scaffolds with random/aligned structures were prepared by electrospun to promote bone regeneration. The integration of CS nanowires improved the biological property of PLLA electrospun scaffolds. Furthermore, in vitro results indicated that aligned 1 wt% CS adopted PLLA (PCA1) electrospun scaffolds with better physical properties and facilitated cell adhesion, improved alkaline phosphate (ALP) activity and the expression of osteogenic genes (Osteopontin (OPN), Collagen type 1 (Col-1) and Bone morphogenetic protein-2 (BMP-2)) compared with random 1 wt% CS adopted PLLA (PCR1) electrospun scaffolds. In conclusion, the prepared PCA1 electrospun scaffolds might be a potential candidate for bone regeneration in defect areas.

The topographical features and material composition of scaffolds have a powerful influence on cell behaviors such as proliferation and differentiation. Here, scaffolds consisting of aligned fibers with incorporated bioactive collagen I were tested for their ability to enhance osteogenesis in vitro. Rat adipose-derived mesenchymal stem cells (ADSCs) were seeded on the scaffolds and their morphology, proliferation, and osteogenic differentiation were examined. Aligned scaffolds with collagen I showed the best osteogenic properties. Also, adhesion-related genes showed the higher expression on aligned scaffolds with collagen I. Our findings indicate that fiber alignment combined with incorporation of collagen I increases the capacity of electrospun scaffolds to induce enhanced and directed osteogenesis. Such scaffolds may, therefore, have potential for improving guided oral bone regeneration.

Introduction The high incidence of skin injuries and the limitations of conventional dressings highlight the need for advanced wound care materials. Electrospun nanofibrous scaffolds, with their extracellular matrix-like architecture, offer potential to enhance healing. Methods A bilayer nanofibrous scaffold of thermoplastic polyurethane (TPU) and chitosan loaded with zinc oxide nanoparticles (ZnO) (TPU/CS@ZnO) was fabricated via electrospinning. The scaffold consisted of a hydrophobic TPU outer layer for waterproof protection and a hydrophilic TPU/CS@ZnO inner layer for bioactivity. Physicochemical properties were characterized by morphology, mechanical strength, and wettability. Cytocompatibility was evaluated in vitro, and wound healing efficacy was tested in vivo using a full-thickness skin defect model. Results The scaffold displayed uniform fibres with a base-layer diameter of 231.81 ± 44.85 nm, tensile strength of 8.42 ± 0.58 MPa, and Young’s modulus of 17.96 ± 0.78 MPa. Water contact angles confirmed hydrophilic and hydrophobic layer characteristics (52.68° ± 4.46° vs. 113.60° ± 2.85°). In vitro studies showed enhanced cell proliferation and adhesion, while in vivo experiments demonstrated over 90% wound closure at day 14, significantly faster than untreated groups. Histological analysis indicated contributions from cellular adhesion, angiogenesis, and immunomodulation. Discussion The bilayer TPU/CS@ZnO scaffold integrates structural protection with biological activity, accelerating wound repair through multiple mechanisms. These findings support its potential as a multifunctional wound dressing, while further studies are needed to clarify molecular pathways and advance clinical application.

To study the effects of polyethylene glycol (PEG) content on the mechanical properties and degradation of silk fibroin, nano-hydroxyapatite, and PEG (SF/nHAP/PEG) electrospun scaffolds, and according to the PEG ratio in the scaffold (SF : nHAP : PEG), test groups were divided as follows: PEG-0 (10 : 2), PEG-0.5 (10 : 2 : 0.5), PEG-1 (10 : 2 : 1), and PEG-2 (10 : 2 : 2). A series of tests to determine the mechanical properties, degradation rates, and osteogenic characteristics was undertaken. PEG facilitated SF degradation (PEG-1 > PEG-0.5 > PEG-0 > PEG-2), and the mass loss of the scaffolds in PEG-1 was more than 30%, while in PEG-2 it was less than 20% after 8 days (P < 0.05). The addition of PEG strengthened the mechanical properties of the scaffold (PEG-1 > PEG-2 > PEG-0.5 > PEG-0), as the Young's modulus increased from 41.72 ± 3.40 MPa for PEG-0 to 76.12 ± 3.73 MPa for PEG-1 (P < 0.05). PEG was favorable for the osteogenic differentiation of BMSCs (PEG-0.5 > PEG-1 > PEG-2 > PEG-0). The enhancements were attributable to the increased hydrophilicity and nHAP dispersion, as well as to the secondary structure transformation of SF. The PEG content was deemed to be optimal when the SF/nHAP/PEG ratio was equal to 10 : 2 : 1.

Application of synthetic or biological meshes is the main therapy for the repair and reconstruction of abdominal wall defects, a common disease in surgery. Currently, no ideal materials are available, and there is an urgent need to find appropriate ones to satisfy clinical needs. Electrospun scaffolds have drawn attention in soft tissue reconstruction. In this study, we developed a novel method to fabricate a composite electrospun scaffold using a thermoresponsive hydrogel, poly (N-isopropylacrylamide)-block-poly (ethylene glycol), and a biodegradable polymer, polylactic acid (PLA). This scaffold provided not only a high surface area/volume ratio and a three-dimensional fibrous matrix but also high biocompatibility and sufficient mechanical strength, and could simulate the native extracellular matrix and accelerate cell adhesion and proliferation. Furthermore, rat adipose-derived stem cells (ADSCs) were seeded in the composite electrospun scaffold to enhance the defect repair and regeneration by directionally inducing ADSCs into endothelial cells. In addition, we found early vascularization in the process was regulated by the hypoxia inducible factor-1α (HIF-1α)/vascular endothelial growth factor (VEGF) pathway. In our study, overexpression of HIF-1α/VEGF in ADSCs using a lentivirus system promoted early vascularization in the electrospun scaffolds. Overall, we expect our composite biomimetic scaffold method will be applicable and useful in abdominal wall defect regeneration and repair in the future.

Purpose Electrospun scaffolds have been studied extensively for their potential use in bone tissue engineering. However, their hydrophobicity and relatively low matrix stiffness constrain their osteoinduction capacities. In the present study, we studied polymer electrospun scaffolds coated with hydrophilic hematite nanoparticles (αFeNPs) constructed using layer-by-layer (LbL) assembly to construct a bioactive interface between the scaffolds and cells, to improve the osteoinduction capacities of the scaffolds.Materials and methods The morphology of the αFeNPs was assessed. Surface properties of the scaffolds were tested by X-ray photoelectron spectroscopy (XPS), surface water contact angle, and in vitro protein adsorption test. The stiffness of the coating was tested using an atomic force microscope (AFM). In vitro cell assays were performed using rat adipose-derived stem cells (ADSCs).Results Morphology characterizations showed that αFeNPs assembled on the surface of the scaffold, where the nano assemblies improved hydrophilicity and increased surface roughness, with increased surface stiffness. Enhanced initial ADSC cell spread was found in the nano assembled groups. Significant enhancements in osteogenic differentiation, represented by enhanced alkaline phosphatase (ALP) activities, elevated expression of osteogenic marker genes, and increased mineral synthesis by the seeded ADSCs, were detected. The influencing factors were attributed to the better hydrophilicity, rougher surface topography, and harder interface stiffness. In addition, the presence of nanoparticles was believed to provide better cell adhesion sites. Conclusion The results suggested that the construction of a bioactive interface by LbL assembly using αFeNPs on traditional scaffolds should be a promising method for bone tissue engineering.

To improve bone regeneration in oral microenvironment, we generated a novel biodegradable, antibacterial, and osteoconductive electrospun PLGA/PCL membrane as an ideal osteogenic scaffold. The novel three-layer membranes were structured with serial layers of electrospun chlorhexidine-doped-PLGA/PCL (PPC), PLGA/PCL (PP), and β-tricalcium phosphate-doped-PLGA/PCL (PPβ). To characterize osteoconductive properties of these membranes, MC3T3-E1 (MC) cultures were seeded onto the membranes for 14 days for evaluation of cell proliferation, morphology and gene/protein expression. In addition, MC cells were cultured onto different surfaces of the three-layer membranes, PPC layer facing MC cells (PPβ-PP-PPC) and PPβ layer facing MC cells (PPC-PP-PPβ) to evaluate surface–material effects. Membrane properties and structures were evaluated. Antibacterial properties against Streptococcus mutans and Staphylococcus aureus were determined. Scanning electron microscope demonstrated smaller interfiber spaces of PPC and PPβ-PP-PPC compared to PPβ, PPC-PP-PPβ, and PP. PPC and PPβ-PP-PPC exhibited hydrophilic property. The three-layer membranes (PPC-PP-PPβ and PPβ-PP-PPC) demonstrated significantly higher Young’s modulus (94.99 ± 4.03 MPa and 92.88 ± 4.03 MPa) compared to PP (48.76 ± 18.15 MPa) or PPC (7.92 ± 3.97 MPa) (p < 0.05). No significant difference of cell proliferation was found among any groups at any time point (p > 0.05). Higher expression of integrins were detected at 12 h of cultures on PPC-PP-PPβ compared to the controls. Promoted osteoconductive effects of PPC-PP-PPβ were revealed by alkaline phosphatase assays and Western blot compared with the controls at 7 and 14 days. PPC, PPC-PP-PPβ and PPβ-PP-PPC exhibited a significantly wider antibacterial zone against the tested bacteria compared to PP and PPβ (p < 0.05). These results suggested that the three-layer electrospun membranes demonstrated superior properties: higher strength, better cell adhesion, and promoted osteoconductive properties compared to single-layer membrane: however, antibacterial properties were exhibited in three-layer electrospun membranes and chlorhexidine-doped single-layer membrane. We concluded that the novel three-layer membranes could be used as a biocompatible scaffold for intraoral bone regeneration due to its enhanced osteoconductive activity and antibacterial effect.

Purpose Successful intestinal tissue engineering requires specialized biocompatible scaffolds and a vibrant vascularization microenvironment. A pre-vascularized chamber can provide both in vivo, but there is little report on using it to improve intestinal regeneration. Besides, researchers have found that gelatin is highly biocompatible and graphene oxide (GO) can be used to improve mechanical properties. Thus, applying a pre-vascularized chamber fabricated gelatin and GO into intestinal tissue engineering is worth a try.Materials and Methods In this study, an investigation into the physicochemical and mechanical properties as well as biocompatibility of the electrospun graphene oxide-gelatin (GO-Gel) scaffolds were conducted in vitro. Meanwhile, a pre-vascularized GO-Gel (V-GO-Gel) chamber model was built by implanting the scaffold around the mesenteric vessels in rat. After vascularization process, the chamber was used to repair the perforation and then assessed by histology and immunofluorescence analyses. Results These porous scaffolds were mechanical improved with GO incorporated into gelatin. Further, the cell adherence, viability and morphology on the scaffolds were maintained. The V-GO-Gel chamber model was successfully built and effective enhanced the repair of the intestinal wall than the other group without recurrence or complications. Conclusion The V-GO-Gel chamber shows promising therapeutic potential in the repair of intestinal wall defects.

Purpose In this study, we aimed to develop a unique N-acetyl cysteine (NAC)-loaded polylactic-co-glycolic acid (PLGA) electrospun system with separate compartments for the promotion of osteogenesis.Materials and methods We first prepared solutions of NAC-loaded mesoporous silica nanoparticles (MSNs), PLGA, and NAC in N, N-dimethylformamide and tetrahydrofuran for the construction of the electrospun system. We then fed solutions to a specific injector for electrospinning. The physical and chemical properties of the scaffold were characterized through scanning electron microscopy, transmission electron microscopy, and Fourier transform infrared spectroscopy. The release of NAC and Si from different PLGA scaffolds was estimated. The cell viability, cell growth, and osteogenic potential of rat bone marrow-derived stroma cell (rBMSCs) on different PLGA scaffolds were evaluated through MTT assay, live/dead staining, phalloidin staining, and Alizarin red staining. The expression levels of osteogenic-related markers were analyzed through real-time PCR (qRT-PCR).Results NAC was successfully loaded into MSNs. The addition of MSNs and NAC decreased the diameters of the electrospun fibers, increased the hydrophilicity and mechanical property of the PLGA scaffold. The release kinetic curve indicated that NAC was released from (PLGA + NAC)/(NAC@MSN) in a biphasic pattern, that featured an initial burst release stage and a later sustained release stage. This release pattern of NAC encapsulated on the (PLGA + NAC)/(NAC@MSN) scaffolds enabled to prolong the high concentrations of release of NAC, thus drastically affecting the osteogenic differentiation of rBMSCs.Conclusion A PLGA electrospun scaffold was developed, and MSNs were used as separate nanocarriers for recharging NAC concentration, demonstrating the promising use of (PLGA + NAC)/(NAC@MSN) for bone tissue engineering.

Nanofibers (NFs) have been widely used in tissue engineering such as wound healing. In this work, the antibacterial ZnO quantum dots (ZnO QDs) have been incorporated into the biocompatible poly (ε-caprolactone)/collagen (PCL/Col) fibrous scaffolds for wound healing. The as-fabricated PCL-Col/ZnO fibrous scaffolds exhibited good swelling, antibacterial activity, and biodegradation behaviors, which were beneficial for the applications as a wound dressing. Moreover, the PCL-Col/ZnO fibrous scaffolds showed excellent cytocompatibility for promoting cell proliferation. The resultant PCL-Col/ZnO fibrous scaffolds containing vascular endothelial growth factor (VEGF) also exhibited promoted wound-healing effect through promoting expression of transforming growth factor-β (TGF-β) and the vascular factor (CD31) in tissues in the early stages of wound healing. This new electrospun fibrous scaffolds with wound-healing promotion and antibacterial property should be convenient for treating wound healing.
