Synergistic effects of electroactive antibacterial material and electrical stimulation in enhancing skin tissue regeneration: A next-generation dermal wound dressing

Xi-Liang Zang,Fei Gao,Zhao Zhang,Lin-Hua Shen,Yue-Hai Pan

Objective We aimed to develop an electroactive antibacterial material for the treatment of skin wound diseases. Method To this aim, we modified chitosan (CS), a biocompatible polymer, by coupling it with graphene (rGO) and an antimicrobial polypeptide DOPA-PonG1. The material's effect on skin injury healing was studied in combination with external electrical stimulation (EEM). The structure, surface composition, and hydrophilicity of the modified CS materials were evaluated using scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FTIR), and contact angle measurements. We studied NIH3T3 cells cultured with modified materials and subjected to EEM to assess viability, adhesion, and tissue repair-related gene expression. Results SEM data demonstrated that rGO was distributed uniformly on the surface of the CS material, increasing surface roughness, and antimicrobial peptides had minimal impact on surface morphology. FTIR confirmed the uniform distribution of rGO and antibacterial peptides on the material surface. Both rGO and DOPA-PonG1 enhanced the hydrophilicity of CS materials, with rGO also improving tensile strength. The dual modification of CS with rGO and DOPA-PonG1 synergistically increased antibacterial efficacy. Cellular events and gene expression relevant to tissue repair process were enhanced by these modifications. Furthermore, EEM accelerated epidermal regeneration more than the material alone. In a rat skin wound model, DOPA-PonG1@CS/rGO dressing combined with electrical stimulation exhibited accelerated healing of skin defect. Conclusion Overall, our results demonstrate that CS materials modified with rGO and DOPA-PonG1 have increased hydrophilicity, antibacterial characteristics, and tissue regeneration capacities. This modified material in conjunction with EEM hold promise for the clinical management for dermal wounds.

Modulation of macrophages by a phillyrin-loaded thermosensitive hydrogel promotes skin wound healing in mice

Chenggang Huang,Jiajia Teng,Wei Liu,Junzhe Wang,An Liu

Background   Impaired wound healing in traumatic skin injuries remains a severe clinical challenge due to impaired re-vascularization, harmful bacteria infection, and inflammation dysregulation. Macrophages are recognized as prominent immune cells in tissue regeneration and wound healing. Consequently, the modulation of macrophages provides a promising therapeutic target for wound healing disorders. Here, we aimed to explore whether a novel constructed combination of thermosensitive hydrogel Pluronic F-127 (PF-127) and phillyrin (PH, the main active compound of forsythia suspensa) could improve skin wound healing. Methods   Firstly, the biological effects of pH on the phenotype and inflammation of macrophages were assessed by flow cytometry and ELISA. The biocompatibility of the PF-127 plus PH combination was investigated on keratinocytes and red blood cells. The biological effect of PF-127/PH hydrogel on the migratory ability of keratinocytes in vitro was evaluated using the scratch and transwell migration assays. In addition,   S. aureus   and   E. coli   were employed to test the antibacterial properties of the PF-127 plus PH combination. Finally, PF-127 plus PH scaffold was applied   to the full-thickness skin defect in mice. Histomorphological evaluation and immunochemistry were performed to explore the wound-healing activity of PF-127/PH hydrogel. Results   PH can promote the polarization of macrophages from the M1 (pro-inflammatory) phenotype to the M2 (anti-inflammatory) phenotype. The PF-127/PH hydrogel was highly biocompatible and showed a potent stimulative effect on the migration of keratinocytes   in vitro. The combination of PF-127 and PH exerted a pronounced antibacterial activity on   S. aureus   and   E. coli in vitro.   PF-127/PH hydrogel potently accelerates the healing of full-thickness skin defects by promoting skin cell proliferation, accelerating angiogenesis, and inhibiting inflammation. Conclusions Our study suggests that PF-127/PH hydrogel has excellent potential for treating traumatic skin defects.

Bionic Bovine Achilles Tendon Collagen Composite Membrane Loaded with Anti-Inflammatory Kukoamine B Promotes Skin Wound Healing

Ruting Luo,Yujie Mu,Le Zhao,Jinglin Hua,Lixin Cao,Danting Chen,Kun Li,Zhenkai Jin,Yanchuan Guo,Bing Zhang,Min Wang

Skin is the first line of defence between the human body and the outside world, and it is constantly exposed to external injuries and wounds for a variety of reasons. Collagen is a structural protein of the extracellular matrix and an important component of the dermis. As a wound dressing, collagen not only provides nutrients to wounds but also enhances the immune response in the pre-healing phase, making it an excellent biomaterial for healing. In this study, we used electrospinning and freeze-drying technology to prepare a Bovine Achilles Tendon Collagen (BATC) electrospun composite membrane and a BATC freeze-dried composite membrane using BATC as a substrate supplemented with 16.7% Polyethylene oxide (PEO) and 0.2% Kukoamine B (KuB). The physicochemical properties and biocompatibility of the BATC composite membrane were verified via scanning electron microscopy, Fourier-transform infrared spectroscopy, and DSC analysis and by measuring the DPPH radical-scavenging capacity, water absorption, water retention, in vitro drug release, and extract cytotoxicity. The BATC composite membrane was found to have a significant effect on skin wound healing, especially in the middle stage of healing, in a mouse full-thickness skin injury model. The BATC/PEO/KuB electrospun composite membrane (EBPK) had the best capacity for promoting wound healing and can be used as a wound dressing for in-depth research and development, and KuB, a monomer component with a clear structure and mechanism of action, can be used as a candidate component of composite dressings.

Double-alternating injectable multifunctional hydrogel based on chitosan for skin wound repair

Yongyi Yang,Rong Liu,Li Xie,Yang Li,Mengqi Chen,Huang Zhu,Qingqing Pan

Injectable hydrogels can be suitable for any irregular wounds to play an important role in wound healing, which have attracted much attention from researchers. Here, a chitosan-based double alternating injectable hydrogel had been proposed, which aligned with characteristics of wound healing. First of all, carboxymethyl chitosan (CMCS) and oxidized gellant (OGG) were used to fabricate above double-alternating injectable hydrogel skeleton (COG) through Schiff base bond. Then, dipotassium glycyrrhizinate (DPG) and bioactive glass (BG) were loaded respectively into COG, forming COG2-D10 and COG2-B0.5 hydrogel. Various properties of above hydrogels such as rheological behaviors, injectable capability, cytocompatibility, antibacterial and anti-inflammatory activity were systematically investigated. The results indicated that above hydrogel not only had a stable network structure, good toughness, injectability, and excellent adhesive properties, but also exhibited efficient antibacterial, anti-inflammatory, hemostatic, and biocompatible properties. The investigation of in vivo full-thickness skin wound healing showed that compared with COG2-D10 or COG2-B0.5 hydrogel alone, the double-alternating COG2-D10/COG2-B0.5 hydrogel could better enhance the efficacy of drugs, promote cell migration and proliferation, accelerate collagen fiber deposition and neovascularization, and significantly improve wound healing ( p  < 0.01). Therefore, the double-alternating therapy strategy of hydrogel will provide a new approach for the clinical treatment of skin wound healing.

SIKVAV-Modified Chitosan Hydrogel as a Skin Substitutes for Wound Closure in Mice

Xionglin Chen,Xiaoming Cao,He Jiang,Xiangxin Che,Xiaoyuan Xu,Baicheng Ma,Jie Zhang,Tao Huang

Skin wound healing is a complex and dynamic process that involves angiogenesis and growth factor secretion. Newly formed vessels can provide nutrition and oxygen for skin wound healing. Growth factors in skin wounds are important for keratinocytes and fibroblasts proliferation, epithelialization, extracellular matrix remodeling, and angiogenesis, which accelerate skin wound healing. Therefore, treatment strategies that enhance angiogenesis and growth factors secretion in skin wounds can accelerate skin wound healing. This study investigated the effects of a SIKVAV (Ser-Ile-Lys-Val-Ala-Val) peptide-modified chitosan hydrogel on skin wound healing. Hematoxylin and eosin (H&E) staining demonstrated that the SIKVAV-modified chitosan hydrogel accelerated the re-epithelialization of wounds compared with that seen in the negative and positive controls. Masson’s trichrome staining showed that more collagen fibers were deposited in the skin wounds treated with the SIKVAV-modified chitosan hydrogel than in the negative and positive controls. Immunohistochemistry assays demonstrated that more myofibroblasts were deposited and more angiogenesis occurred in skin wounds treated with the SIKVAV-modified chitosan hydrogel than in the negative and positive controls. In addition, ELISA assays showed that the SIKVAV-modified chitosan hydrogels promoted the secretion of growth factors in skin wounds. Taken together, these results suggest that the SIKVAV-modified chitosan hydrogel has the potential to be developed as synthesized biomaterials for the treatment of skin wounds. Keywords:SIKVAV;chitosan hydrogel;skin wound healing;angiogenesis;growth factor

Effective wound dressing based on Poly (vinyl alcohol)/Dextran-aldehyde composite hydrogel

Ce Zheng,Chunyu Liu,Hailiang Chen,Ning Wang,Xia Liu,Guozhen Sun,Weihong Qiao

Wound dressing is of significant importance to promote cutaneous wound healing process. To develop an effective wound dressing, a PVA/DA hydrogel was prepared using a Poly (vinyl alcohol)/Dextran-aldehyde solution blend, followed by crosslinking via freeze-thaw method and freeze-drying. We characterized the hydrogel by infrared spectroscopy, mechanical property tests, swelling behavior test and biocompatibility test. Results showed that the PVA/DA hydrogels had a 3-dimensional, highly porous structure with uniformly distributed pores of 5–10 μm, strong tensile strength of 5.6 MPa, efficient ability to absorb fluid of 6 time its weight and suitable water vapor transmission rate of 2100 g m −2 day −1 to keep a moist environment and good biocompatibility shown by very low hemolysis and no cytotoxicity. In wound healing tests using a full-thickness skin wound model, macroscopic observations showed that the wound covered by the PVA/DA hydrogel almost reached complete healing faster by 10 days, while histological analysis indicated a faster regeneration of skin. Thus, the PVA/DA hydrogel was suitable for application as a wound dressing and may have potential for use in various biomedical applications.

Carbon Nanotube Reinforced Carbonized Cellulose Aerogels for Enhanced Hemostasis and Accelerated Skin Wound Healing

Jiang Tan,Yongjin Fang,Kaicheng Wang,Miaomiao Wei,Zihan Zhang,Lei Wang,Shixuan Chen,Jingye Pan

Trauma from car accidents and other causes can result in severe bleeding and significant skin defects. Current biomaterials still face challenges in achieving rapid hemostasis to stabilize life-threatening conditions and subsequently promoting the complete repair of skin tissue. This study develops a carbon nanotube (CNT)-reinforced carbonized cellulose aerogel for managing fatal, uncontrolled bleeding and enhancing skin wound healing. These aerogels demonstrate high porosity with large pore sizes, appropriate hydrophilicity, and excellent shape recovery after compression. In vitro, the composite aerogels show good cytocompatibility and blood compatibility. Not only do they absorb substantial amounts of adhered red blood cells and platelets, but they also activate platelets. In vivo, the composite aerogels outperform clinically used gauze and gelatin sponges in hemostatic capacity in rat tail amputation and liver perforation hemorrhage models. Moreover, this work discovers that the photothermal effect of the composite aerogel could accelerate blood clot formation, enhance the mechanical properties of the formed clot. Additionally, they promote skin wound healing, achieving results comparable to the widely used commercial wound dressing, PELNAC. The appropriate temperature generated by the photothermal effect can promote the formation of a local blood supply. Overall, this composite aerogel presents a promising strategy for treating uncontrollable bleeding and promoting wound healing.

A Programmable Handheld Extrusion-Based Bioprinting Platform for In Situ Skin Wounds Dressing: Balance Mobility and Customizability

Chenmin Wang,Chengwei Hu,Haojin Cheng,Weichen Qi,Liangliang Wang,Tianchi Wu,Jun Wu,Xu Cui,Jiake Xu,Haobo Pan,Shaoquan Bian,Weijia William Lu,Xiaoli Zhao

Bioprinting technology plays a crucial role for constructing tissue substitutes. However, the mismatched scaffold shapes and the poor treatment timeliness limit its clinical translational application. In situ printing technology that prints bioregenerants directly inside patient's body can meet the needs of specific tissue repair. This study develops a smartphone controlled handheld bioprinter for in situ skin wounds dressing. The mini bioprinter can be handheld and placed on any printing surface to create strips, complex patterns, and 3D structures, and can be equipped with microchannel needles to expand functionality. The size of the strips as well as the printing path can be programmed and controlled by the smartphone to ensure the precision of the printed product quality. Furthermore, the device not only allows for smooth switching between different bioinks for printing heterogeneous structure, but also allows for fast and uniform coverage of large wound surfaces. When dealing with complex wounds in vitro & vivo, the printer can effectively fill and precisely close wounds, promoting wound healing. The programmable handheld bioprinter can balance mobility and customizability in the management of skin wounds and is expected to realize its potential for emergency medical treatment in condition-constrained scenarios, such as battlefields or disaster areas.

PDA/Cu Bioactive Hydrogel with “Hot Ions Effect” for Inhibition of Drug-Resistant Bacteria and Enhancement of Infectious Skin Wound Healing

Qing Xu,Mengling Chang,Yu Zhang,Endian Wang,Min Xing,Long Gao,Zhiguang Huan,Feng Guo,Jiang Chang

Quick and effective sterilization of drug-resistant bacteria inevitably became an ever-growing global challenge. In this study, a multifunctional composite (PDA/Cu-CS) hydrogel mainly composed of polydopamine (PDA) and copper-doped calcium silicate ceramic (Cu-CS) was prepared. It was confirmed that PDA/copper (PDA/Cu) complexing in the composite hydrogel played a key role in enhancing the photothermal performance and antibacterial activity. Through a unique “hot ions effect”, created by the heating of Cu ions through the photothermal effect of the composite hydrogel, the hydrogel showed high-efficiency, quick, and long-term inhibition of methicillin-resistant Staphylococcus aureus and Escherichia coli. In addition, the hydrogel possessed remarkable bioactivity to stimulate angiogenesis. The in vivo results confirmed that the “hot ions effect” of the composite hydrogel removed existing infection in the wound area efficiently and significantly promoted angiogenesis and collagen deposition during infectious skin wound healing. Our results suggested that the design of multifunctional hydrogels with “hot ions effect” may be an effective therapeutic approach for the treatment of infectious wounds.

Electrospun PLGA/PCL Nanofiber Film Loaded with LPA Promotes Full-Layer Wound Healing by Regulating the Keratinocyte Pyroptosis

Xinqi Huang,Jianghuiwen Lu,Yumei An,Mingyuan Xu,Xueshi Chen,Chao Liu,Xuefeng Zhou,Haiyan Shan,Yunzhu Qian,Mingyang Zhang

Electrospun nanofibers have a number of qualities that make them a suitable choice for skin wound healing. Lysophosphatidic acid (LPA) stimulates the keratinocytes and fibroblasts to proliferate, differentiate, and migrate and enhances skin wound healing. Here, we developed the electrospun scaffolds contained in polycaprolactone (PCL) and polylactic-co-glycolic acid (PLGA). The scaffolds loaded with LPA nanoparticles retained a porous nanofiber structure and showed better physicochemical properties and biocompatibility. The scaffold continuously releases LPA to quickly initiate cell signaling and maintain long-term anti-inflammatory activity. In this study, we found that PP scaffold with LPA reduces the disordered collagen deposition and the thickness of the newborn epidermis, improves skin healing, and reduces scar formation. Explaining the mechanism of LPA mineralized tissue regeneration in skin wound healing, LPA inhibited the pyroptosis of keratinocyte, a cell death process that induces inflammation and scar formation by inhibiting the expression of TNF-α and β-catenin proteins. Thus, the electrospun PP scaffold with LPA can be potentially developed as a therapeutic avenue to target skin wound healing.

Injectable Adhesive Self-Healing Multicross-Linked Double-Network Hydrogel Facilitates Full-Thickness Skin Wound Healing

Bo Yang,Jiliang Song,Yuhang Jiang,Ming Li,Jingjing Wei,Jiajun Qin,Wanjia Peng,Fernando López Lasaosa,Yiyan He,Hongli Mao,Jun Yang,Zhongwei Gu

The development of natural polymer-based hydrogels, combining outstanding injectability, self-healing, and tissue adhesion, with mechanical performance, able to facilitate full-thickness skin wound healing, remains challenging. We have developed an injectable micellar hydrogel (AF127/HA-ADH/OHA-Dop) with outstanding adhesive and self-healing properties able to accelerate full-thickness skin wound healing. Dopamine-functionalized oxidized hyaluronic acid (OHA-Dop), adipic acid dihydrazide-modified HA (HA-ADH), and aldehyde-terminated Pluronic F127 (AF127) were employed as polymer backbones. They were cross-linked in situ using Schiff base dynamic covalent bonds (AF127 micelle/HA-ADH network and HA-ADH/OHA-Dop network), hydrogen bonding, and π–π stacking interactions. The resulting multicross-linked double-network design forms a micellar hydrogel. The unique multicross-linked double-network structure endows the hydrogel with both improved injection abilities and mechanical performance while self-healing faster than single-network hydrogels. Inspired by mussel foot adhesive protein, OHA-Dop mimics the catechol groups seen in mussel proteins, endowing hydrogels with robust adhesion properties. We also demonstrate the potential of our hydrogels to accelerate full-thickness cutaneous wound closure and improve skin regeneration with reduced scarring. We anticipate that our hydrogel platform based on a novel multicross-linked double-network design will transform the future development of multifunctional wound dressings.

Fabrication of Curcumin-Loaded Silk Fibroin and Polyvinyl Alcohol Composite Hydrogel Films for Skin Wound Healing

Ruofan Wang,Liming Ruan,Guohua Jiang,Pengfei Li,Uladzislau E. Aharodnikau,Khaydar E. Yunusov,Xiaofei Gao,Sergey O. Solomevich

Skin regeneration of full-thickness wounds remains a challenge, requiring a well-regulated interplay of cell–cell and cell–matrix signaling. Herein, the composite hydrogel films composed of silk fibroin (SF) and polyvinyl alcohol (PVA) as scaffolds loaded with curcumin nanoparticles (Cur NPs) were developed for skin wound healing. The structure and physicochemical properties of hydrogel films were first evaluated by scanning electron microscopy (SEM), water contact angle, and chemical and mechanical measurements. In addition, the as-fabricated composite hydrogel films have a unique 3D structure and excellent biocompatibility that facilitates the adhesion and growth of cells. Antimicrobial tests in vitro showed that they could inhibit the growth of bacteria due to the incorporation of Cur NPs into composite hydrogel films. The efficacy of the curcumin-loaded SF/PVA composite hydrogel films for skin wound healing was investigated on the skin defect model in vivo. Immunological analysis showed that the as-fabricated Cur NP-loaded SF/PVA composite hydrogel films inhibited inflammation at the wound sites, while promoting angiogenesis during the wound healing process.
