The Effect of Sleep Enhancement Educational Program on Maternal Sleep Quality for Nulliparous Pregnant Women: A Randomized-Controlled Trial

Bay Ozcalik Habibe,Sogukpinar Neriman

Background Pregnancy is a critical period during which women usually do not prefer taking medication. Therefore, non-pharmacological and safe methods are needed to improve sleep quality during pregnancy. This study aims to identify whether an educational program on sleep enhancement for pregnant women has any effect on maternal sleep quality. Method This was a randomized-controlled trial. Pregnant women ( N  = 181) were divided into two groups, an intervention group ( n  = 90) and a control group ( n  = 91). A two-session educational program for the enhancement of sleep quality was provided to the intervention group, whereas solely the routine practices of the hospital were put in place for the control group. Results The intervention group had a significantly lower mean Pittsburgh Sleep Quality Index score, and accordingly, better sleep quality than the control group. Participation in the education program was significantly related to sleep quality and accounted for 6% of the total variance in sleep quality ( R 2  = 0.055) ( p  < 0.001). Conclusion It was concluded that the educational program on sleep enhancement implemented to improve the sleep quality of pregnant women enhanced maternal sleep quality. Stretchable organic electrochemical transistors for sustained high-fidelity electrophysiology and deep learning-assisted sleep monitoring

Yuncong Pang,Yang Li,Yuzhe Gu,Benfei Xu,Zihan Zhu,Xiaotian Wang,Yuan Liao,Liya Huang,Qiang Zhao

Good-quality sleep is essential for health, yet obstructive sleep apnea (OSA) underscores the limitations of traditional polysomnography, which is costly, complex, and often uncomfortable. Organic electrochemical transistors (OECTs) offer a promising solution for sleep monitoring due to their high transconductance; however, limitations in stretchability, long-term stability, and intelligent data analysis hinder their broader application. Here, a high-performance stretchable OECT that combines a biocompatible ionic liquid-modified conducting polymer channel with an ionogel electrolyte is developed, addressing the trade-off between performance and wearability. This OECT achieves exceptional transconductance (∼2.1 mS), mechanical resilience (30% strain), and long-term stability (>6 months), enabling high-fidelity electrocardiography (ECG) monitoring with a signal-to-noise ratio (SNR) of 35.7 dB. Through the integration of circuit boards and deep learning algorithms, we have established a wearable, stable, and highly accurate wireless system capable of detecting OSA events from single-lead ECG signals, presenting a novel approach for reliable and portable sleep monitoring.

Low Levels of Adenosine and GDNF Are Potential Risk Factors for Parkinson’s Disease with Sleep Disorders

Li Wang,Zheng Gao,Gang Chen,Deqin Geng,Dianshuai Gao

Sleep disturbances are the most prevalent non-motor symptoms in the preclinical stage of Parkinson’s disease (PD). Adenosine, glial-derived neurotrophic factor (GDNF), and associated neurotransmitters are crucial in the control of sleep arousal. This study aimed to detect the serum levels of adenosine, GDNF, and associated neurotransmitters and explored their correlations with PD with sleep disorders. Demographic characteristics and clinical information of PD patients and healthy participants were assessed. Serum concentrations of adenosine, GDNF, and related neurotransmitters were detected by ELISA and LC-MS. The correlation between serum levels of adenosine, GDNF, and associated neurotransmitters and sleep disorders was explored using logistic regression. PD patients with sleep disorders had higher scores of HAMA, HAMD, ESS, UPDRS-III, and H-Y stage. Lower levels of adenosine, GDNF, and γ-GABA were observed in PD patients who had sleep problems. Logistic regression analysis showed adenosine and GDNF were protective factors for preventing sleep disorders. Adenosine combined with GDNF had a higher diagnostic efficiency in predicting PD with sleep disorders by ROC analysis. This study revealed low adenosine and GDNF levels may be risk factors for sleep disorders in PD. The decrease of serum adenosine and GDNF levels may contribute to the diagnosis of PD with sleep disturbances. Keywords:Parkinson’s disease;sleep disorders;adenosine;GDNF;neurotransmitters

Chlordane exposure impairs the growth and behavior of Drosophila

Boyang Li,Luxi Chen,Feng Li,Qiaoyao Cao,Chenyan Yan,Xin Wu,Kexin Wang,Mingjiang Wu,Yitian Gao,Haibin Tong

Chlordane, a previously extensively utilized insecticidal pesticide, has since been prohibited, however, owing to its limited degradability, it continues to persist significantly in soil and water reservoirs, subsequently accumulating within plant and animal organisms, representing a substantial threat to human health. Despite extensive research conducted over the past few decades to investigate the toxic effects of chlordane, there remains a notable dearth of studies focusing on its impact on sleep activity. Therefore, in this study, the effects of short-term and long-term exposure to chlordane on the activity and sleep of Drosophila were investigated. When exposed to chlordane at a concentration of 1 μM, Drosophila lost body weight, decreased body size and resulted in lipid metabolism disorders. In addition, chlordane exposure altered the arousal and sleep behaviors of Drosophila . Short-term exposure to chlordane resulted in an increase in night-time sleep duration, while long-term exposure to chlordane resulted in an increase in activity and a decrease in sleep, as evidenced by a decrease in the duration of each sleep session and the appearance of sleep fragmentation. Under conditions of long-term chlordane exposure, reactive oxygen species levels were significantly up-regulated in Drosophila . Our results suggest that long-term chlordane exposure triggers oxidative stress damage in Drosophila , leading to sleep disruption. This study offers novel insights into the harmful impacts of environmental pollutants on human sleep patterns and proposes that mitigating the presence of chlordane in the environment could potentially contribute to the reduction of global sleep disorder prevalence.

Gastrodia elata blume ameliorates circadian rhythm disorder-induced mice memory impairment

Hong Huang,Ning Jiang,Yi Wen Zhang,Jing Wei Lv,Hai Xia Wang,Cong Lu,Xin Min Liu,Guang Hua Lu

Circadian rhythm disorder (CRD) in space flight can lead to memory impairment, performance decrements and adverse health outcomes, the main manifestations of which are circadian desynchronization, sleep loss and insomnia. Sleep deprivation (SD) provide the means to evaluate these effects and the risks associated with CRD on ground. Gastrodia elata Blume (GEB) has beneficial effects on the treatment of sleep disturbances and memory loss. Fresh GEB (FG), an unprocessed raw tuber of GEB, has been used as functional health food in Asian countries for a long time. However, the research report of FG to ameliorate memory impairment caused by insomnia or lack of sleep is meager. In this study, ICR male mice were sleep-deprived continuously and water extract of FG (WFG) was orally administrated (3 and 9 g/kg/d, i.g) during the SD process lasted for 25 days, except control and model groups gavage administration with water, positive control group with modafinil (MOD, 0.1 g/kg/d, i.g). We studied the effect of WFG on CRD-induced learning and memory impairment using a set of behavioral analyses including the object location recognition test (OLRT), novel object recognition test (NORT), and the passive avoidance test (PAT). In addition, oxidative stress parameters were assessed by measuring the malondialdehyde (MDA) and superoxide dismutase (SOD) reactivity in serum and hippocampus. Our results revealed that SD decreased discrimination index (DI) in OLRT and NORT, with shorter latency into the dark chamber in PAT. Both WFG and MOD treatment can reverse these changes ( P < 0.05). We concluded that WFG treatment improve CRD-induced learning and memory impairment and oxidative stress damage which makes FG a promising candidate as herbal health product of memory decline in CRD.

Skin-inspired wearable self-powered electronic skin with tunable sensitivity for real-time monitoring of sleep quality

Ouyang Yue,Xuechuan Wang,Mengdi Hou,Manhui Zheng,Zhongxue Bai,Boqiang Cui,Siyu Cha,Xinhua Liu

In modern times, it is essential to ensure excellent sleep quality and comfort, and a way to scientifically assess these conditions is urgently needed. However, most existing sleep monitoring devices are complicated and uncomfortable to wear. Here, a dermal papillae-bioinspired self-powered multifunctional electronic skin (e-skin) with an all-fibrous multilayer nanostructure based on a triboelectric nanogenerator is proposed, which is sensitive, lightweight, ultrathin, breathable, biocompatible, and low-cost. Owing to the bead polyvinylidene fluoride nanofibers of the e-skin simulating the dermal papillae of natural skin and the N-(2-aminoethyl)-3-aminopropyl trimethoxy silane-modified nature of the collagen aggregate nanofibers, the e-skin exhibits the highest sensitivity (0.32 V kPa −1 ) in the range of 0–135 kPa. Additionally, it has short response and recovery times (20 and 33 s, respectively) in the humidity range of 25–85% RH. More importantly, the e-skin converts mechanical energy into electrical energy for humidity detection via an energy circulation system. The e-skin can collect the heartbeat and respiration signals and perform an analysis on their frequency and stability, which can be used to accurately measure the different sleep phases. By analyzing the proportion of each phase and environmental humidity, the e-skin can be employed to scientifically assess the sleep quality and comfort of insomnia patients and night owls through real-time monitoring.

An endoplasmic reticulum-targeted two-photon fluorescent probe for bioimaging of HClO generated during sleep deprivation

Qineng Xia,Xiaoyan Wang,Yanan Liu,Zhangfeng Shen,Zhigang Ge,Hong Huang,Xi Li,Yangang Wang

With the development of social society, sleep deprivation has become a serious and common issue. Previous studies documented that there is a correlation between sleep deprivation and oxidative stress . However, the information of sleep deprivation related ROS has rarely been obtained. Also, it has been demonstrated that sleep deprivation can induce endoplasmic reticulum (ER) stress. As such, for a better understanding of sleep deprivation as well as its related diseases, it is important to develop probes with ER-targeting ability for detecting ROS generated in this process. Herein, a novel two-photon fluorescent molecular probe , JX-1 , was designed for sensing HClO in live cells and zebrafish. The investigation data showed that in addition to real-time response (about 150 s), the probe also exhibited high sensitivity and selectivity . Moreover, the probe JX-1 demonstrated two-photon fluorescence, low cytotoxicity and ER targeting ability. These prominent properties enabled the utilization of the probe for monitoring exogenous and endogenous HClO in both live cells and zebrafish. Using this useful tool, it was found that sleep deprivation can induce the generation of HClO in zebrafish.

Integrative metabolomics and transcriptomics analysis of hippocampus reveals taurine metabolism and sphingolipid metabolism dysregulation associated with sleep deprivation-induced memory impairment

Ting Chen,Junke Jia,Chenyi Gao,Qi Zhong,Lijuan Tang,Xiaokai Sui,Shuang Li,Chang Chen,Zongze Zhang

Sleep plays a crucial role in restoring and repairing the body, consolidating memory, regulating emotions, maintaining metabolic and so on. Sleep deprivation is known to impair cognitive functions. In this study, we investigated the mechanisms underlying memory impairment induced by sleep deprivation through a combined metabolomic and transcriptomic analysis of hippocampus. Eight-week-old mice were selected as the study subjects and the sleep deprivation chamber was used to establish a sleep deprivation (SD) model. Novel object recognition tests (NOR), and Y-maze tests were used to assess the behavioral outcomes in mice. The hippocampus were extracted and studied using the untargeted metabolomics or transcriptomics high-throughput sequencing method. An integrative analysis was conducted to elucidate the metabolic and genetic changes. Behavioral tests showed that SD group exhibited memory impairment. Metabolomic analysis identified 84 differentially expressed metabolites (DEMs), including 12 under the positive ion mode and 72 under the negative ion mode. The analysis revealed that sleep deprivation caused abnormalities in several metabolic pathways, with particularly pronounced effects observed in glycerophospholipid metabolism, linoieic acid metabolism, alanine, aspartate, glutamate metabolism, taurine and hypotaurine metabolism, and purine metabolism. While transcriptomic analysis releaved 97 differentially expressed genes (DEGs) (51 were down-regulated and 46 were up-regulated DEGs). Integrative analysis of the metabolomic and transcriptomic identified profiles showed that sleep deprivation may regulate taurine and hypotaurine metabolism and sphingolipid metabolism, there by influencing memory. Our results prompt severe metabolic disturbances occur in the hippocampus with SD in mice, which can provide a basis for the mechanism research.

Brief disruption of activity in a subset of dopaminergic neurons during consolidation impairs long-term memory by fragmenting sleep

Lin Yan,Litao Wu,Timothy D Wiggin,Xiaojuan Su,Wei Yan,Hailiang Li,Lei Li,Zhonghua Lu,Yuantao Li,Zhiqiang Meng,Fang Guo,Fan Li,Leslie C Griffith,Chang Liu

Sleep disturbances are associated with poor long-term memory (LTM) formation, yet the underlying cell types and neural circuits involved have not been fully decoded. Dopamine neurons (DANs) are involved in memory processing at multiple stages. Here, using both male and female flies, Drosophila melanogaster, we show that, during the first few hours of memory consolidation, disruption of basal activity of a small subset of protocerebral anterior medial DANs (PAM-DANs), by either brief activation or inhibition of the two dorsal posterior medial (DPM) neurons, impairs 24 h LTM. Interestingly, these brief changes in activity using female flies result in sleep loss and fragmentation, especially at night. Pharmacological rescue of sleep after manipulation restores LTM. A specific subset of PAM-DANs (PAM-α1) that synapse onto DPM neurons specify the microcircuit that links sleep and memory. PAM-DANs, including PAM-α1, form functional synapses onto DPM mainly via multiple dopamine receptor subtypes. This PAM-α1 to DPM microcircuit exhibits a synchronized, transient, post-training increase in activity during the critical memory consolidation window, suggesting an effect of this microcircuit on maintaining the sleep necessary for LTM consolidation. Our results provide a new cellular and circuit basis for the complex relationship between sleep and memory.

Taurine Prevents Impairments in Skin Barrier Function and Dermal Collagen Synthesis Triggered by Sleep Deprivation-Induced Estrogen Circadian Rhythm Disruption

Qi Shao,Zhaoyang Wang,Yifang Li,Xun Tang,Ziyi Li,Huan Xia,Qihong Wu,Ruxue Chang,Chunna Wu,Tao Meng,Yufei Fan,Yadong Huang,Yan Yang

Sleep deprivation is a prevalent issue that disrupts the circadian rhythm of estrogen, particularly estradiol, thereby significantly affecting women’s skin health and appearance. These disruptions can impair skin barrier functionality and decrease dermal collagen synthesis. In this study, our results demonstrate that topical taurine supplementation promotes the expression of tight junction (TJ)-related proteins and enhances collagen production, effectively restoring skin homeostasis in sleep-deprived female mice. Mechanistically, taurine upregulates the expression ofTMEM38B, a gene encoding the TRIC-B trimeric cation channel, resulting in increased intracellular calcium ion levels. This, in turn, promotes the upregulation of TJ-related proteins, such as ZO-1, occludin, and claudin-11 in epidermal cells, while also enhancing the expression of type III collagen in fibroblasts, thus restoring skin homeostasis. These findings suggest that taurine may serve as an alternative to estradiol, effectively improving skin homeostasis disrupted by sleep deprivation while mitigating the potential risks associated with exogenous estrogen supplementation. Collectively, these results provide preliminary insights into the protective mechanisms of taurine against sleep deprivation-induced skin impairments and establish a foundation for its potential application in treating skin conditions related to estrogen imbalances, such as skin aging in menopausal women.

Arctium lappa L. polysaccharides (ALP) ameliorate exercise capacity and modulate gut microbiota composition in mice with acute sleep deprivation via the gut-muscle axis

Haotian Zhao,Wenlong Sun,Wenfei Cai,Chang Liu,Wei Wang,Naiyan Lu

This study investigated the effects of Arctium lappa L. polysaccharides (ALP) on exercise performance and gut microbiota composition in mice subjected to acute sleep deprivation (SD) with a focus on the gut-muscle axis. The results revealed that ALP significantly enhanced the forelimb grip strength and muscle endurance in SD mice, thereby improving their overall exercise capacity. Additionally, ALP regulated the energy metabolism by increasing blood glucose, liver glycogen, and muscle glycogen levels, while simultaneously reducing oxidative stress and inflammation. Histological analysis revealed that ALP strengthened intestinal barrier function by promoting the expression of tight junction proteins ZO-1 and Occludin. In terms of gut microbiota, ALP increased the Alpha diversity of the gut microbiota and optimized its structure. Notably, ALP significantly elevated the abundance of beneficial bacteria such as Akkermansia and Muribaculaceae, while inhibiting the proliferation of harmful bacteria like Colidextribacter and Lachnoclostridium. These findings suggest that ALP improves the exercise capacity and overall health of SD mice by modulating the gut microbiota and intestinal barrier function, providing new insights into the use of dietary interventions to alleviate health issues caused by sleep deprivation.

Transcranial near infrared therapy reprograms metabolism to rescue sleep and place cell functionality

Yue Dong,Yuping Wang,Xiaoyun Liu,Yinchang Wang,Ziteng Yue,Zhanjing Wang,Sheng Wang

Sleep disturbances exacerbate Alzheimer’s disease (AD) pathology, accelerating cognitive decline and impairing hippocampal memory functions. This study explores the therapeutic potential of transcranial near-infrared (tNIR) therapy in mitigating hippocampal dysfunction caused by chronic sleep deprivation (SD) in a tauopathy mouse model of AD. Using targeted 808 nm tNIR therapy, we observed restored cerebral blood flow, enhanced metabolic reprogramming favoring oxidative phosphorylation, and reestablished ionic homeostasis. These effects improved hippocampal oscillatory dynamics, including enhanced theta, gamma, and delta power, synchronized sharp-wave ripples (SWRs), and improved the phase-locking of place cell activity to theta and awake SWR oscillations. tNIR therapy also refined spatial encoding precision and reorganized sleep architecture, promoting slow-wave sleep while reducing REM disturbances. These findings position tNIR therapy as a promising non-invasive approach for addressing SD-driven cognitive and neural impairments, offering a novel intervention for sleep-related neurodegeneration in AD.
