Comparative analysis of antidepressant-like effects of five herbal essential oils on mice with reserpine-induced depression

Minghui Tang,Yong Ai,Ni Song,Xian Xu,Lin Geng,Dongqiu Chen,Lu Ren,Xiaolu Chen,Jingyi Chen,Lanyue Zhang,Tinggang He

The antidepressant-like effects of five herbal essential oils (EOs), Jasminum sambac (Linn.) Aiton essential oil (JSEOs), Magnolia denudata Desr. essential oil (MDEOs), Rosa rugosa Thunb. essential oil (RREOs), Aloysia citriodora Ortega ex Pers. essential oil (ACEOs) and Abies balsamea (L.) Mill. essential oil (ABEOs) were evaluated. By virtue of a reserpine-induced depression mouse model, the antidepression activities and mechanisms of these EOs were explored. The results indicated that the inhalation of EOs significantly ameliorated depression-like behaviors in reserpine-induced depression mice, including a decrease in anxiety and immobility time. JSEOs and ABEOs significantly attenuated reserpine-induced depression-like behaviors, particularly the decreased body weight and immobility time, as determined by tail suspension and forced swimming tests. Furthermore, RT-qPCR and western blotting analyses indicated that the EOs influenced brain-derived neurotrophic factor (BDNF), serotonin1A (5-HT1A), and inflammatory cytokine levels. ACEOs attenuated reserpine-induced hippocampal and neuronal damage, significantly increased glucocorticoid receptor (GR) and 5-HT1A levels, and suppressed the inflammatory response. These results provide new insights into the antidepressant effects of herbal EOs and suggest that the five EOs evaluated in this study are potential nutraceuticals for the treatment of depression.

Development and Characterization of Novel Active Chitosan Films Containing Fennel and Peppermint Essential Oils

Ting Liu,Jingfan Wang,Fumin Chi,Zhankun Tan,Liu Liu

The effects of fennel essential oil (FEO) and peppermint essential oil (PEO) on chitosan-based films were evaluated in this study. The results showed that the addition of FEO and PEO slightly increased the density and thickness, while significantly decreasing the moisture content, water swelling, and solubility properties. The color values (L,a,b, ΔEand whiteness index (WI)) of the composite films containing FEO and PEO changed obviously with a tendency toward yellowness, which was beneficial in resisting food decomposition caused by ultraviolet light. The differential scanning calorimetry (DSC) and fourier-transform-infrared (FTIR) results indicated that the addition of FEO and PEO affected the structure of the chitosan films, while the interaction between the chitosan and polyphenols in FEO and PEO established new hydrogen bonds and improved the thermal stability. The environmental scanning electron microscopy (ESEM) illustrated that the surfaces of the composite films containing FEO and PEO were smooth, but the cross-section was rougher than in pure chitosan film. Furthermore, the composite films containing FEO and PEO exhibited prominent antioxidant activity. In short, the novel active chitosan-based films with incorporated FEO and PEO present broad application prospects in fresh-cut meat or vegetable packaging. Keywords:active packaging;composite film;fennel essential oil;peppermint essential oil;antioxidant activity

Chemical Compositions of Ligusticum chuanxiong Oil and Lemongrass Oil and Their Joint Action against Aphis citricola Van Der Goot (Hemiptera: Aphididae)

Chao Zhang,Runqiang Liu,Jun He,Zhiqing Ma,Xing Zhang

In order to develop novel botanical insecticides, the joint action ofLigusticum chuanxiongoil (LCO) and lemongrass oil (LO) againstAphis citricolavan der Goot was determined systematically indoors and outdoors. The chemical profiles of LCO and LO as determined by gas chromatography–mass spectrometry (GC-MS) analysis revealed that the main compounds from LCO were (Z)-ligustilide (44.58%) and senkyunolide A (26.92%), and that of LO were geranial (42.16%) and neral (32.58%), respectively. The mixture of LCO and LO showed significant synergy againstA. citricola, with a common-toxicity coefficient (CTC) value of 221.46 at the optimal ratio of LCO to LO (4:1,w:w). Based on the results of solvents and emulsifiers screening,L. chuanxiongoil·Lemongrass oil 20% emulsifiable concentrate (20% LCO·LO EC) was developed, and its stability was confirmed with tests of cold and thermal storage. Field trials indicated that the insecticidal activity of the diluted 20% LCO·LO EC (1000 fold dilution) was comparable to conventional pesticide (20% imidacloprid EC) onA. citricolaseven days after application. Thus, the mixture of LCO and LO has the potential to be further developed as a botanical pesticide. Keywords:lemongrass oil;Ligusticum chuanxiongoil;Aphis citricolavan der Goot;botanical aphicides

Antibacterial Activity and Mechanisms of Essential Oil from Citrus medica L. var. sarcodactylis

Ze-Hua Li,Ming Cai,Yuan-Shuai Liu,Pei-Long Sun,Shao-Lei Luo

In this work, antibacterial activity of finger citron essential oil (FCEO,Citrus medica L. var. sarcodactylis) and its mechanism against food-borne bacteria were evaluated. A total of 28 components in the oil were identified by gas chromatography-mass spectrometry, in which limonene (45.36%), γ-terpinene (21.23%), and dodecanoic acid (7.52%) were three main components. For in vitro antibacterial tests, FCEO exhibited moderately antibacterial activity against common food-borne bacteria:Escherichia coli,Staphylococcus aureus,Bacillus subtilisandMicrococcus luteus. It showed a better bactericidal effect on Gram-positive bacteria than Gram-negative. Mechanisms of the antibacterial action were investigated by observing changes of bacteria morphology according to scanning electron microscopy, time-kill analysis, and permeability of cell and membrane integrity. Morphology of tested bacteria was changed and damaged more seriously with increased concentration and exposure time of FCEO. FCEO showed a significant reduction effect on the growth rate of surviving bacteria and lead to lysis of the cell wall, intracellular ingredient leakage, and consequently, cell death. Keywords:finger citron;essential oil;antibacterial;mechanism

A cinnamon and clove essential oil mix microencapsulated with hydroxypropyl-β-cyclodextrin: Study on physicochemical, antibacterial, and low salt pickles preservation properties

Rui Zeng,Ying Liu,Yue Lu,Chao Chang,Jine Wu

In order to overcome the technical challenges of poor stability and weak antibacterial effects of individual essential oil in food preservation applications, the present study aimed to encapsulate cinnamon and clove essential oil compound by using spray-drying technique. The combination of cinnamon and clove essential oils was determined to have good synergistic bacteriostatic effects by the checkerboard dilution method, and the best bacteriostatic effect could be obtained when the volume ratio was 7:3 for compounding. Microcapsules were prepared using hydroxypropyl-β-cyclodextrin (HPCD) as wall material and compound essential oil as core material, the optimal conditions for the microcapsule preparation process through a one-way test were: homogenizing speed of 8000 r/min, wall material addition of 2 %, HPCD to EO ratio of 1:3, EO to T-80 ratio of 1.5:1, and homogenizing time of 8 min. The physicochemical properties of the prepared compound essential oil microcapsules (EOM) were characterized, and the results showed that the EOM was successfully encapsulated in HPCD with good physicochemical properties, and the encapsulation rate of the prepared microcapsules was measured to be 65.82 ± 4.00 %. The thermal stability of the encapsulated EOM was improved, and volatilization of the essential oils was effectively inhibited. In addition, the EOM showed antibacterial activity against the five types of bacteria tested, and the number of surviving bacteria decreased by about 17–18 % after 72 h. The preservation experiment of low salt pickles showed that the EOM was more effective in maintaining the quality and prolonging the shelf life of the pickles compared with commercial sodium benzoate, which also demonstrated the potential application of EOM in preserving low-salt pickles. This study provides a feasible and new technical strategy for more effective application of plant essential oils in food preservation.

Chitosan/zein films incorporated with essential oil nanoparticles and nanoemulsions: Similarities and differences

Zhe Li,Xiaoxia Jiang,Hao Huang,Ao Liu,Hongning Liu,Naeem Abid,Liangshan Ming

The purpose of this study was to prepare chitosan/zein (CS/Zein) edible films reinforced with Mosla chinensis essential oils (EOs) nanoemulsions (NEs) and nanoparticles (NPs) in order to compare their properties. NEs and NPs containing EOs could be used to fabricate films with functional properties, and the films were prepared using a casting method. The influence of EO concentration and mixing methods on the physical, mechanical, and functional properties of the films was investigated. The results indicated that the films formulated with EO NEs generated favorable fundamental and functional characteristics with excellent mechanical properties, moisture barrier capacity, and significant antioxidant and antibacterial activity . In addition, the use of NEs-based films improved the release of bioactive compounds, and the mechanism of EO release was found to follow a first order model. In summary, EO NEs were more effective in preserving the fundamental and functional properties of CS/Zein nanocomposite edible films than NP-based films. These differences may reflect different forms and methods of dispersing EOs in NEs and NPs. This study demonstrated that NEs reinforced films could be used to enhance the effectiveness of EOs in food products and develop new strategies for their delivery and application.

Xiao Yu,Huaibi Zhang,Juan Wang,Junming Wang,Zhenxing Wang,Jinbo Li

Essential oils were extracted from dark-purple, red and yellow petals ofPaeonia delavayiusing Supercritical Carbon Dioxide method. The compositions of essential oils were analyzed using gas chromatography-mass spectrometry (GC-MS). Antioxidant activity assays were carried out using DPPH, ABTS- and FRAP methods. Total polyphenols and total flavonoids were measured to evaluate the in vitro antioxidant activity in addition to the volatile compounds contained in the essential oils extracted from the flower petals ofP. delavayiwith the three flower colors. A total of 194 compounds were detected from essential oils ofP. delavayiflowers, including 83 in dark-purple petals, 90 in red petals and 80 in yellow petals. These compounds mainly include alcohols, aldehydes, ketones, alkenes, alkanes, esters and polyphenols. The results showed that the volatile compounds accumulated differentially among the essential oils from the different colors of flower petals. Principal component analysis (PCA) indicated that essential oils derived from dark-purple and red petals were more closely clustered while the yellow petal essential oil was very different with both the purple-red and red. Antioxidant assays suggested that the radical scavenging activity and the iron reduction antioxidant activity in the essential oils were highly correlated with the flower petal colors. These results suggestP. delavayiflower petals are potentially good resources for high quality essential oils and natural antioxidants. Keywords:Paeonia delavayi;flower colors;essential oil;antioxidant activity

The essential oil (EO) was extracted from aerial parts with insecticidal and fungicidal activity. Herein, the hydro-distilled essential oils ofSeseli maireiH. Wolff roots were determined by GC-MS. A total of 37 components were identified, (E)-beta-caryophyllene (10.49%),β-geranylgeranyl (6.64%), (E)-2-decenal (6.17%) and germacrene-D (4.28%). The essential oil ofSeseli maireiH. Wolff had nematicidal toxicity againstBursaphelenchus xylophiluswith a LC50value of 53.45 μg/mL. The subsequent bioassay-guided investigation led to the isolation of three active constituents: falcarinol, (E)-2-decenal, and octanoic acid. The falcarinol demonstrated the strongest toxicity againstB. Xylophilus(LC50= 8.52 μg/mL). The octanoic acid and (E)-2-decenal also exhibited moderate toxicity againstB. xylophilus(LC50= 65.56 and 176.34 μg/mL, respectively). The LC50of falcarinol for the toxicity ofB. xylophiluswas 7.7 and 21 times than that of octanoic acid and (E)-2-decenal, respectively. Our findings demonstrate that the essential oil fromSeseli maireiH. Wolff roots and their isolates may be developed as a promising natural nematicide. Keywords:Seseli maireiH. Evaluation of the phytotoxic effect of the essential oil from Artemisia absinthium

Chunyu Jiang,Shixing Zhou,Lin Liu,Zokir Toshmatov,Ling Huang,Kai Shi,Chi Zhang,Hua Shao

The chemical profile and the phytotoxicity of Artemisia absinthium essential oil (EO) were investigated to evaluate its potential value as a biopesticide for food safety purposes. A total of 54 compounds were identified in A. absinthium EO, with the most abundant constituents being eucalyptol (25.59%), linalool (11.99%), and β-myrcene (10.05%). The EO, linalool, and a mixture of three major components exhibited potent suppressive activity against four receiver species; however, eucalyptol and β-myrcene showed a much weaker effect. Bioassay-guided fractionation led to the isolation of linalool as the major active compound responsible for the EO’s phytotoxicity. Subsequent scanning electron microscopy (SEM) analysis revealed that linalool significantly inhibited root-hair formation and metaxylem development. This is the first report on the determination of linalool as the major active phytotoxic compound in A. absinthium EO, as well as the elucidation of its mechanism of phytotoxicity from the perspective of root structure changes in the receiver species. Our results suggest that both the EO and its major constituents have potential value as environmentally friendly herbicides.

The Flavor Characterization of ‘Huyou’ (Citrus changshanensis) Essential Oils Extracted by Conventional and Novel Methods

Huan Cheng,Feifei Liu,Yanru Zhang,Zhiyue Ye,Jin Chen,Shiguo Chen,Xingqian Ye

The aroma of citrus is among the most appealing natural flavors. ‘Huyou’ (HY) is a hybrid citrus with a unique flavor compared to grapefruit (GF), but few studies have analyzed its characteristic flavor comprehensively. In this study, we extract six essential oils (EOs) from HY and GF peels by cold pressing (CP) and microwave-assisted hydrodistillation (MADH) and spinning cone column (SCC). Further, the flavor of six EOs was investigated by using flavoromics analysis, including e-nose, GC-MS and GC-O combined with chemometric approaches. The results showed that CP EOs exhibited a stronger citrus characteristic flavor, while MADH and SCC EOs contained more diverse volatiles. A total of 23 key odorants were identified in the GC-O-MS analysis, 12 of which were specific to HY. The flavor wheel and partial least squares regression (PLSR) revealed that floral, sweet and fruity odors were positively correlated with linalool, α-terpineol and geraniol, while fatty, green and woody odors with germacrene D, germacrene B and nootkatone. Additionally, based on orthogonal partial least squares discriminant analysis (OPLS-DA), six aroma-active compounds were screened as aroma markers to distinguish HY from GF; i.e., γ-terpinene, D-limonene, germacrene D, nootkatone, germacrene B and terpinolene. The extraction methods and citrus varieties both impact the flavor characterization of citrus EOs, and our study provides guidance on the extraction and application for citrus EOs.Keywords:‘Huyou’ (Citrus changshanensis);essential oil;flavor;aroma-active compounds

Xiaonan Zhang,Xiaocheng Zhuang,Manxin Chen,Jingrong Wang,Zhuoyu Liu,Daxiong Qiu,Jinna Wang,Yan Huang,Weina Li,Zhiwei Liu

Tannic acid and Fe 3+ were used in this study to create a polyphenol–metal network-based composite film for the preservation of Melaleuca bracteata essential oils. The inhibition rate of ABTS and DPPH free radicals reached more than 90 % at an essential oil concentration of 20 mg/mL. Additive quantities of 2.0 % essential oil nano-emulsion, 0.8 % tannic acid/Fe 3+ solution, 0.4 % microcrystalline cellulose, and 1 % chitosan were used to maximize the characteristics of the composite films. When combined with FTIR analysis, X-ray diffraction and scanning electron microscopy revealed that the composite film containing the essential oil emulsion had a more reticulated structure. The essential oil composite layer on mangoes increased the fruit shelf time to 12 days, decreased weight loss by 10.81±4.70 %, increased the amount of soluble solids by 2.03±0.31 %, and increased the amount of vitamin C by 2.18±0.09 %. A trustworthy technical method for the storage and transportation of agricultural goods is offered by this study. Graphical

Insecticidal activities of the essential oil of Rhynchanthus beesianus rhizomes and its constituents against two species of grain storage insects

Xiujuan Pan,He Xiao,Xinping Hu,Zhi Long Liu

The aim of this research was to evaluate insecticidal activities of the essential oil of Rhynchanthus beesianus rhizomes against adults of Liposcelis entomophila and Tribolium castaneum . Gas chromatography-mass spectrometry analyses revealed the presence of 44 compounds with β -eudesmol (19.1%), elemol (8.1%), α -terpineol (8.0%), methyl eugenol (6.5%), and caryophyllene (4.8%) being the major constituents. Bioactivity-directed chromatographic separation of the oil led to the isolation of four constituents, elemol, β -eudesmol, methyl eugenol, and α -terpineol. The essential oil exhibited fumigant toxicity against the adults of L. entomophila and T. castaneum with LC 50 values of 0.57 and 4.96 mg/L air while the two isolates, methyl eugenol and α -terpineol possessed fumigant toxicity against the booklice (LC 50  = 0.15 and 0.48 mg/L air, respectively) and the beetles (LC 50  = 1.81 and 4.96 mg/L air, respectively). The oil also possessed contact toxicity against the booklice and the beetles with LD 50 values of 121.56 μg/cm 2 and 54.93 μg/adult, respectively, while the two isolates β -eudesmol and elemol showed contact toxicity against L. entomophila (LD 50  = 99.21 and 35.19 μg/cm 2 , respectively) and T. castaneum (LD 50  = 35.26 and 8.89 μg/adult, respectively). The results indicate that the oil of R. beesianus rhizomes and its isolates have potential as a source for natural insecticides.
