Monoclinic Gd2O3:Eu3+ nanoparticles (NPs) possess favorable magnetic and optical properties for biomedical application. However, how to obtain small enough NPs still remains a challenge. Here we combined the standard solid-state reaction with the laser ablation in liquids (LAL) technique to fabricate sub-10 nm monoclinic Gd2O3:Eu3+ NPs and explained their formation mechanism. The obtained Gd2O3:Eu3+ NPs exhibit bright red fluorescence emission and can be successfully used as fluorescence probe for cells imaging. In vitro and in vivo magnetic resonance imaging (MRI) studies show that the product can also serve as MRI good contrast agent. Then, we systematically investigated the nanotoxicity including cell viability, apoptosis in vitro, as well as the immunotoxicity and pharmacokinetics assays in vivo. This investigation provides a platform for the fabrication of ultrafine monoclinic Gd2O3:Eu3+ NPs and evaluation of their efficiency and safety in preclinical application.

Graphical A NIR-driven Janus Cu 2 O/Au nanomotor had been constructed to explore the synergistical therapeutic effect on hepatoma carcinoma cells. The asymmetric Au modification causes uneven heating on both sides of the Cu 2 O/Au nanomotor, resulting in autonomous motion from high to low temperature, actively targeting hepatocellular carcinoma cells. Under the synergistic effects of photothermal therapy, photodynamic therapy and Cu 2 O′s own nanotoxicity, Cu 2 O/Au nanomotors exhibit highly effective killing effect on liver cancer cells. We presented a NIR-driven Janus Cu 2 O/Au nanomotor. The nanomotor has a truncated octahedral structure. By asymmetric Au evaporation, the light response range of Cu 2 O nanomotor is extended to near-infrared range, and the speed of Cu 2 O/Au nanomotors under NIR is significantly increased. In promoting apoptosis of hepatocellular carcinoma, except the nanotoxicity of Cu 2 O itself, the Au layer enhances the photothermal properties, allowing Cu 2 O/Au nanomotors to induce apoptosis in hepatocellular carcinoma cells by heating them. On the other hand, a Schottky barrier formed at the interface of Cu 2 O and Au, preventing the recombination of electrons, which makes more electrons react with biomolecules to produce toxic ROS to kill hepatocellular cells. The killing rate of hepatocellular carcinoma cells reached 87 % by the combined effect of nanotoxicity inhibition of proliferation and photothermal & photodynamic therapy (PTT & PDT). Nanomotors in combination with multiple approaches are explored as a new treatment to tumor in this article.

Nanoparticles (NPs) are extensively being used in state-of-the-art nano-based therapies, modern electronics, and consumer products, so can be released into the environment with enhancement interaction with humans. Hence, the exposures to these multifunctional NPs lead to changes in protein structure and functionality, raising serious health issues. This study thoroughly investigated the interaction and adsorption of catalase (CAT) with HfO2-NPs by circular dichroism (CD), Fourier transform infrared (FTIR), absorption, and fluorescence spectroscopic techniques. The results indicate that HfO2 NPs cause fluorescence quenching in CAT by a static quenching mechanism. The negative values of Vant Hoff thermodynamic expressions (ΔHo, ΔSo, and ΔGo) corroborate the spontaneity and exothermic nature of static quenching driven by van der Waals forces and hydrogen bonding. Also, FTIR, UV-CD, and UV–visible spectroscopy techniques confirmed that HfO2 NPs binding could induce microenvironment perturbations leading to secondary and tertiary conformation changes in CAT. Furthermore, synchronous fluorescence spectroscopy confirmed the significant changes in the microenvironment around tryptophan (Trp) residue caused by HfO2 NPs. The time depending denaturing of CAT biochemistry through HfO2-NPs was investigated by assaying catalase activity elucidates the potential toxic action of HfO2-NPs at the macromolecular level. Briefly, this provides an empathetic knowledge of the nanotoxicity and likely health effects of HfO2 NPs exposure.

Graphene family nanomaterials (GFNs) have attracted significant attention due to their unique characteristics and applications in the fields of biomedicine and nanotechnology. However, previous studies highlighted the in vitro and in vivo toxicity of GFNs with size and oxidation state differences are still elusive. Therefore, we prepared graphene (G) and graphene oxide (GO) of three different sizes (S-small, M-medium, and L-large), and characterized them using multiple surface-sensitive analytical techniques. In vitro assays using HEK 293T cells revealed that the small and large sizes of G and GO significantly reduced the cell viability and increased DNA damage, accompanying with activated reactive oxygen species (ROS) generation and induced various expressions of associated critical genetic markers. Moreover, the bacterial assays highlighted that G and GO caused strong acute toxicity on Tox2 bacteria. Effects of G were higher than GO and showed size dependent effect: L > M > S, while the medium size of GO induced mild genetic toxicity on RecA bacteria. In vivo assays revealed that exposure to G and GO caused the developmental toxicity, induced ROS generation, and activated related pathways (specifically GO) in zebrafish. Taken together, G showed stronger ability to decrease the survival rate and induce the acute toxicity, while GO showed obvious toxicity in terms of DNA damages, ROS generation, and abnormal gene expressions. Our findings highlighted that G and GO differentially induced toxicity based on their varying physical characteristics, especially sizes and oxidation state, and exposure concentrations and sensitivity of the employed in vitro and in vivo models. In short, this study provided deep insights on the negative effects of GFNs exposure.

The toxic effects of nanoparticles on individual organisms have been widely investigated, while few studies have investigated the effects of nanoparticles on ubiquitous multicommunity microbial aggregates. Here, periphyton as a model of microbial aggregates, was employed to investigate the responses of microbial aggregates exposed continuously to Fe2O3 nanoparticles (5.0 mg L–1) for 30 days. The exposure to Fe2O3 nanoparticles results in the chlorophyll (a, b, and c) contents of periphyton increasing and the total antioxidant capacity decreasing. The composition of the periphyton markedly changes in the presence of Fe2O3 nanoparticles and the species diversity significantly increases. The changes in the periphyton composition and diversity were due to allelochemicals, such as 3-methylpentane, released by members of the periphyton which inhibit their competitors. The functions of the periphyton represented by metabolic capability and contaminant (organic matter, nitrogen, phosphorus and copper) removal were able to acclimate to the Fe2O3 nanoparticles exposure via self-regulation of morphology, species composition and diversity. These findings highlight the importance of both physiological and ecological factors in evaluating the long-term responses of microbial aggregates exposed to nanoparticles.

Background Cardiovascular diseases (CVDs) have the highest mortality worldwide. Human pluripotent stem cells (hPSCs) and their cardiomyocyte derivatives (hPSC-CMs) offer a valuable resource for disease modeling, pharmacological screening, and regenerative therapy. While most CVDs are linked to significant over-production of reactive oxygen species (ROS), the effects of current antioxidants targeting excessive ROS are limited. Nanotechnology is a powerful tool to develop antioxidants with improved selectivity, solubility, and bioavailability to prevent or treat various diseases related to oxidative stress. Cerium oxide nanozymes (CeONZs) can effectively scavenge excessive ROS by mimicking the activity of endogenous antioxidant enzymes. This study aimed to assess the nanotoxicity of CeONZs and their potential antioxidant benefits in stressed human embryonic stem cells (hESCs) and their derived cardiomyocytes (hESC-CMs).Results CeONZs demonstrated reliable nanosafety and biocompatibility in hESCs and hESC-CMs within a broad range of concentrations. CeONZs exhibited protective effects on the cell viability of hESCs and hESC-CMs by alleviating excessive ROS-induced oxidative stress. Moreover, CeONZs protected hESC-CMs from doxorubicin (DOX)-induced cardiotoxicity and partially ameliorated the insults from DOX in neonatal rat cardiomyocytes (NRCMs). Furthermore, during hESCs culture, CeONZs were found to reduce ROS, decrease apoptosis, and enhance cell survival without affecting their self-renewal and differentiation potential. Conclusion sCeONZs displayed good safety and biocompatibility, as well as enhanced the cell viability of hESCs and hESC-CMs by shielding them from oxidative damage. These promising results suggest that CeONZs may be crucial, as a safe nanoantioxidant, to potentially improve the therapeutic efficacy of CVDs and be incorporated into regenerative medicine.Graphical Abstract

Inorganic nanoparticles (NPs), particularly iron oxide (IO) and gold (Au) NPs, are widely used in a variety of biomedical applications, such as diagnosis and cancer therapy. As an important component of host defense in organisms, macrophages play a crucial role in responding to foreign substances, such as nanoparticles. Thus, it is of utmost importance to understand the nanotoxicity effects on the immune system by investigating the influences of such nanoparticles. In this study, we found that macrophages can take up large amounts of amphiphilic polymer (PMA)-modified Au and IO NPs, which will induce macrophage cell vacuolization and enhance macrophage polarization. This mechanism is an essential part of the immune response in vivo. In addition, we report that smaller-sized nanoparticles (ca. 4 nm) show more significant effects on the macrophage polarization and caused lysosomal damage compared to larger nanoparticles (ca. 14 nm). Moreover, the amount of NP uptake in macrophages decreases upon trapping the PMA with PEG, resulting in reduced vacuolization and a reduced immune response. We hypothesize that vacuoles are formed in large amounts during NP uptake by macrophages, which enhances the immune response and induces macrophages toward M1 polarization. These findings are potentially useful for disease treatment and understanding the immune response when NPs are used in vitro and in vivo.

The sterilization effect of photocatalysis and biotoxicity of nanomaterial catalysts have attracted high attention. In this study, the novel visible-driven defective carbon nitride (VL/DCN) system exhibits non-photoreactivation, non-toxic superior performance compared with traditional ultraviolet radiation (UV) and UV/titanium dioxide (UV/TiO 2 ). The inactivation of antibiotic-resistant bacteria (ARB) by novel VL/DCN still reached 7 log within 4 h, and the reduction rates of aminoglycoside gene strB and tetracycline gene tetA exceeded 0.8 log and 1.2 log, respectively. Further, the sterilization mechanism and nanotoxicity were contrastively and systematically analyzed among above three systems as following. Firstly, in the VL/DCN system, reactive oxygen species (ROSs) generated from photocatalytic process leads to the destruction of cell membranes, resulting in dissolving out of potassium ion (K + ), protein and cell membrane ATP content. Thus, resistant bacteria were completely inactivated and photoreactivation disappears. In contrast, the UV only acted on bacterial DNA and existed the light resurrection. The UV/TiO 2 strictly dependent on ultraviolet light and can be used in limited scenarios. Secondly, in cell viability analysis by human lung cell line BEAS-2B experiments, the 10% inhibition of cell growth when DCN was 600 mg/L much lower than 28% inhibition of cell growth when TiO 2 was only 200 mg/L. The expression of pro-inflammatory cytokines (( Interleukin, IL) -6), IL-8, IL-1β ) under the effect of DCN was 1.5-fold, 5.7-fold and 3.7-fold lower than TiO 2 , respectively. Meanwhile, DCN induced cells to produce less ROSs, malondialdehyde (MDA), and more superoxide dismutase (SOD). Above results demonstrated that DCN has far lower cytotoxicity than TiO 2 . This study provides theoretical support for the application of photocatalytic sterilization technology and the exploration of the toxicity of nanomaterials. Environmental Implication In this study, the novel visible-driven defective carbon nitride (VL/DCN) system exhibits non-photoreactivation, non-toxic superior performance compared with traditional ultraviolet radiation (UV) and UV/titanium dioxide (UV/TiO 2 ). The VL/DCN exhibited excellent sterilization efficiency and a high rate of reduction of the E. coli resistance gene strB. The inactivation of ARB reached 7 log within 4 h without the occurrence of photoreactivation. The reduction rates of the aminoglycoside gene strB and tetracycline gene tetA exceeded 0.8 log and 1.2 log, respectively. This study provides a valuable theoretical basis for the development of a new sustainable and environmentally friendly visible light catalytic sterilization technologies.

The biological effect of soilborne nanoparticles (NPs) is a manifestation of soil-NMs-bio interactions. Soil factors are known to restructure NPs surfaces and thus influence the nanotoxicity. However, the mechanisms by which environmental factors affecting nano-bio interactions to aggravate or alleviate nanotoxicities are poorly understood. Herein, we compared the toxicity of TiO 2 NPs (nTiO 2 ) in five soils using the model nematode ( Caenorhabditis elegans ), and investigated the variation of nano-bio interactions under different conditions. A correlation analysis showed that pH and dissolved organic matter (DOM) were dominant regulators of nTiO 2 toxicity. At the nano-bio interface, low pH (5.0) led to nTiO 2 adhesion to micron-sized furrows and aggravated dermal wrinkling, while humid acid (HA) alleviated these impacts. Mechanically, low pH increased nTiO 2 adhesion through enhanced electrostatic attraction and subsequent stimulation of mucin and collagen synthesis, resulting in a positive feed cycle of pH-dependent contact nanotoxicity. HA not only prevented nTiO 2 adhesion onto the epidermis due to its negative charge, but also relieved the overstimulation of stress response pathways, thereby alleviating nanotoxicity. These findings broaden our knowledge of how NPs induce contact toxicity in soil invertebrates through specific biointerfacial interactions, and highlight the important role of DOM in alleviating the combined hazards of NPs and soil acidification.

Background Despite the increasing interest in combination therapy for the treatment of cancer, controlled delivery of different therapeutics with high body-clearance efficacy and cancer cell specificity remained a great challenge. In this study, a novel codelivery system was synthesized through one-pot coordination-driven self-assembly of 2-methylimidazole, zinc ion and chemotherapeutic drug (doxorubicin, DOX), followed by a surface decoration of photothermal agent (indocyanine green, ICG). To improve the targeting specificity performance, folic acid-conjugated polyethylene glycol (FA-PEG) antennas was connected on the surface of nanoparticles. Results The hybrid nanoparticles keep stable under neutral physiological condition but decompose when exposed to acidic environment, resulting in the on-demand release of DOX and ICG for chemo-photothermal combined therapy. Moreover, by switching the initial large size (~ 94 nm) to an ultrasmall size (∼10 nm) in cancer cells, the nanoparticles hold great potential to avoid nanotoxicity for clinical applications. Conclusion sThis work provides a new strategy for co-delivery of different therapeutics for combined cancer therapy with high cancer cell specificity and low nanotoxicity.

In recent years, the release of nanomaterials pollutants to water bodies, to a great extent, attributed to anthropogenic activities. Their impacts on aquatic organisms as well as nanomaterial monitoring and bioremediation using organism have drawn much attentions. However, studies on relationship of nano-contaminants and aquatic organisms are very scarce. Our results showed that titanium dioxide nanoparticles (TiO 2 -NPs) and Multi-walled carbon nanotubes (MWCNTs) caused an obvious cell decreases on the whole, but a significant increase at 48 h TiO 2 -NPs exposure, indicating a resistant mechanism in ciliates for nano-toxic. Besides, MWCNTs was more toxic to Pseudocohnilembus persalinus than that of TiO 2 -NPs in terms of EC 50 value. It is firstly found that P. persalinus ingested and released TiO 2 -NPs through cytostome and cytoproct, which might be the reason that TiO 2 -NPs less toxic than MWCNTs. The significantly increased superoxide dismutase (SOD) and glutathione S-transferase (GST) enzyme activities and expression levels were evaluated by reactive oxygen species ROS generation, which demonstrated that P. persalinus antioxidant defense enzyme played roles on nano-toxic resistant in ciliates. Moreover, the integrated biomarker response (IBR) was also determined, which demonstrated that MWCNTs had comparatively higher values than those of TiO 2 -NPs after higher concentration exposure to ciliates. In addition, it was confirmed by the present work that sod , gst and cat played different roles on immunity, and the sensitivity of cat gene expression to these two nanomaterials exposure was dissimilar. Damages of shrunk as well as losses of cilia on the cell surface caused by TiO 2 -NPs and MWCNTs exposure in P. persalinus using SEM revealed possible physical hazards of aggregated nanomaterials. Our findings will be helpful to understand the effect mechanisms of NPs on ciliates, and also demonstrated the possibility of P. persalinus as bio-indicator of nanomaterials in aquatic and potentials on bioremediation.

Understanding the environmental health and safety of nanomaterials (NanoEHS) is essential for the sustained development of nanotechnology. Although extensive research over the past two decades has elucidated the phenomena, mechanisms, and implications of nanomaterials in cellular and organismal models, the active remediation of the adverse biological and environmental effects of nanomaterials remains largely unexplored. Inspired by recent developments in functional amyloids for biomedical and environmental engineering, this work shows their new utility as metallothionein mimics in the strategically important area of NanoEHS. Specifically, metal ions released from CuO and ZnO nanoparticles are sequestered through cysteine coordination and electrostatic interactions with beta-lactoglobulin (bLg) amyloid, as revealed by inductively coupled plasma mass spectrometry and molecular dynamics simulations. The toxicity of the metal oxide nanoparticles is subsequently mitigated by functional amyloids, as validated by cell viability and apoptosis assays in vitro and murine survival and biomarker assays in vivo. As bLg amyloid fibrils can be readily produced from whey in large quantities at a low cost, the study offers a crucial strategy for remediating the biological and environmental footprints of transition metal oxide nanomaterials.
