Hydroxy-α-sanshool (HAS), hydroxy-β-sanshool (HBS), hydroxy-γ-sanshool (HRS), and γ-sanshool (RS) are the key components from theZanthoxylumgenus, processing a range of pharmacological activities. The present study investigated the protective capacities of four sanshools on a dextran sulfate sodium (DSS)-induced model of ulcerative colitis (UC). The results showed that sanshool administration alleviated the colitis symptoms by reducing body weight loss and disease activity index (DAI) score, increasing the colon length, and improving colonic injury and the change in immune organ weight. Furthermore, sanshools enhanced the antioxidant enzyme activities, and RS exhibited the lowest effect on the improvement in total antioxidative capacity (T-AOC) and antioxidant abilities compared to the other three sanshools. The p65 nuclear factor κB (p65 NFκB) signaling pathway was inhibited to prevent hyperactivation and decreased the production of inflammatory factors. The gut barrier function in DSS-induced mice was restored by increasing goblet cell number and levels of tight junction proteins (zonula occludens-1, occludin, and claudin-1), and the levels of protein in HAS and HRS groups were higher than that in the HBS group, significantly. The analysis of gut microbiota suggested that sanshool administration significantly boosted the abundance ofLachnospiraceae,Muribaculaceae,Oscillospiraceae, andAlistipesand reduced the level ofBuchnerain colitis mice. Collectively, the sanshool treatment could ameliorate colitis by resisting colon injury and regulating intestinal barrier dysfunction and gut microbiota dysbiosis; meanwhile, HRS and HAS have better improvement effects. Keywords:sanshool;colitis;intestinal barrier;p65;intestinal flora

In actual production, the restraint stress caused by crowded and narrow spaces, as well as operations such as capture and transportation, can lead to intestinal inflammation and affect animal growth performance, damaging the economic benefits of livestock farms. Isovaleric acid, a type of short-chain fatty acid (SCFA), has been found to be useful in enhancing calf digestion and inhibiting the expression of inflammatory factors. In this study, we first constructed a mouse model of chronic restraint stress (CRS) and fed a diet supplemented with sodium isovalerate to mice with CRS to investigate the mitigating effect of exogenously added isovalerate on mice with chronic restraint stress. An inflammation model was then constructed using piglet jejunal epithelial cell line-J2 (IPEC-J2) to simulate intestinal inflammation after stress, and to investigate the mitigating effect of isovaleric acid on inflammation as well as the repairing effect on intestinal barrier. The results showed that feeding sodium isovalerate significantly increased the weight gain as well as the body weight growth rate of CRS mice from 1-28 days ( P < 0.05), and extremely significantly decreased the feed intake and feed to gain ratio of CRS mice ( P < 0.01). In addition, feeding sodium isovalerate ameliorated the decrease in organ index in mice caused by CRS and significantly increased leg muscle index in CRS mice ( P < 0.05). Meanwhile, feeding sodium isovalerate improved depressive behavior and promoted 5-HT secretion in the brain and hypothalamus of CRS mice. Besides, feeding sodium isovalerate ameliorated intestinal inflammation in CRS mice, as evidenced by extremely significantly decreased the expression of colonic inflammatory factors and increased the expression of tight junction proteins ( P < 0.01). It was found that the addition of isovaleric acid extremely significantly reduced the expression of inflammatory factors and the phosphorylation level of NF-κB p65 in inflammatory cells ( P < 0.01), extremely significantly increased the survival rate of inflammatory cells ( P < 0.01) and the expression of tight junction proteins ( P < 0.05), improved the morphology of inflammatory cells. After the addition of short-chain fatty acid receptor GPR41 and GPR43 antagonists, the resistance effect of isovaleric acid on inflammation was inhibited. The above results suggest that isovaleric acid has an ameliorating effect on CRS in mice and inhibits NF-κB activation thereby exerting its anti-inflammatory effect.

Objective: The aims of the present study were to determine the efficacy of edible traditional Chinese medicines (ETCMs) in treating constipation, verify their laxative effects, and conduct preliminary investigations into their mechanisms of action. Methods: ICR mice were treated with loperamide to induce constipation, and various fecal parameters, including fecal volume, water content, and intestinal transport function, were measured in these constipation model mice to screen for ETCMs with laxative properties. The mechanism of action was preliminarily explored by examining changes in the intestinal mucosal structure, protein expression levels, and alterations in intestinal flora composition. Results: In ICR mice with loperamide-induced constipation, Elsholtzia ciliata aqueous extract (ECAE) and Hovenia dulcis aqueous extract (HDAE) significantly ameliorated constipation symptoms, mitigated colonic pathological tissue damage, significantly increased the expression levels of proteins associated with the promotion of intestinal peristalsis [Stem Cell Factor Receptor (c-Kit) and Stem Cell Factor (SCF)] and the maintenance of the intestinal barrier [Zonula Occludens-1 (ZO-l), Occludin and Claudin-l], and promoted beneficial intestinal bacterial colonization. Conclusion: ECAE and HDAE ameliorated constipation in mice, and their mechanism of action may be related to the increased abundance of intestinal bacteria such as Turicibacter, Olsenella, and Odoribacter, which contribute to higher butyrate production. This increase in butyric acid reduces inflammation, improves intestinal barrier function, and increases the abundance of beneficial intestinal bacteria.

The aim of the present study was to evaluate the effect of chlorogenic acid (CGA) on the mucosal barrier function and tight junction proteins in colitic rats. CGA (60 mg/kg body weight) significantly decreased the ratio of lactulose to mannitol (L/M) (P < 0.05), and endotoxin level (P < 0.01) in CGA + colitis group, compared to colitis group. CGA supplementation in CGA + colitis group increased the expression of occludin (P < 0.05), claudin-1 (P < 0.01), ZO-1 (P < 0.05), decreased the expression of myosin light chain kinase (MLCK), compared to colitis group. CGA also increased the phosphorylation level of occludin (P < 0.01) at tight junction, and decreased the phosphorylation level of occludin (P < 0.05) and claudin-1 (P < 0.05) on the basolateral membranes. These results indicate that CGA supplementation attenuates colonic barrier damage by decreasing MLCK expression and promoting molecular dynamic distribution of tight junction proteins in colitic rats.

Ethnopharmacological relevance Bazi Bushen Capsule (BZBS), a traditional Chinese medicine formulation composed of multiple bioactive herbal components, has been validated in multicenter randomized double-blind controlled trials for its potent anti-aging properties. Previous studies from our group have demonstrated that BZBS effectively restores gut microbiota homeostasis and attenuates the impairment of intestinal barrier function, thereby ameliorating age-related cognitive decline. However, the specific molecular mechanisms by which BZBS modulates key microbial-metabolite networks to delay brain aging remain poorly understood and warrant further investigation. Aim of the study This study aims to elucidate the key microbiota-metabolite networks through which BZBS improves cognitive function and delays brain aging in senescence-accelerated mouse-prone 8 (SAMP8) mice. Materials and methods Eight-week-old male SAMP8 mice were used as experimental models, randomly divided into Model, BZ-low (0.5 g/kg/d BZBS), BZ-high (1 g/kg/d BZBS), and RAPA (2 mg/kg/d rapamycin) groups. Senescence-accelerated mouse resistant 1 (SAMR1) mice served as the control group. Cognitive function was assessed using the Barnes Maze test and the three-chamber social test. The structural damage and pathological changes in the brain tissue were evaluated through transcranial Doppler, micro-computed tomography, Nissl staining, and Western blot analysis. Next, the intestinal barrier function was detected by hematoxylin-eosin (HE) staining, periodic acid-Schiff (PAS) staining, and immunofluorescence (IF) staining. Characteristic bacteria were identified by 16S rRNA sequencing, and metabolomic profiling was performed using non-targeted metabolomics. Akkermansia muciniphila (Akk) was cultured, and fecal microbiota transplantation (FMT) was employed to evaluate its contribution to intestinal barrier function. Results The study revealed that BZBS therapy not only enhances cognitive capabilities but also restores the intestinal barrier function. Akk was identified as a key regulatory agent mediating the therapeutic effects of BZBS. BZBS administration significantly increased the abundance of Akk and modulated its metabolite profile, particularly components associated with spermidine, thereby reinforcing the intestinal barrier and mitigating age-related cognitive decline. Furthermore, this study demonstrated that Akk, administered via fecal microbiota transplantation, alleviated dextran sulfate sodium (DSS)-induced colitis.. Conclusion The results showed that BZBS capsule, a traditional Chinese medicine, may delay brain aging in SAMP8 mice by modulating Akk and its spermidine production.

Quorum sensing (QS) is an essential pathway for bacterial communication through autoinducers (AIs). The luxS gene involved in QS has been proven to regulate different behavior of Lactobacillus rhamnosus GG (LGG), whereas it is unclear whether the AI-2/LuxS system have beneficial effects the host by regulating LGG. Our aim was to investigate whether luxS mutation affects the ability of LGG to maintain intestinal microbiota homeostasis, intestinal barrier integrity and inflammatory immune responses in mice infected with Escherichia coli ( E. coli ). This study showed that both wild-type (WT) and luxS mutant (ΔluxS) of LGG can alleviate changes in the microbiota caused by E. coli by down-regulating the relative abundance of Parasturtella . Supplementation with WT and ΔluxS increased the CD4 + /CD8 + ratio in spleen, improved goblet cell numbers and enhanced Muc 2 secretion. Moreover, WT and ΔluxS decreases proinflammatory mediator levels by inhibiting MAPK and NF-κB signal transduction.

Inflammatory bowel diseases (IBDs) are characterized by dysregulated inflammatory responses, increased reactive oxygen species (ROS), and compromised intestinal barrier function. Current clinical treatments often yield suboptimal responses and significant side effects. Given the great potential of Akkermansia muciniphila (AKK) bacterial membrane components in IBD treatment and the possible pathogenicity of live bacteria, to essentially address these challenges, we develop a natural membrane-based single-cell coating, incorporating a self-assembled AKK bacterial membrane (AKM) and astaxanthin (AST), forming a protective biological barrier on probiotics. Utilizing Escherichia coli Nissle 1917 (EcN) for the oral delivery system, we engineer AKM-AST@EcN nanocoated probiotics, demonstrating exceptional gastrointestinal (GI) tract tolerance and robust bioactivity. Critically, our findings highlight the potential of AKM-AST@EcN as a promising therapeutic approach for IBD treatment while also presenting a distinctive strategy for probiotic oral delivery systems.

N-glycolylneuraminic acid (Neu5Gc) is a specific factor in red meat that induces intestinal disease. Our aim was to investigate the effect of Neu5Gc on the intestinal barrier as well as its mechanism of endocytosis and exocytosis. Ten specific inhibitors were used to explore the mechanism of Neu5Gc endocytosis and exocytosis by Caco-2 cells. Amiloride hydrochloride and cytochalasin D had the strongest inhibitory effect on the endocytosis of Neu5Gc. Sodium azide, dynasore, chlorpromazine hydrochloride, and nystatin also inhibited Neu5Gc endocytosis. Dynasore exhibited a stronger inhibitory effect than that of chlorpromazine hydrochloride or nystatin alone. Exocytosis inhibitors, including nocodazole, brefeldin A, monensin, and bafilomycin A, inhibited the transmembrane transport of Neu5Gc. Monensin promoted the exocytosis of Neu5Gc from Caco-2 cells. In another experiment, we observed no significant inhibitory effects of monensin and brefeldin A. Dietary concentrations of Neu5Gc induced prominent damage to intestinal tight junction proteins zonula occludens-1 (ZO-1), occludin, and claudin-1 and promoted the phosphorylation of IκB-α and P65 to activate the canonical Nuclear Factor kappa-B (NF-κB) pathway. Neu5Gc increased the RNA levels of pro-inflammatory factors IL-1β, IL-6, and TNF-α and inhibited those of anti-inflammatory factors TGF-β and IL-10. BAY, an NF-κB signaling pathway inhibitor, attenuated these changes. Reductions in the levels of ZO-1, occludin, and claudin-1 were recovered in response to BAY. Our data reveal the endocytosis and exocytosis mechanism of Neu5Gc and prove that Neu5Gc can activate the canonical NF-κB signaling pathway, regulate the transcription of inflammatory factors, thereby damaging intestinal barrier function.Keywords:Neu5Gc;transport mechanisms;intestinal barrier function;NF-κB;cytotoxicologyKey Contribution:Systematically studied the absorption and transportation mechanisms of Neu5Gc in intestinal epithelial cells. Neu5Gc could activate the canonical NF-κB signaling pathway, regulate the transcription of inflammatory factors, and destroy the intestinal barrier function.

Morchella esculenta polysaccharides (MEPs) are known to have multiple bioactive properties, including immunomodulatory, anticancer, antioxidant, anti-inflammatory, etc. In this study, we assessed the impact of MEPs on dextran sulfate sodium (DSS)-induced colitis in mice, focusing on intestinal barrier and microbiota modulation. Using NMR analysis, MEPs were found to predominantly consist of 1,4,6- α - d -Glc p and 1,4- α - d -Glc p . MEPs were able to significantly alleviate weight loss, reduce colon length shortening, and mitigate colon pathology in mice. Notably, MEPs suppressed elevated pro-inflammatory cytokines levels but boosted anti-inflammatory cytokine levels. Besides, MEPs modulated gut microbiota, enhancing microbial diversity and promoting homeostasis compared to untreated. Furthermore, there was a higher relative abundance of beneficial bacteria, indicating that MEPs could enhance gut microbiota composition. Collectively, MEPs have promising therapeutic potential in preventing colitis, and therefore could be developed as a novel nutraceutical strategy.

Probiotics offer a promising strategy to address the dysfunction of the intestinal mucosal barrier and dysregulation of the gut microbiota in inflammatory bowel disease (IBD). However, the low viability and poor adhesion of probiotics in complex gastrointestinal environments pose significant challenges. To tackle these issues, we designed a specialized protective nano-coating (PDA@CeO 2 ) using biointerfacial phenolic assembly combined with nanozymes for Bifidobacterium bifidum (B.B.). Characteristic peaks of CeO 2 nanoparticles were detected on B.B. via XRD analysis, while SEM and TEM images confirmed the successful attachment of CeO 2 nanoparticles to the probiotic surface. The nano-coating (PDA@CeO 2 ) simultaneously provides B.B. with high adhesion in the intestine, strong tolerance in complex gastrointestinal environments, and the ability to scavenge excess reactive oxygen species (ROS) due to its excellent mucoadhesive ability and high nanozyme activity. Specifically, the protection provided by nano-coating against simulated gastric fluid (SGF, pH 1.2) resulted in cell survival rates approximately 9.4 times higher than those of unprotected B.B. after 1 h of exposure. In IBD mouse models, the combination of PDA@CeO 2 and B.B. demonstrated excellent therapeutic effects, promoting gut barrier repair. Additionally, an increase in Muribaculaceae and Prevotellaceae_UCG-001 and a decrease in Desulfovibrionaceae reshaped the intestinal flora, reducing recurrence. This study highlights the potential of enhancing probiotic functionality through targeted design of protective nano-coatings for IBD therapy.

Constipation is a common gastrointestinal condition. Lactiplantibacillus plantarum T34 has been demonstrated efficacy in relieving loperamide-induced constipation in mice model. Oral intervention of L. plantarum T34 (1 × 10 8 CFU/mouse/day) resulted in improved defecation and enhanced intestinal morphology. Mechanistic studies revealed that L. plantarum T34 strengthend the intestinal barrier through the upregulation of tight junction protein genes, specifically Claudin-1 , as well as mucosal barrier proteins such as Muc2 and Muc3 . Additionally, L. plantarum T34 modulated gut homeostasis by influencing the expression of aquaporin genes, levels of the gut neurotransmitter serotonin (5-HT), and oxidative stress markers. Furthermore, L. plantarum T34 effectively restored the composition of the gastrointestinal microbiota, addressing dysbiosis linked to constipation. These findings suggested that L. plantarum T34 may serve as a promising probiotic intervention for managing constipation, offering a safe and effective alternative to traditional treatments.

Constipation is a prevalent intestinal disorder that significantly diminishes patients' quality of life and predisposes them to various serious health conditions. Current therapeutic approaches for constipation frequently result in drug dependence and adverse effects. In this study, we present evidence that licorice water extract (LWE), a naturally derived compound from Glycyrrhiza uralensis Fisch, effectively ameliorates constipation in mice induced by diphenoxylate (DIP). Our findings indicate that oral administration of LWE (82 mg/kg/d) substantially reduces defecation time, enhances intestinal motility, and increases fecal water content in constipated mice. Furthermore, LWE elevates the levels of excitatory neurotransmitters such as serotonin (5-HT) and motilin (MTL), while concurrently decreasing the levels of inhibitory neurotransmitters including vasoactive intestinal peptide (VIP), nitric oxide (NO) as well as Lipopolysaccharide (LPS). Mechanistically, compared to the model control group, the LWE treatment upregulates the expression of ZO-1, Occludin , and MUC2 genes; repairs DIP-induced damage to the intestinal barrier; increases aquaporin 3 ( AQP3 ) protein expression; downregulates Toll-like receptor 4 ( TLR4 ); mitigates low-grade intestinal inflammation; and restores colonic goblet cell populations. Additionally, LWE is metabolized by gut microbiota, thereby restoring dysbiosis associated with DIP-induced constipation. In conclusion, our results suggest that LWE alleviates constipation through mechanisms involving intestinal barrier repair and gut microbiota modulation, highlighting its potential as an alternative therapeutic option for this condition. In response to this, we have developed a convenient gel-type candy (LWEI) with licorice extract and inulin as the main ingredients, which has shown good efficacy in relieving constipation.
