This work aims to resolve the environmental pollution caused by petroleum ether in the production of pure nuciferine from lotus leaf by applying bio-based solvent (D-limonene) and green solvent (ethyl acetate). Some obvious advantages were observed, over 97% of nuciferine could be extracted by vortex assisted two-phase solvent extraction based on multi-stage strategy (MS-VTSE), over 90% of nuciferine could be recovered by solid phase extraction (SPE), and over 97% of nuciferine could be obtained by pH-dependent liquid-liquid extraction (pH-LLE). It follows that the newly developed method can not only avoid the use of petroleum ether, but also increase the yield of the whole process by 14% (57%) in comparing with the result of the original one (50%) reported in our previous study. As D-limonene could by recycled conveniently, solvents replacement has little effect on the cost of production. In a word, the newly proposed method has advantages in environmental protection and solvent saving, thus, is fitting for the green and low-cost production of pure nuciferine from lotus leaf.

To achieve high catalytic activity and stability with low noble-metal loadings on green and natural supports has attracted tremendous attention in the past few years. Herein, a facile strategy is exploited to synthesize Au nanoclusters (with size of ∼1.4 nm) which are homogeneously distributed on carbonized natural lotus leaf sheets with less mass. Compared to the pure AuNCs, the AuNCs@carbonized lotus leaf (AuNCs@C-lotus leaf) micro/nanohybrid with the unique papillae structures and large surface area not only exhibits better catalytic activity for the reduction of 4-nitrophenol (4-NP), but also prevent the aggregation of AuNCs. In addition, the catalytic performance of AuNCs@C-lotus leaf catalyst increased with the temperature increasing of the reaction temperature. Moreover, the AuNCs@C-lotus leaf catalyst can be packed into a filtering platform to fabricate a fixed-bed system, with which the catalytic conversion could reach 71.7% for 0.2 mM 4-NP solution that pass through at flow rate of 1 mL/min. Furthermore, the AuNCs@C-lotus leaf catalyst shows excellent recyclability and only slight loss of catalytical activity is observed after recycling for five successive times. This mild and environmental friendly methodology could be generalized to the synthesis of other metallic nanoparticles@carbonized botanical leaf micro/nanohybrids as promising heterogeneous catalysts for varied catalytic reactions.

Polysaccharides exist as one of the most abundant components in lotus leaves, which attract increasing attention owing to their promising health-promoting benefits. In this study, the digestive and microbial degradation characteristics of lotus leaf polysaccharides (LLP) were studied by using an in vitro gastrointestinal model. The results suggested that LLP was stable in the human upper gastrointestinal tract in vitro according to its digestive stabilities at different simulated digestion stages. Conversely, the indigestible LLP (LLPI) could be remarkably utilized by intestinal microbiota in human feces during in vitro fermentation, and its fermentability was 58.11% after the in vitro fermentation of 48 h. Indeed, the microbial degradation characteristics of LLPI during in vitro fermentation by human fecal inoculum were revealed. The results showed that the content of reducing sugars released from LLPI obviously increased from 0.498 to 2.176 mg/mL at the initial fermentation stage (0–6 h), and its molecular weight sharply decreased from 4.08 × 10 4 to 2.02 × 10 4 Da. Notably, the molar ratios of arabinose (Ara), galactose (Gal), and galacturonic acid (GalA) in LLPI decreased from 2.89 to 1.40, from 5.46 to 3.72, and from 21.24 to 18.71, respectively, suggesting that the utilization of arabinose and galactose in LLPI by intestinal microbiota was much faster than that of galacturonic acid at the initial fermentation stage. Additionally, LLPI could remarkably regulate gut microbial composition by increasing the abundances of several beneficial microbes, including Bacteroides, Bifidobacterium, Megamonas, and Collinsella , resulting in the promoted generation of several short-chain fatty acids, especially acetic, propionic, and butyric acids. The findings from the present study are beneficial to better understanding the digestive and microbial degradation characteristics of LLP, which indicate that LLP can be used as a potential prebiotic for the improvement of intestinal health.

Interfacial solar vapor generation is promising and sustainable for seawater desalination. However, the performance degradation caused by salt accumulation has hindered the development of the interfacial solar vapor generator severely. In this paper, we have designed a lotus leaf like vertical hierarchical solar vapor generator (LSG) for stable evaporation of high-salinity brine. The LSG is assembled by Si/PPy-PVA sponge and multi-root columnar PVA sponge by mimicking the structure of the lotus leaf. The Si/PPy-PVA sponge is fabricated by successively depositing the PPy layer and silane layer on the PVA sponge and acts as the absorber of LSG. LSG enables a stable evaporation rate of 1.35 ± 0.02 kg m −2 h −1 out of 26.6 wt% brine under 1 sun for over 15 days. Besides, LSG demonstrates high efficiency of 92.6% under 0.25 sun. LSG is also tested to treat different industrial dyeing wastewater with the highest evaporation rate of 1.38 ± 0.01 kg m −2 h −1 . The condensed water produced by LSG has reached the Class IV Surface Water Standards. Our work provides a new guideline to design high-efficiency solar vapor generators for stable evaporation of high-salinity brine.

Ethnopharmacological relevance Dopamine receptors are long-standing primary targets in the treatment of mental diseases and there is growing evidence that suggests relationships between obesity and the dopamine system, especially dopamine D1 and D2 receptors. Leaves of Nelumbo nucifera Gaertn. (lotus leaves) have been medically used for helping long-term maintenance of weight loss. Whether and how components of lotus leaves function through the dopamine receptors remains unclear. Aim of the study This work aimed to discover dopamine receptor-active alkaloids isolated from the lotus leaves, to evaluate their potencies and to analyze their structure activity relationship . Materials and methods Dried lotus leaves were prepared and total extract was divided into alkaloids and flavones . Eight alkaloids were separated and characterized by a combination of high-performance liquid chromatography, quadrupole time-of-flight mass spectrometry and nuclear magnetic resonance, and assayed by a fluorometric imaging plate reader platform. Human embryonic kidney 239 cell lines expressing dopamine D1, D2 and serotonin 2A (5-HT2A) receptors, respectively, were cultured and used in the assay. Results Alkaloids in the lotus leaves were the bioactive phytochemicals and inhibited dopamine from accessing the D1 and D2 receptors. All eight compounds functioned as D1-receptor antagonists and except N-nornuciferine, seven alkaloids functioned as D2-receptor antagonists. (S)-coclaurine and (R)-coclaurine are optical isomers and antagonized both D1 and D2 with equivalent potencies, suggesting that the optical rotation of the methylene linker in the monobenzyl isoquinoline backbone did not influence their activity. Among the eight alkaloids, O-nornuciferine was the potent antagonist to both receptors (the lowest IC 50 values, D1: 2.09 ± 0.65 μM and D2: 1.14 ± 0.10 μM) while N-nornuciferine was found to be the least potent as it moderately antagonized D1 and was inactive on D2. O-nornuciferine was also a 5-HT2A antagonist (IC 50 ~20 μM) while N-nornuciferine had no activity. These hinted the importance of a methyl group attached to the nitrogen atom in the aporphine backbone. Armepavine showed a nearly 10-fold selectivity to D2. Conclusions In this work, eight alkaloids were isolated from the leaves of Nelumbo nucifera Gaertn. and assayed on the D1 and D2 receptors. They were D1/D2 antagonists with IC 50 values in the mid- to low-micromolar range and O-nornuciferine was the most potent alkaloid among the eight. This family of alkaloids was biochemically evaluated on the dopamine receptors by the same platform for the first time.

Leaf of Nelumbo nucifera Gaertn. ( N. nucifera ) has been widely used as the main ingredient in lipid-lowering herbal teas and some prescriptions in China due to their excellent hypoglycemic and hypolipidemic effects. However, the active components responsible for these beneficial properties and their mechanisms remain unexplored. In this work, the N. nucifera leaf extracts significantly promoted the glucose consumption of HepG2 cells, and also exhibited remarkable inhibitory activities against α-glucosidase, pancreatic lipase, and COX-2. Furthermore, the top four potential active compounds ( N -nornuciferine, Nuciferine, 2-Hydroxy-1-methoxyaporphine, and Isorhamnetin 3-O-glucoside) targeting the above three enzymes were screened out by bioaffinity ultrafiltration with multiple targets coupled with HPLC-MS/MS. The enzyme inhibitory activities of candidate compounds were verified by enzyme inhibition assay and molecular docking. In addition, molecular docking revealed the binding information between the candidate molecules and enzymes. The current study provided valuable information in discovering functional active ingredients from complex medicinal plant extracts.

Dynamic high pressure microfluidization (DHPM)-assisted extraction (DHPMAE) of lotus ( Nelumbo nucifera ) leaves polysaccharides (LLPs) was optimized by response surface methodology. The optimal extraction conditions were: liquid/solid ratio of 35:1 (v/m, mL/g), processing pressure of 180 MPa, processed two times, extraction temperature of 76 °C, extraction time of 50 min. Under the optimal extraction conditions, DHPMAE produced a higher polysaccharides yield (6.31%) than leaching (2.95%). Scanning electron microscope (SEM) analysis revealed that DHPM could reduce the particles size and make the surface more unconsolidated. The LLPs prepared by both methods showed similar FT-IR spectrum, and were consisted of the same monosaccharides , including rhamnose , fucose , arabinose , xylose , mannose , glucose and galactose . The content of each monosaccharide in extracts, however, was quite different. The average molecular weight of LLPs prepared by DHPMAE is 550 kDa, smaller than 578 kDa obtained by leaching. The LLPs prepared by DHPMAE exhibited stronger DPPH scavenging ability (IC 50 value of 0.38 mg/mL), HO scavenging ability (IC 50 value of 0.61 mg/mL) and reducing power. Therefore, DHPMAE can be a promising alternative to traditional extraction techniques for polysaccharides from plants, and lotus leaves might be a potential resource of natural antioxidants.

Nuciferine is an important drug candidate for the treatment of many diseases. However, there is no general method for its low - cost production. In this work, a feasible method for the production of nuciferine from lotus leaf, using ultrasonic-assisted extraction-solid phase extraction (UAE-SPE) as extraction and cleanup procedure, was developed. Petroleum ether and silica gel have been successfully used as extraction solvent and adsorbent to integrate UAE with SPE, respectively. Except for filtration, no treatment (e.g. concentration and redissolution, etc) was needed on UAE extract before SPE and the effluents obtained in the loading process of SPE could be used as UAE extraction solvent without purification. No obvious decline in the extraction efficiency of UAE and adsorption capacity of SPE was observed at least for 5 runs, which provides a feasible way for the continuous production of nuciferine in industry, i.e. Cyclic UAE-SPE. Moreover, SPE column could be conveniently regenerated and reused without significant decline in its adsorption capacity at least for 5 cycles, which can be used to reduce the cost of the whole system further. In comparison with other cleanup procedures, Cyclic UAE-SPE showed apparent advantages in energy conservation and emission reduction. LLE and crystallization were applied to separate nuciferine from other impurities further. Under optimum conditions, the total recovery rate of nuciferine with a purity over 90.0% from lotus leaf reached 50.1%. All in all, the developed method has advantages in convenient operation, low cost, and high efficiency, thus, is fitting for the production of high purity nuciferine.

In this work, a superparamagnetic Fe 3 O 4 -graphene oxide (MGO) nanocomposite was prepared by one-step chemical co-precipitation method, and characterized by transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), vibrating sample magnetometer (VSM), powder X-ray diffraction (PXRD), Raman spectroscopy and nitrogen adsorption–desorption curve. The as-prepared MGO was used to adsorb nuciferine, and the adsorption kinetic, isotherm and reusability of MGO were also investigated. The results showed that the adsorption of nuciferine on MGO reached its equilibrium very quickly (within 10 min) due to the two-dimensional carbon nanostructure of GO. In comparison with MGO, five conventional sorbents, i.e., macroporous resin D-101, silica gel, reverse phase silica gel (RP-C18) and cation exchange resin and polyamide, were also used to evaluate their adsorption capabilities. Therefore, MGO combined the advantages of both superparamagnetic particle and GO, i.e., easy separation and high absorption capacity. Finally, MGO was successfully applied to enrichment and separation of nuciferine in the extract of Nelumbinis Folium (Lotus leaf).

Enzyme immobilization by metal organic frameworks (MOFs) is an efficient way for screening active constituents in natural products. However, the enzyme’s biocatalysis activity is usually decreased due to unfavorable conformational changes during the immobilization process. In this study, sodium cholate was firstly used as the modifier for zeolitic imidazolate framework-8 (ZIF-8) immobilized lipase to increase both the stability and activity. More importantly, with the help of solubilization of sodium cholate, a total of 3 flavonoids and 6 alkaloids candidate compounds were fished out. Their structures were identified and the enzyme inhibitory activities were verified. In addition, the binding information between the candidate compound and the enzyme was displayed by molecular docking. This study provides valuable information for the improvement of immobilized enzyme activity and functional active ingredients in complicated medicinal plant extracts.

The development of versatile surface-enhanced Raman scattering (SERS) platforms facilitates the rapid identification of contaminants in diverse application scenarios. Herein, a dual-functional SERS substrate comprising silver nanoislands-decorated micropillar/wrinkle hierarchical structure (Ag NIs@PWHS) on a polydimethylsiloxane (PDMS) film was fabricated through an amalgamation of fabrication techniques: soft lithography, surface wrinkling, and magnetron sputtering. The PDMS hierarchical architecture was constructed by replicating mastoid structures of lotus leaves through a two-step molding process, followed by the formation of periodic wrinkles via oxygen plasma treatment. This hierarchical design integrating micropillars and wrinkles not only provided a multiscale surface architecture conducive to the formation of three-dimensional (3D) hot spots, but also conferred hydrophobicity and bending insensitivity on the SERS substrate. The optimal Ag NIs@PWHS substrate had an analytical enhancement factor of 1.99 × 10 6 , outstanding signal uniformity (relative standard deviation, RSD = 2.98 %) and reproducibility (RSD = 4.54 %), and robust mechanical stability. Furthermore, the Ag NIs@PWHS substrate enabled the rapid detection of thiram on pear surface and malachite green in lake water, with a detection sensitivity of 10 −7 M for both pesticides. Thus, the developed Ag NIs@PWHS SERS substrate holds significant potential for utilization in realms of food safety assessments and water pollution surveillance.

Lotus leaf is an optimal raw material for the preparation of foods with the function of regulating glycolipid metabolism. However, further investigation is required to elucidate the synergistic regulatory effects of lotus leaf polysaccharide (LLP), flavonoids, and alkaloids, which act as the main active components in lotus leaves. This study optimally prepared LLP and whole functional components (LLE) containing LLP, flavonoids and alkaloids by high-pressure homogenization-assisted dual enzyme extraction with and without pre-/post treatment. LLP and LLE exerted regulatory roles in glycolipid metabolism and alleviated liver, kidney, and pancreas damages of db/db mice. LLP was utilized by gut bacteria, promoting the production of beneficial metabolites i.e. lithodeoxycholic and butyric acids, which reached the liver through blood circulation, activating insulin signaling, and regulating glycolipid metabolism. The combination of polysaccharide, flavonoids and alkaloids promoted the production of butyric acid but different secondary bile acids i.e. deoxycholic and ursodeoxycholic acids, acted in the intestine and liver, and intervened in more metabolic pathways, showing a synergistic effect on glycolipid metabolism. This study provided insights into the rational processing of lotus leaf for hypoglycaemic foods.
