Chemically synthesized poly(4-hydroxybutyrate) (P4HB) is a new generation of biomass-derived and degradable semi-crystalline polymer with good comprehensive properties, but high costs limit its application. Starch, as an inexpensive natural polymer , can reduce the cost of P4HB products. However, starch lacks thermoplastic behavior and has poor compatibility with P4HB, thus its extensive use will inevitably impair the mechanical properties of P4HB. In this study, the ball-milling starch grafting process is adopted, which can simultaneously solve the two major deficiencies of starch, and the prepared ball-milling starch-g-polycaprolactone (BSt-g-PCL) has thermoplasticity and better compatibility with P4HB. BSt-g-PCL can melt near 55 °C, and the interweaving of its molecular chains with P4HB reduces the binding energy ( E interaction ) of both, making the phase interface blurred or even disappear. Therefore, the elongation at break retention ( R EB ) of P4HB/BSt-g-PCL can increase from 37.1 % to 74.3 % compared to P4HB/starch at the same filling (70 Phr). Additionally, BSt-g-PCL can exert the effect of accelerating P4HB degradation and still make it maintain excellent anti-aging ability. The ball-milling starch graft process provides a simple and effective method for the preparation of inexpensive fully biodegradable P4HB composite films with excellent mechanical properties.

Owing to inherent issues like interfacial stress concentration, open-cell, a low volume expansion ratio ( VER ) and cell density, the impact resistance and thermal insulation of supercritical foamed poly(lactic acid) (PLA) blend foams remain unsatisfactory. To overcome these challenges, an in-situ cross-linking strategy is employed to enhance the compatibility between PLA and poly(4-hydroxybutyrate) (P4HB), while improving the melt strength and crystallization behavior of the blends. The modified PLA/P4HB blend exhibits a blurred interfacial boundary with fewer stress concentrations. As a result, the specific impact strength increases from 9.76 to 26.17 kJ·m −2 /(g·cm −3 ) after foaming. The ambiguous interface and enhanced melt strength reduce the risk of cell wall rupture and collapse, while the increased crystallization provides additional nucleation sites. Consequently, the modified PLA/P4HB foam transitions from an open-cell to a closed-cell structure, achieving a VER of 35.4 and a cell density of 2.0 × 10 7 cells/cm 3 , representing respective increases of 16.1-fold and 9.1-fold compared to unmodified PLA/P4HB foam. At this time, the thermal conductivity and thermal diffusivity of the PLA/P4HB foam reach 31.5 mW/m·K and 8.1 × 10 −7 m 2 /s, respectively. In conclusion, this strategy provides a method for producing PLA/P4HB foams with high impact resistance and thermal insulation, and addresses common issues associated with PLA-based blends foam.

A blend of poly(lactic acid) (PLLA) and poly(4-hydroxybutyrate)-based polyurethane prepolymer (P4HBU) was prepared through reactive blending in this study. Thermal properties, mechanical properties, rheological properties, and the degradation behavior of the PLLA/P4HBU blends were investigated as varying the P4HBU content. Dynamic mechanical analysis indicated that the glass transition temperatures of PLLA and P4HBU got closer to each other after melt blending, implying that the compatibility between them was improved. With the increase of P4HBU content increased, the elongation of the PLLA/P4HBU composite enhanced obviously, as well as the notched impact strength. When the P4HBU content reached 20 wt%, the elongation increased to 196.2% and the notched impact strength increased to 8.4 kJ/m 2 , respectively, about 26.2 and 2.4 times those of PLLA homopolymer. Meanwhile, the tensile strength decreased to 30.5 MPa. Scanning electron microscopy results proved that the shear yielding of the PLLA matrix and cavitation were the primary toughening mechanisms, resulting in an enhanced ductility of the PLLA/P4HBU blends. The degradation rate of the PLLA/P4HBU blends was improved for the incorporation of the P4HBU. Highlights P4HBU improved PLLA elongation (196.2%) and impact strength (8.4 kJ/m 2 ). Tensile strength of PLLA/P4HBU was reduced to 30.5 MPa. The compatibility between PLLA and P4HBU was enhanced. Shear yielding and cavitation were the primary toughening mechanisms. The degradation rate of the PLLA/P4HBU blends was improved.

Lignin can be employed as a sustainable functional additive and reinforcement filler for polymers. In this work, melamine phosphate modified lignin (MAP-lignin) was synthesized as a bio-based halogen-free flame-retardant agent. The synergetic effects between MAP-lignin and rice husk ash (RHA) on the thermal stability, dynamic mechanical performance and flame-retardant properties of the poly(3-hydroxybutyrate-co-4-hydroxybutyrate) (P34HB) biocomposites were investigated. The results revealed that MAP-lignin and RHA could significantly increase the char residue and MAP-lignin had a good miscibility with P34HB. The incorporation of MAP-lignin and RHA as stiff fillers could enhance the storage modulus of the P34HB matrix. Moreover, the tensile strength and elongation at break of the P34HB/RHA composite could be improved due to the MAP-lignin could serve as a compatibilizer to improve the interaction between RHA and P34HB. Cone calorimetry data demonstrated that the addition of 30 wt% MAP-lignin and 5 wt% RHA reduced the peak heat release rate ( PHRR ) and total heat release ( THR ) of P34HB noticeably by 42.8 and 24.3%, respectively. In addition, the CO 2 production rate and CO concentration could be significantly suppressed by the addition of MAP-lignin. The enhanced flame retardancy of P34HB composite should be ascribed to the barrier effect of the increased char residue with compact and intact structure, as well as the released gases from the decomposition of melamine.

Considering the current research background of environmental problems and shortages of petroleum resources aiming at proposing the “green” polymer blend with good degradability and properties similar to oil-based merchandise currently on the market, in the present study, two series of fully biodegradable blends consisting of PBS/P34HB (PBP) and PBS-g-GMA/P34HB (PGP) have been successfully fabricated by a simple and environmental-friendly processing methodology. In this work, the resulting biodegradable blends are in detail studied from the mechanical, thermal, crystallization, and morphology points of view. To improve the miscibility and enhance the interfacial adhesion between both phases of poly(butylene succinate) (PBS) as the matrix and poly (3-hydroxybutyrate-co-4-hydroxybutyrate) (P34HB) as the modifier, with consequent improvement in material mechanical properties, glycidyl methacrylate (GMA) is grafted on PBS via reactive blending in the process. The incorporation of 10wt% P34HB into the blend promotes a dramatic increase of 614% in the Izod impact strength. The crystallization temperature (Tc) of PGP is 87.78℃ when 30wt% P34HB is added, which is 25% higher than that of pure PBS and 11% higher than that of the PBP blend. Meanwhile, the crystallinity of PGP is 30.12%, 47%, and 37% decline compared to pure PBS and PBP, respectively. Rheological results show that storage modulus, loss modulus, and complex viscosity improve significantly with the increase of P34HB amount, indicating that the enhancement of melt strength is beneficial to the processing and forming of the materials.

Poly(butylene-adipate-co-terephthalate) (PBAT) is recognized for its advantages as a biodegradable thermoplastic polyester. However, its application is often limited due to low strength and poor processability. In contrast, poly(4-hydroxybutyrate) (P4HB) emerges as a fully biodegradable polyester characterized by high strength, excellent toughness, and facile processing. In this study, P4HB was employed to reinforce PBAT, and glycidyl methacrylate grafted onto poly(4-hydroxybutyrate) (P4HB-g-GMA) was introduced as an effective compatibilizer to address their inherent incompatibility. The study explored the influence of P4HB-g-GMA content on the mechanical, thermal, morphological, and rheological properties of the blends. The findings revealed that P4HB-g-GMA significantly enhanced the compatibility, mechanical properties, and melt strength of PBAT/P4HB blends. Furthermore, various PBAT/P4HB foams were prepared through a batch foaming process, employing supercritical CO 2 as a physical foaming agent. The volume expansion ratio and cell size of PBAT/P4HB foams increased with higher levels of the compatibilizer.

The use of physical barriers is an effective strategy for preventing postoperative adhesion, a prevalent complication in clinical abdominal surgeries. However, the current anti-adhesion barriers face limitations in precisely regulating key cellular responses during tissue repair. In this study, a multi-modal synergistic degradable anti-adhesion membrane was developed via coaxial electrospinning of poly(4-hydroxybutyrate) (P4HB), polyglycolic acid (PGA), and diclofenac sodium (DS). The designed membrane features a densely packed fibrous architecture, exhibiting over 97 % inhibition efficiency against fibroblasts interlayer penetration in vitro. The rapid initial release of DS synergizes with PGA-mediated biological functions to suppress fibroblast-to-myofibroblast differentiation, and to significantly mitigate early-stage pathological changes. This cascade effect ultimately leads to reduced collagen deposition and significantly diminished adhesion formation, as demonstrated in a mouse cecal and peritoneal abrasion model. Furthermore, the membrane demonstrates exceptional flexibility and enhanced hydrophilicity, ensuring conformal coverage of complex three-dimensional wounds. The integration of structural design, controlled drug release, and dynamic biological modulation renders this multifunctional barrier as a promising candidate for next-generation clinical anti-adhesion applications.

Abdominal wall defect caused by surgical trauma, congenital rupture, or tumor resection may result in hernia formation or even death. Tension-free abdominal wall defect repair by using patches is the gold standard to solve such problems. However, adhesions following patch implantation remain one of the most challenging issues in surgical practice. The development of new kinds of barriers is key to addressing peritoneal adhesions and repairing abdominal wall defects. It is already well recognized that ideal barrier materials need to have good resistance to nonspecific protein adsorption, cell adhesion, and bacterial colonization for preventing the initial development of adhesion. Herein, electrospun poly(4-hydroxybutyrate) (P4HB) membranes infused with perfluorocarbon oil are used as physical barriers. The oil-infused P4HB membranes can greatly prevent protein attachment and reduce blood cell adhesion in vitro. It is further shown that the perfluorocarbon oil-infused P4HB membranes can reduce bacterial colonization. The in vivo study reveals that perfluoro(decahydronaphthalene)-infused P4HB membranes can significantly prevent peritoneal adhesions in the classic abdominal wall defects’ model and accelerate defect repair, as evidenced by gross examination and histological evaluation. This work provides a safe fluorinated lubricant-impregnated P4HB physical barrier to inhibit the formation of postoperative peritoneal adhesions and efficiently repair soft-tissue defects.

In this study, hydrophilic pullulan, which is favorable for cell adhesion, proliferation, and differentiation, was selected as a modifier for the preparation of poly(3-hydroxybutyrate-co-4-hydroxybutyrate) (P(3HB-co-4HB))/pullulan nanofibers via electrospinning to improve the biocompatibility of P(3HB-co-4HB) and increase the drug loading of composite fibers. Alkyl polyglycoside was used as the emulsifying agent to promote emulsification and stabilize the P(3HB-co-4HB)/pullulan composite solution. Drug-loading property of the nanofibers with a shell-core structure is increased because gelatin was not formed into fibers via electrospinning, thereby forming a stable drug-containing gelatin solution in the core layer. Finally, P(3HB-co-4HB)/pullulan-gelatin shell-core nanofibers were prepared. The intermolecular interaction, morphology, crystallization properties, mechanical properties, morphology, sustained release, and biocompatibility of composite nanofibers were characterized. Results show that the crystallization property of P(3HB-co-4HB)/pullulan composite nanofibers increases continuously with an increase in the pullulan content. As the pullulan content increases, the strain and stress of P(3HB-co-4HB)/pullulan nanofibers increase initially and decrease later. At the mass ratio of P(3HB-co-4HB) to pullulan of 10:2, P(3HB-co-4HB)/pullulan composite nanofibers exhibit a uniform morphology with an average diameter of 590 nm and porosity of 70.71%. At this mass ratio, the P(3HB-co-4HB)/pullulan-gelatin/drug shell-core structure, which sustained a release effect for more than 180 h, has potential applications as biomaterials without cytotoxicity.

Antibacterial and hemostatic properties are of great importance to wound dressing in treating trauma. Poly(4-hydroxybutyrate) (P4HB), an U.S. Food and Drug Administration (FDA)-approved bioabsorbable polyester, is used as dermal substitutes to prevent sever wound contraction and improve wound healing. However, P4HB fibrous mats are not efficient in halting bleeding and preventing bacterial associated infection. Here the authors prepare a new kind of wound dressing by incorporating poly(ethylene glycol) (PEG) and zinc oxide (ZnO) nanoparticles into the P4HB matrix by using electrospinning method. The in vitro results reveal that the P4HB/PEG/ZnO dressings can synergistically help blood clotting, prevent bacteria adhesion, and kill bacteria efficiently. It's found that the bactericidal activity of the bioactive P4HB/PEG/ZnO dressings is related to the release of Zn 2+ ions rather than reactive oxygen species (ROS) under dark condition. The in vivo antibacterial evaluation indicates that the bioactive P4HB/PEG/ZnO dressings can successfully prevent wound infections in Sprague–Dawley (SD) rats’ skin defect model. The in vivo hemostatic experiments evaluated in the rabbit ear injury model further confirm that the bioactive P4HB/PEG/ZnO dressings have excellent hemostatic performance. Hence, this study demonstrates that the bioactive P4HB/PEG/ZnO dressings with hemostatic and bactericidal properties have great potential in possible clinical applications.

Objective: Alcohol dependence (AD) is a chronic recurrent mental disease caused by long-term drinking. It is one of the most prevalent public health problems. However, AD diagnosis lacks objective biomarkers. This study was aimed to shed some light on potential biomarkers of AD patients by investigating the serum metabolomics profiles of AD patients and the controls. Methods: Liquid chromatography-mass spectrometry (LC–MS) was used to detect the serum metabolites of 29 AD patients (AD) and 28 controls. Six samples were set aside as the validation set (Control: n = 3; AD group: n = 3), and the remaining were used as the training set (Control: n = 26; AD group: n = 25). Principal component analysis (PCA) and partial least squares discriminant analysis (PCA-DA) were performed to analyze the training set samples. The metabolic pathways were analyzed using the MetPA database. The signal pathways with pathway impact >0.2, value of p <0.05, and FDR < 0.05 were selected. From the screened pathways, the metabolites whose levels changed by at least 3-fold were screened. The metabolites with no numerical overlap in their concentrations in the AD and the control groups were screened out and verified with the validation set. Results: The serum metabolomic profiles of the control and the AD groups were significantly different. We identified six significantly altered metabolic signal pathways, including protein digestion and absorption; alanine, aspartate, and glutamate metabolism; arginine biosynthesis; linoleic acid metabolism; butanoate metabolism; and GABAergic synapse. In these six signal pathways, the levels of 28 metabolites were found to be significantly altered. Of these, the alterations of 11 metabolites changed by at least 3-fold compared to the control group. Of these 11 metabolites, those with no numerical overlap in their concentrations between the AD and the control groups were GABA, 4-hydroxybutanoic acid, L-glutamic acid, citric acid and L-glutamine.Conclusion: The metabolite profile of the AD group was significantly different from that of the control group. GABA, 4-hydroxybutanoic acid, L-glutamic acid, citric acid, and L-glutamine could be used as potential diagnostic markers for AD.

The understanding of cellular energy metabolism activation by engineered scaffolds remains limited, posing challenges for therapeutic applications in tissue regeneration. This study presents biosynthesized poly(3-hydroxybutyrate- co -4-hydroxybutyrate) [P(3HB- co -4HB)] and its major degradation product, 3-hydroxybutyrate (3HB), as endogenous bioenergetic fuels that augment cellular anabolism, thereby facilitating the progression of human bone marrow-derived mesenchymal stem cells (hBMSCs) towards osteoblastogenesis. Our research demonstrated that 3HB markedly boosts in vitro ATP production, elevating mitochondrial membrane potential and capillary-like tube formation. Additionally, it raises citrate levels in the tricarboxylic acid (TCA) cycle, facilitating the synthesis of citrate-containing apatite during hBMSCs osteogenesis. Furthermore, 3HB administration significantly increased bone mass in rats with osteoporosis induced by ovariectomy. The findings also showed that P(3HB- co -4HB) scaffold substantially enhances long-term vascularized bone regeneration in rat cranial defect models. These findings reveal a previously unknown role of 3HB in promoting osteogenesis of hBMSCs and highlight the metabolic activation of P(3HB- co -4HB) scaffold for bone regeneration.
