This study aimed to evaluate the antioxidant potential of Artemisia selengensis Turcz (AST) leaves, a byproduct when processing AST stalk, and identify the antioxidant constituents by using HPLC-QTOF-MS 2 . The total phenolics content (TPC), total flavonoids content (TFC) and antioxidant abilities of fractions resulted from the successively partition of chloroform, ethyl acetate and n -butanol were compared. Ethyl acetate fraction (EAF) exhibited the highest TFC (65.44 mg QuE/g fraction), n -butanol fraction ( n BuF) showed the highest TPC (384.78 mg GAE/g fraction) and the best DPPH scavenging ability, ABTS + scavenging ability and reducing power. Totally, 57 compounds were identified or tentatively identified in n BuF and EAF, 40 of them were reported in AST for the first time. The major constituents in EAF were flavonoids, and the major constituents in n BuF were phenolic acids and organic acids. Thus, AST leaves might be a potential low-cost resource of natural antioxidants .

Nowadays, obesity is a global health risk factor, and its development is closely related to the absorption and metabolism of lipids. In this study, the main chemical constituents and antiobesity effect of Artemisia selengensis Turcz extract (ASTE) were investigated by HPLC-QTOF-MS 2 and obese mice models. Twenty-three compounds were identified from ASTE, and caffeoylquinic acid and dicaffeoylquinic acid are the dominant bioactive compounds. ASTE administration reduced body weight (9.8%), improved glucose tolerance (14.2%), corrected dyslipidemia (the levels of total cholesterol, total triglyceride, and low-density lipoprotein cholesterol dropped by 19.8%, 24.4%, and 27.2%, respectively, and the high-density lipoprotein cholesterol level rose by 27.6%), and alleviated hepatic lipid accumulation. ASTE improved the gut microbiota dysbiosis caused by High-Fat Diet (HFD), mainly by increasing the relative abundance of Odoribacter , Candidatus_Saccharimonas , Bacteroides , and unclassified_f__Lachnospiraceae , and reducing the relative abundance of Faecalibaculum . Gene expression heatmaps and pathway enrichment analyses based on transcriptomics indicated that ASTE significantly reduced HFD-induced increases in fatty acid uptake, triglyceride synthesis, and cholesterol synthesis. Our findings indicated that ASTE holds significant potential as a candidate for modulating lipid metabolism and preventing or treating obesity, meriting further investigation.

Effects of dynamic high pressure microfluidization (DHPM), microwave, ultrasound, maceration and high temperature, on the phenolic acids and antioxidant activity of Artemisia selengensis Turcz (AST) were investigated. Microwave exhibited the best extraction efficacy on phenolic compounds and flavonoids with the content of 40.30 μg GAE/mg Extract, and 37.75 μg RTE/mg Extract, respectively. Microwave extract (MWE) also showed the strongest DPPH· and HO· scavenging ability (IC 50 value of 0.23 and 0.50 mg/mL, respectively), and reducing power (Ab 0.3 value of 0.23 mg/mL). Quinic acid, 3 caffeoylquinic acids (CQAs), 2 coumaroylquinic acids, 4 dicaffeoylquinic acids (diCQAs) were identified as the major phenolic acids in AST using HPLC-QTOF-MS/MS, and 3-CQA (63.71 μg/mL) showed the highest content followed by 1,5-diCQA (17.91 μg/mL) and 3,5-diCQA (13.50 μg/mL). MWE had the highest 3-CQA and diCQAs. While, high temperature could promote the degradation of diCQAs, 3- and 4-caffeoyl are more liable to decompose as compared with 1- and 5-caffeoyl. Practical Application Over production of reactive oxygen species (ROS) in body will induce various chronic diseases. AST is an underutilized vegetable with many health benefits. This research indicates that AST possessed appreciating antioxidant activities and abundant phenolic acids, it is could be a potentail antioxidants resource and for prevention of chronic diseases associated with ROS. It also suggested that microwave could be a promising method for extracting the antioxidants in AST. Therefore, this research could lay a foundation for the high-value application of AST in food, health care and natural therapy industry, and provide some methodological references for the development of antioxidants in AST.

Macroporous resin was used to enrich flavonoids in the ethyl acetate extract of Artemisia Selengensis Turcz. Based on a single factor experiment, the enrichment process was optimized using the response surface method. The optimal parameters of the enrichment process were a sample concentration of 0.3 mg/mL, a loading rate of 1 mL/min, an elution flow rate of 2 mL/min, and a total flavonoid content of 155.38±0.97 mg/g. The flavonoids enriched by AB-8 macroporous resin demonstrated significant scavenging activities against DPPH, ABTS + , and hydroxyl free radicals, and also exhibited certain inhibitory effects on α -amylase and α -glucosidase. Among them, the scavenging ability of the flavonoids enriched by AB-8 macroporous resin on hydroxyl free radical (IC 50 =30.31±1.92 μg/mL) was the closest to Vc, and the inhibitory effect on α -glucosidase (IC 50 =16.19±1.35 μg/mL) was the best. These findings confirmed the potential of Artemisia Selengensis Turcz. was a natural antioxidant and hypoglycemic drug. Graphical

Caffeioyl quinic acids and polysaccharides from Artemisia selengensis Turcz are considered potential bioactive substances for hyperuricemia (HUA) treatment. While the mechanism of multi-component combined intervention of polysaccharides and dicaffeoylquinic acids (diCQAs) is not yet clear. In this study, we investigated the effect of A. selengensis Turcz leaves polysaccharides (APS) on the HUA treatment with diCQAs in vitro by direct inhibition of XOD activities and in vivo by using animal model. The results showed that APS had almost no inhibitory effect on XOD activities in vitro , but the inhibitory activity of diCQAs on XOD was affected by changes in inhibition type and inhibition constant. Compared to APS and diCQAs alone, high-dose APS and diCQAs in combination (ADPSh) could significantly reduce the production of uric acid (16.38 % reduction compared to diCQAs group) and oxidative stress damage. Additionally, this combined therapy showed promise in restoring the gut microbiota balance and increasing the short-chain fatty acids levels. The results suggested that APS and diCQAs in combination could be a potential inhibitor for HUA treatment.

In this study, the caffeoylquinic acid (CQA) extract solution of the Artemisia selengensis Turcz (AST) leaves prepared by 70% ethanol reflux method was separated with petroleum ether, ethyl acetate, and water-saturated n -butanol to obtain the proper solvent to purify CQAs. Results showed that the ethyl acetate fraction (EAF) exhibited the highest total chlorogenic acid content (TCC) (241.46 ± 0.00 mg/g dry fraction), followed by the water-saturated n -butanol fraction ( n BuF) (TCC = 62.30 ± 0.82 mg/g dry fraction). Ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry and high-performance liquid chromatography photodiode array were used to qualitatively and quantitatively determine the CQAs in these two fractions. In EAF, four CQAs, namely, 3-CQA, 3,5-diCQA, 3,4-diCQA, and 4,5-diCQA, were identified with the contents of 5.69 ± 0.03, 106.65 ± 0.83, 15.65 ± 0.24, and 28.08 ± 0.82 mg/g, respectively. Six CQAs, namely, 3-CQA, 4-CQA, 1,3-diCQA, 3,5-diCQA, 3,4-diCQA, and 4,5-diCQA, with contents of 16.57 ± 0.23, 3.22 ± 0.55, 4.49 ± 0.01, 16.86 ± 0.24, 7.35 ± 0.11, and 6.41 ± 0.13 mg/g, respectively, were identified in n BuF. Quantitative analysis of the six CQAs in AST leaves have not been reported, and 1,3-diCQA has been found in AST leaves for the first time. Moreover, either EAF or n BuF and the six CQA reference compounds showed certain inhibitory activity on xanthine oxidase (XOD) in vitro . EAF demonstrated a stronger XOD inhibitory effect (IC 50 = 1.67 mg/mL) than n BuF. Among the six CQA compounds, 3,4-diCQA had the lowest IC 50 of 12.80 μM, whereas 1,3-diCQA and 3-CQA had IC 50 values of 23.69 and 28.29 μM, respectively. The inhibitory activity of 4-CQA was weak. These findings indicated that CQAs are the major components inhibiting the XOD activity in vitro in AST leaves.

Background: Lactobacillus fermentation has become a typical food processing method with the development of the modern food industry. Aims: This study aimed to assess the effects of Lactobacillus acidophilus NCUF202.2 fermentation on Artemisia selengensis Turcz (AST) in relieving hyperuricemia (HUA) and the mechanisms involved. Results: The fermented Artemisia selengensis Turcz extracts (FASTE) could significantly increase the content of free polyphenol and enhance the inhibitory effect on xanthine oxidase (XOD) in vitro. The result of ultrafiltration coupled with UPLC-MS/MS screened out the components that might be combined with XOD in the caffeoylquinic acids (CQAs) of Artemisia selengensis Turcz extracts (ASTE) as 5-CQA, 3-CQA, 4-CQA, 1-CQA, 3,4-diCQA, 3,5-diCQA, 1,5-diCQA and 4,5-diCQA as well as caffeic acid (CA) of FASTE except the above. Our results also showed that ASTE and FASTE could alleviate HUA in rats. In contrast, FASTE had a better ability to reduce serum adenosine deaminase (ADA) activity than ASTE. Furthermore, FASTE could restore catalase (CAT) in HUA model rats in vivo, and superoxide dismutase (SOD) activity to a better degree, thereby inhibiting the production of excess malondialdehyde (MDA). Under the intervention of ASTE and FASTE, the levels of serum pro-inflammatory and anti-inflammatory factors in rats tended to be normal. However, FASTE could increase the expression of the uric acid secretion protein OAT1 and decrease the expression of reabsorption proteins URAT1 and GLUT9 in model rats, thereby reducing the serum uric acid (SUA) level in model rats. ASTE and FASTE can increase the abundance of beneficial bacteria, such as Bifidobacterium and Akkermansiaceae. Conclusion: Overall, part of diCQAs and CQAs was decomposed after fermentation. FASTE had a more substantial HUA-relieving effect than ASTE. It is expected to be applied to functional foods and contribute to the research and development of uric acid lowering functional foods.

Artemisia selengensis is a medicinal and edible plant, whose tender stems are widely consumed as a folk vegetable in China. However, due to their bitterness, Artemisia selengensis leaves (ASL) are often discarded as a waste material, resulting in resource wastage. This study aimed to characterize ASL's chemical composition, develop a “green” extraction method, and explore its potential bioactivity. Using LC-MS, 72 compounds were identified. To extract 7 representative caffeoylquinic acids (CQAs), the mixture of choline chloride and ethylene glycol (1:6) was selected as the optimal deep eutectic solvent (DES), and the extraction parameters were optimized as follows: extraction time of 21 min, temperature of 44°C and ultrasonic power of 420 W. Based on molecular docking analysis, both 3,5-diCQA and 1,5-diCQA exhibited strong binding affinity to xanthine oxidase (XO), consistent with their potent XO inhibitory effects (IC 50 = 3.69 and 3.35 nmol/mL, respectively). In addition, a greater XO inhibition was observed for the DES extract (IC 50 = 6.05 μg/mL) compared with the methanol extract (IC 50 = 22.5 μg/mL). This study not only helps to reduce ASL disposal's environmental load and boost its agricultural value, but also sets a green model for similar vegetable waste to aid circular agriculture.

Artemisia selengensis Turcz. (AST) is a common edible and medicinal herb with multiple health benefits. Our previous research indicated that AST leaves, by-products of AST, possess strong in vitro xanthine oxidase (XOD) inhibitory activity, making them a potential treatment for hyperuricemia (HUA). In this study, we investigated the anti-HUA effect of AST leaves extract (ASTLE) in HUA mice and explore the underlying mechanisms. Chemical analysis showed that ASTLE is rich in caffeoylquinic acids (CQAs), including four mono-CQAs (1-CQA, 3-CQA, 5-CQA, and 4-CQA) and six di-CQAs (1,3-diCQA, 1,4-diCQA, 3,4-diCQA, 1,5-diCQA, 3,5-diCQA, and 4,5-diCQA). In HUA mice, ASTLE reduced serum uric acid level and increased urine uric acid level and fraction excretion of uric acid. ASTLE also protected against liver and kidney damage, as evidenced by decreased serum levels of glutamic oxaloacetic transaminase, glutamic-pyruvic transaminase, creatinine, and blood urea nitrogen. After ASTLE intervention, the hepatic XOD activity was inhibited. Additionally, the mRNA expression of mURAT1 and mGLUT9 was down-regulated, while the mRNA expression of mABCG2, mOAT1, and mOAT3 was up-regulated. Metabolomic analysis further suggested that ASTLE mainly regulated amino acids and their metabolites, which are associated with renal uric acid excretion function. These results demonstrate that ASTLE rich in CQAs, has beneficial effects against HUA in mice by inhibiting hepatic XOD activity, modulating the mRNA expression of renal uric acid transporters, and improving metabolic disorders in HUA mice. Our findings suggest that AST leaves may be a promising natural resource for ameliorating HUA.
