Investigation of active biomolecules involved in the nucleation and growth of gold nanoparticles by Artocarpus heterophyllus Lam leaf extract

Jiang Xinde,Sun Daohua,Zhang Genlei,He Ning,Liu Hongyu,Huang Jiale,Odoom-Wubah Tareque,Li Qingbiao

The effects of different biomolecules in Artocarpus heterophyllus Lam leaf extract on the morphology of obtained gold nanoparticles were investigated in this study. The results indicated that reducing sugars, flavones, and polyphenols consisting of about 79.8 % dry weight of the leaf extract were mainly involved in providing the dual function of reduction and the size/shape control during the biosynthesis. The gold nanoparticles present included 64 ± 10 nm nanospheres, 131 ± 18 nm nanoflowers, and 347 ± 136 nm (edge length) nanoplates and they were synthesized using the main content of reducing sugars, flavones, and polyphenols, respectively, after they were desorbed by the AB-8 macroporous adsorption resin column. Particularly, flower-like and triangular/hexagonal gold nanoparticles with a yield more than 80 % were obtained. Possible shape-directed agents for the nucleation and growth were characterized by FTIR, it can be seen that ketones were bound on the surface of the spherical and flower-like GNPs, while both the ketones and carbonyls bound on the Au {111} plane this may have favored the formation of the twin defects, which are very essential for nanoplates’ formation.

Comparison of microwave, ultrasound and ultrasound-microwave assisted solvent extraction methods on phenolic profile and antioxidant activity of extracts from jackfruit (Artocarpus heterophyllus Lam.) pulp

Ming Cheng,Jiali He,Huaijie wang,Chuan Li,Gang Wu,Kexue Zhu,Xiaoai Chen,Yanjun Zhang,Lehe Tan

This study aimed to investigate the impact of different solvents (water, 60% methanol, 60% ethanol and 60% acetone) and methods (conventional solvent extraction, ultrasound-assisted extraction, microwave-assisted extraction and ultrasound-microwave assisted extraction) on the phenolic profile and antioxidant activity of jackfruit ( Artocarpus heterophyllus Lam.) pulp. The Folin-Ciocalteu assay was used to determine the total phenolic content (TPC), and antioxidant activity was measured by DPPH and ABTS assays. The phenolic profiles in different extracts were identified using the UPLC-ESI-Q-TOF-MS/MS technique. The results showed that significant differences ( p  < 0.05) in TPC were found among different extraction methods, the microwave-assisted extraction with 60% ethanol had the highest TPC (2.40 mg GAE/g) compared with the extracts obtained from other methods. Meanwhile, the highest DPPH radical and ABTS radical cation was found in the 60% ethanol-MAE extracts. Correlation analysis further showed a significant positive correlation between TPC and antioxidant activity (R 2  > 0.8, p  < 0.001). Seventy-eight phenolic compounds were identified by UPLC-ESI-Q-TOF-MS/MS analysis. The most abundant phenolic compounds in jackfruit pulp were identified in negative mode, with 60% ethanol-MAE being the most effective. The quantity of phenolic compounds extracted varied with different extraction methods indicated that the extraction methods had effects on the phytochemical characteristics.

Polysaccharide from Artocarpus heterophyllus Lam. Pulp Ameliorates Cyclophosphamide-Induced Intestinal Damage by Regulating Gut Microbiota and Co-Metabolites

Zhenyu He,Yunlong Li,Jun Yang,Chuan Li,Wei Wang,Yanjun Zhang,Huawei Chen,Jianjie Li,Jun Cao,Kexue Zhu

Background: Polysaccharides modulate host health by interacting with gut microbiota and reshaping the host–microbial metabolome, potentially facilitating immune regulation. Methods: This study evaluated the modulatory effect ofArtocarpus heterophyllusLam. (jackfruit) polysaccharide (JFP-Ps) against cyclophosphamide (Cy)-induced immunosuppression in mice, focusing on gut microbiota modulation and metabolite changes. Results: JFP-Ps effectively increased the beneficial bacteria ratio, such asLactobacillusandLachnospiraceae, while inhibiting some species likeAkkermansia. Metabolomic analysis showed that JFP-Ps notably regulated gut microbe-associated metabolites, including short-chain fatty acids (SCFAs), amino acids, bile acids, indoles, and derivatives. These metabolites were involved in various metabolic pathways, including primary bile acid synthesis and biosynthesis of phenylalanine, tyrosine, and tryptophan, along with tryptophan catabolism, purine metabolic processes, and unsaturated fatty acid production. Additionally, significant correlations between microbial groups and functional metabolites were identified. Overall, JFP-Ps exerted an immuno-modulatory effect by reshaping gut microbiota and enhancing co-metabolism with the host. Conclusions: These results provided valuable insights into host–microbiota interactions and gut microbiota-targeted intervention strategies of tropical natural bioactive polysaccharides.

Chemical markers of pulp, shreds and inflorescence axis of Artocarpus heterophyllus Lam. and their xanthine oxidase inhibitory activity

Wang Yu,Wang Yueping,Zhao Wenli,Li Yan,Zhang Qipeng,Liang Wenjuan

To understand the chemical markers and their xanthine oxidase (XO) inhibitory activities in the pulp, shreds and inflorescence axis of A. heterophyllus , an investigation guided by UHPLC-OE-MS tracking, in vitro activity screening and molecular docking was carried out. The inflorescence axis exhibited an XO inhibition rate of 31.87%, significantly higher than that of the pulp (6.69%) and shreds (3.75%), which was closely associated with its abundant content of flavonoids. Further analysis identified flavonoids such as luteolin ( Fl2 ), kaempferol ( Fl3 ), quercitrin ( Fl4 ) and 8-prenylluteone ( Fl16 ) serve as potential contributors to the XO inhibitory activity. Molecular docking with XO revealed binding energies below − 5.0 kcal/mol for above compounds. 8-Prenylluteone ( Fl16 ) showed the strongest interaction, with the lowest binding energy of -8.8 kcal/mol, suggesting its enhanced activity was attributed to the isopentenyl side chain attached to the flavonoid skeleton. This study provided a new perspective for discovering XO inhibitors from A. heterophyllus by-products.

The characteristic VOCs of different parts of Artocarpus heterophyllus fruit based on HS-SPME-GC–MS and PTR-TOF-MS

Yueping Wang,Yequn Huang,Lianping Song,Hong Wang,Li Wan,Jinqian Pang,Wenjuan Liang

Insight investigation on both edible pulps and inedible parts involving inflorescence axis and shreds of Artocarpus heterophyllus Lam were carried out, a total of 98 VOCs and 201 masses were identified by the combination of HS-SPME-GC–MS and PTR-TOF-MS. Among them, according to the consistency of OAV and results of VIP > 1, p  < 0.05, compounds methyl isovalerate ( A2 ), 3-methylbutyl acetate ( A5 ) and octanoic acid, ethyl ester ( A21 ) were recognized as aroma markers to distinguish the pulps, shreds and inflorescence axis. Meanwhile, the inflorescence axis (IC 50 : 1.82 mg/mL) and shreds (IC 50 : 16.74 mg/mL) exhibited more excellent antioxidant potency than pulps (IC 50 : 17.43 mg/mL) in vitro . These findings validated the feasibility of coupling HS-SPME-GC–MS and PTR-TOF-MS for rapid detection of characteristic VOCs of this plant, and offered new prospect of fragrance utilization and waste management of the edible and inedible parts of A. heterophyllus fruit.

Dynamic Changes in Jackfruit (Artocarpus heterophyllus Lam.) Polyphenols During Maturation and In Vitro Digestion and Their Effects on the Gut Microbiota

Liru Ma,Zhen Feng,Chao Zhang,Chunhe Gu,Yihong Bao

Jackfruit (Artocarpus heterophyllusLam.) phenolic compounds are chemically diverse. Previous studies have predominantly focused on easily extractable free polyphenols, with limited attention on bound (conjugated) polyphenols. The transformation mechanisms of these polyphenols during maturation, digestion, and colonic fermentation remain unclear, limiting the full utilization of jackfruit resources. This study provides a comprehensive portrait of how jackfruit polyphenols shift in abundance, chemical composition, and bioactivity throughout maturation, in vitro digestion, and colonic fermentation. It also quantifies their selective reshaping of the human gut microbiota and their enhancement of short-chain fatty acid production. The results indicate a significant conversion of bound to free polyphenols during maturation, with distinct behaviors observed across different digestion stages. More importantly, jackfruit polyphenols selectively enrich beneficial gut bacteria, promote short-chain fatty acid production, and positively regulate gut health.

Jackfruit (Artocarpus heterophyllus Lam.) peel: A better source of antioxidants and a-glucosidase inhibitors than pulp, flake and seed, and phytochemical profile by HPLC-QTOF-MS/MS

Lu Zhang,Zong-cai Tu,Xing Xie,Hui Wang,Hao Wang,Zhen-xing Wang,Xiao-mei Sha,Yu Lu

Jackfruit ( Artocarpus heterophyllus Lam.) peel is an underutilized by-product in both, the production and processing of jackfruit . This research compared the antioxidant and hypoglycemic potential of jackfruit peel with jackfruit pulp, flake and seed for the first time. The phytochemical profile of peel extract was characterized with HPLC-QTOF-MS/MS. Results revealed that peel extract exhibited the highest total phenolic and total flavonoid content, and the phenolics was 4.65, 4.12 and 4.95 times higher than that of pulp, flake and seed extract, respectively. The strongest DPPH and ABTS + scavenging ability, α -glucosidase inhibition were also found in peel extract, and the α -glucosidase inhibition was about 11.8-fold of that of acarbose. The HPLC-QTOF-MS/MS analysis led to the tentative identification of 53 compounds, prenylflavonoids, hydroxycinnamic acids and glycosides are the predominant bioactive compounds. Above results reveal promising potential of jackfruit peel as a new source of natural antioxidants and hypoglycemic agents.

Xingjie Zhang,Kexue Zhu,Jianhua Xie,Yi Chen,Lehe Tan,Shuai Liu,Ruihong Dong,Yuting Zheng,Qiang Yu

The study presented the first comparison of content and composition along with the antioxidant activities of non-extractable polyphenols (NEPP) in the dietary fiber of jackfruit pulp (JDF) released by alkaline, acid and enzymatic hydrolysis . Firstly, the NEPP extraction was optimized by response surface methodology (RSM), and the alkaline hydrolysis extract contained the most abundant content. Besides, the composition of extracts were characterized by UPLC-ESI-QTOF-MS/MS and the alkaline hydrolysis extract possessed most diverse NEPP (4 phenolic acids and 13 flavonoids). Moreover, the antioxidant activities of NEPP were tested by ABTS ·+ , ORAC and PSC scavenging ability assays, the antioxidant activities of alkaline hydrolysis extract were significantly highest among three extracts. This study provided available evidence for the NEPP released from dietary fiber (DF) by different methods, and revealed alkaline hydrolysis extract promising potential as a natural source of health care products.
