Hyaluronanized gold nanoparticles functionalized with CTX-II antibody for early diagnosis and treatment of knee osteoarthritis

Cheng Chen,Cong Tang,Kaiquan Chen,Muhai Deng,Xiangling Pu,Tao Hu,Zhiyu Chen,Ling Zhong,Yunsheng Jiang,Yang Huang

Knee osteoarthritis (KOA) is a prevalent degenerative joint disease that leads to disability. Early diagnosis and intervention can significantly delay KOA progression and improve therapeutic outcomes, yet there is still a lack of effective strategies for the early diagnosis and treatment of KOA. The C-terminal cross-linked telopeptides of type II collagen (CTX-II) is a degradation product of type II collagen in the cartilage matrix under the action of matrix metalloproteinases, serving as an important early diagnostic biomarker of KOA. However, the application of CTX-II is hindered by its fluctuating levels in peripheral body fluids. In this study, we utilized the in situ accumulation of CTX-II in early degenerative cartilage, to develop an integrated diagnostic and therapeutic agent, hyaluronic acid-gold nanoparticles conjugated with CTX-II antibody (HAuNP-CAb). HAuNP-CAb can target and accumulate in the damaged cartilage, enhance imaging contrast, thus enabling the visualization of otherwise invisible (damaged) cartilage in X-ray or CT scans. Additionally, we modified HAuNP-CAb with Cy3 (Cy3-HAuNP-CAb) to achieve fluorescent tracing of early degenerative cartilage regions, offering a novel strategy for the detection of early-stage degenerative cartilage during arthroscopic examinations. The aggregation of Cy3-HAuNP-CAb in degenerative cartilage also exerts an anti-inflammatory therapeutic effect by modulating the IL-17 signaling pathway and downregulating the expression of MMP-13 in chondrocytes, preventing the degradation of the cartilage matrix. Intra-articular injection of Cy3-HAuNP-CAb in rat OA model achieved better therapeutic efficacy as compared to other treatment groups, manifested by lower OARSI and synovitis scores, and more type II collagen deposition. In summary, we reported a feasible approach for in situ targeting of cartilage CTX-II, providing potential new strategies for the clinical diagnosis and treatment of early KOA.

Manganese-induced apoptosis through the ROS-activated JNK/FOXO3a signaling pathway in CTX cells, a model of rat astrocytes

Wan-He Li,Zheng-Ting-Yan Xiang,An-Xin Lu,Su-Su Wang,Chong-Huai Yan

Manganese (Mn) is an essential trace element that maintains many normal physiological functions. However, multi-system disorders would occur once overexposure to Mn, especially neurotoxicity. Despite evidence demonstrating the critical role of ROS-activated JNK/FOXO3a signaling pathway in neuronal survival, the specific mechanisms by which it contributes to Mn-induced neurotoxicity are still unclear. The objectives of this study was to examine the modulation of the JNK/FOXO3a signaling pathway, which is activated by ROS, in Mn-induced apoptosis, using a rat brain astrocyte cell line (CTX cells). This study found that a dose-dependent decrease in cell viability of CTX cells was observed with 150, 200, 250, 300 μmol/L Mn. The results of apoptosis-related protein assay showed that Mn decreased the expression of anti-apoptotic protein Bcl-2 and enhanced the expression of apoptosis-related proteins like Bax and Cleaved-Caspase3. In addition, treatment with Mn resulted in elevated ROS levels and increased phosphorylation levels of JNK. Conversely, phosphorylation of nuclear transcription factors FOXO3a, which regulates expression of transcription factors including Bim and PUMA, was decreased. Depletion of ROS by N-acetyl- L -cysteine (NAC) and inhibition of the JNK pathway by SP600125 prevented Mn-induced JNK/FOXO3a pathway activation and, more importantly, the level of apoptosis was also significantly reduced. Confirmation of Mn-induced apoptosis in CTX cells through ROS generation and activation of the JNK/FOXO3a signaling pathway was the outcome of this study. These findings offer fresh insights into the neurotoxic mechanisms of Mn and therapeutic targets following Mn exposure.

Myofiber directs macrophages IL-10-Vav1-Rac1 efferocytosis pathway in inflamed muscle following CTX myoinjury by activating the intrinsic TGF-β signaling

Liao Zhaohong,Lan Haiqiang,Jian Xiaoting,Huang Jingwen,Wang Han,Hu Jijie,Liao Hua

Background To explore the role of skeletal muscle specific TGF-β signaling on macrophages efferocytosis in inflamed muscle caused by Cardiotoxin (CTX) injection. Methods CTX myoinjury was manipulated in TGF-βr2 flox/flox (control) mice or transgenic mice with TGF-β receptor 2 (TGF-βr2) being specifically deleted in skeletal muscle (SM TGF-βr2 −/− ). Gene levels of TGF-β signal molecules, special inflammatory mediators in damaged muscle or in cultured and differentiated myogenic precursor cells (MPC-myotubes) were monitored by transcriptome microarray or qRT-PCR. TGF-β pathway molecules, myokines and embryonic myosin heavy chain in regenerating myofibers, the phenotype and efferocytosis of macrophages were evaluated by immunofluorescence, immunoblotting, Luminex, or FACS analysis. In vitro apoptotic cells were prepared by UV-irradiation. Results In control mice, TGF-β-Smad2/3 signaling were significantly up-regulated in regenerating centronuclear myofibers after CTX-myoinjury. More severe muscle inflammation was caused by the deficiency of muscle TGF-β signaling, with the increased number of M1, but the decreased number of M2 macrophages. Notably, the deficiency of TGF-β signaling in myofibers dramatically affected on the ability of macrophages to conduct efferocytosis, marked by the decreased number of Annexin-V − F4/80 + Tunel + macrophages in inflamed muscle, and the impaired uptake of macrophages to PKH67 + apoptotic cells transferred into damaged muscle. Further, our study suggested that, the intrinsic TGF-β signaling directed IL-10-Vav1-Rac1 efferocytosis signaling in muscle macrophages. Conclusions Our data demonstrate that muscle inflammation can be suppressed potentially by activating the intrinsic TGF-β signaling in myofibers to promote IL-10 dependent-macrophages efferocytosis. 3D printing and coating to fabricate a hollow bullet-shaped implant with porous surface for controlled cytoxan release

Ning Yang,Huanfei Chen,Huijie Han,Yuanyuan Shen,Song Gu,Yong He,Shengrong Guo

Intratumoral implants have aroused great interests for local chemotherapy of cancer, however, how to efficiently control drug release from implants is still a great challenge. Herein, we designed and prepared a new hollow bullet-shaped implant with porous surface by 3D printing, loaded chemotherapeutic agent cytoxan (CTX) with tetradecyl alcohol or lecithin as matrix and coated it with poly (lactic acid) to obtain a CTX implant, which has a highly tuned drug release property with a drug release time from 4 h to more than 1 month. The drug release from the implant can be easily controlled by changing pore sizes, kinds of matrices, and coating thickness.

Cordycepin attenuated cyclophosphamide (CTX)-induced immunosuppression in mice via EGFR/Nrf2 antioxidant signaling pathway

Mengling Zhong,Mingmei Feng,Xinlin Chen,Yuxin Zhang,Yupeng Wang,Peizhi Yuan,Tianyao Zhang,Zhipeng Han,Chenglun Tang,Qi Zhang,Yang Sun

Immunosuppression causing the host more vulnerable to pathogen invasion, increases the risk of malignant tumors and inflammatory diseases. Cordycepin possesses diverse pharmacological activities, including anti-inflammatory, antioxidant, and antitumor effects. However, the immunomodulatory mechanisms of cordycepin remain poorly understood. This study aimed to explore the immunoregulatory effect of cordycepin and elucidate its underlying mechanisms. In our study, cordycepin restored body weight, organ indices and ameliorated splenic damage in cyclophosphamide (CTX)-induced immunosuppressed mice. Cordycepin enhanced the proliferation of T and B lymphocytes stimulated by ConA and LPS. Cordycepin normalized the counts of white blood cells (WBC), lymphocytes (LYM), neutrophils (NEU) and eosinophils (EOS), and increased the levels of immunoglobulins (immunoglobulin A (IgA), IgM, IgG). Cordycepin also increased the production of pro-inflammatory cytokines IL-2 and IFN-γ, but decreased the level of inhibitory cytokines IL-10. Additionally, cordycepin reduced serum level of malondialdehyde (MDA), enhanced content of glutathione (GSH) and the enzymatic activities of superoxide dismutase (SOD) and catalase (CAT). Moreover, metabolomic analysis of spleen and serum revealed that cordycepin counteracted the metabolic disturbance induced by CTX, particularly regulating pyruvate metabolism pathway. Network pharmacology approaches suggested that cordycepin modulated metabolic and immune pathways by targeting EGFR. Furthermore, cordycepin reduced the protein level of phosphorylated EGFR and upregulated the protein expression of Nrf2, NQO1 and HO-1 in the spleens of immunosuppression mice. In conclusion, this study demonstrated that cordycepin ameliorated CTX-induced immunosuppression of mice by reversing metabolic dysfunction and activating Nrf2 pathway through regulating EGFR, indicating its potential as a therapeutic agent for immunosuppression.

Magnetic imprinted surface enhanced Raman scattering biosensor for the detection of bone loss biomarker CTX-I

Zheng Diantao,Li Qiuyan,Hu Ran,Zhang Baoliang

Serum C-terminal telopeptide of type I collagen (CTX-I) is a key bone turnover biomarker in osteoporosis, and its elevated levels are indicative of increased bone resorption. Current techniques for CTX-I measurement suffer from high operational requirements, complex procedures, and high costs. This study focuses on a novel strategy for detecting serum CTX-I concentration to predict the occurrence and severity of osteoporosis. Using silver-capped Fe 3 O 4 (Fe 3 O 4 @Ag) as the substrate material, an artificial antibody for CTX-I was prepared via magnetic core-shell molecularly imprinted polymers (MIPs), and surface-enhanced Raman scattering (SERS) was used as a signal output device. With crystal violet (CV) at 10 −4 M as the reporter molecule, the proposed biosensor exhibited a good linear response over the range of 10 pg/mL to 2 ng/mL (R 2  = 0.986, LOQ = 4.79 pg/mL), covering the normal reference range required for identifying bone turnover. Moreover, the biosensing platform demonstrated resistance to matrix interference in real serum and urine samples. After six reuse cycles, the SERS intensity of CV decreased by only 10.5%, and after 15 days of storage at 4 °C, it decreased by 15.7%. The Fe 3 O 4 @Ag-MIPs were used to detect CTX-I with excellent uniformity (RSD = 4.1%) and repeatability (RSD = 2.7%), and the overall analysis time was 30 min. The proposed biosensing platform was applied to analyze known real serum samples obtained from human. The proposed biosensor was validated using enzyme-linked immunosorbent assay (ELISA), and the developed biosensor showed good correlation with ELISA. The experimental results indicate that the proposed novel biosensing platform is a fast, low-cost, and reliable tool for the rapid detection of the bone biomarker CTX-I in serum or urine. Yan Zhang,Hui Wang,Qin Ge,Jing Shi,Huiying Zhang,Jiaqi Gao,Jiaxin Han

MLS is a bioactive extract of Melientha longistaminea shoots. In this study, we characterized the preliminary structure of MLS. And establish in vivo immune model experiments. To investigate the potential mechanism of immunomodulatory effects on CTX-induced immunosuppressed mice. From the experimental results, we found that MLS restored immune organ damage in immunosuppressed mice, increased cytokine and immunoglobulin secretion in the spleen, and attenuated oxidative damage to the liver by CTX. Meanwhile, MLS was also effective in ameliorating immune injury in immunosuppressed mice by activating the NF-κB and Nrf2 signaling pathways. MLS effectively improved the composition of the gut flora and increased the production of SCFAs in the gut. The results indicate that MLS has immunomodulatory effects and provides a research basis for developing Melientha longistaminea polysaccharides as immunomodulatory adjuvants.

Effects of Segatella copri-accessible polysaccharides on gut microbiota and intestinal immune function of CTX-induced immunocompromised mice

Yizhen Lyu,Lianghua Wen,Yadong Zhong,Dafang Yin,Mine Yang,Yanlong Wen,Yang Li,Jielun Hu

Segatella copri (formerly Prevotella copri ) is a major indigestible polysaccharides consumer in the lower gut, and its interaction with polysaccharides is closely related to host health. To investigate the effects of S. copri -accessible polysaccharides with different structures (SAPS) on gut microbiota and intestinal immunity, single strain and gut microbial fermentation were performed for accessibility testing, respectively, followed by cyclophosphamide (CTX)-induced immunocompromised mice experiment. During fermentation, xylan, arabinan, arabinoxylan, and arabinogalactan (four SAPS) promoted S. copri and gut microbiota growth with increased OD 600 values, and enhanced the short-chain fatty acids (SCFAs) production, predominantly acetic acid. However, S. copri could not utilize xyloglucan. Interestingly, the four SAPS ameliorated CTX-induced immunocompromise with increased TNF-α, INF-γ, IL-6, IL-17, IL-2 in colon and index of spleen and thymus. Additionally, the four SAPS increased the relative abundance of Lactobacillus , Bacteroides , and S. copri , while inhibiting Oscillospira growth. Furthermore, they significantly promoted colonic levels of SCFAs, as well as altering the fecal metabolomic profile, e.g. Leukotriene C4, 5,6-DHET, N-Methylserotonin and 24-Hydroxycholesterol, which were involved in arachidonic acid metabolism, tryptophan metabolism, and primary bile acid biosynthesis. Notably, arabinoxylan exhibited advantages in enhancing spleen index and MUC2 expression, whereas arabinan was more effective in increasing IL-17 and IL-6. Additionally, arabinogalactan increased colonic Claudin-1 expression, and xylan showed more benefits in promoting SCFAs production. In summary, the current research explored the regulation of gut microbiota and intestinal immunity by SAPS, providing a theoretical basis for the research on the relationship among polysaccharide structure, gut microbiota, and intestinal immunity.

Fabrication of magnetic nanochains linked with CTX and curcumin for dual modal imaging detection and limitation of early tumour

Yuedi Yang,Zhongbing Huang,Ximing Pu,Guangfu Yin,Lei Wang,Fabao Gao

Objective Five-year survival rate at early lung tumour was about 70%; however, its early diagnosis rate was still at a low level, so the enhancement of diagnosis level for early lung tumour is the key factor to increase the survival rate. Diagnosis and therapy of early lung tumour are still challenged. Methods The magnetic nanochains (NCs) with biocompatibility and transverse relaxivity (r 2 = 231 Fe mmol l −1 s −1 ) were fabricated through a co-precipitation method in the assistance of dextran, and then, linked with chlorotoxin (CTX) and curcumin (Cur) via the PEGylation and carbodiimide technique (named as CTX-NCs-Cur). Results The results of cell test indicated that CTX-conjugated NCs could obviously target non–small-cell lung cancer cells and limit their growth. The in vivo results of magnetic resonance imaging and fluorescence imaging indicated that the CTX-NCs-Cur significantly targeted the tumour site and enhanced images contrast of the small-size tumour. Moreover, the results of everyday tail-vein injection confirmed that CTX-NCs-Cur could significantly limit the growth of early tumour, due to blocking Cl ion channels from CTX-NCs-Cur-MMP-2 composite and intracellular ROS increase from Cur treatment. Conclusions We provided a mechanism about the effect of CTX-NCs-Cur on the targeting and limiting early tumour, and these results indicated the application foreground of CTX-NCs-Cur in tumour diagnosis and therapy.

Caragana sinica (Buc'hoz) Rehd. (jin ji er) polysaccharide regulates the immune function and intestinal microbiota of cyclophosphamide (CTX) induced immunosuppressed mice

Yan Zhang,Qirui Xu,Yazi Wang,Chenchen Zhang,Shan Xu,Manhong Luo,Shuhan Yang

s Ethnopharmacological relevance Caragana sinica (Buc'hoz) Rehd. is a plant widely grown in Yunnan, China, for both medicinal and edible purposes. The “National Compilation of Chinese Herbal Medicine” describes its nature as “slightly temperate and sweet”. Caragana sinica is usually medicated with whole herbs, the main function is to replenish the kidneys and stop bleeding. Caragana sinica was used in folk medicine in Chuxiong, Yunnan, to treat deficiency colds, fatigue, fever, cough, hypertension, and other diseases. Aim of the study This article investigates the structural characteristics of Caragana sinica polysaccharide (CSP) and explores its immune-regulatory activity and molecular biological mechanisms in cyclophosphamide-induced immunosuppressed mice, as well as its effects on intestinal bacteria. Methods With the water-extraction and alcohol-precipitation method, Caragana sinica polysaccharide were extracted, obtaining CSP by purification. A variety of methods and techniques have been used to analyze the chemical properties and structural characteristics of CSP. Immunosuppressive mice model was established through intraperitoneal injection of cyclophosphamide (CTX) to study the immune-regulatory effects and mechanisms of CSP. Results The data indicated that CSP is a neutral heteropolysaccharide mainly composed of arabinose and galactose. This article uses immunosuppressive mice induced by cyclophosphamide (CTX) as the model. The results showed that CSP can promote the immune function of CTX treated immunosuppressed mice and regulate the diversity and composition of intestinal microbiota. CSP can increase macrophage phagocytosis, NK cell killing activity, and lymphocyte proliferation activity. It can also repair the index and morphological damage of the thymus and spleen. And by binding to the TLR4 receptor, MyD88 was activated and interacted with TRAF6 to promote the transfer of NF-κB into the nucleus. Thereby promoting cytokine release and increasing the production of IL-1β, IL-6, IL-10, TNF-α, IgA, and IgG in the serum. CSP also effectively alleviated the liver damage caused by CTX through antioxidant activity. Furthermore, CSP can dramatically affect the intestinal microbiota and the body's immunity by boosting the relative presence of Bacteroides and Verrucamicrobiota . Conclusions Research results indicated that CSP can regulate the immune function of mice, providing a basis for developing CSP as a potential immune modulator and functional food.

Blood-Enriching Effects and Immune-Regulation Mechanism of Steam-Processed Polygonatum Sibiricum Polysaccharide in Blood Deficiency Syndrome Mice

Wang Juan,Wang Furong,Yuan Lixia,Ruan Hongsheng,Zhu Zhibiao,Fan Xiaoling,Zhu Lingyan,Peng Xin

Polygonatum sibiricum Red. has been used as a medicinal herb and nutritional food in traditional Chinese medicine for a long time. It must be processed prior to clinical use for safe and effective applications. However, the present studies mainly focused on crude Polygonatum sibiricum (PS). This study aimed to investigate the chemical properties, blood-enriching effects and mechanism of polysaccharide from the steam-processed Polygonatum sibiricum (SPS), which is a common form of PS in clinical applications. Instrumentation analyses and chemistry analyses revealed the structure of SPS polysaccharide (SPSP). A mice model of blood deficiency syndrome (BDS) was induced by acetylphenylhydrazine (APH) and cyclophosphamide (CTX). Blood routine test, spleen histopathological changes, serum cytokines, etc. were measured. The spleen transcriptome changes of BDS mice were detected by RNA sequencing (RNA-seq). The results showed that SPSP consists predominantly of Gal and GalA together with fewer amounts of Man, Glc, Ara, Rha and GlcN. It could significantly increase peripheral blood cells, restore the splenic trabecular structure, and reverse hematopoietic cytokines to normal levels. RNA-seq analysis showed that 122 differentially expressed genes (DEGs) were obtained after SPSP treatment. GO and KEGG analysis revealed that SPSP-regulated DEGs were mainly involved in hematopoiesis, immune regulation signaling pathways. The reliability of transcriptome profiling was validated by quantitative real-time PCR and Western blot, and the results indicated that the potential molecular mechanisms of the blood-enriching effects of SPSP might be associated with the regulating of JAK1-STAT1 pathway, and elevated the hematopoietic cytokines (EPO, G-CSF, TNF-α and IL-6). This work provides important information on the potential mechanisms of SPSP against BDS.
