A novel ratiometric electrochemiluminescent (ECL) biosensor was designed for the detection of concanavalin A (Con A) based on two ECL emitters competing for the dissolved oxygen (O 2 ). In this strategy, CdTe quantum dots (QDs) were used as the cathodic emitter and N-(aminobutyl)-N-(ethylisoluminol) (ABEI) was used as the anodic emitter. With the presence of dissolved O 2 utilized as co-reactant, CdTe QDs showed an ECL emission at − 1.7 V, and ABEI showed an emission at + 0.6 V. Phenoxy-derivatized dextran (Dexp), as a recognition element, was immobilized by graphene oxide functionalized CdTe QDs (G-CdTe QDs) to recognize Con A, and further combine with Dexp, gold and platinum nanoparticles decorated ABEI (Dexp-Au-Pt-ABEI) to form a sandwich structure. With the increasing concentration of analyte Con A, the ECL signal of cathodic CdTe QDs decreased and the anodic response of ABEI increased, thus achieving a ratiometry detection of Con A with a wide linear range from 1.0 × 10 −4 ng/mL to 10 ng/mL. The detection limit was low to 3.0 × 10 −5 ng/mL. This proposed ratiometric ECL biosensor not only conquered the false errors caused by external interferences, but excluded the disability caused by exogenous co-reactant via the application of common dissolved O 2 . It also exhibited an excellent stability, selectivity and reproducibility.

A solid-state electrochemiluminescence (ECL) biosensor for the detection of concanavalin A (Con A) was constructed using graphene quantum dots (GQDs) as luminophore, which were loaded on polyamidoamine (PAMAM) functionalized CeO 2 @Ag NPs by the interaction between the NH 2 of PAMAM and the COOH of GQDs. The electrodepositing gold layers were used as matrix to immobilize glucose oxidase (GOx). GOx served as recognition elements to further capture Con A by the specific interaction between carbohydrate and Con A. With the increase of the concentration of Con A, the amount of GOx-CeO 2 @Ag-GQDs incubated onto the electrode was increased and a higher ECL signal was obtained. The constructed biosensor exhibited a sensitive response towards Con A and a low detection limit of 0.16 pg/mL was obtained. Such a construction strategy provides a new approach for applying GQDs to establish a solid-state ECL biosensor.

Intake of Concanavalin A (Con A), a hazardous lectin protein commonly discovered in legume vegetables, could cause various systemic symptoms including rash, nausea, vomiting, diarrhea, etc. Herein, we present a novel label-free and cost-effective electrochemical Con A aptasensor. [Fe(CN) 6 ] 3−/4- ions in testing buffer not only generate redox peaks during electrochemical scan, but also serve as electron acceptor/donor to enhance electron transfer. Anti-Con A aptamer is immobilized on surface of screen printed electrode to capture Con A molecules and form aptamer-protein complex, which hinders electron transfer near electrode surface and causes a decrease in peak current value. The constructed aptasensor displays a low detection limit of 17.88 ng/mL (around 171.92 pM), as well as good specificity against other legume lectins. The practicality of electrochemical sensors was evaluated using real-world samples, demonstrating their ability to rapid detect Con A in crude protein extracts from jack bean seeds. This work provides novel tools for rapid evaluation of Con A content in massive samples, which is required in food safety inspection and safety breeding of legume vegetables.

In this work, a soft and nondestructive approach was developed to prepare concanavalin A-chelating magnetic nanoparticles (Con A-MNPs) for selective enrichment of glycoproteins. Ethylenediamine tetraacetic acid-modified-MNPs (EDTA-MNPs) were prepared by a one-pot chemical coprecipitation method first, and then, Cu(II) cations were used as bridge groups to immobilize Con A on EDTA-MNPs. The as-prepared absorbents with a mean diameter of 15 nm showed a strong magnetic response to an externally applied magnetic field. The results of thermogravimetric analysis showed the content of immobilized Con A was up to 28 wt %. For glycoprotein ovalbumin, the maximum capacity and equilibrium constant were 72.41 mg/g and 0.6035 L/mg, respectively. The as-prepared nanocomposites exhibited a remarkable selectivity for glycoproteins and can enrich glycoproteins specifically from a mixture of glycoprotein and nonglycoprotein even at a molar ratio of 1:600. It was also successfully applied for the enrichment of glycoproteins from real egg white samples. We expect that our finding will serve as a helpful template for others to design new adsorbents for enriching glycoproteins.

A low-cost, simple, portable and sensitive paper-based electrochemical sensor was established for the detection of K-562 cell in point-of-care testing. The hybrid material of 3D Au nanoparticles/graphene (3D Au NPs/GN) with high specific surface area and ionic liquid (IL) with widened electrochemical windows improved the good biocompatibility and high conductivity was modified on paper working electrode (PWE) by the classic assembly method and then employed as the sensing surface. IL could not only enhance the electron transfer ability but also provide sensing recognition interface for the conjugation of Con A with cells, with the cell capture efficiency and the sensitivity of biosensor strengthened simultaneously. Concanavalin A (Con A) immobilization matrix was used to capture cells. As proof-of-concept, the paper-based electrochemical sensor for the detection of K-562 cells was developed. With such sandwich-type assay format, K-562 cells as model cells were captured on the surface of Con A/IL/3D AuNPs@GN/PWE. Con A-labeled dendritic PdAg NPs were captured on the surface of K-562 cells. Such dendritic PdAg NPs worked as catalysts promoting the oxidation of thionine (TH) by H 2 O 2 which was released from K-562 cells via the stimulation of phorbol 12-myristate-13-acetate (PMA). Therefore, the current signal response was dependent on the amount of PdAg NPs and the concentration of H 2 O 2 , the latter of which corresponded with the releasing amount from cells. So, the detection method of K-562 cell was also developed. Under optimized experimental conditions, 1.5×10 −14 mol of H 2 O 2 releasing from each cell was calculated. The linear range and the detection limit for K-562 cells were determined to be 1.0×10 3 –5.0×10 6 cells/mL and 200 cells/mL, respectively. Such as-prepared sensor showed excellent analytical performance with good fabrication reproducibility, acceptable precision and satisfied accuracy, providing a novel protocol in point-of-care testing of cells.

Glucose and pH dual-responsive microhydrogels based on concanavalin A (Con A) were prepared and used for insulin delivery. The combination of the specific saccharide-binding affinity of Con A and the cationic groups of N-(2-(dimethylamino) ethyl)-methacrylamide (DMAEMA) led to dual-responsive systems. SEM, fluorescence microscopy and particle size analysis showed that the obtained microhydrogels had a dense surface morphology and an average size of 38 μm. The in vitro insulin release study revealed that the microhydrogels could quickly respond to the changes of glucose concentrations in the medium and small change in pH value of the environment. The kinetics of insulin release was analyzed by using empirical equation and the apparent diffusion coefficient was calculated according to a solution of Fick's second law. The released insulin was proved to remain active. The result suggested that this microhydrogel might find potential applications for self-regulated insulin delivery, actuators and separation systems with sensitivity to glucose.

Concanavalin A (Con A) has been widely used as glucose recognition element in glucose sensors, however, these sensors typically detect glucose by fluorescence, and suffer from laborious labeling processes. Here a new label-free optical Con A-based glucose sensor was designed. The sensor uses Con A-containing thin hydrogel films as sensing material and Fabry-Perot cavity simultaneously, which were fabricated by layer-by-layer assembly of Con A and dextran, using lectin-saccharide biospecific interaction as driving force. These films display Fabry-Perot fringes on their reflection spectra, from which optical path length of the films can be calculated. The films swell upon addition of glucose, which causes a shift of Fabry-Perot fringes on the reflection spectra of the film, from which the glucose concentration can be reported. The glucose-induced swelling is reversible, and the increase in optical path length increases linearly with glucose concentration over a wide range of glucose concentration. The sensor works well at physiological temperature, pH and ionic strength. Other sugars, which may present in blood, do not interfere with glucose detection, because of their very low concentrations in the blood. Particularly, unlike other hydrogel-based sensors, this sensor responds quite fast, because the hydrogel films are very thin. The new sensor may have potential for real time, continuous glucose monitoring.

A new signal amplification strategy based on target-induced proximity ligation assay accompanying three-way junction-based rolling chain amplification was designed for ultrasensitive detection of concanavalin A (Con A) by coupling with a sequential injection mode. To construct such a proximity ligation assay system, two types of magnetic sensing probes including glucosamine/DNA1-conjugated magnetic bead (GA-MB-DNA1) and glucosamine/DNA2-labeled magnetic bead (GA-MB-DNA2) were first synthesized and prepared through a typical carbodiimide coupling. In the presence of target Con A, GA-MB-DNA1 and GA-MB-DNA2 were ligated together based on the interaction between Con A and the conjugated glucosamine on the MB, thereby resulting in the formation of a three-way DNA junction because of partial base pairing on the DNA1/DNA2. With the aid of ligase and polymerase, the formed three-way DNA junction could be used as the primer to produce numerous repeated oligonucleotide sequences through rolling circle amplification (RCA) reaction. The formed long oligonucleotide strand could cause the intercalation of numerous positively charged methylene blue molecules with a negatively charged DNA backbone. During the electrochemical measurement, each of the intercalated indicators could produce an electrochemical signal within the applied potentials, resulting in the amplification of detectable electronic signal. By monitoring the change in the signal, we could indirectly determine the concentration of target Con A in the sample. Under the optimal conditions, the developed sensing platform exhibited high sensitivity for detection of Con A with a wide dynamic range of 1.96 pM to 98 nM and a low limit of detection (LOD) of 1.5 pM at the 3sB level. Intra-assay and interassay coefficients of variation were less than 8.9% and 9.7%, respectively. In addition, the methodology was validated by assaying Con A spiked samples including newborn cattle serum and peanuts, and the recovery in all cases was 88.8–134.7%.

Herein, a three-dimensional carbohydrate modified polyacrylamide hydrogel microarray (3D carbohydrate hydrogel microarray) has been fabricated and employed as micro-reactor for capturing Escherichia coli ( E. coli ) by multivalent binding of concanavalin A (Con A) with O-antigen on the cellular surface of E. coli and immobilized monosaccharides on hydrogel spot, and the interactions of type 1 fimbriae of E. coli with immobilized monosaccharides. Because of the transparent performance of polyacrylamide hydrogel, the captured E. coli can be directly observed by a conventional microscope under a bright-field mode. The experimental result demonstrates that α-D-mannopyranoside (Man-α) modified hydrogel surface shows high efficiency of E. coli capturing. The 3D Man-α hydrogel microarray-based assay shows reasonable low detection limit (1.0×10 4 cells/mL) and large dynamic range (1.0×10 5 to 1.0×10 9 cells/mL) for detecting E. coli . In addition, bacterial adhesion inhibition assay has been demonstrated by the interactions of E. coli with ten saccharides , and satisfactory results have been obtained.

A novel signal-on electrochemiluminescence (ECL) biosensor for detecting concanavalin A (Con A) was fabricated with phenoxy dextran-graphite-like carbon nitride (DexP-g-C 3 N 4 ) as signal probe . In this construction strategy, the nanocomposites of three-dimensional graphene and gold nanoparticles (3D-GR-AuNPs) were used as matrix for high loading of glucose oxidase (GOx), which served as recognition element for bounding Con A. Con A further interacted with DexP-g-C 3 N 4 through a specific carbohydrate-Con A interaction to achieve a sandwiched scheme. With the increase of Con A incubated onto the electrode, the ECL signal resulted from DexP-g-C 3 N 4 would enhance, thus achieving a signal-on ECL biosensor for Con A detection. Due to the integration of the virtues of 3D-GR-AuNPs and the excellent ECL performance of DexP-g-C 3 N 4 , the prepared biosensor exhibits a wide linear response range from 0.05 ng/mL to 100 ng/mL and a low detection limit of 17 pg/mL (S/N=3).

A novel “signal-on” biosensor for the detection of concanavalin A (Con A) was designed utilizing an enzymatic reaction to in situ generate coreactant of poly(9,9-di- n -octylfluorenyl-2,7-diyl) (PFO). Firstly, PFO polymers were wrapped with graphene oxide nanosheets to obtain GO@PFO nanocomposite, which was modified onto the surface of the electrode as a matrix for immobilizing phenoxy-derivatized dextran (Dexp) through π-π stacking interaction. Then Con A interacted with Dexp through the specific carbohydrate-protein interaction. Finally, glucose oxidase (GOx) and lactate oxidase (LOx) decorated gold nanoparticles@C 60 -Polyamidoamine (Au@C 60 -PAMAM) was bound onto Con A via a biospecific interaction between Con A and the internal residues of GOx, thus achieving a sandwich structure. In this construction strategy, LOx catalyzed the oxidation of lactate to in situ generate H 2 O 2 , which could act as the coreactant of PFO to enhance the ECL intensity. Due to the excellent ECL property of PFO–H 2 O 2 system, our prepared biosensor showed a wide linear range from 0.001 ng/mL to 10 ng/mL, and a low detection limit down to 0.0003 ng/mL. The integration of excellent ECL property of PFO–H 2 O 2 system and excellent catalytic activity of Au@C 60 -PAMAM would open a new and promising platform for fabricating ECL biosensors and biocatalysts.

Diabetes mellitus is a long-term chronic disease characterized by abnormal high level blood glucose (BG). An artificial closed-loop system that mimics pancreatic β-cells and releases insulin on demand has potential to improve the therapeutic efficiency of diabetes. Herein, a lectin Concanavalin A modified oxidized starch nanogel was designed to regulate glucose dynamically according to different glucose concentrations. The nanogels were formed by double cross-linking the Concanavalin A and glucose units on oxidized starch via specific binding and amide bonds to achieve the high drug loading and glucose responsiveness. The results showed that oxidized starch nanogels prolonged the half-life of antidiabetic peptide drug exenatide and released it in response to high BG concentrations. It could absorb BG at a high level and maintain glucose homeostasis. Besides, the oxidized starch nanogels performed well in recovering regular BG level from hyperglycemia state and maintaining in euglycemia state that fitted in a biological rhythm. In addition, the nanogels showed high biocompatibility in vivo and could improve plasma half-life and therapeutic efficacy of exenatide. Overall, the nanogels protected peptide drugs from degradation in plasma as a glucose-responsive platform showing a high potential for peptide drugs delivery and antidiabetic therapy.
