Cynanchum auriculatum Royle ex Wight, commonly known as “Baishouwu,” has been traditionally used in China for its medicinal and dietary benefits. Despite its long history of use, the potential therapeutic effects of C. auriculatum in the treatment of colitis have not been fully investigated. This study aims to evaluate the effects of the water extract of C. auriculatum root on colitis and elucidate its potential mechanisms of action. The water extract of C. auriculatum root (CW) was prepared and characterized using UPLC-Q-TOF-MS, identifying thirty-two distinct compounds, including saponins, organic acids, fatty acid derivatives, and alkaloids. The therapeutic efficacy of CW was assessed in a colitis mouse model. CW significantly alleviated colitis symptoms, evidenced by increased colon length, reduced disease activity indices, and decreased colon tissue damage. CW reduced colonic inflammatory cytokine production and enhanced the expression of tight junction proteins, including claudin-1, occludin, and ZO-1, thereby strengthening intestinal barrier integrity. Additionally, CW modulated the gut microbiota by increasing microbial diversity, promoting beneficial Lactobacillus growth, reducing pathogenic Pseudomonas levels, and enhancing short-chain fatty acid production. The results suggest that CW exhibits significant therapeutic potential in the management of colitis by attenuating inflammation, restoring gut barrier function, and modulating the gut microbiota. These findings provide a basis for further exploration of C. auriculatum as a functional food for prevention and treatment of colitis.

The crude polysaccharides CAPS and CAP of Cynanchum Auriculatum , which were prepared by degrading starch by single-enzymatic method (α-amylase) and double-enzymatic method (α-amylase and glucoamylase) respectively, were compared. CAP had good water solubility and higher non-starch polysaccharide content. A homogeneous neutral polysaccharide CAP-W, with the degree of acetylation about 17 %, was obtained from CAP by anion exchange column chromatography. Its detailed structure was identified by various methods. CAP-W, with the weight average molecular weight of 8.4 kDa, was composed of mannose, glucose, galactose, xylose, and arabinose in a molar ratio of 1.27:1.00:0.25:0.10:1.16. The backbone included β -1,4-Man p , β -1,4,6-Man p , β -1,4-Glc p and β -1,4,6-Glc p residues, with branches at the O-6 position of β -1,4,6-Man p and β -1,4,6-Glc p residues, consisting of α -T-Ara f , α -1,5-Ara f , α -1,2,5-Ara f , α -1,3,5-Ara f , T-Xyl p ,1,4-Xyl p , β -T-Man p and β -T-Gal p residues. In vitro immunological experiments suggested that CAP-W improved the phagocytic ability of macrophages, stimulated the release of NO, TNF-α and IL-6 from RAW264.7 cells, promoted the expression of NF-κB and caused nuclear translocation of NF-κB p65.

A novel polysaccharide fromCynanchum auriculatumRoyle ex Wight was isolated, structurally characterized, and its antioxidant activity was evaluated. The crude extract was purified by ion exchange and size exclusion chromatography to obtain a homogeneous fraction, CAP2-1. CAP2-1 displayed a weight-average molecular mass of 184.17 kDa and is mainly composed of galactose, arabinose, and galacturonic acid. Structural analysis revealed that CAP2-1 is a highly branched acidic arabinogalactan-type polysaccharide with a backbone of →6)-β-D-Galp-(1→, →3,6)-β-D-Galp-(1→, and →4)-α-D-GalpA-(1→ units, and side chains enriched in α-L-arabino furanose residues. Ultrasonic degradation produced a lower-molecular-weight derivative, UCAP2-1, which exhibited significantly stronger free radical scavenging ability compared with CAP2-1 (p< 0.01). These findings suggest that molecular weight reduction enhances antioxidant properties by improving electron-donating capacity and accessibility to reactive sites. This study reveals the structure–antioxidant relationship of CAP2-1 and UCAP2-1 and highlights UCAP2-1 as a promising natural antioxidant.
