Antioxidant and neuroprotective effects of Dictyophora indusiata polysaccharide in Caenorhabditis elegans

Ju Zhang,Ruona Shi,Haifeng Li,Yanxia Xiang,Lingyun Xiao,Minghua Hu,Fangli Ma,Chung Wah Ma,Zebo Huang

Ethnopharmacological relevance Dictyophora indusiata is a medicinal mushroom traditionally used in China for a variety of conditions, including inflammatory and neural diseases. D. indusiata polysaccharides (DiPS) are shown to have in vitro antioxidant activity but in vivo evidence is lacking. This study aimes to explore the antioxidant capacity and related neuroptotective activities of DiPS using wild-type and neurodegenerative Caenorhabditis elegans models. Materials and methods The antioxidant capacities of DiPS were first determined using paraquat survival and Pgst-4 ::GFP expression assays in wild-type and transgenic C. elegans models, respectively, and then further investigated by determining reactive oxygen species (ROS) level, malondialdehyde (MDA) content and superoxide dismutase (SOD) activity as well as functional parameters of mitochondria. The activation of stress response transcription factors and neuroptotective activities were examined using nuclear localization and chemosensory behavioral assays in transgenic nematodes, respectively. Results DiPS was shown not only to increase survival rate and reduce stress level under paraquat-induced oxidative conditions but also to decrease ROS and MDA levels and increase SOD activity in C. elegans models. Moreover, DiPS was also able to restore the functional parameters of mitochondria, including membrane potential and ATP content, in paraquat-stressed nematodes. In addition, nuclear translocation assays demonstrate that the stress response transcription factor DAF-16/FOXO was involved in the antioxidant activity of the polysaccharide. Further experiments reveal that DiPS was capable of reducing ROS levels and alleviating chemosensory behavior dysfunction in transgenic nematode models of neurodegenerative diseases mediated by polyglutamine and amyloid-β protein. Conclusions These findings demonstrate the antioxidant and neuroprotective activities of the D. indusiata polysaccharide DiPS in wild-type and neurodegenerative C. elegans models, and thus provide an important pharmacological basis for the therapeutic potential of D. indusiata in neurodegeneration .

Comprehensive Analysis of Volatile and Nonvolatile Compounds in Dictyophora indusiata Hydrolysates

Haibin Yuan,Tianyang Wang,Lan Xiao,Hongfeng Jia,Huachang Wu,Ping Dong

Although enzymatic hydrolysates of edible fungi are widely used as flavoring agents in the food industry, the effects that different enzymes have on the flavor profile of Dictyophora indusiata remain unclear. In this study, neutrase, papain, cellulase-neutrase, and cellulase-papain were used to hydrolyze D. indusiata , and the taste and odor profiles then investigated via HPLC, electronic tongue sensor, HS-GC-IMS, and HS-SPME-GC-MS analyses. Multivariate statistical analyses of PLS-DA and variable importance projection values were employed to evaluate the volatile compounds. The enzymatic hydrolysate treated with neutrase contained a higher amount of free amino acids, whereas the cellulase-papain enzymatic hydrolysate demonstrated a more pronounced umami taste. Electronic tongue sensors exhibited greater sensitivity to nucleotides. HS-GC-IMS and HS-SPME-GC-MS identified 78 and 92 volatile aroma compounds, respectively. Dual-enzyme groups had a more significant impact on the variety and content of characteristic volatile organic compounds than the single-enzyme groups did. Overall, this study elucidated changes in the flavor profiles of different enzyme hydrolysates, which could provide a better understanding of the scientific processing and condiment development of D. indusiata .

Structural characterization and bifidogenic activity of polysaccharide from Dictyophora indusiata

Lei Pan,Lin Wang,Fan Zhang,Yi Zhang,Baodong Zheng

Dictyophora indusiata is an edible and medicinal fungi with various nutritious, especially higher in carbohydrates . In this work, polysaccharide from Dictyophora indusiate (DIP) was extracted by the hot-water, and four polysaccharide fractions (DIP0, DIP1, DIP2 and DIP3) were obtained using cellulose DE-52 column. The objective of this study was to investigate the structure and bifidogenic activity of four polysaccharide fractions from Dictyophora indusiate . Structure of DIP was evaluated by HPLC , FT-IR and SEM, and the results indicated that four polysaccharide fractions from Dictyophora indusiate were consisted of mannose , glucose and galactose with almost the same functional groups. Remarkably, the DIP3 had significant differences in monosaccharide compositions, which additionally contains glucuronic acid . In addition, DIP0 exhibited a three-dimensional network structure formed by crosslinked chains, while DIP1, DIP2 and DIP3 displayed the irregular flake structure. During the periods of fermentation in vitro , a strong increase of Bifidobacterium longum was observed in the presence of DIP0, DIP1 and DIP2. In particular, Bifidobacterium longum produced more acetic acid in the medium with DIP0 as the carbon source during the fermentation, probably due to the fact that DIP0 contained higher mannose and lower glucose. These results suggest that DIP0 promoted the growth of Bifidobacterium longum and its metabolic synthesis of acetic acid, which would provide necessary information for the prebiotic studies of DIP.

Hierarchical structured TiO2 templated by dictyophora indusiata for enhanced photocatalytic activity

Zi-Chang Wu,Yi Lu,Yong Wang,Yuan-Zhou Li,Li He,Xiu Cheng,Wei Geng,Jie Ying,Gang-Gang Chang

A hierarchical structured TiO 2 @C photocatalyst has been fabricated by using P25 as model nano-photocatalyst and dictyophora indusiata as a natural hierarchical template, which showed significantly higher photocatalytic performance in comparison with P25 and M-TiO 2 @C. The enhanced catalytic properties can be mainly ascribed to the formation of titanium vacancy and promoted diffusion and transport of substrate molecule benefited from the hierarchical structure.

Dynamic structural evolution and in vitro colonic fermentability of modified Dictyophora indusiata soluble dietary fibers

Mengfan Lin,Meiling Dai,Changrong Wang,Zebin Guo

Steam explosion reduced crystallinity, enriching primary degraders and butyrate. • Cellulase hydrolysis reorganized hydrogen bonds, enriching Bifidobacterium. • Both structural modifications competitively excluded Escherichia-Shigella. • Dynamic structural evolution of SDFs dictates specific prebiotic mechanisms. Understanding the degradation behavior of soluble dietary fibers (SDFs) during gastrointestinal transit is essential for elucidating their prebiotic mechanisms and optimizing their application as functional food ingredients. This study investigated the dynamic structural evolution and in vitro colonic fermentability of Dictyophora indusiata SDFs modified by steam explosion (S-SDF) and cellulase hydrolysis (E-SDF). After surviving simulated upper gastrointestinal digestion, the modified SDFs exhibited distinct structural degradation kinetics during fecal fermentation. Thermomechanical disruption facilitated the progressive decrystallization of S-SDF (reducing crystallinity to 1.9% at 48 h). This time-dependent structural transition was accompanied by an increase in the relative abundance of Bacteroidota to 18.38% and a butyric acid concentration of 1251.2 μg/mL. Conversely, cellulase modification reorganized intermolecular hydrogen-bond networks, which accelerated the early-stage structural breakdown of E-SDF (generating 1630.78 μg/mL of total SCFAs at 24 h) and was associated with higher relative abundances of the [ Eubacterium ] ventriosum group alongside Bifidobacterium . Furthermore, the modified SDF groups exhibited lower relative abundances of Escherichia–Shigella under fermentation conditions characterized by greater SCFA accumulation and lower pH. These findings elucidate how specific processing methods modulate the dynamic degradation trajectories of SDFs in the gut, providing a mechanistic basis for developing tailored functional ingredients from traditional edible fungi. In vitro dynamic digestion properties and fecal fermentation of Dictyophora indusiata polysaccharide: Structural characterization and gut microbiota

Lin Wang,Zhikun Zeng,Yaqing Lin,Baodong Zheng,Yi Zhang,Lei Pan

The in vitro dynamic digestive model more realistically simulates the human digestive system compared to static digestive model. In this study, the dynamic in vitro stomach-intestine digestive system and fecal fermentation was used to investigate the dynamic digestion properties and fermentation properties of Dictyophora indusiata polysaccharide. The results showed that there were no significant changes in molecular weight, functional groups and surface morphology after the in vitro dynamic simulated digestion, indicating that D. indusiata polysaccharide maintained a relatively stable structure during the dynamic in vitro salivary-gastrointestinal digestion. In addition, D. indusiata polysaccharide improved the abundance of beneficial bacteria, including Blautia , Coprobacter and Fusicatenibacter . It is remarkable that D. indusiata polysaccharide significantly increased the level of acetic acid and propionic acid. In conclusion, these results suggested that D. indusiata polysaccharide was a potential source of prebiotics, which provides a basis for the development of D. indusiata polysaccharide in the food or medical field.

Steam-exploded Dictyophora indusiata polysaccharide regulated gut microbiota based on dynamic in vitro stomach-intestine digestion system

Zhikun Zeng,Hangyu Huang,Lin Wang,Yaqing Lin,Binbin Wang,Baodong Zheng,Yi Zhang,Lei Pan

Dictyophora indusiata is a nutritious edible mushroom, which polysaccharide exhibited a variety of biological activities. In this study, the digestion properties of steam-exploded Dictyophora indusiata polysaccharide (DIPS) were investigated by dynamic in vitro stomach-intestine digestive system. Here, the molecular weight of DIPS significantly was decreased and the microscopic surface structure appeared to be loose after 90 min of simulated stomach digestion. Interestingly, the molecular weight of DIPS was restored during the small intestine digestion phase, which was not significantly different compare with DIPS before stomach digestion, the functional groups, thermal stability and antioxidant properties of DIPS remained stable after digestion. It is remarkable that DIPS after dynamic in vitro stomach-intestine digestion regulated the composition of the gut microbiota, including Blautia , Megamonas , Parabacteroides and Phascolarctobacterium . In addition, DIPS promoted the level of acetic acid, propionic acid and butyric acid during the in vitro fermention, which were 2.42, 4.36 and 9.17 folds higher than controls, respectively. This study provides important significance for exploring the effects of edible mushroom polysaccharide on gut health.

Effects of different fractions of polysaccharides from Dictyophora indusiata on high-fat diet-induced metabolic syndrome in mice

Hong Yao,Jingrui Yang,Song Li,Steve W. Cui,Huizi Tan,Shaoping Nie

Dictyophora indusiata is a common edible mushroom with great potential in the field of medicine against metabolic disorders, inflammation, and immunodeficiency. Our previous studies have shown that different fractions of the polysaccharide from Dictyophora indusiata (DIP) have various structural characteristics and morphology. However, the impact of the structural features on the protective effects of DIP against metabolic syndrome remains unclear. In this study, three distinct polysaccharide fractions have been extracted from Dictyophora indusiata and a high-fat diet-induced metabolic syndrome (MetS) was constructed in mice. The effects of these fractions on a range of MetS-associated endpoints, including abnormal blood glucose, lipid profiles, body fat content, liver function, intestinal microbiota and their metabolites were investigated. Through correlation analysis, the potential link between the monosaccharide composition of the polysaccharides and their biological activities was determined. The study aimed to explore the potential mechanisms and ameliorative effects of these polysaccharide fractions on MetS, thereby providing statistical evidence for understanding the relationship between monosaccharides composition of Dictyophora indusiata polysaccharides and their potential utility in treating metabolic disorders.

A dual-network hydrogel patch fabricated by alginate/sulfobetaine methacrylate enriched with Dictyophora indusiata β-glucans promotes diabetic wound repair

Zeguo Feng,Shan Lu,Ying He,Yanting Sun,Lei Ma,Hui Guo,Yiting Kang,Rui Xu,Yifei Song,Lei Huang,Liguo Qin,Jianbao Zhang,Yali Zhang

This study developed a dual-network hydrogel patch loaded with Dictyophora indusiata β-glucans to enhance diabetic wound healing. The hydrogel combines a flexible primary network formed by polymerized sulfobetaine methacrylate with a rigid secondary alginate network crosslinked via metal ions. The resulting material demonstrates favorable mechanical properties for wound care, achieving 600 % elongation at break, 3.12 MPa compressive strength, and 1.5 kPa tissue adhesion strength. These characteristics meet with the physical requirements necessary for effective diabetic wound management. Furthermore, the β-glucans derived from Dictyophora indusiata , which serve as the main bioactive component, endowed the hydrogel patch with significant antioxidant and anti-inflammatory properties. Cellular experiments have demonstrated that the hydrogel patch significantly reduces reactive oxygen species levels in cells and inhibits inflammatory responses. In animal wound model, diabetic wound treated with a hydrogel patch achieved a closure rate of 98.26 % by the second week. Additionally, histological analyses revealed that the hydrogel patch significantly facilitates angiogenesis, collagen deposition, and re-epithelialization in diabetic wound. Consequently, the hydrogel patch based on β-glucans from Dictyophora indusiata appears to be an effective agent for promoting wound healing, thereby offering a novel therapeutic strategy for the repair of diabetic wound.

Quality, improvement of soluble dietary fiber from Dictyophora indusiata by-products by steam explosion and cellulase modification: Structural and functional analysis

Mengfan Lin,Changrong Wang,Wenfei Wu,Qingsong Miao,Zebin Guo

Steam explosion (SE) and cellulase treatment are potentially effective processing methods for Dictyophora indusiata by-products, for use in high-value applications. The treatment conditions were optimized by response surface methodology, increasing the soluble dietary fiber (SDF) yield by 1.52 and 1.16 times after the SE and cellulase treatments, respectively. The both treatments did not affect the functional groups and crystal types of the polysachharides, but both reduced the crystallinity. The SDF had a porous microstructure, which would increase the specific surface area and facilitates the adsorption of water and glucose, thereby improving its functional properties. SE and cellulase treatment significantly improved the hydration capacity of SDF; the glucose adsorption capacity increased by 1.15 and 1.07 times, respectively. Overall, the modified SDF showed different degrees of advantages in terms of yield, physicochemical and functionality. This study demonstrated that SE and cellulase are effective modification methods for SDF made from D. indusiata by-products.
