BACKGROUND Dinotefuran (DNF) is a neonicotinoid insecticide with strong systemic, poisoning and gastric toxicity, which is no cross-resistance with the existing pesticides, but its toxicity to environmental organisms such as bees and earthworms is high. The development of environmentally responsive pesticide controlled-release agents using nanomaterials to process the agents by physical or chemical methods can improve the utilization of the pesticide and better utilize the active molecules of the pesticide. RESULTS To enhance the performance of dinotefuran, UiO-67 was prepared, characterized, and then combined with dinotefuran to form dinotefuran-UiO-67 (DNF@UiO-67) nanocomposites. Compared with dinotefuran, DNF@UiO-67 nanocomposites significantly increased the wettability and photolysis resistance of dinotefuran. Dinotefuran was loaded on UiO-67 without influencing its original activity and was more efficient in the long-term control of pea aphid compared to dinotefuran. The treatment of pea seeds with different concentrations of DNF@UiO-67 did not significantly affect the germination and growth of pea seeds and ensured the normal growth of plants. DNF@UiO-67 reduced the acute toxicity to earthworms and bees. These improvements imply that the DNF@UiO-67 nanocomposites could be used as a potential formulation to improve insecticidal activity and enhance its safety for environmental organisms of dinotefuran. CONCLUSION This study not only develops DNF@UiO-67 nanocomposites to improve the performance of dinotefuran while maintaining its original insecticidal, but also provides a new idea for the application of dinotefuran in agriculture. Our findings will provide more possibilities for the application of DNF@UiO-67 nanocomposites as an effective and environmentally friendly formulation in agriculture. © 2025 Society of Chemical Industry.

As a typical neonicotinoid insecticide, dinotefuran (DIN) has immunotoxicity, but its immunotoxicity effects on aquatic organisms and its molecular mechanisms are not known. In this study, zebrafish embryos were used as model to reveal its immunotoxicity and its mechanism from the perspective of inflammatory response. Zebrafish larvae were exposed to DIN at environmentally relevant concentrations (0, 2, 200, and 2000 mg/L) for 120 h, followed by comprehensive analyses of immune cell populations, immune marker activities, oxidative stress levels, and gene expression profiles. The results showed that the number of neutrophils and macrophages, two types of innate immune cells, was significantly reduced, and the activities of the immune markers, lysozyme (LYS), immunoglobulin M (IgM), and complement protein C3, were significantly inhibited. The level of oxidative stress in zebrafish larvae was significantly elevated and antioxidant enzyme activities were markedly inhibited in a dose-dependent manner after exposure to DIN. In this case, pro-inflammatory cytokines such as TNF-α, IL-6 and IL-1β were also abnormally expressed, and the expression levels of the genes related to the NF-κB pathway and the JNK-STAT pathway, tlr4a, myd88, nf-κb p65, jak1, jak2 , and stat3 , were elevated. The expression levels of genes related to the antioxidant signaling pathway Nrf2-keap1 signaling pathway were suppressed. Taken together, our results suggest that DIN exposure causes damage to the immune system of zebrafish embryos-larvae, generating oxidative stress and inflammatory responses that lead to immunotoxicity.

The Asian citrus psyllid, Diaphorina citri , is a serious pest worldwide that transmits the huanglongbing pathogen Candidatus Liberibacter spp. Previous studies have shown that chemosensory proteins (CSPs) are associated with insecticide resistance in various insect species. Dinotefuran is one of the most poisonous neonicotinoids to D. citri . However, the specific functional role of CSPs in modulating dinotefuran tolerance remains poorly characterized. In this study, we found that sublethal exposure to dinotefuran (LC 50 concentration) dramatically elevated DcCSP4 expression in the surviving D. citri adults. In addition, RNA interference-mediated silencing of DcCSP4 substantially enhanced the susceptibility of D. citri adults to dinotefuran toxicity. The recombinant Dc CSP4 protein was effectively expressed in the Escherichia coli BL21 (DE3) strain and purified with Ni-NTA Resin. Fluorescence competitive binding assays demonstrated a medium-affinity interaction between Dc CSP4 and dinotefuran with a dissociation constant (Ki) of 24.30 μM. Molecular docking results revealed that Dc CSP4 binds to dinotefuran mostly via van der Waals (vdW) forces. Overexpressing Dc CSP4 recombinant protein enhanced E. coli cell viability after 100 mg/L dinotefuran exposure. These findings imply that the overexpression of DcCSP4 in D. citri adults constitutes an adaptive tolerance/resistance mechanism to dinotefuran, which provides novel mechanistic insights into insect xenobiotic detoxification processes.

BACKGROUND Bemisia tabaci (Gennadius) is a serious agricultural pest worldwide. Neonicotinoids are the most important new class of synthetic insecticides used in the management of B. tabaci . However, B. tabaci populations have developed resistance to various active ingredients in neonicotinoids following long-term and widespread application. RESULTS Dinotefuran exhibited high toxicity against most B. tabaci field populations. One population (Din-R) with a high level of resistance to dinotefuran (255.6-fold) was first identified in the field. The Din-R population exhibited medium to high levels of resistance to all the tested neonicotinoid insecticides and a high level of resistance to spinetoram. Genetic inheritance analysis revealed that resistance to dinotefuran was incompletely recessive and polygenic. The synergist piperonyl butoxide significantly increased the toxicity of dinotefuran to Din-R. P450 activity in the Din-R population was 2.19-fold higher than in the susceptible population. RNA-sequencing analysis showed that 12 P450 genes were significantly upregulated in the Din-R population, of which CYP6DW5 , CYP6JM1 and CYP306A1 were found to exhibit more than 3.00-fold higher expression in Din-R when using a reverse transcription quantitative real-time polymerase chain reaction. Expression of eight P450 genes was obviously induced by dinotefuran, and CYP6DW5 showed the highest expression level. After knockdown of CYP6DW5 in Din-R, the toxicity of dinotefuran increased significantly. CONCLUSION P450 had a crucial role in dinotefuran resistance in B. tabaci , and CYP6DW5 was involved in the resistance. These results provide important information for the management of resistance in B. tabaci and improve our understanding of the resistance mechanism of dinotefuran. © 2022 Society of Chemical Industry.

Root exudates are released by plant roots and are important carrier substances for material exchange and information transmission among plants and the rhizosphere. In the present study, the effect of dinotefuran on root exudates of Chinese cabbage ( Brassica rapa var. chinensis ) was investigated. The physiological activities revealed that dinotefuran uptake caused oxidative stress in vegetable tissues even at low dinotefuran exposure levels. The metabolic profile of plant root exudates acquired by LC-QTOF/MS was clearly changed by dinotefuran, where the numbers of both up- and down-regulated MS peaks increased with increasing dinotefuran concentration. Under dinotefuran stress, some osmotic adjustment substances (proline and betaine) and defence-related metabolites (spermidine, phenylalanine and some phenolic acids) were significantly upregulated, which may help plants adapt to adverse environmental conditions. Specifically, the contents of some phenylalanine-derived secondary metabolites increased with increasing dinotefuran concentration, which may increase the external detoxification ability of plants. Moreover, respiration metabolism was significantly affected, where some intermediates in the TCA cycle (succinic acid and malic acid) were upregulated with low-level dinotefuran exposure; however, anaerobic respiration products (lactic acid and 3-phenyllactic acid) were accumulated at high exposure levels. In addition, the release of glucosinolates was significantly inhibited in both dinotefuran treatment groups.

Dinotefuran (DNT) belongs to the third-generation neonicotinoid pesticides, which are among the most common residuals in a variety of food commodities. To guarantee accurate quantification and traceability of results in food samples, certified reference materials (CRMs) are the indispensable benchmark. In this work, a DNT CRM was characterized and its purity was assessed by two independent methods, including mass balance (MB) and quantitative nuclear magnetic resonance spectroscopy (qNMR). The mass fraction of moisture was 0.33 mg/g, the inorganic impurity was 0.01 mg/g, and no detectable organic solvent was detected. Benzoic acid was chosen as the internal standard for qNMR. Its mass fraction was 997.9 mg/g and 992.9 mg/g by MB and qNMR, respectively. Eventually, the DNT CRM was assigned a mass fraction of 995 mg/g, with expanded uncertainty of 5 mg/g (k= 2). This CRM can be used to prepare calibrant solutions and is applicable to national routine monitoring of DNT residuals in agro-products and food. Keywords:dinotefuran;certified reference material;mass balance;quantitative nuclear magnetic resonance spectroscopy

This study investigated the efficacy of three spray adjuvants (Coerce, Wetcit, and Silwet408) in enhancing dinotefuran permeation in rice leaves. Different concentrations of these adjuvants were tested using an external standard method. The rice leaf surface was analyzed by using the van Oss–Chaudhury–Good method to establish a link between surface free energy (SFE) and dinotefuran permeation. All of the adjuvants effectively increased dinotefuran permeation in rice leaves, with the highest permeation of 8.496 mg/kg achieved using Wetcit at 1000 mg/L. The SFE of the rice leaf surface was determined to be 29.28 mJ/m2. A notable correlation was observed between the energy difference (the SFE of the pesticide liquid versus the SFE of the rice leaf surface) and permeation. Various fitting methods, including linear, exponential, logarithmic, polynomial, and power methods, were applied. Polynomial fitting demonstrated the highest coefficient of determination (R2> 0.9000). The relationship between the permeation (y) and energy difference (Δγ) conformed to the polynomial equationy= aΔγ2+ bΔγ + c, where a, b, and c are constants. This model provides a predictive tool for the optimal dosage of spray adjuvants according to target plant characteristics, enhancing the understanding of the interaction between pests and pesticides.

Firstly, the surfactant cetyltrimethylammonium bromide (CTAB) and 1-hexadecyl-3-methylimidazolium chloride monohydrate (HMCM) were intercalated into attapulgite (AP) to obtain CTAB-AP and HMCM-AP complexes, respectively, and then pesticide dinotefuran (DIN) was introduced into the complexes to prepare DIN/CTAB-AP (HMCM-AP) hybrids, finally the hybrids was added into N-isopropylacrylamide (NIPAM) solution to obtain DIN/CTAB-AP(HMCM-AP)/PNIPAM hydrogels through the free radical polymerization. Their structures were systematically characterized by XRD, FTIR, TGA , SEM and BET. The effect of hybrids content, crosslinker concentration and pH on the hydrogel was investigated and analyzed. The existence of DIN and surfactant molecules in the hydrogels was confirmed. Fabrication mechanism of DIN hydrogels was put forward. Release behaviors of the hydrogels were investigated in different release mediums and temperatures. For DIN/CTAB-AP/PNIPAM, DIN release amount increased from 79.5 % at 20°C to 97.7 % at 30°C. For DIN/CTAB-AP/PNIPAM and DIN/HMCM-AP/PNIPAM ( p = 4, 9.3) hydrogels, the release rates are much slower in pH= 5.0 than pH= 9.0. The release behaviors were well fitted by pseudo second-order and Ritger-Peppas models. Bioassays revealed extended efficacy against aphids (73 % mortality on third day for the third inoculation), suggesting a long duration and control effect of the hydrogels on the pest compared to the DIN water dispersible granules. This study provided a new method to improve the utilization efficiency of pesticide, and the hydrogel can be potentially used as a pesticide delivery system.

It is well known that the widespread residue of the neonicotinoids dinotefuran in aquatic environment has adverse effects on non-target organisms. Therefore, the efficient removal of dinotefuran in water environment is an important measure to control its safety risk. In the present study, layered phosphorus-doped carbon nitride (HPCN 0.5 ) was prepared via a simple hydrothermal method and applied for photocatalytic degradation of dinotefuran. With the addition of phosphorous acid, a tube-like structure was observed in HPCN 0.5 and planar sheets were stacked inside the tubes. Compared to bulk g-C 3 N 4 , the degradation efficiency of dinotefuran was 6 times higher. According to the reaction mechanism and product identification analysis, the main photodegradation pathways correspond to demethylation and hydroxylation. The result of the acute toxicity test revealed the toxicity of dinotefuran to earthworm was greatly reduced after photodegradation by HPCN 0.5 . In addition, HPCN 0.5 was capable of simultaneously removing three other commonly used neonicotinoids. This work proved that HPCN 0.5 has great potential to be applied in alleviating negative effects of neonicotinoids on the non-target organisms and environment.

The monitoring and the rapid quantification of pesticides and their residues are becoming increasingly important in the field of food safety. Herein, the polycaprolactone/polypyrrole/β-cyclodextrin (PCL/PPy/β-CD) flexible sensor was developed for the electrochemical determination of new neonicotinoid insecticide Dinotefuran (DNF). The morphology, structure, and hydrophilicity of PCL/PPy/β-CD sensor probes were characterized by SEM, FTIR spectroscopy and static contact angle test. Under optimum conditions, the fabricated PCL/PPy/β-CD sensor exhibited excellent electrochemical sensing performance for DNF with a low detection limit of 0.05 μM in the linear concentration range from 0.2 μM to 50 μM and high sensitivity 14.07 μA·μM −1 ·cm −2 , which attributed to the two-stage porous structure, good electron transfer rate and the adsorption effect. The PCL/PPy/β-CD sensor also showed reproducibility (RSD = 4.76%), stability, and high selectivity towards DNF. In addition, a real samples investigation in rice with recoveries of 96.67 % ∼ 103.65 % implied the good application potential of PCL/PPy/β-CD in DNF monitoring.

The brown planthopper (BPH), Nilaparvata lugens (Stål), is a major insect pest of rice in Asia and is mainly controlled with insecticides. Dinotefuran has been commercialized for control of N. lugens in China since 2013; however, the characteristics and mechanisms of resistance to dinotefuran are still unclear. Here, we conducted a long-term (from 2013 to 2021) and large geographical scale (11 provinces) resistance monitoring program for dinotefuran in the BPH in China. Before 2020, field populations of N. lugens have developed moderate resistance to dinotefuran, while since 2020, most field populations of N. lugens have developed high resistance. To develop a better resistance management strategy, a dinotefuran-resistant (Din-R) strain of N. lugens was selected in the laboratory to investigate the inheritance, possible mechanisms of resistance and potential fitness costs. This Din-R strain developed resistance to imidacloprid, nitenpyram and thiamethoxam. The dinotefuran resistance was controlled by multiple genes. RNA interference (RNAi)-mediated suppression of NlCYP6ER1 , NlCYP6CS1 and NlCYP314A1 expression increased the susceptibility of N. lugens to dinotefuran. Transgenic lines of Drosophila melanogaster expressing NlCYP6ER1 or NlCYP314A1 were less susceptible to all four neonicotinoids. Furthermore, the Din-R strain had a relative fitness of 0.64 with a substantially decreased survival rate, emergence rate and fecundity. Collectively, our results showed that dinotefuran resistance was mediated by multiple CYP genes. The high expression of these genes was related to the cross-resistance of N. lugens to other neonicotinoids. This study provided valuable information for facilitating the development of neonicotinoids resistance management strategies.

Dinotefuran (DIN) contaminates groundwater and is toxic to bees, so designing a novel pesticide loading system for dinotefuran is of great significance. In this paper, dinotefuran was loaded into TpBDMe COF with a mesoporous hollow structure, and the TpBDMe COF loaded with dinotefuran exhibited initial burst and subsequent sustained release behavior. The results showed that the product effectively suppresses wheat aphid development, with virtually no infestation occurring for 7–14 days, whereas other commercially available formulation had lost their usefulness. Our TpBDMe COF carrier effectively immobilizes dinotefuran, reducing pesticide leaching in soil by 70 % and enhancing the application efficiency of neonicotinoid insecticides. The LD 50 of this product for acute exposure of honey bees doubled the LD50 of the original furosemide at 24 h and was equally effective at 48 h, reducing the toxicity to bees and promoting ecological safety. The results of this work show that this nanocarrier system is highly stable, loadable, insecticidal active, and environmentally friendly, showing great potential in the development of new nanocarriers for use.
