Fabrication of hyaluronic acid-inulin coated Enterococcus faecium for colon-targeted delivery to fight Fusobacterium nucleatum

Pengrong Zhu,Lingtong Meng,Yue Shu,Yuqiao Xu,Weiming Liu,Yong Bi,Jian Xu,Lijuan Meng,Yanan Li

The abundance of Fusobacterium nucleatum ( F. nucleatum ) is highly associated with the development and poor prognosis of colorectal cancer (CRC), which is regarded as a promising target for CRC. However, until now, the novel strategy to clear F. nucleatum in the colon and CRC has not been well proposed. Herein, a probiotic strain Enterococcus faecium ( E. faecium , EF47) is verified to secrete various organic acids and bacteriocins to exert superior antimicrobial activity towards F. nucleatum . However, the oral delivery of EF47 is affected by the complex digestive tract environment, so we design the hyaluronic acid-inulin (HA-IN) coated EF47 for colon-targeted delivery to fight F. nucleatum . IN can protect EF47 from the harsh gastrointestinal tract environment and is degraded specifically in the colon, acting as prebiotics to further promote the proliferation of EF47. The exposed HA can also enhance the targeting effect to the tumor area via the interaction with the CD44 receptor on the tumor cells, which is confirmed to increase the adhesive ability in tumor tissues and inhibit the growth of F. nucleatum . Therefore, this colon-targeted delivery system provides a novel platform to realize high-activity and adhesive delivery of probiotics to assist the therapeutic efficiency of CRC.

Application of ion-exchange resin as solid acid for buffer-free production of γ-aminobutyric acid using Enterococcus faecium cells

Hui Zhu,Faizan A. Sadiq,Yun Li,ShengYuan Yang,Fei Zhou

In this study, ion-exchange resin was applied as solid acid catalyst for the γ-aminobutyric acid (GABA) production using a strain of Enterococcus faecium (GZ2) by whole-cell bioconversion. It was observed that the accumulated GABA resulted in the inhibition of the bioconversion activity. To improve the productivity, a cation-exchange resin-based in situ product removal (ISPR) approach was developed. Three cation-exchange resins were tested to select a suitable adsorbent used in the bioconversion. The strong acidic cation-exchange resin IRP-69 exhibited the highest GABA production and remained helpful in reducing the free GABA concentration in the reaction solution. Compared to the bioconversion in sodium acetate buffer, the GABA production, with the addition of resin, was significantly improved by 2.12-fold. Furthermore, a fed-batch bioconversion strategy with the feeding of L-monosodium glutamate (L-MSG) was tested. The maximum production of GABA, 406.1 mM, was achieved after 12 h reaction with an initial substrate concentration of 200 mM, and a triple feeding of 100 mM L-MSG afterwards at 4 h, 6 h and 8 h. The result shows that the operation of bioconversion reaction in a feeding substrate batch mode is an effective approach to improve the efficiency and productivity of GABA.

Metabolome responses of Enterococcus faecium to acid shock and nitrite stress

Zhong Yu,Dongchen Shi,Wencong Liu,Yabing Meng,Fangang Meng

Enterococcus faecium is gaining increasing interest due to its virulence and tolerance to a range of stresses (e.g., acid shock and nitrite stress in human stomach). The chemical taxonomy and basic structural features of cellular metabolite can provide us a deeper understanding of bacterial tolerance at molecular level. Here, we used hierarchical classification and molecular composition analysis to investigate the metabolome responses of E. faecium to acid shock and nitrite stress. Our results showed that considerable high biodegradable compounds (e.g., dipeptides) were produced by E. faecium under acid shock, while nitrite stress induced the accumulations of some low biodegradable compounds (e.g., organoheterocyclic compounds and benzenoids). Complete genome analysis and metabolic pathway profiling suggested that E. faecium produced high biodegradable metabolites responsible for the proton-translocation and biofilm formation, which increase its tolerance to acid shock. Yet, the presence of low biodegradable metabolites due to the nitrite exposure could disturb the bacterial productions of surface proteins, and thus inhibiting biofilm formation. Our approach uncovered the hidden interactions between intracellular metabolites and exogenous stress, and will improve the understanding of host-microbe interactions.

Gut Enterococcus faecium derived leucic acid mediates the antiobesity effect of dietary fiber

Baifei Hu,Qianru Gao,Junmin Hui,Mengqiong Wang,Haiyan Gao,Kai Li,Junping Zheng,Zhe Dai,Songlin Liu,Hongtao Liu

Obesity is a globally prevalent metabolic disorder closely associated with gut microbiota dysbiosis. However, the precise roles of gut microbiota and microbial metabolism in this disease remain unclear. Through fecal metagenomic data mining and clinical cohort validation, we identified a gut bacterial strain with anti-obesity potential, Enterococcus faecium ( E. faecium ). It inhibited adipocyte enlargement, dyslipidemia, and hepatic steatosis in obese mice by metabolizing leucine to produce leucic acid (LEA). Colonization with E. faecium or exogenous LEA supplementation improved obesity-associated metabolic phenotypes. Mechanistically, E. faecium metabolized leucine into LEA via putative branched-chain amino acids metabolic enzymes. The bacterium-derived LEA regulated mitochondrial oxidative phosphorylation and thus alleviated obesity. Furthermore, using a high-content screening system and an in vitro fermentation model, we found that the dietary fiber glucomannan exerted anti-obesity effects by promoting the growth of E. faecium . This study reveals the critical role of microbial metabolism and LEA production in ameliorating the onset and progression of obesity, providing a theoretical basis for dietary fiber intervention strategies targeting the gut microbiota. Graphical This study identified Enterococcus faecium as a gut bacterium with anti-obesity activity, which alleviated adipocyte hypertrophy, dyslipidemia, and hepatic steatosis by converting leucine into leucic acid (LEA) to regulate mitochondrial oxidative phosphorylation. In addition, dietary glucomannan effectively enriched this bacterium, providing a theoretical basis for developing precision dietary fiber-based anti-obesity strategies targeting the gut microbiota. Highlights Metagenomic and clinical data verified E. faecium as an anti-obesity gut bacterium. E. faecium alleviated obesity via leucine conversion to leucic acid (LEA). LEA modulated mitochondrial oxidative phosphorylation against obesity. Glucomannan inhibited obesity by enriching E. faecium for microbiota intervention.

Effect of Coreopsis tinctoria microcapsules on tyramine production by Enterococcus faecium in smoked horsemeat sausage

Yazhuo Li,Honghong Yu,Zonggui Tang,Jingyun Wang,Tao Zeng,Shiling Lu

This work investigated the inhibitory effects of Coreopsis tinctorius essential oil (CTEO) and Coreopsis tinctorius microcapsules (CTM) on tyramine accumulation by Enterococcus faecium in pure culture and during the ripening of smoked horsemeat sausage, in order to provide a novel natural additive for the control of tyramine in smoked horsemeat sausage. To evaluate the expression of related-genes in the tyrosine decarboxylation (TDC) pathway, we used reverse transcription-quantitative polymerase chain reaction (RT-qPCR) to determine the expression of tyrosine decarboxylase (tyrDC) and tyrosine/tyramine antiporter (tyrP) genes. Furthermore, the pH, moisture content, microorganisms and tyramine accumulation of sausage were assessed. The results revealed that CTEO and CTM markedly ( P < 0.05) decreased tyramine accumulation by inhibiting E. faecium growth and the expression of tyrDC and tyrP genes. Furthermore, tyramine accumulation in sausage was evidently inhibited by CTEO and CTM, and the inhibitory effect of CTM was more significant ( P <0.05). Hence, CTM addition is a desirable approach to inhibit tyramine accumulation, and enhance the safety of smoked horsemeat sausages.

Immobilization and enzymatic properties of glutamate decarboxylase from Enterococcus faecium by affinity adsorption on regenerated chitin

Yang Sheng-Yuan,Liu Shu-Min,Wu Yan-Yan,Lin Qian,Liang Gui-Lian,Liu Jiao-Fen,Liang Zi-Zhan,Liang Jia-Rong

Glutamate decarboxylase (GAD, EC 4.1.1.15) is an important enzyme in gamma-aminobutyric acid biosynthesis and DL -glutamic acid resolution. In this study, the Enterococcus faecium -derived GAD was successfully immobilized by regenerated chitin (RC) via specific adsorption of cellulose-binding domain (CBD). The optimal binding buffer was 20 mmol/L phosphate buffer saline (pH 8.0), and the RC binding capacity was 1.77 ± 0.11 mg cbd-gad /g rc under this condition. The ratio of wet RC and crude enzyme solution used for immobilization was recommended to 3:50 (g/mL). To evaluate the effect of RC immobilization on GAD, properties of the immobilize GAD (RC-CBD-GAD) were investigated. Results indicated RC-CBD-GAD was relatively stable at pH 4.4–5.6 and temperature − 20–40 °C, and the optimal reaction pH value and temperature were pH 4.8 and 50 °C, respectively. When it was reacted with 5 mmol/L of follow chemical reagents respectively, the activity of RC-CBD-GAD was hardly affected by EDTA, KCl, and NaCl, and significantly inactivated by AgNO 3 , MnSO 4 , MgSO 4 , CuSO 4 , ZnSO 4 , FeCl 2 , FeCl 3 , AlCl 3 , CaCl 2, and Pb(CH 3 COO) 2 . The apparent K m and V max were 28.35 mmol/L and 147.06 μmol/(g RC-CBD-GAD ·min), respectively. The optimum time for a batch of catalytic reaction without exogenous pH control was 2 h. Under this reaction time, RC-CBD-GAD had a good reusability with a half-life of 23 cycles, indicating that it was very attractive for GABA industry. As a novel, efficient, and green CBD binding carrier, RC provides an alternative way to protein immobilization.

Characterization of novel exopolysaccharide of Enterococcus faecium WEFA23 from infant and demonstration of its in vitro biological properties

Kaiying Jia,Xueying Tao,Zhengqi Liu,Hui Zhan,Weijun He,Zhihong Zhang,Zheling Zeng,Hua Wei

In this study exopolysaccharide (EPS) of Enterococcus faecium WEFA23 from healthy infant's feces was yielded as high as 130 mg/L by fermentation. By purification the EPS was further fractioned into A23-1, A23-2, A23-3 and A23-4 on HiTrap Q HP and Superdex G-200 column. As the major purified fractions, A23-2 and A23-4 were analyzed for the preliminary structures and investigated for the biological properties in vitro. The molecular weight of A23-2 and A23-4 was 2.50 × 10 4  Da and 3.23 × 10 4  Da, respectively. A23-2 was composed of mannose, glucose and galactose with a ratio of 1.38:1.00:1.42, while A23-4 consisted of only mannose. The antioxidant ability of A23-2 was significantly higher than that of A23-4, as proved by scavenging test of DPPH radical, hydroxyl radical and superoxide radical. Both A23-2 and A23-4 exhibited strong inhibition against the adhesion of L . monocytogenes CMCC54007 on HT-29 cells no matter for the styles of competition, displacement and exclusion. Notably, for exclusion they had a maximal inhibition rate of 89.85 ± 0.46% and 90.81 ± 2.29%, respectively, at 200 μg/mL.

Effect of host-derived Enterococcus faecium L6 on growth performance, intestinal health and antibacterial activities of juvenile grass carp

Xiaonan Sun,Hongzhou Xu,Yanzhen Song,Jingfei Long,Chenyang Yan,Xiaoyu Qi,Lixin Wang,Yuanjiang Jin,Haixia Liu

The various diseases outbreaks and infections caused by pathogens have brought great losses to the aquaculture industry of grass carp. Probiotics are one of the important ways in water quality management and inhibition of bacterial diseases. Thus, more and more attention is being paid to potential indigenous probiotics. The present study isolated a probiotic strain of Enterococcus faecium ( E. faecium ) L6 from the intestinal content of healthy grass carp, and evaluated the probiotic effect in vitro. Subsequently, a feeding trial of 56 days was performed to evaluate the effect of E. faecium (1 × 10 8  CFU/g) on growth performance in juvenile grass carp and infection resistance to Aeromonas hydrophila ( A. hydrophila ). Our data showed that diet supplementation with E. faecium L6 for 56 days significantly increased final body weight (FBW), weight gain rate (WGR), specific growth rate (SGR) of juvenile grass carp and decreased feed conversion ratio (FCR) compared to that of basal diet group without probiotic supplemented ( P  < 0.05). The expression of inflammatory factors TNF-α , IL-6 and IL-1β in the liver, spleen and kidney showed an early increase and later decrease, and ultimately restored a nearly normal level on 56 days of feeding, and tight junction proteins genes zonula occludens-2 ( ZO-2 ), zonula occludens-3 ( ZO-3 ), occludin and claudin were significantly up-regulated from 21 days after feeding E. faecium L6 supplementary diet ( P  < 0.05). Following a challenge by A. hydrophila infection, there was an increase of 26.67 % in the survival rate of diet supplementation of E. faecium L6 group, and the bacterial load in the liver, spleen and kidney was significantly reduced after E. faecium L6 feeding ( P  < 0.01). High-throughput sequencing of 16S rRNA gene revealed that E. faecium L6 feeding increased the relative abundance of the phylum Fusobacteriota and Bacteroidota, and Cetobacterium and Bacteroides at genus level in the intestinal microbiota composition, as well as improved the functional pathways related to metabolism of purine, biotin, cysteine, methionine, d -Glutamine and D-glutamate, and biosynthesis of aminoacyl-tRNA and lipopolysaccharide. Analysis of microbial community-level phenotypes revealed that aerobic, contains mobile elements, facultatively anaerobic, biofilm-forming, Gram-negative, potential-pathogenicity and stress-tolerance phenotypes were remarkably decreased in E. faecium L6 supplemented diet group, while anaerobic and Gram-positive phenotypes were highly increased. Collectively, E. faecium strain L6 as a prospective probiotic can be used in the healthy breeding of grass carp production.

Effects of Live Combined Bacillus subtilis and Enterococcus faecium on Gut Microbiota Composition in C57BL/6 Mice and in Humans

Pi Xionge,Teng Weilin,Fei Dibo,Zhao Gang,Liu Wei

Probiotics, prebiotics, and synbiotics can alleviate metabolic syndrome by altering the composition of the gut microbiota. Live combined Enterococcus faecium and Bacillus subtilis has been indicated to promote growth and reduce inflammation in animal models. However, the modulatory effects of live combined B. subtilis R-179 and E. faecium R-026 (LCBE) on human microbiota remain unclear. The current study examined the growth of these two strains in the presence of various oligosaccharides and assessed the effects of this probiotic mixture on human and murine gut microbiota in vitro and in vivo. Oligosaccharides improved the growth of E. faecium R-026 and B. subtilis R-179 as well as increased their production of short-chain fatty acids. E. faecium R-026 or B. subtilis R-179 co-incubated with Bifidobacterium and Clostridium significantly increased the number of the anaerobic bacteria Bifidobacterium longum and Clostridium butyricum by in vitro fermentation. Moreover, LCBE significantly reduced plasma cholesterol levels in mouse models of hyperlipidemia. LCBE combined with galacto-oligosaccharides led to a significant decrease in the Firmicutes/Bacteroidetes ratio and a significant increase in the relative abundance of Akkermansia and Bifidobacteria after treating mice with LCBE (0.23 g/day) for eight weeks. Furthermore, in vitro fermentation also showed that both the single strains and the two-strain mixture modulated human gut microbiota, resulting in increased Lactobacillus and Bifidobacteria, and decreased Escherichia-Shigella. Overall, these results suggest that LCBE can improve host health by reducing the level of cholesterol in mouse models by modifying the composition of the gut microbiota.

Peach gum polysaccharide protects intestinal barrier function, reduces inflammation and oxidative stress, and alleviates pulmonary inflammation induced by Enterococcus faecium E745

Shugang Li,Guopeng Li,Dongjie Liu,Mingxin Li,Huanhuan Liu,Wenxiu Zhu,Ke Wang,Xiaomeng Ren

Peach gum polysaccharide (PGP) is a natural compound known for its anti-inflammatory and antioxidant properties. This study aimed to investigate the potential protective effect of PGP against Enterococcus faecium E745-induced pneumonia and elucidate its underlying mechanism. It demonstrated that PGP exert antioxidant effects by activating the Nrf2 signaling pathway, as evidenced by the normalization of MDA, SOD and GSH levels. Notably, PGP inhibited the NF-κB signaling pathway, reduced IL-6 and TNF-α levels in serum, colon and lung, and enhanced the expression of tight junction proteins. In addition, PGP affected the synthesis of short-chain fatty acids (SCFAs), especially increased the content of propionate and butyrate. These results suggest that PGP alleviates pneumonia by restoring the integrity of the intestinal barrier, increasing the production of SCFAs, reducing the inflammatory response and oxidative stress. In conclusion, our study provides valuable insights into the potential application of PGP in the treatment of inflammatory diseases.

Postbiotic cell-free supernatant of Enterococcus faecium L6 enhances growth performance, intestinal homeostasis and disease resistance in largemouth bass (Micropterus salmoides)

Xiaonan Sun,Tian Yang,Hongzhou Xu,Di Wang,Yuanjiang Jin,Huanjie Li,Yuxin Shen,Xiaoqiang Liu,Haixia Liu

E. faecium L6 harbors biosynthetic gene clusters encoding diverse bioactive metabolites. • Dietary CFS supplementation improved growth performance by enhancing antioxidant capacity, innate immunity, and intestinal barrier integrity in largemouth bass. • CFS reshaped the intestinal microenvironment by enriching beneficial bacteria and functional metabolites. • CFS significantly increased survival following A. veronii challenge through modulation of inflammatory responses. Probiotics are widely used to promote fish health. However, the application of live probiotics is limited by poor stability and potential safety concerns. Postbiotics have drawn increasing attention because they may provide beneficial effects without the limitations associated with live bacteria. This study investigated the effects of probiotic (live Enterococcus faecium L6, LEF) and postbiotics (cell-free supernatant, CFS, and heat-killed E. faecium L6, HKEF) on growth performance, intestinal health, and disease resistance in Micropterus salmoides ( M. salmoides ). A 56-day feeding trial was conducted, followed by an Aeromonas veronii ( A. veronii ) challenge test. Growth parameters, intestinal morphology, antioxidant and non-specific immune enzyme activities, inflammatory cytokine expression, intestinal microbiota composition, and metabolomic profiles were analyzed. Both LEF and CFS significantly improved final body weight, weight gain rate, and specific growth rate while decreasing the feed conversion ratio, whereas HKEF had no significant effect. LEF and CFS improved intestinal barrier integrity by increasing villus height and the villus height:crypt depth ratio, and by up-regulating ZO -1, Claudin -1, and Occludin . Antioxidant (SOD, CAT) and non-specific immune (ACP, AKP, LZM) activities were also significantly elevated in the LEF and CFS groups. After A. veronii challenge, survival rates were significantly higher in the LEF (55.0%) and CFS (60.0%) groups compared with the PBS control (30.0%). The improved resistance was associated with reduced intestinal pathogen loads and a balanced inflammatory response, characterized by down-regulated TNF-α and IL-1β and up-regulated IL-10 . Furthermore, CFS reshaped the intestinal microbiota and increased functional metabolites. In conclusion, E. faecium L6-derived CFS conferred growth-promoting and disease-protective effects comparable to, or stronger than, its live probiotic counterpart, supporting its potential as a stable and effective postbiotic strategy for enhancing fish health.
