In this study, iron-based metal-organic framework@platinum nanoparticles (FeMOF@PtNPs) with outstanding peroxidase-like catalytic activity was synthesized using a simple one-pot hydrothermal method. Leveraging this catalytic platform, a cost-effective colorimetric aptasensor was developed for the detection of aflatoxin B1 (AFB1) residues in feed. The sensor exhibited a linear response to AFB1 concentrations ranging from 10 to 1000 nM, with a detection limit as low as 2.18 nM under optimized conditions. Furthermore, the sensor demonstrated excellent selectivity and stability. The reliability of the proposed aptasensor was validated by testing real feed samples, with results closely matching those obtained using the standard ELISA method. This suggests that the constructed colorimetric aptasensor holds great potential for practical applications. In addition to AFB1 detection, the proposed sensing platform can be readily adapted for various analytes by replacing the corresponding aptamer. This offers broad applicability in food safety, environmental monitoring, and medical diagnostics.

Matrix metalloproteinase-9 (MMP-9) has been implicated in various tumor cell invasions and metastases. In light of the limitations of traditional methods for MMP-9 detection, we have constructed a novel biosensor depending on cucurbit[8]uril (CB[8]) -mediated host-guest interactions and a sacrificial iron metal-organic framework (FeMOF). Herein, MMP9-specific peptides modified on the gold bare electrode are bonded to the [email protected] @peptide complex through CB[8] addition. The connection between MMP9-specific peptides and signal peptides via CB[8] provides stability as well as enables the immobilization of FeMOF on the electrode surface . When Fe 3+ from the FeMOF interacts with electrochemical buffer K 4 Fe(CN) 6 , Prussian blue will be generated on the gold electrode surface, and a significantly enlarged current response can be detected. However, in the presence of MMP-9, their peptide substrates are specifically cleaved at the site between serine (S) and Leucine (L), which causes an abrupt decrease in the electrochemical signal. The change of signal can reflect MMP-9 concentration. This sensor can reach an ultrahigh sensitivity with a wide detection range of 0.5 pg⋅mL −1 to 500 ng⋅mL −1 and a low detection limit of 1.30 pg⋅mL −1 . Importantly, this sensor is very simple, relying solely on self-sacrificial label of FeMOF, rather than complex functional materials. Additionally, it has been well used in serum samples, showing attractive potential for practical applications.

Postoperative immunosuppression, which is closely associated with the oxidative-stress-driven expansion of regulatory T cells (Tregs), increases susceptibility to severe postoperative outcomes, including infectious complications, multiple organ dysfunction syndrome (MODS), and tumor progression. Here, we report a zirconium-based metal–organic framework (MOF) cascade nanozyme, designated “Pt@PCN-224-Fe”, coated with poly(ethylene glycol) (Pt@FeMOF–PEG) that simultaneously mimics superoxide dismutase (SOD) and catalase (CAT) to neutralize excessive reactive oxygen species (ROS). Mechanistically, scavenging ROS reactivates the PI3K–AKT–mTOR signaling axis, suppresses Foxp3 expression, and thereby inhibits both the differentiation of naı̈ve CD4+ T cells into Tregs under Treg-polarizing conditions and their TGF-β1 secretion in vitro. In a murine model of surgical trauma, the single intraperitoneal administration of Pt@FeMOF–PEG reversed immunosuppression within 72 h, evident as a reduced frequency of Tregs, expanded Th17 cells, normalized CD4+/CD8+ ratio, and suppressed M2 macrophage polarization (all p < 0.05). Concurrently, proinflammatory cytokines (IL-1β, IL-6, TNF-α, IFN-γ) increased, whereas anti-inflammatory cytokines (IL-4, IL-10) decreased, indicating the systemic reinvigoration of the innate and adaptive immunity. The nanozyme showed excellent biocompatibility, with negligible hemolysis (<5%), no histological organ damage, and minimal systemic toxicity. In this work, we have developed a dual-enzyme-mimicking nanozyme (Pt@FeMOF–PEG) that precisely targets the ROS–PI3K–Treg axis in a single-dose strategy to reverse postoperative immune dysfunction, thus addressing a long-unmet clinical requirement for surgical recovery.
