Protective Effects of Hippophae rhamnoides L. Phenylpropanoids on Doxorubicin-Induced Cardiotoxicity in Zebrafish

Gang Li,Ming Chu,Yingying Tong,Yuexin Liang,Shenghui Wang,Chengjun Ma,Zhenhua Wang,Wenna Zhou

Hippophae rhamnoidesL. is a deciduous shrub that contains many unique bioactive substances. This sea buckthorn possesses anticancer, antioxidant, anti-inflammatory, and cardiovascular protective properties. Herein, the effects of phenylpropyl compounds extracted fromH. rhamnoidesL. on doxorubicin (Dox)-induced cardiotoxicity were evaluated in zebrafish. Cardiac injury in zebrafish was induced using 35 μM Dox for 96 h, and 30 μM phenylpropanoid compounds were used as the protective treatment. The cardioprotective effects and mechanisms of the four phenylpropanoids were investigated using microscopy, behavioral analysis, acridine orange staining, western blotting, flow cytometry, and real-time quantitative polymerase chain reaction. The extracted phenylpropanoids could significantly relieve Dox-induced cardiac injury in zebrafish and inhibit cardiomyocyte apoptosis. The mechanisms of action were mainly related to the stability of mitochondrial biogenesis and function maintained by phenylpropanoids in zebrafish. To our knowledge, this is the first report on the protective effect of sea buckthorn against myocardial injury in zebrafish. Our findings provide support for the further research and development of sea buckthorn and its components. Keywords:sea buckthorn;zebrafish;doxorubicin;cardiotoxicity;mitochondria

Structural characterization and intestinal protection activity of polysaccharides from Sea buckthorn (Hippophae rhamnoides L.) berries

Chen Shen,Teng Wang,Feng Guo,Kunlai Sun,Bin Wang,Jie Wang,Zefeng Zhang,Xing Zhang,Yuqin Zhao,Yin Chen

The sea buckthorn ( Hippophae rhamnoides L.) berries are rich in various bioactive components and widely used as fruit and traditional medicine. In this study, a novel heteropolysaccharide fraction (SP0.1-1) was isolated from Sea buckthorn berries. SP0.1-1 is composed of mannose , glucose, galactose , and arabinose in the molar ratio of 1:2.3:1.9:11.2 with a core structure containing 1,4-linked-α- d -Glc p , 1,4,6-linked-α- d -Glc p and 1,4-linked-α- d -Man p residues as the backbone. And the side-chains comprised of 1,3,5-linked-α- l -Ara f , 1,5-linked-α- l -Ara f , terminal α-Ara f and 1,4-linked-β- d -Gal p . Furthermore, a diet supplemented with SP0.1-1 extended the mean lifespan, enhanced antioxidant enzyme (superoxide dismutase, SOD; glutathione peroxidase , GSH-Px; and catalase , CAT) activities, and decreased the malondialdehyde (MDA) level and hydrogen peroxide (H 2 O 2 )-induced mortality rate in fruit flies ( Drosophila melanogaster ). To summarize, the study's findings will provide evidence for the development of sea buckthorn polysaccharide products.

Succinylation-driven structural modification of Hippophae rhamnoides L. protein enhances interfacial properties and emulsion performance

Cheng Xuehuan,Ge Shuya,Liu Wanlu,Zhang Junhang,Wang Bing,Blanchard Chris,Strappe Padraig,Zhou Zhongkai,Zhao Guohua

Sea buckthorn protein (SBP) is an underutilized plant protein with limited aqueous dispersibility and interfacial functionality. In this study, SBP was modified by succinylation, and the resulting changes in structural, physicochemical, and functional properties were systematically characterized. Succinylation markedly improved the dispersibility of SBP, as evidenced by an increase in solubility from 25.25% to 68.99%, a shift in zeta potential from − 33.82 to − 51.62 mV, and reduced particle aggregation. Structural analysis indicated that succinylation induced moderate conformational rearrangement, including decreased α-helix content, increased random coil content, and changes in the tertiary microenvironment. These changes enhanced the emulsifying performance of SBP, with the emulsifying activity index increasing from 25.02 to 35.71 m²/g. β-carotene-loaded emulsions stabilized by succinylated SBP showed smaller and more uniform droplets, with droplet size decreasing from 2085.47 to 700.53 nm, together with improved encapsulation efficiency and storage stability. During in vitro digestion, free fatty acid release increased from 20.92% to 26.02%, while β-carotene bioaccessibility increased from 10.17% to 27.67%. These results suggest that succinylation improves the functional performance of SBP through a charge-conformation-interface mechanism, thereby enhancing its potential as a plant-based emulsifier for β-carotene-loaded emulsion systems. Phenolic compounds, antioxidant activity and sensory evaluation of sea buckthorn (Hippophae rhamnoides L.) leaf tea

Qian He,Kailin Yang,Xinyan Wu,Chunhong Zhang,Chunnian He,Peigen Xiao

Sea buckthorn leaf tea, an emerging potential functional beverage product, has not yet had appropriate product standards and corresponding quality evaluation methods, and its poor taste directly affects the acceptance of the population, thus limiting its market consumption potential. In this study, two major packaging forms of sea buckthorn leaf tea available in the Chinese market were selected. The contents of total phenolics, total flavonoids, and 10 phenolic compounds, as well as the in vitro antioxidant capacity and sensory characteristics of sea buckthorn leaf tea were analyzed. Results showed that the quality of sea buckthorn leaf tea in the Chinese market varied widely. The total phenolic content, total flavonoid content, antioxidant activity, and consumer acceptance of bagged sea buckthorn leaf tea were higher than those of bulk sea buckthorn leaf tea. Multifactorial statistical analysis showed that the taste astringency of sea buckthorn leaf tea was closely related to ellagic acid and isorhamnetin-3-O-neohesperidin. Furthermore, isorhamnetin-3-O-neohesperidin had a greater effect on the antioxidant activity of sea buckthorn leaf tea. Therefore, ellagic acid and isorhamnetin-3-O-neohesperidin can be used as potential quality markers for sea buckthorn leaf tea. This work provides a reference for taste improvement and quality control of sea buckthorn leaf tea.

Phytic acid enhances the oral absorption of isorhamnetin, quercetin, and kaempferol in total flavones of Hippophae rhamnoides L.

Yan Xie,Huilin Luo,Jingze Duan,Chao Hong,Ping Ma,Guowen Li,Tong Zhang,Tao Wu,Guang Ji

Aim Total flavones of Hippophae rhamnoides L. (TFH) have a clinical use in the treatment of cardiac disease. The pharmacological effects of TFH are attributed to its major flavonoid components, isorhamnetin , kaempferol , and quercetin . However, poor oral bioavailability of these flavonoids limits the clinical applications of TFH. This study explores phytic acid (IP 6 ) enhancement of the oral absorption in rats of isorhamnetin, kaempferol, and quercetin in TFH. Methods In vitro Caco-2 cell experiments and in vivo pharmacokinetic studies were performed to investigate the effects of IP 6 . The aqueous solubility and lipophilicity of isorhamnetin, quercetin, and kaempferol were determined with and without IP 6 , and mucosal epithelial damage resulting from IP 6 addition was evaluated by MTT assays and morphology observations. Results The P app of isorhamnetin, kaempferol, and quercetin was improved 2.03-, 1.69-, and 2.11-fold in the presence of 333   μg/mL of IP 6 , respectively. Water solubility was increased 22.75-, 15.15-, and 12.86-fold for isorhamnetin, kaempferol, and quercetin, respectively, in the presence of 20   mg/mL IP 6 . The lipophilicity of the three flavonoids was slightly decreased, but their hydrophilicity was increased after the addition of IP 6 in the water phase as the logP values of isorhamnetin, kaempferol, and quercetin decreased from 2.38   ±   0.12 to 1.64   ±   0.02, from 2.57   ±   0.20 to 2.01   ±   0.04, and from 2.39   ±   0.12 to 1.15   ±   0.01, respectively. The absorption enhancement ratios were 3.21 for isorhamnetin, 2.98 for kaempferol, and 1.64 for quercetin with co-administration of IP 6 (200   mg/kg) in rats. In addition, IP 6 (200   mg/kg, oral) caused neither significant irritation to the rat intestines nor cytotoxicity (400   μg/mL) in Caco-2 cells. Conclusions The oral bioavailability of isorhamnetin, kaempferol, and quercetin in TFH was enhanced by the co-administration of IP 6 . The main mechanisms are related to their enhanced aqueous solubility and permeability in the presence of IP 6 . In summary, IP 6 is a potential absorption enhancer for pharmaceutical formulations that is both effective and safe.

Characterizing and Decoding the Key Odor-Active Compounds in Fresh, Pasteurized, and High Pressure Processing Sea Buckthorn (Hippophae rhamnoides L.) Juice

Xiao Jia,Jingnan Ren,Zhifeng Zhang,Nawei Zhang,Qi An,Mingzhe Li,Siyi Pan,Gang Fan,Jinchu Yang,Yingjie Feng,Zongcan Yang,Yongming Xu

ABSTRACT The non-thermal and thermal effects on aroma of sea buckthorn juice have rarely been investigated. In this study, 57 odor compounds were identified in fresh sea buckthorn juice (FSBJ), high pressure processing sea buckthorn juice (HSBJ), and pasteurized sea buckthorn juice (PSBJ), including 29 esters, 8 aldehydes, 1 ketone, 5 alcohols, 5 acids, 6 terpenoids, and 3 others. Ethyl 2-methylbutanoate, ethyl 3-methylbutanoate, ethyl hexanoate, and ethyl 2-hydroxy-3-methylbutanoate with flavor dilution factors ranging from 729 to 59049 contributed to the fruity odors of FSBJ and HSBJ. Besides, the formation of off-odor compounds including hexanal, nonanal, furfural, 3-methylbutanoic acid, and dimethyl disulfide with odor activity values ≥ 1, imparts fatty, roasted, sweaty, and cooked odor in PSBJ. The variations of vitamin C and reducing sugar are significantly associated with changes in odor-active compounds during pasteurized processing. These findings provide new insights that high pressure processing minimizes the adverse effects of pasteurization.

Impact of Drying Methods on Phenolic Components and Antioxidant Activity of Sea Buckthorn (Hippophae rhamnoides L.) Berries from Different Varieties in China

Yue Li,Pei Li,Kailin Yang,Qian He,Yue Wang,Yuhua Sun,Chunnian He,Peigen Xiao

Sea buckthorn berries are rich in bioactive compounds and can be used for medicine and food. The variety and drying method used have an important influence on quality. In this study, different sea buckthorn varieties from China were selected and dried with four common drying methods. The total phenolic content (TPC), total flavonoids content (TFC), contents of 12 phenolic compounds and antioxidant capacity in vitro were analyzed. The results showed that the TPC, TFC and antioxidant activity of two wild sea buckthorn berries were higher than those of three cultivated berries, and for the same varieties, measured chemical contents and antioxidant activity of the freeze-dried fruit were significantly higher than those obtained with three conventional drying methods. In addition, forty-one compounds in sea buckthorn berry were identified by UPLC-PDA-Q/TOF-MS, most of which were isorhamnetin derivatives. Multivariate statistical analysis revealed narcissin and isorhamnetin-3-O-glucoside varied significantly in sea buckthorn berries of different varieties and with different drying methods; they were potential quality markers. Strong correlations were found between TPC, gallic acid and antioxidant capacity (p< 0.05). The results revealed how components and antioxidant activity varied in different sea buckthorn, which provides a valuable reference for quality control and further development and utilization of sea buckthorn. Keywords:sea buckthorn;drying method;variety;phenolic compounds;UPLC-Q-TOF-MS;antioxidant activity

Fabrication of Nanosuspensions to Improve the Oral Bioavailability of Total Flavones from Hippophae rhamnoides L. and their Comparison with an Inclusion Complex

Tian Rui,Wang Hui,Xiao Yi,Hu Pengyue,Du Ruofei,Shi Xiufeng,Wang Zhengtao,Xie Yan

The aim of this work was to increase the solubility and oral bioavailability of isorhamnetin, kaempferol, and quercetin in the total flavones of Hippophae rhamnoides L. (TFH) by preparing their nanosuspensions (NSs) and an inclusion complex. Based on the particle size and zeta potential, P407, Soluplus, SDS, PEG-6000, and HP-β-CD were selected as stabilizers. TFH NSs and a TFH/HP-β-CD inclusion complex were prepared, and their morphology, crystallinity, molecular interactions, and cytotoxicity were investigated. Furthermore, the saturation solubility, dissolution, and pharmacokinetics of the three flavonoids in the TFH, TFH NSs, and TFH/HP-β-CD inclusion complex were compared. The five obtained TFH NSs were physically stable, and their particle sizes were all below 200 nm. The solubility and dissolution of the three active components were obviously enhanced by the formation of the TFH NSs and TFH/HP-β-CD inclusion complex. Correspondingly, the oral bioavailability of isorhamnetin, kaempferol, and quercetin increased up to 4.11-, 3.85-, and 6.73-fold, respectively, in the TFH NSs and 2.89-, 3.71-, and 9.51-fold, respectively, in the TFH/HP-β-CD inclusion complexes compared to those in the raw TFH. In brief, both NSs and inclusion complexes can improve the oral bioavailability of the three flavonoids in TFH. Taking the drug loading and the stable ratio of the multiple components into consideration, the NSs is a more promising strategy than the inclusion complex for increasing the oral bioavailability of multiple water-insoluble components in herbal extracts. The effect of Hippophae rhamnoides L and Alkekengi officinarum Moench fruits extracts on the Changes of acrylamide content in roasted chicken breast during storage

Linze Liu,Yawen Deng,Hangqi Ren,Ying Ren,Taizheng Tang,Eerdun bayaer,Shaobo Li,Tungalag Dong,Xueyan Yun

Plant extracts with antioxidant activity are frequently employed to inhibit acrylamide (AM) production, but there has been limited focus on their inhibitory effects on AM during the extended storage of foods. In this study, impregnation solutions were prepared using Hippophae rhamnoides L (Hp) and Alkekengi officinarum Moench (Ak) fruits extracts to investigate their protective effects on roasted chicken breasts (RCB) during processing and storage (vacuum packed at 4°C and stored at 25°C for 90 days). In a 90-day storage experiment with RCB, these extracts also slowed the increase of AM in RCB. However, these extracts exhibit inadequate protective effects on RCB during long-term storage. It is needed to develop new methods for monitoring and inhibiting AM formation in RCB.

Structural characterization of a distinct neutral polysaccharide fraction (SP0–1) from sea buckthorn (Hippophae rhamnoides L.) and its antioxidant-related intestinal protection in Drosophila melanogaster

Su Jinle,Ge Lingrui,Niu Chunyu,Chen Yan,Zhang Leifang,Chen Yin

SP0–1, a neutral polysaccharide fraction isolated from sea buckthorn berries ( Hippophae rhamnoides L.), was purified and structurally characterized, and its antioxidant-related intestinal protective effects were evaluated in Drosophila melanogaster . HPGPC analysis showed a single symmetrical peak and indicated a relatively homogeneous fraction with a moderate molecular weight (Mw, 5.59 × 10 4  Da; ≈55.9 kDa). PMP-HPLC analysis revealed that SP0–1 was mainly composed of galactose, xylose, and arabinose (Gal/Xyl/Ara-rich; Gal:Xyl:Ara = 1:0.38:0.93). Methylation analysis combined with 1D/2D NMR spectroscopy supported the presence of major →4)- and →4,6)-linked Galp motifs, →5)- and →3,5)-linked Araf motifs, and terminal Xylp residues. The consecutive Gal–Gal arrangement, the residue adjacent to Gal O-6, the overall connection of the Ara/Xyl-containing region to the Gal-containing region, and the O-3 substituent of residue D remain unresolved. Functionally, SP0–1 exhibited concentration-dependent radical-scavenging activities against DPPH, ABTS• + , and hydroxyl radicals in vitro. In a Drosophila oxidative-stress model, dietary SP0–1 supplementation significantly improved survival and climbing performance, enhanced endogenous antioxidant enzyme activities, and reduced malondialdehyde levels. Moreover, SP0–1 markedly alleviated H 2 O 2 -induced intestinal oxidative injury, as evidenced by decreased reactive oxygen species accumulation and reduced intestinal epithelial cell death. These findings suggest that the structural characteristics of SP0–1, including its proposed Gal-rich linkage features and Ara/Xyl-containing residue motifs, may be associated with the observed antioxidant-related intestinal protection. Collectively, this study provides a structural and functional basis for further investigation of SP0–1 as a potential bioactive polysaccharide fraction.
