Photothermal material was one of the hot topics in nanomedicine in recent years, which had aroused widespread attention of scholars. In this paper, a kind of core-shell drug nanocarriers with good photothermal properties were prepared, and its photothermal therapeutic effect on tumor cells in vitro was studied. First, fluorescein isothiocyanate (FITC) was encapsulated in silica nanoparticles to form silica fluorescent (F-SiO 2 ) nanoparticles. Then mesoporous dopamine (MPDA) layer were built on the F-SiO 2 nanoparticles, the core-shell nanoparticles (F-SiO 2 @MPDA) with mesoporous shell structure were obtained. Finally, Au nanoparticles (AuNPs) were wrapped on the surface of the layer of MPDA shell by the citrate reduction method, and the F-SiO 2 @MPDA-AuNPs composite nanoparticles were formed. Then, the fluorescence property, adsorption performance, sustained release property, photothermal property and cytotoxicity of the F-SiO 2 @MPDA-AuNPs nanoparticles were researched. It was found that F-SiO 2 @MPDA-AuNPs nanoparticles had good fluorescence performance, high adsorption capacity (DOX, 111.275 mg/g) and pH responsive release performance. When pH = 5.2, the DOX release rate was 39.5% within 7 h under the action of near-infrared light (NIR). In addition, it also had a high photothermal conversion rate (36.95%), and the DOX loaded F-SiO 2 @MPDA-AuNPs nanoparticles had obvious inhibitory effect on tumor cells in vitro. As a kind of mesoporous drug nanocarriers with photothermal properties, it has potential application value.

The magnetically core-shell Fe 3 O 4 @mesoporous polydopamine (Fe 3 O 4 @MPDA) nanospheres were prepared by a facile and efficient self-assembly approach, the key mechanism for generating the mesoporous structure of PDA is the π-π stacking interactions between dopamine molecules and 1,3,5-trimethylbenzene (TMB) molecules. Because of the mesoporous structure of the surface, the Fe 3 O 4 @MPDA particles exhibited a large specific surface area of 17.27 m 2 /g and total pore volumes of 0.057 cm 3 /g. Besides, it maintained good magnetic properties . The adsorption properties and mechanism of organic dyes methylene blue (MB) and rhodamine B (RhB) on Fe 3 O 4 @MPDA nanoparticles were studied. And a model anticancer drug of doxorubicin hydrochloride (DOX) was used as a target drug to research the delivery and sustained-release properties of the Fe 3 O 4 @MPDA particles. The average loading capacities toward DOX could reach 295.2 mg/g, and the release amounts were about 41% within 60 h. Meanwhile, the in vitro cytotoxicity of the DOX loaded Fe 3 O 4 @MPDA nanoparticles against hela cells and osteoblast cells were also being researched. The DOX-loaded Fe 3 O 4 @MPDA nanoparticles showed greater toxicity than Fe 3 O 4 nanoparticles and Fe 3 O 4 @MPDA nanoparticles, even than free DOX. Compared with osteoblasts, the inhibitory effect on hella cells was more obvious. The above properties are of great significance for its application in the field of biological medicine, especially the target drug carriers.

In recent years, there has been a notable focus on the development and design of multifunctional drug delivery systems tailored for photothermal, photodynamic, and chemotherapy triple-modality cancer treatment. The paper presents the utilization of gold nanorods (AuNRs) for photothermal effect, zinc oxide (ZnO) for photodynamic effect, mesoporous polydopamine (mPDA) loaded with curcumin (Cur), and 1-tetradecanol (TD) as a temperature-sensitive gating switch to achieve integration of functional organic/inorganic nanomaterials. The core-shell structural nanocarriers (AuNRs@ZnO@mPDA-Cur/TD) achieved a drug loading rate of 178 mg/g. It is important to note that the pH-responsive release properties of Cur have been observed due to the acid response of mPDA. Furthermore, the photothermal effect of AuNRs and mPDA facilitates the thermal-induced melting of TD and subsequent release of Cur upon near-infrared (NIR) irradiation, thereby achieving an effective integration of photothermal therapy and chemotherapy. Simultaneously, under NIR excitation, AuNRs inject thermal electrons into the conduction band of ZnO, leading to the generation of singlet oxygen species that confer remarkable photodynamic healing capabilities to the as-prepared carriers. The combined treatment involving photothermal, photodynamic, and chemotherapy demonstrated synergistic effects and superior efficacy compared to individual treatments. The core-shell nanodrug carrier AuNRs@ZnO@mPDA developed in this study exhibits pH-, NIR-, and temperature-responsive drug release properties, highlighting the synergistic impact of photothermal, photodynamic, and chemotherapy on tumor therapy. This research provides a novel concept and preparation strategy for a multifunctional smart drug delivery platform with integrated therapeutic efficacy.

Endoscopic submucosal dissection (ESD) is one of the most important strategies for early gastrointestinal tumor treatment. However, the ideal healing of artificial ulcers produced after ESD procedure are still hard to achieve. In a gastric ulcer environment, the inherent peristole, acid condition and excessive oxidative stress seriously affect the therapeutic outcome of traditional medication. Curcumin (Cur) is a promising anti-inflammatory drug for gastric ulcer treatment, but its long-term use is still challenging. A polymer-assisted drug delivery system in gastrointestinal disease treatment is a new direction but is still at an initial stage. Herein, we report a hybrid hydrogel consisting of catechol motif-modified methacrylated gelatin (GelMAc) and methacrylated poly (vinyl alcohol) (PVAMA). GelMAc brought significant adhesive property and biocompatibility. PVAMA could effectively provide stability and mechanical strength. Moreover, mesoporous polydopamine nanoparticles were prepared as the drug carrier to load Cur via π-π stacking and hydrogen bonding interactions, and they were encapsulated in the hybrid GelMAc/PVAMA hydrogel. The prepared sample was proved to have a unique stability on gastric tissue even bearing acid (pH 2) and oxidative stress (H 2 O 2 ) conditions. The slowly released Cur from the hydrogel was capable of inducing M2 polarization of macrophages. In an in vivo gastric ulcer model, the prepared sample remarkably improved gastric ulcer healing by curbing the pro-inflammatory response and alleviating oxidative stress. An accelerated reepithelization and angiogenesis were also observed. Thus, this study provides a promising hydrogel for the treatment of ESD-induced ulcers, which may attract more attentions to explore the deeper combination between biomaterials and gastrointestinal research.

Hepatic ischemia-reperfusion injury (IRI) is a common pathological process during hepatectomy and liver transplantation and the two primary reasons for hepatic IRI are reactive oxygen species (ROS)-mediated oxidative stress and excessive inflammatory responses. Herein, a novel antioxidant nanodrug (A-MPDA@Fe3O4@PVP) is prepared by employing L-arginine-doped mesoporous polydopamine (A-MPDA) nanoparticles as the carrier for deposition of ultra-small ferric oxide (Fe3O4) nanoparticles and further surface modification with polyvinylpyrrolidone (PVP). A-MPDA@Fe3O4@PVP not only effectively reduces the aggregation of ultra-small Fe3O4, but also simultaneously replicates the catalytic activity of catalase (CAT) and superoxide dismutase (SOD). A-MPDA@Fe3O4@PVP with good antioxidant activity can rapidly remove various toxic reactive oxygen species (ROS) and effectively regulate macrophage polarization in vitro. In the treatment of hepatic IRI, A-MPDA@Fe3O4@PVP effectively alleviates ROS-induced oxidative stress, reduces the expression of inflammatory factors, and prevents apoptosis of hepatocytes through immune regulation. A-MPDA@Fe3O4@PVP can further protect liver tissue by activating the PPARγ/NF-κB pathway. This multiplex antioxidant enzyme therapy can provide new references for the treatment of IRI in organ transplantation and other ROS-related injuries such as fibrosis, cirrhosis, and bacterial and hepatic viral infection.

Novel highly soluble phenylethynyl-endcapped oligoimides with different calculated molecular weights were prepared by using mellophanic dianhydride and bis(4-amino-2-trifluoromethylphenyl)ether in the presence of 4-phenylethynyl phthalic anhydride as the reactive endcapping agent. The effect of molecular weights of the aromatic oligoimides on their solubility and processability as well as the thermal and mechanical properties of the thermal-cured polyimides was fully investigated. This kind of phenylethynyl-terminated imides exhibited excellent comprehensive properties. Most importantly, a big progress has been made in the solubility of the oligoimides in this paper. Most of the oligoimides showed excellent solubility (more than 33 wt%) even in acetone , which was reported for the first time. And oligoimides with appropriate molecular weights also showed very good processability with minimum melt viscosities no higher than 120 Pa·s by the rheological study. After thermally curing at 370°C for 1 hour, the cured flexible films showed very high glass transition temperatures up to 392°C by dynamic mechanical analysis, and the temperature of 5% weight loss was higher than 500°C in both air and N 2 atmosphere by thermogravimetric analysis. The cured films also exhibited very good toughness with elongation up to 9%.

Periodontitis, a bacteria-induced inflammatory and osteolytic disease, remains a major cause of tooth loss and is commonly treated through mechanical debridement combined with antibiotics. However, conventional antibiotic therapy often causes drug resistance and fails to achieve simultaneous antibacterial and anti-inflammatory effects. In this work, a dual-functional punicalagin–mesoporous polydopamine (PUN–MPDA) hydrogel nanoplatform was developed based on an injectable hyaluronic acid–phenylboronic acid–poly(vinyl alcohol) (HA–PBA–PVA, HPP) matrix for combined photothermal antibacterial and anti-inflammatory therapy. The nanoplatform exhibited excellent photothermal performance and broad-spectrum antibacterial activity. Under 808 nm near-infrared irradiation, antibacterial efficiencies against Porphyromonas gingivalis and Fusobacterium nucleatum reached 99.1 % and 99.2 %, respectively, with efficient eradication of mixed-species biofilm. The hydrogel also showed good biocompatibility and anti-inflammatory capacity by downregulating pro-inflammatory genes ( Il-6 , Tnf-α ) and upregulating anti-inflammatory genes ( Il-10 , Tgf-β ), thereby enhancing alkaline phosphatase activity and promoting mineralized nodule formation. In a rat periodontitis model, the nanoplatform effectively alleviated inflammation, reduced collagen degradation, and inhibited alveolar bone resorption. Collectively, this PUN–MPDA hydrogel nanoplatform offers a promising light-controllable therapeutic strategy integrating antibacterial, anti-inflammatory, and osteogenic effects for advanced periodontitis therapy.

The combination of chemotherapy and photothermal therapy can provide better tumor treatment effect than monotherapy , while the combination of imaging technology can achieve diagnostic effect during the treatment. In this paper, biocompatible mesoporous polydopamine (MPDA) were combined with the near-infrared fluorescent dye IR780 under alkaline condition to form an integrated drug delivery platform for tumor diagnosis and photothermal co-therapy. The particles size and mesoporous pore size of the nanospheres were adjusted by changing the preparation conditions of MPDA, so the platform had strong permeability and large drug loading capacity in tumor cells as drug carriers. The loading and release results of doxorubicin hydrochloride (DOX) proved that the carrier had a high drug loading and sustained release rate, as well as a faster dependent release under acidic and lighting conditions. Photothermal experiments showed that the carrier had high photothermal conversion efficiency, which was very critical in its application in photothermal therapy and fluorescence imaging of tumors. The vitro cytotoxicity test evaluated the survival rates of LX-2 cells and HepG2 cells after drug loading. The results showed that the MPDA@IR780-DOX + NIR carrier had a significant inhibitory effect on HepG2 cells (12.57%), but had little effect on LX-2 cells, which was beneficial for its application in integration of biological diagnosis and treatment.

Hepatic fibrosis, primarily driven by the sustained activation of hepatic stellate cells (HSCs), continues to pose a significant global health challenge due to the scarcity of effective therapeutic interventions. Existing single-target approaches are inadequate in addressing the complex multicellular regulatory network within the fibrotic niche, where injured hepatocytes, Kupffer cells (KCs), and capillarized liver sinusoidal endothelial cells (LSECs) collectively perpetuate HSC activation through paracrine signaling mechanisms. To address this challenge, a novel nanoplatform has been developed, comprising a co -delivery system of quercetin (Que) and simvastatin (Sim) within a mesoporous polydopamine (MPDA)-based matrix, designed to achieve the synchronized modulation of multicellular interactions. The MPDA carrier possesses intrinsic liver-targeting and reactive oxygen species (ROS)-scavenging properties, facilitating preferential accumulation in fibrotic lesions and the mitigation of oxidative stress originating from hepatocytes and KCs. In the acidic fibrotic environment, Que and Sim are sustained within the system due to the acid-responsive properties of MPDA. The released Que reprograms pro-inflammatory M1 macrophages into anti-inflammatory M2 phenotypes, while Sim reverses LSEC capillarization, collectively disrupting the microenvironmental factors driving HSC activation. Moreover, all components—MPDA, Que, and Sim—can directly inhibit HSC activation through complementary pathways. This multi-target strategy resulted in a more significant reduction in collagen deposition compared to monotherapies in preclinical models, alongside a notable restoration of liver function. By integrating microenvironmental reprogramming with the interception of multicellular networks, our study establishes a paradigm-shifting nanotherapeutic approach for reversing fibrosis and underscores the potential of engineered multidrug coordination in treating complex stromal diseases.

Catalytic nanomedicine with high oxygen-generation efficiency may find applications in alleviating tumor hypoxia and improving the efficiency of photodynamic therapy (PDT). In this study, a catalytic nanosystem (Ce6-Rh@MPDA) was developed using mesoporous polydopamine nanoparticles (MPDA) to encapsulate catalase-like rhodium nanoparticles (Rh NPs) and photosensitizer chlorine6 (Ce6) for photoacoustic/fluorescence dual imaging-guided tumor therapy. The Rh NPs can catalyze the production of O2 from tumor-enriched H2O2in situ, in which the mesoporous structure of MPDA plays an important role via improving the catalytic efficiency of Rh NPs. Moreover, the hyperthermia generated by both MPDA and Rh NPs under laser irradiation accelerates the O2 generation to promote the PDT. The Ce6-Rh@MPDA nanoparticles described herein represent a multifunctional metal-based catalytic nanomedicine which not only alleviates tumor hypoxia but also enables a synergistic antitumor treatment using PTT and PDT.

Oral nano-drug delivery systems offering combination therapy have garnered significant interest in colon cancer treatment due to their precision in targeting tumors and minimizing peripheral tissue exposure. However, challenges such as the complex gastrointestinal environment and effective retention of nanoparticles in the colon have impeded further advancement. We developed a novel oral drug delivery system designed for localized treatment of colon cancer via chemotherapy and photothermal therapy (PTT). This system utilized mesoporous polydopamine (MPDA) as a photothermal carrier for doxorubicin hydrochloride (DOX), with surface modification using folic acid (FA) to enhance systemic tumor targeting. Additionally, to ensure gastrointestinal retention and precise colon localization, the nanoparticles were coated with an enteric-soluble material, ES100, resulting in the formulation MPDA-FA-DOX/ES100. This formulation exhibited high photothermal conversion efficiency, robust photothermal stability, and responsive drug release under near-infrared (NIR) laser stimulation. FA modification significantly enhanced the cellular uptake of nanoparticles by CT26 cells, promoting greater cytotoxic effects through combined chemotherapy and PTT. In vivo , MPDA-FA-DOX/ES100 demonstrated superior accumulation in colon tumor tissues and substantial photothermal effects, and notably, the CT/PTT group demonstrated significant tumor growth inhibition along with excellent biocompatibility. Collectively, these findings highlight the clinical potential of MPDA-FA-DOX/ES100 as an effective platform for localized and synergistic CT/PTT of colon cancer.

The discovery of gold nanoparticles as precious-metal photothermal materials has aroused great interest in morphology regulation, especially in the emerging anisotropic morphology of gold nanoparticles. In this study, the influence of the local surface plasmon resonance effect on the photothermal conversion ability of gold nanoparticles with different morphologies is investigated. This study is based on gold nanobipyramids—a nanostructure with excellent photothermal conversion efficiency. It compares two strategies for preparing gold nanobipyramids and explores how morphology and growth mechanisms are regulated by varying the gold seed concentration and the ascorbic acid-to‑silver nitrate molar ratio. Subsequently, the biocompatibility of the nanoparticles was enhanced by capping mesoporous polydopamine onto the periphery of the gold nanobipyramids, and their chemotherapeutic capability was endowed via loading the drug doxorubicin (DOX) into the mesoporous structure; the drug loading capacity and encapsulation efficiency were 45.1% and 82.0%, respectively. In order to improve the therapeutic efficacy of the nanocarriers in the tumor microenvironment, the pH-responsive property of polyethylenimine (PEI) was incorporated into AuNBP@mPDA to confer targeted drug release capability. Surprisingly, the PEI layer effectively seals the mesoporous structure and prevents premature drug release, thereby enabling precise drug delivery. After a series of structural characterizations and performance evaluations, the final AuNBP@mPDA-DOX@PEI nanocarrier was confirmed to exhibit excellent thermochemotherapeutic performance (reduction of Hela S3 cells to 27.1%) and pH-responsive drug release (38.60% cumulative release at pH 5.0), demonstrating its potential for cancer therapy. This combination of photothermal–chemotherapeutic efficacy, pH-responsive release, and high biocompatibility renders the nanocarrier promising for cancer treatment.
