Fiber SPR biosensor sensitized by MOFs for MUC1 protein detection

Rui Wang,Chunlan Liu,Yong Wei,Ze Ran,Tianci Jiang,Chunbiao Liu,Chen Shi,Zhuo Ren,Xingkai Wang,Zhihai Liu,Yu Zhang

The concentration of tumor markers is low, which needs a highly sensitive, stable and fast detection method. In this paper, we proposed and demonstrated a U-shape fiber SPR biosensor sensitized by MOFs materials. The surface of the U-shape SPR sensor was modified with MOFs materials to enhance the sensitivity, and the nucleic acid aptamer was immobilized on the sensor surface because of the biocompatibility of MOFs materials. By the high specificity of the nucleic acid aptamer, the MUC1 protein was recognized and detected. The testing results indicate that the sensor has a logarithmic linear response in the MUC1 protein concentration detection range of 1 pg/ml-100 μg/ml, its sensitivity and detection limit are 5.33 nm/log(μg/ml) and 0.16 pg/ml respectively. After being sensitized by MOFs, the detection sensitivity of the sensor can be increased by 1.62 times, the LOD can be decreased by 0.75 times. The sensor has high sensitivity and specificity, which has broad application prospects in clinical detection of tumor markers.

A portable multicolor aptasensor for MUC1 detection based on enzyme-mediated cascade reaction

Jin-Hong Sui,Yun-Yun Wei,Jin Li,Zhang-Run Xu

Mucin 1 (MUC1) is an important tumor biomarker, and the determination of MUC1 is of great significance in clinical diagnostics, prognosis assessment and therapy monitoring. Developing point-of-care testing method of MUC1 is necessary in resource-limited settings. Herein, a visual multicolor aptasensor for MUC1 detection was designed based on an alkaline phosphatase (ALP)-triggered cascade reaction. In the presence of MUC1, ALP-decorated complementary DNA and MUC1 aptamer on magnetic beads were dissociated due to the recognition between MUC1 and aptamer. With the assistance of a magnet, ALP-decorated complementary DNA was separated and collected to catalyze the hydrolysis of l -ascorbic acid-2-phosphate generating l -ascorbic acid, which etched the manganese dioxide nanosheets to mediate the oxidation reaction of 3,3′,5,5′-tetramethylbenzidine sulfate, resulting in different products with distinct visible multicolor. The naked eye detection of MUC1 was realized and the detection limit was estimated to be 0.18 U mL −1 by using the colorimetric method. This proposed aptasensor possesses the merit of vivid multicolor change, and provides a new way for MUC1 detection by naked eyes.

NSUN2 affects diabetic retinopathy progression by regulating MUC1 expression through RNA m5C methylation

Wang Runze,Xue Wei,Kan Feifei,Zhang Huiying,Wang Di,Wang Lei,Wang Jianwen

Background Diabetic retinopathy (DR) is the leading cause of blinding eye disease among working adults and is primarily attributed to the excessive proliferation of microvessels, which leads to vitreous hemorrhage and retinal traction, thereby significantly impairing patient vision. NSUN2-mediated RNA m 5 C methylation is implicated in various diseases, and in this investigation, we focused on elucidating the impact of NSUN2 on the regulation of the expression of the downstream gene MUC1, specifically through RNA m 5 C methylation, on the progression of DR. Method Utilizing Microarray analysis, we examined patient vitreous fluid to pinpoint potential therapeutic targets for DR. Differential expression of NSUN2 was validated through qRT-PCR, Western blot, and immunofluorescence in human tissue, animal tissue, and cell model of DR. The relationship between NSUN2 and DR was explored in vitro and in vivo through gene knockdown and overexpression. Various techniques, such as MeRIP-qPCR and dot blot, were applied to reveal the downstream targets and mechanism of action of NSUN2. Results The levels of both NSUN2 and RNA m 5 C methylation were significantly elevated in the DR model. Knockdown of NSUN2 mitigated DR lesion formation both in vitro and in vivo. Mechanistically, NSUN2 promoted MUC1 expression by binding to the RNA m 5 C reader ALYREF. Knockdown of ALYREF resulted in DR lesion alterations similar to those observed with NSUN2 knockdown. Moreover, MUC1 overexpression successfully reversed a series of DR alterations induced by NSUN2 silencing. Conclusions NSUN2 regulates the expression of MUC1 through ALYREF-mediated RNA m 5 C methylation, thereby regulating the progression of DR and providing a new option for the treatment of DR in the future.

Ultra-responsive label-free electrochemical aptamer sensor based on 3D-CdCo-ONSs@AuNPs for detection of tumour marker MUC1

Longbin Que,Hehua Zhang,Xue Qi,Xin Gu,Hongmin Gao,Yang Zhou,Dong Chang,Hongzhi Pan

Mucin 1 (MUC1), a pivotal tumor biomarker overexpressed in breast cancer, is conventionally detected using laborious, costly, and technically demanding methods. To overcome these limitations, we present a novel label-free electrochemical aptasensor for rapid, sensitive, and cost-efficient MUC1 quantification. The sensor employs a nanohybrid platform composed of gold nanoparticle-decorated three-dimensional cadmium-cobalt oxide nanostructures (3D-CdCo-ONSs@AuNPs), which synergistically amplify the differential pulse voltammetry (DPV) signal. The 3D-CdCo-ONSs provide a high-surface-area scaffold for dense AuNP deposition, enabling efficient immobilization of thiolated aptamers via Au–S bonds. Comprehensive material characterization via scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), and X-ray photoelectron spectroscopy (XPS) confirmed the structural and compositional integrity of the nanocomposite. Under optimized conditions, the sensor exhibited a broad linear detection range from 10 pg/mL to 100 μg/mL (R 2  = 0.998) with an ultralow limit of detection (LOD) of 0.96 pg/mL. The assay demonstrated excellent reproducibility, stability, and selectivity in complex matrices, facilitating precise MUC1 quantification in clinical samples. This work advances the development of portable, high-performance biosensors for early cancer diagnosis and monitoring.

Au-Luminol-decorated porous carbon nanospheres for the electrochemiluminescence biosensing of MUC1

Jing-Wen Gao,Miao-Miao Chen,Wei Wen,Xiuhua Zhang,Shengfu Wang,Wei-Hua Huang

Electrochemiluminescence (ECL) nanomaterials are usually deposited compactly on the surface of electrodes, which may cause poor mass transfer of reactants, thereby resulting in low ECL efficiency. In this work, we developed a novel kind of luminescent material denoted as C–Au-luminol nanospheres (C–Au-Lum NSs) by high dispersion of luminophores on porous carbon nanospheres (PCNSs). C–Au-Lum NSs were facilely prepared by the in situ reduction of chloroauric acid with the luminescent reagent luminol (Lum) on the nano-pores of PCNSs. Plenty of luminescent Au-Lum NPs were dispersedly concentrated inside the numerous pores and hollow interiors of PCNSs, effectively increasing the mass transfer of reagents and accelerating the electron transport inside the porous nanospheres. This greatly improved the availability of luminophores and endowed C–Au-Lum NSs with excellent ECL emission. After further integrating with enzymatic circulation and strand displacement, an ultrasensitive ECL biosensor was achieved for the ultrasensitive detection of an important tumor biomarker, mucin1. The logarithmically linear range from 0.1 pg mL−1 to 1 ng mL−1 with the detection limit of 47.6 fg mL−1 (S/N = 3) was achieved, demonstrating the superior performance of C–Au-Lum NSs. This work would provide new ideas for the construction of high-performance ECL sensing platforms for diverse applications.

A Sensitive Electrochemical MUC1 Sensing Platform Based on Electroactive Cu-MOFs Decorated by AuPt Nanoparticles

Ya Li,Lina Hou,Zhiling Liu,Wenbo Lu,Man Zhao,He Xiao,Tianjun Hu,Zhanfeng Zheng,Jianfeng Jia,Haishun Wu

Mucin 1 protein (MUC1) is a well-known tumor marker overexpressed in a variety of human malignancies, and thus its highly selective and sensitive detection is significantly desired for clinical diagnostics. Herein, a sensitive electrochemical MUC1 sensing platform has been fabricated based on copper-based metal-organic frameworks (Cu-MOFs) decorated with gold-platinum alloy nanoparticles (AuPt NPs). Besides serving as a redox probe to avoid introducing extra redox media, the electroactive Cu-MOFs function as catalysts in the hydrogen peroxide reduction, resulting in electrochemical signal amplification. Meanwhile, AuPt NPs loaded on the Cu-MOFs mimic horseradish peroxidase to promote the reduction of hydrogen peroxide, giving rise to the secondary-amplified electrochemical signal. Moreover, the target MUC1 triggered catalytic hairpin assembly (CHA) further improves the sensitivity of the developed biosensor via the target recycling amplification. The constructed sensing platform displayed a desirable dynamic range between 10 fM and 10 nM in the MUC1 detection, with a sufficiently low limit-of-detection (3.33 fM). The current strategy of utilizing hybrid nanocomposite consisting of MOFs and alloy NPs and enzyme-free CHA provides a promising platform for the detection of biomarkers in the clinical diagnosis with high selectivity and sensitivity.

An electrochemical method based on CRISPR-Cas12a and enzymatic reaction for the highly sensitive detection of tumor marker MUC1 mucin

Zhenhuan Jin,Wei Xiao,Lin Shen,Xiaoxue Shi,Jianping Li

Anti-cancer therapy is crucial in cancer prevention and anti-cancer, and thus, highly sensitive methods for detecting cancer biomarkers are essential for cancer early diagnosis. Herein, an electrochemical aptamer biosensor based on the CRISPR-Cas12a system was constructed for the detection of cancer tumor biomarker MUC1 mucin. The sensitivity was significantly prompted by enzyme-catalyzed signal amplification, and the selectivity was improved by the dual recognition of the aptamer to MUC1 and crRNA-Cas12a system to the aptamer. Glucose oxidase (GOD) was loaded on the surface of magnetic Fe3O4@Au (MGNP) via probe single-stranded DNA (pDNA) with the terminal modification of mercapto (–SH) to form GOD-pDNA/MGNP. The corresponding aptamer of MUC1 (MUC1 Apt) binds to its complementary ssDNA (cDNA) to form the activator Apt/cDNA, which is specifically recognized by crRNA-Cas12a and excites the trans-cleavage function of Cas12a, thus in turn trans-cleaves pDNA and detaches GOD from the magnetic particles. The magnetic beads were separated and transferred into a glucose solution, and the oxidation current of H2O2 produced by the catalytic reaction of GOD was measured on a Pt-modified magnetically-controlled glassy carbon electrode, resulting in an indirect determination of MUC1. The current change was linear with the logarithm of MUC1 concentration in the range from 1.0 × 10−17 g mL−1 to 1.0 × 10−10 g mL−1. The detection limit was as low as 7.01 × 10−18 g mL−1. The method was applied for the detection of MUC1 in medical samples.

A new approach to sensitively analyze glycoprotein with MUC1 as an example for the diagnosis of ovarian cancer

Shiao Yang,Zheying Mu,Jiazhu Pu,Weikang Ge,Kai Hu,Xuemei Jia,Genxi Li

Glycoproteins require sensitive detection for both biomedical research and clinical examination. Although existing strategies exploit glycosylation sites for recognition or signal labeling, they have not taken full advantage of glycoproteins’ potential as natural signal amplification platforms. To overcome the limitation, this study proposes a highly sensitive and modification-free electrochemical detecting strategy. Which employs a boronate derivative featuring dual-boronic acid groups. Through boronic acid-cis-diol specific interactions, the derivative simultaneously and directly co-immobilizes target glycoproteins and glycan-modified cascade enzymes via covalent polymerization. This forms an enzyme cascade amplification network, inducing methylene blue (MB) redox cycling to generate a differential pulse voltammetry (DPV) signal output. Using mucin 1 (MUC1) as a model target, the constructed biosensor achieves a detection limit of 116 fM with a wide linear range (0.2 pM-20 nM), meeting clinical requirements for glycoprotein quantification. The method also delivers promising results for MUC1 detection in clinical serum samples. Collectively, this work fully leverages the intrinsic potential of natural glycosylation sites on glycoproteins. The innovative dual-boronic acid “molecular grasper” strategy enables efficient and specific signal amplification, offering advantages including operational simplicity, cost-effectiveness, and rapid response. Ultimately, it provides a novel pathway for developing universal glycoprotein detection platforms, demonstrating significant value for disease diagnosis and biological research applications.

Highly sensitive electrochemical biosensor for MUC1 detection based on a DNA functionalized CdTe quantum dots as signal enhancers

Bao-Kang Jin,Jiajia Wang,Chun Kan

In this paper, a proportional electrochemical biosensor based on cadmium telluride/ polypyrrole (CdTe/PPy) nanocomposite is developed for the detection of MUC1 with high selectivity and sensitivity. The result indicates that CdTe/PPy nanocomposite is modified on the surface of glassy carbon electrode (GCE), which supplies a large surface area for immobilizing cap-DNA, ensuring its high selectivity and sensitivity. Whereafter, CdTe linked sig-DNA (MUC1 aptamer) is introduced, allowing MUC1 aptamer to hybridize with cap- DNA. CdTe as signal amplification elements in order to generate the differential pulse voltammetry (DPV) signal. Conceivably, the target MUC1 detection was based upon the current signal change due to the quantities change of the signal amplification elements CdTe. In the presence of MUC1, the MUC1 aptamer specifically binds to MUC1, resulting in the release of CdTe-sig-DNA from the electrode surface and the decrease of peak current. Under the optimized experimental conditions, the electrochemical biosensor present a highly selective, sensitive, stability and reproducibility for the MUC1 from the range of 0.1 nM to 100 nM with detection limit of 0.05 nM (S/N = 3). Therefore, the electrochemical biosensor has a potential application in medical disease diagnosis application detection.

Silencing MUC1 to regulate intracellular lipid metabolism: Overcoming sunitinib resistance and inhibiting metastasis in renal cell carcinoma

Xianhu Zeng,Tian Liu,Yi Teng,Zhipeng Li,Yan Liang,Dengshuai Wei,Guiming Zhang,Yong Sun,Shangcong Han

Renal cell carcinoma (RCC) has been extensively studied and is a paradigmatic example of a malignancy distinguished by metabolic reprogramming. Reprogramming lipid metabolism is crucial for determining the response and resistance to chemotherapy. To overcome this, we developed a modified nanomicelle delivery system comprising polyethyleneimine (PEI) as a positive charge donor and ibuprofen as the hydrophobic end. The overall system, denoted as SPPI, is PEGylated, conjugated with sialic acid, and plays a targeting role. This system successfully delivered siRNA to cancer cells by overcoming systemic and cellular barriers. The resulting conjugates form uniform, spherical nanomicelles (37.8 ± 0.9 nm; 25.5 ± 1.4 mV) with targeting ability, stability, efficient delivery, and low toxicity. The complicated and indirect way cholesterol affects ATP-binding cassette transporters was investigated. Administering SPPI/siMUC1 resulted in significant suppression of MUC1 expression and a 3.25-fold decrease in cholesterol synthesis compared to the control, and it subsequently reversed resistance to sunitinib by modulating P-GP, ABCG2, and MRP1 activities. Importantly, SPPI/siMUC1 nanomicelles demonstrated outstanding growth inhibition (72.2 %) of sunitinib-resistant 786-O cell xenografts and remarkable inhibitory effects on tumor metastasis (45.0 %) without observable systemic toxicity according to tissue analysis and weight change evaluation. The efficacy and safety of SPPI/siMUC1 in the treatment of ccRCC were demonstrated, especially after drug resistance develops. Thus, this study introduces a novel strategy for addressing drug resistance in metabolically active cancers by targeting the lipid metabolism pathway.

Nondestructive analysis of tumor-associated membrane protein MUC1 in living cells based on dual-terminal amplification of a DNA ternary complex

Xiaohao Liu,Dongsheng Mao,Guoli Deng,Yuchen Song,Fan Zhang,Shiqi Yang,Genxi Li,Feng Liu,Wei Cao,Xiaoli Zhu

Non-destructive analysis of cells at the molecular level is of critical importance for cell research. At present, immunoassay-based and aptamer-based methods can achieve non-structural destructive cell analysis, but still lead to changes in cells at the molecular level. Here, we have proposed a dual-terminal amplification (DTA) strategy, which enables nondestructive analysis of membrane protein MUC1 without the effect on protein expression and cell viability in living cells. Methods : A fluorophore (Cy5)-labeled DNA ternary complex consisting of three oligonucleotides is designed. It can recognize MUC1 through its aptamer region, and thus make the MUC1 of cells visible under a fluorescence microscope. When DNA polymerase is added, dual-terminal amplification is performed. One direction dissociates aptamer from MUC1, and the other direction, also known as rolling circle amplification (RCA), produces long linear DNA strands, which can be further adopted for quantitative analysis of MUC1. In this way, all reagents are removed from the surface of the cells after the analysis, which allows nondestructive analysis. We named this strategy dual-terminal amplification (DTA) analysis. Results : By using the DTA analysis, both in situ fluorescence imaging analysis and ex situ fluorescence quantitative analysis of MUC1 were achieved. In addition, the aptamer-containing DNA ternary complex stays on cell surface only during the analysis and leaves the cell after the analysis is complete. The cells can be maintained in a non-interfering state for the rest of the time. So after the analysis, it is found that there are no effect on the physiological activity of cells and the expression of target protein even after two rounds of repeatable imaging and quantitative analysis. Conclusion : In summary, we have successfully constructed a strategy for nondestructive analysis of membrane protein in living cells. We believe that this method provides a promising way for the analysis of the key membrane proteins of cells and the versatile utilization of precious cell samples.

Enhanced photoelectrochemical sensing for MUC1 detection based on TiO2/CdS:Eu/CdS cosensitized structure

Xing-Pei Liu,Pan-Pan Li,Chang-jie Mao,He-Lin Niu,Ji-Ming Song,Bao-Kang Jin

A novel ultrasensitive photoelectrochemical aptasensing was developed to detect mucin 1 (MUC1) based on TiO 2 /CdS:Eu nanocomposite. The TiO 2 /CdS:Eu nanocomposite modified fluorine-doped tin oxide (FTO) electrode was prepared by successive ionic layer adsorption and reaction technique (SILAR) and then was employed as matrix for immobilization of SH-DNA (DNA1). After 6-mercaptohexanol (MCH) blocked unbound sites of the electrode surface, the MUC1 aptamer(DNA2) was introduced, allowing MUC1 aptamer to hybridize with DNA1. Subsequently, CdS-COOH QDs as signal amplification elements were successively labeled on the terminal of MUC1 aptamer. In this case, the target MUC1 detection was based upon the photocurrent change originated from quantities change of the signal amplification elements CdS. In the presence of MUC1, the MUC1 aptamer was specifically bound with MUC1, and meanwhile MUC1/DNA2/CdS was released from the electrode surface, resulting in significantly decreased photocurrent intensity. The novel ultrasensitive photoelectrochemical aptasensing presented a highly selective, sensitive and reproductive detection of MUC1 from 1 nM to 5 μM with a lower detection limit of 0.3 nM. At the same time, it also provided a good specificity, reproducibility, stability and promising strategy to detect a variety of other protein targets for early clinical diagnosis of different diseases.
