Antioxidant and Anti-Inflammatory Activities of the Crude Extracts of Moringa oleifera from Kenya and Their Correlations with Flavonoids

Ming-Quan Guo

Moringa oleiferaLam. (M. oleifera) is commonly distributed and utilized in tropical and sub-tropical areas. There has been a large number of reports on the antioxidant and anti-inflammatory activity of its leaves, but only a few about its seeds and roots. Hence, in this work we aimed to systematically compare the antioxidant and anti-inflammatory activities of the ethanol crude extracts of leaves, seeds, and roots ofM. oleiferafrom Kenya, and further correlate the differential activities with the chemical constituents from these three parts. The antioxidant activities were measured by using three different assays (DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2′-azinobis-(3-ethylbenzthiazoline-6-sulfonic acid) and FRAP (Ferric-Reducing Antioxidant Power), respectively). Results showed that the leaf extracts displayed the highest DPPH radical scavenging and FRAP total reducing power activities with IC50values of 1.02 ± 0.13 mg/mL and 0.99 ± 0.06 mM Fe2+/g, respectively; the leaf and root extracts exhibited potential ABTS radical scavenging activities with the IC50values of 1.36 ± 0.02 and 1.24 ± 0.03 mg/mL. Meanwhile, the leaf and seed extracts (11.1–100 µg/mL) also exerted obvious anti-inflammatory activities, as indicated by the inhibition of NO production. To further reveal correlations between these differential activities with the chemical constituents in the three organs, the total flavonoids content (TFC) of the three different extracts were evaluated, and the TFC of leaves, seeds and roots were found to be 192.36 ± 2.96, 5.89 ± 0.65 and 106.79 ± 2.12 mg rutin equivalent (RE)/g, respectively. These findings indicated the important impacts of the total flavonoid contents on antioxidant and anti-inflammatory activities. Additionally, we further determined the phytochemical profiles ofM. oleiferaby HPLC-UV/ESI-MS/MS, and identified most of the chemical constituents of leaves as flavonoids. In summary, the leaves ofM. oleiferaare a better potential natural source of antioxidants and anti-inflammatory agents, and very promising for development into the health promoting dietary supplements. Keywords:Moringa oleifera;antioxidant;anti-inflammatory;flavonoids;phytochemical profile

Yao-Ying Wang,Chao Peng,Yue Zhang,Zi-Rong Wang,Yi-Ming Chen,Jin-Feng Dong,Ming-Long Xiao,Dong-Li Li,Wu Li,Qing-Jian Zou,Kun Zhang,Ping Wei

Moringa oleifera leaves ( MO L) attract extensive attention in agriculture, pharmaceutical and food industry because of its rich biological activity. In this study, ChCl/LA as one of the deep eutectic solvents (DESs) was selected to extract flavonoids from Moringa oleifera leaves (F MO L). The extraction rate of F MO L reached 54.8 mg/g. After separation of F MO L, its biological activities were systematically studied. The IC50 values of F MO L for DPPH radical, ABTS radical scavenging activity, FRAP and ORAC were 27.3 μg/mL, 41.2 μg/mL, 18.7 mg TE/g and 6008.4 mg TE/g, respectively, indicating F MO L had excellent antioxidant activity . F MO L not only had better antibacterial activity than the positive controls, but also had inhibitory activity to tumor cells. The IC50 values of F MO L on B16 cells, HCT116 cells and Hep G-2 cells were 530.7, 328.0, and 533.7 μg/mL, respectively. Nine flavonoid compounds were identified by UPLC-Q-Orbitrap MS/MS, and four of them were confirmed from MO L for the first time. Therefore, F MO L extracted by ChCl/LA had rich bioactive components and is expected to be used in food and pharmaceutical industries.

The structural characterization of polysaccharides from three cultivars of Moringa oleifera Lam. root and their effects on human intestinal microflora

Linlin Jia,Xiaoqiang Peng,Zeyuan Deng,Bing Zhang,Hongyan Li

Moringa oleifera Lam. is an edible plant . In this experiment, three kinds of small molecular weight Polysaccharides (MRP-D, MRP-P, MRP-H) were extracted from Dai-Jia ecology, Pkm-3 and Hong-he 1 Moringa oleifera root that were cultivated in three regions of Yunnan Province. The Moringa oleifera root Polysaccharides (MRPs) were identified to contain β-(1 → 3)-Glucose structure, without triple-helical conformation. In vitro fermentation experiment showed the MRPs increased the content of short-chain fatty acids (3–5-fold) and the abundance of beneficial microflora such as Bacteroidetes and decreased the abundance of harmful microflora such as Proteobacteria at the same time. The composition and effects on the human intestinal microflora of MRPs were different, indicating that the planting regions would affect the composition and function of the bioactives. These results provide an experimental basis for the industrial layout of Moringa oleifera and the application of MRPs in food processing.

An integrated fecal microbiome and metabolomics in type 2 diabetes mellitus rats reveal mechanism of action of Moringa oleifera Lamarck seeds polysaccharides to alleviate diabetes

Hongru Yang,Chongbin Lei,Dongyao Li,Lei Ma,Na Zhang,Yumiao Lang,Liping Wu,Miaoshu Wang,Hongtao Tian,Chen Li

Moringa oleifera Lamarck seeds (MOS) have been traditionally used in folk medicine and documented for their potential to alleviate type 2 diabetes symptoms, but the potential mechanisms are still unknown. The purpose of this article is to investigate the effects of MSAP (alkali-extracted polysaccharide from MOS) on diabetic rats by assessing its impact on the gut microbiome, diabetes-related biochemical markers, and fecal metabolomics. The results demonstrated that the fasting blood glucose, glucose tolerance, insulin tolerance, insulin level and lipopolysaccharides (LPS) level in the rats treated with MSAP were all improved. Specifically, MSAP was found to modulate the composition and diversity of the gut microbiota, increasing the ratio of Firmicutes / Bacteroidetes , which enhanced the quantity of probiotic Lactobacillus and butyrate-producing bacteria, such as Roseburia , thereby reinforcing the intestinal epithelial barrier. Furthermore, fecal metabolomics indicates that MSAP actively regulates pathways closely associated with diabetes, including sphingolipid metabolism, amino acid synthesis and catabolism, retrograde endogenous cannabinoid signaling, and the modulation of TRP channels by inflammatory mediators. By integrating microbiome and metabolomics data, this study elucidated the mechanisms through which MSAP alleviates diabetes. In conclusion, the findings suggest that polysaccharides from MOS hold potential as a medicinal and edible homologous food for diabetes management.

Optimization and validation of folate extraction from Moringa oleifera leaves powder

Du Huan,Li Tao,Xue Qiaoli,Tian Yang,Hu Yongjin

Moringa oleifera leaves are potentially benign sources of natural folates, thus a reliable folate extraction protocol is necessary, which directly affects the accuracy of subsequent determination of total folates or various folate derivatives. With this aim, process variables including antioxidants, extraction buffer pH, Moringa oleifera leaves powder (MOLP)/extraction buffer ratio, heating extraction time, amounts of α- amylase, protease and rat serum, and also incubation time of rat serum were investigated and optimized by response surface methodology (RSM). It is interesting that no positive influence of α- amylase or protease solution was observed in this study, consequently a single-enzyme treatment was used. An average folate level of 1190.06 µg/100 g dry matter was achieved by microbial assay under the optimized conditions of extraction buffer pH 7.0, MOLP/extraction buffer ratio 1:24 (g/mL), rat serum 350 µL, digest time 2.0 h, in good agreement with the value predicted by quadratic model. In addition, the validity of protocol established in this study was confirmed. The intra-day precision (relative standard deviation) varied between 1.42% with 1.84%, inter-day precision varied in the range of 1.34–2.25%. The average recoveries were in the range of 101.56–103.27%. Besides, the matrix effects ranged from 101.41 to 103.34%. The protocol could provide a reference to fairly evaluate folates in Moringa oleifera leaves from diverse sources, and even in other leafy plants. Meanwhile, Moringa oleifera leaves with high folate level seem to have tremendous potential to increase nutritional value of foods as well as to enhance regional economies.

Moringa oleifera ameliorates streptozotocin-induced hyperglycemia in T2DM rats via gut microbiota

Liu Ye,Fan Minxia,Xu Yongbing,Muema Felix Wambua,Chen Guilin,Guo Mingquan

Moringa oleifera have been widely consumed in China and African countries to treat DM. This study investigated the arresting hypoglycemic effects of Moringa oleifera leaves extracts (MOLE) on T2DM via the gut microbiota pathway. The results displayed that MOLE treatment for 30 days significantly improved the plasma FBG and lipids (TC, TG, LDL-C and HDL-C) levels in T2DM rats in a dose-dependent manner ( p  < 0.05, p  < 0.01), respectively. Also, MOLE treatment increased the abundance of Bacteroidetes and Firmicutes/Bacteroidetes ratio and decreased the abundance of Firmicutes, and dramatically increased the contents of the acetic acid, propionic acid, and total SCFAs in the feces of T2DM rats ( p  < 0.05, p  < 0.01), respectively. KEGG signaling pathway further suggested that MOLE strongly participated in the carbohydrate metabolism in T2DM rats. This work indicates the noteworthy improvement of MOLE on metabolic disorders in T2DM rats via the regulation of gut microbiota and SCFAs.

Synergistic inhibition of isolated flavonoids from Moringa oleifera leaf on α-glucosidase activity

Yahya Saud Hamed,Mohamed Abdin,Ahmed Mohamed Rayan,Hafiz Muhammad Saleem Akhtar,Xiaoxiong Zeng

Flavonoids are phenolic substances naturally distributed in plants with potential antidiabetic activity through the inhibition of α-glucosidase. Therefore, the present study investigated the inhibitory effects of α-glucosidase activity by purified flavonoids (PF) isolated from Moringa oleifera leaves before and after simulated gastric digestion in the absence or presence of nitrate. Furthermore, the combined effects of PF, chlorogenic acid and acarbose to reduce the acarbose dosage that caused 50% inhibition of α-glucosidase activity were studied by using CompuSyn software. Five flavonoids (rutin, kaempferol acetyl glycoside , kaempferol 3-O-glucoside, quercetin-3-glucoside and quercetin-3-acetyl-glucoside) were identified by HPLC-ESI-MS. The results showed that at a concentration of 100 μg/mL, flavonoids and acarbose inhibited α-glucosidase activity by 54.41% and 69.17%, respectively. Lineweaver-Burk plots revealed uncompetitive inhibition of α-glucosidase by PF. Flavonoids caused an 8.17 times reduction in acarbose concentration which was required to inhibit α-glucosidase. In the absence or presence of nitrate, the inhibitory activity decreased by 48.44% and 72.12%, respectively, after simulated gastric digestion when compared with undigested flavonoids. Rutin and kaempferol 3- O -glucoside showed the highest stability in the simulated gastric liquid, while quercetin-3-acetyl-glucoside and kaempferol 3- O -glucoside exhibited the highest stability in the simulated gastric liquid in the presence of nitrate.

Nutritional compositions of Indian Moringa oleifera seed and antioxidant activity of its polypeptides

Lili Liang,Cong Wang,Shaoguang Li,Xuemei Chu,Kunlai Sun

To study the nutritional composition of Indian Moringa oleifera seed and the antioxidant activity of M. oleifera seed polypeptide, Indian M. oleifera seed was used as raw material for composition analysis and content determination. After extraction of the seed protein, enzymatic hydrolysis with flavourzyme, dispase, papain, pepsin, and alcalase was conducted for different time, and the optimal enzymatic hydrolysis conditions was determined with DPPH scavenging capacity as an indicator. The seed polypeptides obtained by enzymatic hydrolysis were ultrafiltered, and the active peptide fragments were tracked with DPPH, HO (•OH), ABTS and superoxide anion (O 2 • − ) free radical scavenging ability and lipid oxidation inhibition rate as indicators. The results showed that the protein content in Indian M. oleifera seed was high to 40.34%, containing seven essential amino acids. The content of macroelements such as potassium, sodium, and magnesium is high, with the potassium content as high as 2,357.71 mg/kg, among the microelements, the iron content as high as 36.2 mg/kg. The optimum enzymatic hydrolysis conditions were as follows: enzymatic hydrolysis with flavourzyme (50°C, pH 6.7) for 300 min, and DPPH scavenging capacity was 84.76%. Activity tracing found that the polypeptide fragment with molecular weight <3.5 kDa had the strongest antioxidant capacity, and the EC 50 values of DPPH, •OH, ABTS, and O 2 • − free radical scavenging rates were 4.0, 4.2, 5.3, and 4.3 mg/ml, respectively. The above results show that Indian M. oleifera seed not only has high nutritional value, but its protease enzymatic hydrolyzate also has significant antioxidant activity, which can be further developed into nutrition products, healthcare products, functional foods, beauty and skin care products, liver protection drugs, etc.

Effects of Heat Treatment on Physicochemical Properties of Moringa oleifera Lam. Leaf Protein

Chuyu Xi,Wenjie Li,Zhiguo Xu,Jing Xie,Xiaoyu Gao,Dan Feng,Yang Tian,Shuang Song

M. oleiferaleaves represent a novel and nutritious food. Prior research has demonstrated thatM. oleiferaleaves can elicit allergic responses in BALB/c mice. Based on these findings, further studies were conducted to investigate the effects of heat treatment on the allergenicity, particle size, zeta potential, total sulfhydryl (TSH) content, hydrophilicity and hydrophobicity, ultraviolet spectrum, and intrinsic fluorescence spectrum ofM. oleiferaleaf protein. Additionally, in vitro digestion experiments were carried out to gain further insights into the protein’s behavior under these conditions. The experiment simulated the alterations inM. oleiferaleaf protein during the processes of cooking and digestion. The findings of this experiment can provide certain guidance for the processing ofM. oleiferaleaf products. The hydrophilicity, hydrophobicity, transmembrane region, antigen index, calcium binding site, spatial structure, and homology ofM. oleiferaleaf fructose 1,6 bisphosphate aldolase (FBA) were simulated and calculated based on the amino acid sequence of the 36 kDa allergen. These parameters collectively serve to indicate the allergenic activity of the peptide. The findings of the analysis align with the outcomes of the sensitization experiments, suggesting that the FBA ofM. oleiferaleaves is indeed consistent. In conjunction with the heat treatment experiments, this research can inform the preparation ofM. oleiferaleaf foods and provide a foundation for further investigation intoM. oleiferaleaf allergens.

Microwave-assisted extraction of polysaccharides from Moringa oleifera Lam. leaves: Characterization and hypoglycemic activity

Chun Chen,Bin Zhang,Qiang Huang,Xiong Fu,Rui Hai Liu

Microwave-assisted extraction of Moringa oleifera Lam. leaf polysaccharides was investigated using a response surface methodology. The following conditions were found to generate an experimental yield of 2.96   ±   0.11%, which closely agreed with the predicted value of 2.89%: time 70   min, microwave power 700   W, temperature 70   °C and a liquid/solid ratio of 35   mL/g. After the MLPs were treated with graded precipitation of 40%, 60% and 80% (v/v) ethanol solution, the respective average molecular weight polysaccharide fractions, namely MLP-1 (9.68   ×   10 3   kDa), MLP-2 (6.29   ×   10 3   kDa) and MLP-3 (4.86   ×   10 3   kDa), were obtained. These three fractions were composed of xylose, mannose, glucose, galactose and arabinose but with differing molar ratios. They had similar Fourier transform infrared spectroscopy but their surface morphology differed as determined by scanning electron microscopy. The Congo red assay indicated that the three fractions were not helical polysaccharides. The three polysaccharide fractions exhibited different rheological properties. MLP-1 exhibited pseudoplastic flow behaviors, while MLP-3 displayed near-Newtonian flow behaviors over the entire accessible frequency range. In addition, MLP-3 had stronger inhibitory effects on α-amylase and α-glucosidase than MLP-1 and MLP-2. The results suggest that MLP-3 has potential for the treatment of diabetes.

Extraction of phenolic compounds from Moringa oleifera Lam. leaves with ultrasonic-assisted deep eutectic solvents

Wang Zilin,Tian Yang,Yang Min,Yang Junyan,Wang Yifan,Tao Liang,Sheng Jun,Shi Chongying

Background: In this study, deep eutectic solvents (DESs) combined with ultrasound-assisted extraction (UAE) were used to extract bioactive compounds from the leaves of Moringa oleifera Lam.Methods: The FT-IR method was used to analyze the structural characteristics of the DESs, and the extraction efficiencies of the DESs for total phenolic content (TPC) and total flavonoid content (TFC) were evaluated. The stability of the extracts under high temperature and UV radiation was assessed, and their antioxidant activity was investigated after undergoing in vitro simulated digestion. Results: The results show that the seven DESs extracted more TPC and TFC than did the 70% ethanol (36.27 ± 1.58 mg GAE/g, 23.09 ± 1.47 mg RT/g), and the extraction process of UAE-DES was optimized by selecting choline chloride: citric acid as the DES solvent, which has the highest extraction of TPC (86.92 ± 1.34 mg GAE/g) and TFC (49.73 ± 0.85 mg RT/g). The stability results indicated that the DES phenolic extracts were less stable when exposed to high temperature and UV radiation, indicating that DES extracts have better bioactivity. Moreover, after in vitro simulated digestion, the DES extract shows a higher DPPH free radical scavenging capacity (12.79 ± 3.88 mmol Trolox/g of DES extracts, 6.99 ± 4.02 mmol Trolox/g of ethanol extracts) and ferric ion reducing antioxidant power (62.61 ± 1.71 mmol Trolox/g of DES extracts, 55.07 ± 1.66 mmol Trolox/g of ethanol extracts) than ethanol extracts. Conclusion: This study confirmed that DESs are a new and environmentally friendly solvent that can be used for the extraction of phenolic compounds.
