Bioresorbable hydrogels prepared by photo-initiated crosslinking of diacrylated PTMC-PEG-PTMC triblock copolymers as potential carrier of antitumor drugs

Yuandou Wang,Laishun Xi,Baogang Zhang,Qingzhen Zhu,Feng Su,Katarzyna Jelonek,Arkadiusz Orchel,Janusz Kasperczyk,Suming Li

PTMC-PEG-PTMC triblock copolymers were prepared by ring-opening polymerization of trimethylene carbonate (TMC) in the presence of dihydroxylated poly(ethylene glycol) (PEG) with Mn of 6000 and 10,000 as macro-initiator. The copolymers with different PTMC block Lengths and the two PEGs were end functionalized with acryloyl chloride. The resulting diacrylated PEG-PTMC-DA and PEG-DA were characterized by using NMR, GPC and DSC. The degree of substitution of end groups varied from 50.0 to 65.1%. Hydrogels were prepared by photo-crosslinking PEG-PTMC-DA and PEG-DA in aqueous solution using a water soluble photo-initiator under visible light irradiation. The effects of PTMC and PEG block lengths and degree of substitution on the swelling and weight loss of hydrogels were determined. Higher degree of substitution leads to higher crosslinking density, and thus to lower degree of swelling and weight loss. Similarly, higher PTMC block length also leads to lower degree of swelling and weight loss. Freeze dried hydrogels exhibit a highly porous structure with pore sizes from 20 to 100 µm. The biocompatibility of hydrogels was evaluated by MTT assay, hemolysis test, and dynamic clotting time measurements. Results show that the various hydrogels present outstanding cyto- and hemo-compatibility. Doxorubicin was taken as a model drug to evaluate the potential of PEG-PTMC-DA and PEG-DA hydrogels as drug carrier. An initial burst release was observed in all cases, followed by slower release up to more than 90%. The release rate is strongly dependent on the degree of swelling. The higher the degree of swelling, the faster the release rate. Finally, the effect of drug loaded hydrogels on SKBR-3 tumor cells was evaluated in comparison with free drug. Similar cyto-toxicity was obtained for drug loaded hydrogels and free drug at comparable drug concentrations. Therefore, injectable PEG-PTMC-DA hydrogels with outstanding biocompatibility and drug release properties could be most promising as bioresorbable carrier of hydrophilic drugs.

Drug-Loaded Janus Dressings Precipitated from PEG–PTMC Thermosensitive Hydrogel for Oral Ulcer Treatment

Geng Chen,Haoming Wu,Xingchen Zhao,Zhen Zhang,Xin Hu,Peijie Hou,Chengdong Xiong,Likun Guo,Lifang Zhang

Oral ulcers, prevalent lesions of the oral mucosa, often stem from trauma, viral infections, and autoimmune diseases, necessitating effective treatment strategies to address both their physical and psychological impacts. Current treatments are often limited by issues such as poor adhesion and rapid dilution within the dynamic oral environment. To overcome these challenges, we have explored the application of thermosensitive hydrogels, specifically aqueous solutions of poly(ethylene glycol)–poly(trimethylene carbonate) (PEG–PTMC) copolymers. These copolymers are noted for their biocompatibility and temperature-responsive phase transition properties. We engineered a thermosensitive drug delivery system by fine-tuning the molecular weight and segment lengths of PEG–PTMC, which gels near 32 °C and precipitates at 37 °C. Utilizing the slight temperature differential between the oral mucosa and ambient air, the system forms a Janus structure with distinct gel and precipitate layers, ensuring sustained drug release and enhanced tissue adhesion. This innovative formulation not only acts as a protective, drug-laden dressing that promotes healing but is also designed for ease of use, allowing for self-administration. This study presents a significant advancement in the treatment of oral ulcers, offering a promising solution through the in situ formation of a bilayered, drug-loaded dressing, leveraging the unique properties of thermosensitive hydrogels.

Investigation on the influence of different block ratios on the properties of biodegradable ABA–type block copolymer PTMC–b–PCL–b–PTMC

Zhen Zhang,Xin Hu,Geng Chen,Peijie Hou,Qi Zhang,Dahong Zhang,Chengdong Xiong,Xiliang Liu,Lifang Zhang

In this study, a series of ABA–type block copolymers, PTMC–b–PCL–b–PTMC, with varying block ratios were synthesized via sequential ring–opening polymerization. A systematic investigation was conducted to assess the mechanical properties, thermal behavior, crystallization characteristics, enzymatic degradation under lipase–rich conditions, and biological performance of the copolymers with different compositions. The results demonstrated that increasing the PTMC block content significantly influenced the physicochemical properties of the copolymers. Specifically, a higher proportion of PTMC blocks resulted in decreased crystallinity and enhanced toughness. Furthermore, lipase degradation studies revealed that during the early stages, all polymer compositions exhibited the characteristic surface erosion behavior of polyesters. However, in the later stages, PCL–OH exhibited more pronounced collapse degradation, whereas the block copolymers PTCT6040 and PTCT7030, with higher crystallinity, maintained surface erosion–dominated degradation while preserving their macroscopic structure and mechanical stability. Additionally, biocompatibility assays confirmed the non–cytotoxicity of these materials, highlighting their potential for applications in tissue engineering.

Preparation of temperature-controlled shrinkage PTMC/PVP core-shell nanofibrous membrane with spindle-knotted structure for accelerating wound closure

Liping Liu,Ting Li,Maolei Sun,Wenyuan Jia,Kun Jiao,Shaoru Wang,Yun Liu,Kongzhao Jiang,Yuheng Yang,Zhihui Dai,Changli Xia,Zhiqiang Cheng,Guomin Liu,Yungang Luo

Temperature-controlled shrinkage polytrimethylene carbonate (PTMC)/polyvinylpyrrolidone (PVP) core–shell nanofibrous membranes were successfully prepared by uniaxial electrospinning to accelerate wound closure. The characterization and temperature-controlled shrinkage of the PTMC/PVP nanofibrous membranes were investigated. The results showed that the PTMC/PVP core–shell nanofibers had a periodic distribution of porous spindle-knotted structure, and EDS elemental mapping of N elements indicated the core–shell structure with PTMC shell and PVP core. The PTMC/PVP (1:3) nanofibrous membrane had the maximum shrinkage in PBS for 120 min, which area percentage were decreased to 68.0% at 20 °C and 49.4% at 37 °C. The results indicated that the PTMC/PVP nanofibrous membranes shrinkage would be increased with the temperature increased. Further experiments confirmed the PTMC/PVP (1:3) nanofibrous membrane could reduce the wound area of rat skin to approximately 70% in 30 min.

Elecrtospinning PTMC/Gt/OA-HA composite fiber scaffolds and the biocompatibility with mandibular condylar chondrocytes

Ting Yang,Xuejun Cui,Yanbing Kao,Hongyan Wang,Jihong Wen

Hydroxyapatite (HA) nanoparticles were modified by oleic acid (OA) to improve their lipophilicity and the dispersibility in polymer. XRD and FTIR demonstrated that the surface modification did not change the chemical properties of HA. OA modified HA (OA-HA) nanoparticles were introduced into polytrimethylene carbonate/gelatin (PTMC/Gt) composite fiber scaffolds (CFSs) by electrospinning. Scanning electron microscope (SEM) showed that the distribution of OA-HA nanoparticles was more uniform than that of HA nanoparticles in the CFSs. Comparing PTMC/Gt CFSs, PTMC/Gt/HA CFSs and PTMC/Gt/OA-HA CFSs, PTMC/Gt/OA-HA CFSs showed the best mechanical property and more hydrophilic. Cell adhesion and proliferation assay (MTT assay) and confocal laser scanning microscopy (CLSM) demonstrated that the PTMC/Gt/OA-HA CFSs could provide a higher cell adhesion and proliferation environment for mandibular condylar chondrocytes(MCCs). Overall the PTMC/Gt/OA-HA CFSs have nontoxicity and good biocompatibility with MCCs and would be potential to be used in the field of bone and cartilage tissue engineering scaffolds in the future.

Construction of biobased PEF/PTMC composite vitrimer with advanced performance in mechanical properties, reprocessability and foamability via catalyst-free method

Weiqiang Han,Xia Liao

In this work, we reported a facile approach for fabricating external catalyst-free poly (ethylene 2,5-furandicarboxylate)/poly (trimethylene carbonate) (PEF/PTMC) blend vitrimer through two-step reactive blending by the addition of 2,2-bis(hydroxymethyl) 2,2′,2″-nitrilotriethanol (BIS-TRIS) and N, N′-tetraglycidyldiaminodiphenyl methane (TGDDM) for the first time. This method breaks through the difficulties of the traditional preparation method of vitrimer materials that must require the addition of metal catalyst, thus realizing the concept of green chemistry. At the same time, it also breaks through the difficulty of poor compatibility in the traditional blend preparation methods. Swelling and FTIR tests confirmed the presence of the dynamic cross-linking structure consisting of PEF and PTMC chains, while rheological analysis shows that the formed cross-linked network in PEF/PTMC blend can significantly improve its melt strength and make it could be foamed by supercritical carbon dioxide. An expansion ratio about 10-fold and microcellular foam with a uniform cell morphology can be obtained for PEF/PTMC composite vitrimer when the PTMC content and the gel content was less than 20% and 50%, respectively. Moreover, PEFMC foams possess greater compressive strength than that of commercial PET foams, exhibiting excellent mechanical properties. TGA and tensile experiments revealed that PEF/PTMC composite vitrimer exhibited excellent mechanical performance and thermal stability. Meanwhile, PEF/PTMC composite vitrimer still maintain the ability of excellent reprocessability. This work provides a practical green method to guide the design of novel polymer blends with improved miscibility, mechanical properties and sustainability by a dynamic cross-linking network.

3D printed gelatin/PTMC core/shell scaffolds with NIR laser-tuned drug/biomolecule release for cancer therapy and uterine regeneration

Shangsi Chen,Jiahui Lai,Jizhuo Chen,Liwu Zheng,Min Wang

Surgical resection is an efficient treatment for cancerous tissues and uterine fibroids in the women uterus. However, the insufficiency of clinical interventions could result in tumor recurrence, and the defective tissues remained would cause intrauterine adhesions (IUAs) and further affect reproduction capacity. In this study, 3D printed hydrogel/poly( l -lactide- co -trimethylene carbonate) (PLLA-co-TMC, “PTMC” in short) core/shell scaffolds with NIR-tuned doxorubicin hydrochloride (DOX) and estradiol (E2) dual release were designed and fabricated for cancer therapy and uterine regeneration. Gelatin (Gel) and DOX were homogeneously mixed and then 3D printed to form Gel-DOX scaffolds. Gel-DOX scaffolds were then immersed in PTMC-PDA@E2 solution to fabricate Gel-DOX/PTMC-PDA@E2 core/shell scaffolds. Consequently, Gel-DOX/PTMC-PDA@E2 scaffolds could release DOX and E2 in a chronological manner, firstly delivering DOX assisted by phototherapy (PTT) to effectively kill Hela cells and then sustainably releasing E2 to promote uterine tissue regeneration. In vitro experiments showed that core/shell scaffolds exhibited excellent anticancer efficiency through the synergy of DOX release and hyperthermia ablation. Moreover, E2 could be sustainably released for over 28 days in vitro to promote the proliferation of bone marrow-derived mesenchymal stem cells (BMSCs). The novel Gel-DOX/PTMC-PDA@E2 core/shell scaffolds have therefore exhibited potential promise for the treatment of cancer therapy and uterine regeneration.

Construction of PVA/OHA-Gs@PTMC/PHA double-layer nanofiber flexible scaffold with antibacterial function for tension free rectal in-situ reconstruction

Bingxu Zhang,Xujian Li,Chuan Jiang,Chuanguang Wang,Haifeng Que,Cheng Zheng,Zhixiao Ji,Xudong Tao,Hongtao Xu,Changcan Shi

The effective prevention and treatment of anastomotic leakage after intestinal anastomosis for colorectal diseases is still a major clinical challenge. In order to assist intestinal anastomosis healing and avoid anastomotic leakage caused by high tension, low blood supply or infection, we designed a double-layer nanofiber intestinal anastomosis scaffold, which was composed of electrospun PTMC/PHA nanofibers as the main layer, and electrospun PVA/OHA-Gs nanofibers with antibacterial properties as the antibacterial surface layer. This double-layer scaffold has good toughness, its maximum tensile force value could reach 8 N, elongation could reach 400 %, and it has hydrophilic properties, and its contact angle was about 60°. On the basis of reducing anastomotic tension and isolating intestinal contents, this double-layer nanofiber anastomotic scaffold not only played an antibacterial effect in the short term after surgery to reduce inflammatory response, but also had the characteristics of multiple three-dimensional network structure like extracellular matrix which could promote tissue healing. The PVA/OHA-Gs@PTMC/PHA scaffold was implanted into a rabbit model simulating mechanical intestinal obstruction, and the results showed that the nanofibers of the scaffold could be degraded in vivo while maintaining a certain stability, that is, the overall structure of the PVA/OHA-Gs@PTMC/PHA scaffold would not shrink and deform due to degradation in a certain period of time. Therefore, the treatment with this scaffold showed better healing at the anastomotic site. Compared to the direct anastomosis group and pure PTMC scaffold group, the double-layer scaffold group promoted a faster return to normal anastomotic strength within 7 days. This PVA/OHA-Gs@PTMC/PHA double-layer nanofiber flexible scaffold appears to be a promising therapeutic strategy to prevent anastomotic leakage after intestinal anastomosis.

The High Molecular Weight PTMC Enhances PLGA and Incorporates Inorganic Particles Into Composite Microsphere Scaffolds to Promote Bone Defect Repair

Gang Chen,Hai Lan

To address the issues of collapse-type degradation and poor toughness observed in PLGA-based bone repair scaffolds during degradation, this study blends PLGA with poly (trimethylene carbonate) (PTMC) and fabricates microsphere scaffolds through the emulsion method followed by sintering to form solid microsphere scaffolds. Additionally, to enhance the osteogenic potential of the scaffold, β-tricalcium phosphate (β-TCP) and varying amounts of titanium dioxide (TiO 2 ) are incorporated into the polymer. Performance analysis of the composite microsphere scaffolds reveals that the scaffold exhibits a continuous pore structure with a compressive modulus comparable to that of cancellous bone. After the incorporation of inorganic particles, the compressive modulus exceeded 70 MPa. In vitro degradation experiments confirm that the composite microsphere scaffold prepared by blending PTMC exhibited a degradation rate of 9.1% at week 12 while maintaining stable macroscopic morphology. Moreover, the pH at the final stage of degradation effectively mitigated the issue of acidic degradation associated with PLGA (pH = 6.32). In vitro cell experiments demonstrate that the prepared microsphere scaffolds possess good biocompatibility, and the incorporation of β-TCP and TiO 2 enhances the composite scaffold's osteogenic potential and antibacterial properties. This scaffold holds promise as a novel approach for the treatment of bone defects.

Preparation and design of intelligent multi-response PTMC/PVP composite nanofibrous dressing to promote wound closure and its applications in diabetic wounds

Liping Liu,Tao Yang,Shaoru Wang,Wenyuan Jia,Yun Liu,Kun Jiao,Yongzheng Yan,Yuheng Yang,Xuanzuo Jiang,Chiyu Li,Zhiqiang Cheng,Guomin Liu,Yungang Luo

The healing process of diabetic wounds is characterized by a slow and arduous progression, making it challenging to achieve wound closure and complete healing through self-contraction, as observed in normal wounds. In this study, an intelligent multi-responsive composite of polytrimethylene carbonate (PTMC)/polyvinylpyrrolidone (PVP) nanofibrous dressing (C-PPZS) loaded with simvastatin-loaded ZIF-8 nanoparticles (ZIF-8@SIM NPs) was fabricated via electrospinning technology. The dressing exhibited liquid-triggered and temperature-controlled shrinkage characteristics, as well as pH-responsive drug release properties. When subjected to a temperature of 37 ℃ for 60 min, the area shrinkage rate of PPZS in PBS was determined to be 78.36 ± 1.14 %, and C-PPZS also reduced the area of isolated mouse skin defects to 38.01 ± 2.33 % of its original size. Furthermore, in vitro studies demonstrated that C-PPZS exhibited favorable biocompatibility and proangiogenic properties, along with significant inhibitory effects against S. aureus and E. coli . In vivo experiments conducted on diabetic mice revealed that C-PPZS facilitated wound closure and healing, attenuated scar formation, stimulated collagen fiber deposition, and promoted vascular regeneration. In conclusion, C-PPZS has great potential as an advanced dressing for the treatment of chronic wounds.

Underwater self-healing of microphase-separated PLLA–b–PTMC biocompatible elastomers

Xin Hu,Shibo Luo,Zhen Zhang,Peijie Hou,Jun Wang,Lifang Zhang

Waterproof and self-healing materials are crucial for preventing leakage in implantable devices subjected to cyclic mechanical stress. Herein, we investigate how block composition governs the underwater self-healing behavior of biodegradable elastomers composed of poly(L-lactic acid) (PLLA) and poly(trimethylene carbonate) (PTMC). By tuning the PLLA/PTMC ratio, we modulated crystallinity and segmental mobility, with soft PTMC domains disrupting PLLA ordering and enhancing chain dynamics. Molecular dynamics simulations and thermal analysis revealed increased diffusivity and looser chain packing in PTMC-rich regions. In parallel, electrostatic potential mapping and surface energy analysis indicated reduced interfacial polarity, favoring hydrophobic polymer–polymer interactions under wet conditions. This work demonstrates a design strategy that integrates segmental dynamics and interfacial modulation to achieve water-tolerant self-healing in biodegradable polyesters, offering promise for biomedical sealing and tissue repair.

Dual sustained-release PTMC/PCL porous microspheres for lipid-soluble drugs

Liang Qiao,Fuhua Deng,Xulin Hu,Tianfang Huang,Junlin An,Dong Pan,Junqiang Yan,Gaofeng Liang,Jian He

It is still a considerable research challenge to design biodegradable drug delivery systems that efficiently transport lipid-soluble drugs with controlled release. In the present study, a microsphere delivery system, γ-cyclodextrin (γ-CD)/glutathione (GSH)-atorvastatin (AT) fibrous nanoparticles (γ-CD/GSH-AT FNs) loaded polytrimethylene carbonate (PTMC)/polycaprolactone (PCL) microspheres (PTMC/PCL/γ-CD/GSH-AT MPs), was constructed through an exquisite method. The results show that a large number of γ-CD/GSH-AT FNs can be encapsulated during the formation of microspheres and released in a controllable manner. Notably, the high dispersibility and drug loading of FNs can load with lipid-soluble drugs and protect the drugs from the solvent during microsphere preparation. FNs and MPs can achieve dual release. First, with the degradation of the microspheres, the encapsulated FNs were gradually released into the local microenvironment. Subsequently, the FNs began to dissolve and release the loaded AT over a period of more than 3 days. Therefore, the dual sustained-release drug delivery system can maintain the blood drug concentration for a longer time and reduce the number of administrations and the side effects of the drug. This study provides new ideas for the on-demand manufacture of controllable degradable drug delivery platforms to treat diseases.
