Polygonatum sibiricum Red., a kind of edible and medicinal plant, has a long history of use in traditional Chinese medicine and as a nutritional food additive. Water-soluble crude polysaccharide (PS50) was obtained from P. sibiricum rhizome. After deproteinization and dialysis, PS50 was isolated and purified via DEAE-52 cellulose and Sephadex G-75 gel filtration chromatography to obtain two novel polysaccharides (PSP50-2-1 and PSP50-2-2). Chemical analysis and nuclear magnetic resonance (NMR) studies indicated that PSP50-2-1 was composed of β-D-Glcp-(1→, →2)-β-D-Galp-(1→, →2,6)-β-D-Galp-(1→, β-D-Fruf-(2→, and PSP50-2-2 was composed of β-D-Glcp-(1→, β-D-Galp-(1→, →2)-β-D-Galp-(1→, →6)-β-D-Galp-(1→, →2,6)-β-D-Galp-(1→, β-D-Fruf-(2→. The morphological analysis suggested that PSP50-2-1 and PSP50-2-2 both had a rough surface with cracks. Pharmacological studies showed that PSP50-2-2 at concentrations of 2.59 and 5.19 μM significantly promoted the differentiation and mineralization of MC3T3-E1 cells in vitro, which indicated that PSP50-2-2 had a function of osteogenic activity. Above all, this study provides evidence that PSP50-2-2 may be used as an anti-osteoporotic agent, with applications in health-care and pharmaceutical industries.

Steam-processed Polygonatum sibiricum (PS) has been used as food for thousands of years. However, fewer studies concentrated on the effects of polysaccharides (SPSP) from the steamed PS on the intestinal tract. With fermentation in vitro , we investigated the impact of SPSP on the fatty acids and microbiotas. Results showed significant increases in short-chain fatty acids (SCFAs) like acetic acid and propionic acid, and long-chain fatty acids (LCFAs) like cis , cis , cis -9,12,15-linolenic acid, cis -6-octadecenoic acid, and cis -9-octadecenoic acid after 12 h. The positive-associated beneficial microbiotas were observed with proliferation like Parabacteroides and Bifidobacterium . Harmful microbiota like Shigella showed decreased abundance. Further, a small molecule polysaccharide was separated from the SPSP with the structure of one glucose and ten fructose, which significantly increased SCFAs and LCFAs contents during fermentation. The potential benefits of SPSP were proved by the analysis of fatty acid levels and the intestinal microbiotas during fermentation.

Polysaccharides are the major bioactive composition of Polygonatum sibiricum ( P. sibiricum ). However, the structural and functional identifications of these polysaccharides were still limited. Herein, we isolated a novel P. sibiricum polysaccharides (PSPF) and explored its potential function and mechanism in alleviating hyperlipidemia. PSPF were purified by diethylaminoethyl-sepharose fast flow (DEAE-Sepharose FF) and cross-linked dextran gel LH-20 (Sephadex LH-20) column chromatography, and identified by gel-permeation chromatography, methylation analysis, fourier transform infrared spectrometer (FT-IR), nuclear magnetic resonance (NMR), and gas chromatography–mass spectrometry (GC–MS). Their molecular weight (Mw), polysaccharide composition, and chemical structure were characterized. Furthermore, egg yolk emulsion-induced acute hyperlipidemia mouse model was constructed to evaluate the lipid-lowering efficacy and the underlying mechanism of PSPF. It was found that PSPF, with the Mw of 3592 Da, were prepared and mainly consisted of fructan with →1)-β-D-Fru f -(2 → main chain and →6)-β-D-Fru f -(2 → side chains. In addition, PSPF supplements efficiently reduced liver lipid accumulation, alleviated hepatocyte steatosis, and upregulated the AMP-activated protein kinase (AMPK) pathway, thereby enhancing fatty acid oxidation and decomposition. These results indicate that PSPF may serve as the potential dietary supplements for lipid reduction.

Although our previous research has indicated that the ultrasound-assisted extraction–deep eutectic solvent method possessed the highest extraction yield forPolygonatum sibiricumPolysaccharide, it is uncertain whether thePolygonatum sibiricumPolysaccharide that was extracted by the ultrasound-assisted extraction–deep eutectic solvent method possesses the same performance as that extracted by other methods and whether separation and purification affect the performance ofPolygonatum sibiricumPolysaccharide. This paper aimed to compare the differences in performance among thePolygonatum sibiricumPolysaccharides extracted using different methods, including the ultrasound-assisted extraction–deep eutectic solvent method, which was the first method used by our team to extractPolygonatum sibiricumPolysaccharide. This paper also aimed to compare the differences inPolygonatum sibiricumPolysaccharides before and after they were separated and purified. The extraction yield, structural composition, and antioxidant activity ofPolygonatum sibiricumPolysaccharides were compared, respectively. The extraction yield (45.08%) obtained by the ultrasound-assisted extraction–deep eutectic solvent method was the highest (p< 0.05), the structural compositions ofPolygonatum sibiricumPolysaccharides extracted using different methods were similar, and the separated and purifiedPolygonatum sibiricumPolysaccharide was a neutral polysaccharide. The 2,2–diphenyl–1–picrylhydrazyl and 2,20–azino–bis (3–ethylbenzothiazoline–6–sulfoniacid) levels in thePolygonatum sibiricumPolysaccharide extracted by the ultrasound-assisted extraction–deep eutectic solvent method were significantly higher than those obtained using other methods (p< 0.05), and the antioxidant activity of thePolygonatum sibiricumPolysaccharide extracted by this method was significantly higher after it was separated and purified (p< 0.05). This study not only proved that the ultrasound-assisted extraction–deep eutectic solvent method could increase the extraction yield and keep the structural composition and antioxidant activity ofPolygonatum sibiricumPolysaccharide at the maximum levels but also confirmed that the extractedPolygonatum sibiricumPolysaccharide should not be further separated and purified, providing a potential technique to extractPolygonatum sibiricumPolysaccharide. This study will further promote the application ofPolygonatum sibiricumPolysaccharides in the health product industry.

Four polysaccharide fractions (P-1: 71.40%, P-2: 1.95%, P-3: 1.14%, P-4: 1.64%) were isolated from crude Polygonatum sibiricum polysaccharide (PSP), processed by water extraction, ethanol precipitation, and further separated with diethylaminoethyl cellulose-52 anion-exchange chromatography. Their molecular weights and monosaccharide compositions were characterized by high performance gel chromatography with evaporative light scattering detector and ultraviolet–visible detector. The antioxidant activity of four polysaccharides fractions were assessed by the electron transfer menchanism (DPPH, ferric reducing power, and ABST assays) and chelation of transition metals (Fe 2+ and Cu 2+ chelation ability). The highest content fraction P-1 exhibited the lowest antioxidant activity, and the ranking of antioxidant capacity was P-4 > P-3 > P-2 > PSP > P-1. After processed by microwave-assisted degradation, the molecular weight of P-1 was decreased from 2.99 × 10 5 to 2.33 × 10 3 Da, while the antioxidant activity of degraded P-1 was about eightfold higher than natural P-1. These results indicated that the proposed microwave-assisted degradation approach was an efficacious methodology to improve their bioactivity by lower the molecular weight of polysaccharides. Practical Application This study provided an environmentally friendly, convenient and efficient microwave-assisted degradation technology to process the neutral polysaccharides from Polygonatum sibiricum . The results could be used for the development and utilization of various plant polysaccharides as a kind of food supplement in our daily life.

The main active component ofPolygonatum sibiricum(P. sibiricum) rhizome isPolygonatum sibiricumPolysaccharide (PsP) with antioxidant function. At present, only the mature rhizome ofP. sibiricumis used to extract PsP, while the young rhizome of by-product is discarded directly as waste, resulting in significant wastage ofP. sibiricumresources. We used ultrasound-assisted extraction-deep eutectic solvents (UAE-DESs) method to extract PsP of young and mature rhizomes, respectively. The extraction rate, structure composition and antioxidant ability of PsP between young and mature rhizomes were compared, so as to provide references for comprehensive utilization ofP. sibiricumresources. The PsP extraction rate (33.88 ± 1.95%) of young rhizome was close to that (45.08 ± 1.92%) of mature rhizomes. The main component (PsP-2) of the PsP in young rhizome contained six kinds of monosaccharides, which belonged to acidic polysaccharides. The above characteristics of the PsP of young rhizome were similar to those of mature rhizome. The PsP of young rhizome also exhibited similar biological activity to that of the mature rhizome, which indicated even more advantages in DPPH free radical scavenging ability. The results of this study support the utility of the young rhizome, consequently helping to avoid unnecessary waste and provide reference for comprehensive utilization ofP. sibiricum.

Polygonatum sibiricum, famous for its physiological activities, is a popular Chinese food and also a traditional Chinese herb containing many carbohydrates as the main component. However, except the most reported polysaccharides, other detailed compositions of carbohydrates were still not clear. To verify the controversial existence of starch and investigate other components especially about oligosaccharides, we used iodine-potassium iodide colorimetric detection and enzymatic hydrolysis to determine starch. Then, oligosaccharides were analyzed by thin-layer chromatography (TLC), gel permeation chromatography (GPC), high-performance anion exchange chromatography with pulsed amperometric detection (HPAEC-PAD), and hydrophilic interaction chromatography-electrospray tandem mass spectrometry (HILIC-ESI-MS/MS). Results showed that the rhizome of Polygonatum sibiricum lacked starch, and fructo-oligosaccharides were the main component, accounting for about 28.95%. Oligosaccharides with degrees of polymerization above 10 were the most abundant components. This study clearly illustrated the unknown carbohydrate components of Polygonatum rhizome, promoting its functional value with new evidence.

The purpose of this study was to investigate the effects ofPolygonatum sibiricum(P. sibiricum) on microbial fermentation, physicochemical properties, and functional properties of fermented soymilk. Three types of fermented soymilk were prepared. The first type was fermented directly from regular soymilk (fermented soymilk, FSM), and the other two were fermented after addingP. sibiricum(P. sibiricumfermented soymilk, P-FSM) orP. sibiricumpolysaccharides (P. sibiricumpolysaccharides fermented soymilk, PP-FSM). The differences in physical and chemical indexes such as pH value, acidity, and water-holding capacity were mainly compared, and the differences in the contents of functional components such as total phenols, total flavonoids, soy isoflavones, γ-aminobutyric acid, and organic acids were compared. The functionalities of the three samples in terms of antioxidant activity were evaluated, and the relevance of each active substance was explored. Compared with the FSM group, the addition ofP. sibiricumandP. sibiricumpolysaccharides could not only significantly promote the fermentation ofLactobacillusbut also significantly improve the stability of the finished products during storage and prolong the shelf life of the finished product. The conversion rates of glycoside soybean isoflavones in the PP-FSM and P-FSM groups were 73% and 69%, respectively, which were significantly higher than those in the FSM group (64%). At the end of fermentation, the γ-aminobutyric acid contents of the PP-FSM and P-FSM groups were 383.66 ± 1.41 mg/L and 386.27 ± 3.43 mg/L, respectively, while that of the FSM group was only 288.66 ± 3.94 mg/L. There were also great differences in the content and types of organic acids among the three samples, especially lactic acid and acetic acid. By comparing the antioxidant capacity of DPPH (1,1-Diphenyl-2-picrylhydrazyl free radical), AB-TS (2,2′-Azinobis-3-ethylbenzthiazoline-6-sulphonate), and iron chelation, it was found that both PP-FSM and P-FSM were superior to FSM, and the antioxidant capacity had a certain correlation with the contents of total phenols and total flavonoids.Keywords:P. sibiricum;polysaccharides;fermented soymilk;physicochemical properties;isoflavones;antioxidant activity;organic acid

In this study, steamed Polygonatum sibiricum (SPS) was utilized as a novel source of polysaccharides and saponins to improve the glycemic response of extruded rice by combining rice flour with various amounts of SPS (0 %, 3 %, and 5 % w /w), to reform extruded rice-SPS complexes using twin-screw extrusion. The effects of SPS on cooking and textural properties, in vitro starch digestion, thermal properties, crystalline structure, and morphology were thoroughly investigated. The results indicated that SPS enhanced cooking performance, leading to longer cooking times, improved water absorption, and increased cooking loss. In vitro digestion experiments revealed a significant increase in slowly digestible starch (SDS) and resistant starch (RS) levels with adding SPS, indicating a deceleration in starch hydrolysis. Structural analyses using X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR) showed that SPS interacted with rice starch during extrusion, resulting in decreased crystallinity due to complex formation. The expected glycemic index (eGI) of the extruded rice decreased significantly from 90.39 to 83.72 with adding SPS. These findings suggest that SPS is a valuable supplement for enhancing the glycemic response of extruded rice by lowering its eGI and improving water absorption.

Polygonatum sibiricum(P. sibiricum) is a significantly health-promoting plant unique in medicine and food. Currently, research on the bioactive components ofP. sibiricumprimarily focuses on polysaccharides. According to theChinese Pharmacopoeia, the polysaccharide content inP. sibiricummust be at least 7.0%, which is the only criterion used for determining its content. In contrast, this study aims to thoroughly investigate and clarify the various components that contribute to the therapeutic and functional properties ofP. sibiricum. We seek to broaden the focus beyond polysaccharides to identify other potentially significant bioactive substances. We established High-Performance Liquid Chromatograph (HPLC) fingerprints for wine-processedP. sibiricumfrom various regions and identified 17 common peaks. The antioxidant activities of these components were assessed using ABTS and DPPH methods. The spectrum–effect relationship was elucidated through partial least squares regression and grey relational analysis. The results revealed that the antioxidant active components in wine-processedP. sibiricuminclude 5-hydroxymethylfurfural,p-hydroxycinnamic acid, myricetin, caffeic acid, vanillic acid, and adenosine. This research not only clarifies the antioxidant material basis of wine-processedP. sibiricumbut also establishes a scientific foundation for enhanced quality control in future applications.

Polygonatum sibiricumpolysaccharide (PsP), one of the main components ofPolygonatumsibiricumused in traditional Chinese food and medicine, has important bioactive functions, but it is difficult to fully utilize PsP because of the degradative effect of digestive gastric juices. This study aimed to innovatively synthesize a new food formulation for PsP, namely, a PsP–hydroxyapatite (HAP) sustained-release system, so as to reduce its degradation. The new food formulation was optimized and evaluated by the response surface method (RSM) and by in vitro experiments. The optimal stirrer temperature, reaction pH, etching pH, and loading time for synthesizing PsP-HAP were 85.62 °C, pH 11.12, pH 8.40, and 5.10 h, respectively, all of which were different from the findings of other similar research studies. The average encapsulation rate of PsP-HAP reached (40.16 ± 1.54)%, and the content of PsP was 8.98%. Additionally, PsP-HAP appeared to be pH-responsive, and its continuous antioxidative effect was first proven by the DPPH assay and then cytologically by a total antioxidative capacity assay. The CCK-8 assay indicated that PSP-HAP did not induce toxicity. This study successfully developed a new food formulation for PsP which appears to have the potential to reduce the degradative effect of digestive gastric juices. Thus, it is possible to achieve full utilization of PsP by using this new sustained-release food formulation.

Recently, the cubosomes have been widely studied as drug carriers. It has been described that cubosomes could further stimulate the immune response after carrying the immune enhancer. Polygonatum sibiricum polysaccharide (PSP), one of the most important biologically active ingredients of Polygonatum sibiricum , has been reported as an immunostimulant to improve immune responses. This study was aimed to observe the immunomodulation effects of ovalbumin (OVA) absorbed cetyltrimethylammonium bromide-modified Polygonatum sibiricum polysaccharide cubosomes (CTAB-modified PSP-Cubs/OVA). Firstly, the antigen uptake of CTAB-modified PSP-Cubs/OVA by macrophages was determined in vitro. After that, mice were immunized with CTAB-modified PSP-Cubs/OVA. The activation of dendritic cells in lymph nodes, activation of lymphocyte, ratios of CD4 + to CD8 + , the concentrations of OVA-specific IgG in serum and the cytokines concentrations were analyzed. As the results showed, CTAB-modified PSP-Cubs/OVA could promote the production of OVA-specific IgG in serum. The ratio of CD4 + to CD8 + in CTAB-modified PSP-Cubs/OVA group was significantly increased compared with other groups. CTAB-modified PSP-Cubs/OVA could significantly activate dendritic cells and promote lymphocyte proliferation. The results indicated that CTAB-modified PSP-Cubs/OVA could promote the secretion of related cytokines and the proliferation of lymphocytes, stimulate the cellular immune response and increase the level of humoral immunity. Above all, CTAB-modified PSP-Cubs had good adjuvant activity.
